Integrating Basic Sciences into Global Health by Implementing the Translational Research

Abstract

An emphasize on herbal medicine in cancer therapy is also included as an example of translational research implementation. A translational research in Faculty of Medicine, Universitas Padjajaran, has been conducted to develop a novel genomic approach (herbal genomic) on herbal crude extracts. This study focuses on the effects of the extracts on specific gene expressions. In this aspect, the concept of molecular targets is slightly different from pharmaceutical approaches  that aim to find compounds that interact with a specific molecule or macromolecule with known function. In this research strategy, the herbal genomic approaches involve screening of herbal extracts that affect particular gene expression related to cell responses rather than a single proteins. The determination of herbal concentration is based on differential gene expression before and after treatment which is analyzed by real time PCR. The results are being compared to the existing small molecule (active molecule) on the gene expressions. All these academic activities require biomedical research workforce to implement the translational research which, in the Faculty of Medicine, Universitas Padjadjaran, is fulfilled by involving the postgraduate students. New crude extract formulation plus specific gene expression profile is a concept required for developing chemotherapeutic agent in Indonesia. Hopefully, this review can support increased cooperation between hospital and academic institutions in implementing translational research in Indonesia.    DOI: 10.15850/ijihs.v1n1.86

Antiviral and Immunostimulant Activities of Andrographis paniculata

Abstract

Andrographis paniculata (Burm. f.) Nees is a medicinal plant which was reported to have anti HIV, anti pathogenic bacteria and immunoregulatory activities. The research purpose was to investigate the activity of Andrographis paniculata ethanol extract as antiviral and immunostimulant. A. paniculata leaves oven-dried, then grinded and macerated with ethanol 90%, and the extract then analyzed using High Performance Liquid Chromatography (HPLC) to determine the content of active compounds andrographolide. The antiviral activity of the extract was determined by observing its ability on inhibiting virus load in A549 cells transfected with Simian Retro Virus (SRV) by Real Time – Polymerase Chain Reaction (RT-PCR) analysis. The immunostimulant activity of extract was determined by its ability to induce lymphocytes cell proliferation using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Our result indicated that the A. paniculata ethanol extract inhibited the SRV virus titer similar to the positive control Lamivudine, and it was not toxic to the A459 cell line. Furthermore, low concentration (1 μg/mL) of A. paniculata extract could stimulated lymphocyte cell proliferation about 38% compared to the control lymphocyte cell without any treatment.

Abstract

Test on cytotoxic activity of marine fungi from Nipah waters have been conducted. Each solate was cultured on malt extract liquid for 15 days, and extracted by using ethyl acetate to both pellet and supernatant. Brine Shrimp Lethality Test (BSLT) was applied for cytotoxic test against Artemia salina Leach at concentrations of 10; 100; 500; and 1000 μg/ml. Cytotoxic activity showed that the percentage mortality of the brine shrimp was obtained by 1000 μg/ml at 75-100%, and the LC50 of each extract was varied from 117.21 to 531.65 μg/ml. Furthermore, the lowest LC50 was showed by sample with code number NA 14.3 at 117.21 μg/ml.
