Free Radicals Scavenger Potency of Betel Leaves (Piper betel L.) Extract and Various Fractions

Abstract

The imbalance between free radicals and antioxidants in the biology system can cause various diseases. Betel leaves (Piper betel L.), parts of a medicinal plant, are popularly used as a herbal remedy for diseases, but the scientific basis especially of their antioxidant properties remains unknown. To evaluate free radicals scavenger activity of ethanol extract and various fractions (hexane, ethyl acetate, butanol and water fraction), 1,1-diphenyl-2-picryl-hydrazyl (DPPH) and hydroperoxide (H2O2) scavenging activity were determined. To determine the DPPH and H2O2 scavenging activity, betel leaves extract and fractions were prepared in 10 concentrations. The result of this research showed that betel leaves extract and fractions had higher H2O2 scavenger and lower DPPH scavenger activities than gallic acid. The highest DPPH scavenging activity with Inhibitory Concentration (IC50) was found in gallic acid 0.732 μg/mL, ethyl acetate fraction 3.156 μg/mL, and ethanol extract 5.489 μg/mL. The highest H2O2 scavenging activity with IC50 was found in butanol fraction 0.223 μg/mL, gallic acid 0.597 μg/mL, and ethyl acetate fraction 0.783 μg/mL. In conclusion, betel leaves extract and fractions were potential free radicals scavenger; they could be potential candidates to inhibit oxidative stress. Gallic acid and ethyl acetate fraction were the highest free radicals scavenger both in DPPH and H2O2 free radical scavenging activity.Keywords: free radical, betel leaves, DPPH, H2O2, antioxidant

In Vitro Antidiabetic Activities of Extract and Isolated Flavonoid Compounds from Artocarpus altilis (Parkinson) Fosberg

Abstract

The antidiabetic activity test through a mechanism of inhibition of α-glucosidase enzyme was studied against ethanol, n-hexane, ethyl acetate and n-butanol fractions of ethanol extract of Artocarpus altilis (Parkinson) Fosberg (Moraceae) leaves and four flavonoid compounds isolated from ethyl acetate extracts of A. altilis. Ethyl acetate fraction has strongest antidiabetic activity compared to ethanol, n-hexane, and n-butanol fractions with IC50values5.98,6.79, 440.18and14.42μg/mL, respectively. Four flavonoid compounds (1-(2,4-dihydroxyphenyl)-3-[8-hydroxy-2-methyl-2-(4-methyl-3-pentenyl)-2H-1-benzopyran-5-yl]-1-propanone (AC-31), 2-geranyl-2,3,4,4-tetrahydroxy dihydrochalcone (AC-51), 8-geranyl-4,5,7-trihydroxyflavone (AC-33) andcyclocommunol (AA-3), have been isolated from ethylacetate fraction. AC-31 was the strongest antidiabetic compound compared to AC-51, AC-33 and cyclocommunolwithIC50values are 15.73, 24.41,49.49,and72.20μg/mL. Kineticstudies of AC-31 using Lineweaver-Burk method showed that inhibition mechanism of enzymeα-glucosidase was anon-competitivetype.

Apoptosis and Antioxidant Activities of Catharanthus rosues [L] G.Don Extract on Breast Cancer Cell Line

Abstract

Tapak dara or Madagascar Periwinkle (Catharanthus roseus [L] G.Don), a natural plant, is empirically reported to have promising anticancer activity. To elucidate its mechanism, a research was conducted to investigate the possible ethanol extract of  C.  roseus in inducing apoptosis on breast cancer cell line (T47D). Antioxidant activity of C. roseus was investigated as well. Sub-G1 flowcytometric apoptotic analysis result showed that extract of C. roseus at 6.25 μg/mL induced apoptosis for 26.365%. Increasing extract concentration resulted an increasing apoptotic level as well, extract at concentration of 12.5 μg/mL induced apoptosis for 22.235%.  Meanwhile doxorubicin at concentration of 10  μg/mL induced apoptosis for 36.055%. The antioxidant activity was determined by using  in vitro assay: inhibition of  2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging  activity. Antioxidant activity of  C. roseus extract were compared to quercetin and butylated hydroxyanisol (BHA), as positive controls.  The results showed that DPPH IC50 of C. roseus extract, quercetin and BHA were 358.411 μg/mL, 19.200 μg/mL  and  94.178  μg/mL, respectively. We suggest that  C. roseus extract had a potential anticancer activity by inducing apoptosis.

