Abstract

Sorghum is a cereal crop that could be use as an alternative source in food diversification strategy. Ithas high carbohydrate content similar to rice. Its protein content is higher than that of rice and this crophas the potential to be produce in increasing quantities in Indonesia. Nowadays, the demand of foodindustry is not only concern for nutritious food but also for health benefits. Several research reports haveshowed that sorghum contains bioactive compounds that are beneficial to human health such as in decreasingthe risk of degenerative diseases. In vitro studies have shown that this cereal crop has the capacity toincrease lymphocytes proliferation. The objective of this study is to study the biological potency of sorghumin lymphocytes cell proliferation in rats. Rats were grouped into three: i) control group animals; ii) animalswere fed 100% of sorghum diets, and iii) animals were fed 50% of sorghum diets. The results showed thatan increase in lymphocytes cells proliferation activity by 63% and 70% were seen in animals that were fed100% and 50% sorghum diets, respectively.

Abstract

ABSTRACTBuah merah (Pandanus conoideus Lam) has been assumed to contain a great number of substances which havepotential effect in increasing immune responses. Buah merah contain carotenoid 55.000 ppm (β-caroten 9.185,α-tocopherol 3.685 and tocopherol total 42,009 ppm).This research was aimed to evaluate cytotoxic effect ofbuah merah. This study was carried out by treating human lymphocyte cell culture with methanol, n-hexana,water extracts and oil of buah merah with various dosages 8,3; 33,3 and 66,7 μg/ml, then incubated at 37°C,for 72 hours. The lymphocyte cells were counted manually with triphan blue and analized by MTT method.Theresult showed that cell mortality decreased dramatically with the increase of dosage of all extracts and oil ofbuah merah. Cells cultured with the addition of 8.3 μg/ml water extracts resulted decreasing of cell mortalityfrom134x104 to 22 x104 sel/ml, with the addition of 33.3 μg/ml oil decreased from 100x104 to 38x104 cell/ mL andthe addition of 8.3 μg/ml methanol extracts decrease cell mortality from 78x104 to 64x104 cell/ml, and with 8.3μg/ml n-hexana extracts decreased from 92x104 to 66x104 cell/ml.It can be concluded that extracts and oil ofPandanus conoideus Lam were not toxic to human lymphocyte cells. The cell toxicity depends on the polarity anddosage of the extracts/oil, the highest polarity and dose indicate the lowest cell toxicity.ABSTRAK Buah merah (Pandanus conoideus Lam) diduga memiliki sejumlah senyawa yang memiliki efek potensial dalammeningkatkan respon imun. Buah merah mengandung karotenoid sebesar 55.000 ppm (9.185 β-karoten,3.685 α-tokoferol, dan 42.009 tokoferol total). Penelitian ini bertujuan untuk mengevaluasi efek sitotoksikbuah merah. Uji menggunakan kultur sel limfosit manusia yang diberi perlakuan ekstrak metanol, n-heksan,air, dan minyak atsiri buah merah dengan konsentrasi 8.3; 33.3; dan 66.7 μg/mL. Inkubasi perlakuan selama72 jam 370C. Sel limfosit dihitung manual dengan triphan blue dan dianalisa dengan metode MTT [3-(4,5-dimetiltiazol-2-il)2,5-difeniltetrazolium]. Hasil penelitian menunjukkan bahwa mortalitas sel menurun secaradramatis dan berkorelasi positif dengan besaran konsentrasi ekstrak dan minyak atsiri buah merah. Kultur seldengan perlakuan ekstrak air 8.3 μg/ml menurunkan mortalitas sel dari 134x104 menjadi 22x104 sel/ml, perlakuanminyak atsiri 33.3 μg/ml dari 100x104 menjadi 38x104 sel/ml, ekstrak metanol 8.3 μg/ml dari 78x104menjadi 64x104 sel/ml, dan ekstrak n-heksan 8.3 μg/ml dari 92x104 menjadi 66x104 sel/ml. Dapat disimpulkanbahwa ekstrak dan minyak atsiri buah merah tidak toksik terhadap sel limfosit manusia. Toksisitas sel tergantungpada polaritas dan dosis ekstrak/minyak atsiri, polaritas dan konsentrasi tertinggi mengindikasikantoksisitas sel yang paling rendah.Kata kunci: buah merah (Pandanus conoideus Lam), sitotoksik sel limfosit, respon imun