Abstract

Various diseases including cancer and cardiovascular diseasesare induced by free radicals via lipid peroxidation, protein peroxidation, DNA damage, and cellular degeneration in thecells.Piper betle L.,commonly called betel,belongs to thePiperaceae family.It originated from South and South Eas tAsia,and has been used as a traditional medicine by people theredue to its medicinal properties.In the present study, free radicalscavenging activity of ethanolic leaves extract of Piper betle L.together with its different solvent fractions(hexane,ethylacetate,butanol,and water)were evaluated by DPPH freeradical scavenging assay.The ethanolic extract and the ethylacetate fraction had shown very significant DPPH (1,1)diphenyl)2)picryl)hydrazyl) radical scavenging activitycompared to the other fractions.In DPPH free radical scavengingassay,the IC50 value of the ethanolic extract and the ethyl acetate fraction were found to be 17.431g/mL and 11.53 1g/mL, respectively. The free radical scavenging activity was assumed due to the total phenolic content.The total phenoliccontent was found to be highest in the ethanol extract(277.68µgEGCG/mg)and the ethyl acetate fraction(559.38µgEGCG/mg).The results concluded that the ethanolic leaves extract of Piper  betle L. and its ethyl acetate fraction have a consider ably freeradical scavenging activity.

Cytotoxicity Assay From Fractions of Hedyotis corymbosa Extract Against Breast Cancer Cell Line T47D

Abstract

Drug discovery for cancer medication is the most important effort that researcher do  at  this time. Indonesia bio diversities  have  possibility  as  a cancer medicine  sources. Finding a herbal medicine for cancer treatment is a first step to find a right cancer medicine in the future.This research has already completed for the earlier another research.Some fractions of Hedyotis corymbosa extract has been analyzed using Sulforhodamine B method with  UV wavelength  515 nm against T47D cell line, a human breast cancer. There are Hexane extract, Methylene chloride extract and Ethyl acetate extract,and give inhibitory concentration 50 (IC50) of 33.45 1g/ml, 54.59 1g/ml and 52.58 1g/ml, respectively. Ethanolic extract,  itself  has  IC50 of 61.57 1g/ml, whereas IC50 value of  Cisplatinum is 9.63 1g/ml. There is a difference between the ethanolic extracts with the other fraction.

Abstract

Nicotinil Dibuthyl Glutamate Ester (NDBGE) is a synthetic compound that is an analog of antibiotic UK3A from mycelium of Streptomyces sp.517-02. This compound was estimated to have anticancer activity similar to UK-3A. The aim of this research was to know the NDBGEs apoptotic inducing potency to human breast cancer T47D in vitro. DMEM + 1% PS + 10% FBS was added to human breast cancer T47D cells until cell concentration reached 1-5x/ 05eells/ml. NDBGE then was diluted by DMSO until 100ppm concentration. Propidium iodide with 50pg/m1 concentration was added to the T47D cells. The analysis was conducted by flow cytometry method. The result was that the apoptosis percentage of the cells was 12,35%.

Abstract

Apoptosis  is  an  important  target  on  anticancer  mechanism. The  purpose  of  this  research  is  to  investigate  apoptosis  induction of Selaginella plana Hieron active fractions on T47D cells. Absolute ethanol  was  used  to  extract Selaginella  plana powders.  Ethanolic extract  was  dilluted  by  methanol:water  (4:1)  and  then fractionated  by  hexane  (S_Hex),  methylene  chloride  (S_MTC), ethyl acetate (S_EA), and buthanol (S_BuOH). The proliferation of T47D  cell  line  was  detected  by  SRB  (Sulforhodamine  B) assay which  was  measured  at  a  wavelength  of  515nm.  Flowcytometry analysis  to  determine  apoptosis  was  examined  by  Propidium Iodide  (PI)  and  Annexin  V  assay  using  T47D  breast  cancer  cell line.  The  result  showed  that  the  IC50 value  of  S_Hex,  S_MTC, S_EA,  and  S_BuOH  on  T47D  cells  were  107  µg/mL,  4  µg/mL,  6 µg/mL,  and  17  µg/mL  respectively.  The  active  fractions  (S_MTC and  S_EA)  at  its  IC50 concentration  significantly  (P<0.05) increased  the  total  number  of  early  apoptotic  cells  in  the  T47D cells  3.39%  and  4.1%  respectively  compared  to  that  of  control (1.95%).  Based  on  the  result,  methylene  chloride  and  ethyl acetate  fraction  of Selaginella  plana induced  apoptosis  on  T47D cell.Keywords: apoptosis, breast cancer, Selaginella

Abstract

Selaginella sp belongs to the Selaginellaceae family. It has been used in China and Indonesia as a traditional medicine. It hasseveral medicinal properties including antibacterial, anticardiovascular, and anticancer agent. The aim of the present study was to access the anticancer property of the ethanolic extracts of Selaginella plana Hieron. on T47D breast cancer cell line. The proliferation of T47D cell line was detected by SRB (Sulforhodamine B) assay which was measured at a wavelength of 515 nM. The result showed that the IC50 of the ethanolic extract was determined at 7.03 pg/mL. This significant activity was assumed due to its high total flavonoid content. The totalflavonoid content of the ethanolic extract was 23.04%. Flow cytometry analysis indicated that the extract may undergo the cell death via apoptosis pathway. In conclusion, the ethanolic extract of Selaginella plana Hieron: has considerable activity in inhibiting T47D cell line proliferation.Key words: Selaginella plana Hieron., SulforhodamineB, T47Dcells, anticancer flow cytometry

Abstract

Sintesis nanofiber komposit Zn-PVDF kopolimer dengan metoda elektrospinning telah berhasil dilakukan. Proses pembuatan nanofiber komposit serta  morfologi yang terbentuk dipengaruhi oleh penambahan Zn-asetat dengan perubahan diameter rata-rata serat dari 357,13 nm menjadi 777,24 nm. Analisis FTIR menunjukkan bahwa struktur kristal nanofiber komposit Zn-PVDF kopolimer didominasi oleh strukturβ-phase, dengan bilangan gelombang 1190,08 cm-1 dan 487,99 cm-1 untuk struktur α-phase dan 1404,18 cm-1; 1280,73 cm-1; 1074,35 cm-1; 881,47 cm-1; dan 840,96 cm-1 untuk struktur β-phase.Kata kunci :nanofiber komposit, Zn-PVDF kopolimer komposit, elektrospinning,kristal struktur, morfologi, diameter fiber The fabrication of Zn-PVDF copolymer nanofiber composite has been investigated in this research study by using electrospinning method. Abstract

Silver nanoparticle-bacterial cellulose bionanofiber nanocomposite (nano Ag-BSB) has been synthesized by using trisodium nitrate as reducing agent. The arrangement offiber in nano Ag-BSB nanocomposite has random shape after 26 confirmed by scanning electron microscopy (SEM)analysis. Nano Ag-BSB nanocomposite has average 64.1 nm in nanofiber diameter. Fourier infra red spectroscopy and X-ray diffraction analysis showed crystal structure in Ag-BSB nanocomposite is cellulose type L Crystalinity of BSB and Ag-BSB are 94 % and 89 %, respectively.This result showed that presence of silver nanoparticles in BSB decreased crystalinity of BSB.
