Abstract

Effect of vitamin E addition (alfa-tokoferol) into sperm washing medium by centrifuge on the quality of Brahman cattle spermatozoa ABSTRACT. The aims of study to determine the effectiveness of the addition of vitamin E in the washing medium by centrifugation on sperm quality Brahman cattle. frozen semen of Brahman cattle, divided into 4 treatment groups addition of vitamin E in the washing medium: 0.0gr/100 ml medium (K0), 0.1gr /100 ml medium (K1); 0.2gr/100 ml medium (K2) and 0.3 g / 100 ml medium (K4), each group was repeated 5 times. Examination of motility, viability and integrity of sperm membrane performed according to WHO standards. The data obtained were analyzed with one-way ANOVA and Duncan test. The average percentage of motility, viability and membrane integrity of spermatozoa in the addition of vitamin E were significantly different (P <0.05) compared to the control. Percentage of motility, viability and membrane integrity of spermatozoa in the group K2 significantly higher (P <0.05) compared with the group K3: K1 and K0. Percentage of motility, viability and sperm capacitation and sperm live on group K3 significantly higher (P <0.05) compared with the K1 and K0. While the percentage motility of spermatozoa in the group K1 higher were not significant (P> 0.05) compared with the group K0. The addition of vitamin E in the medium on the process of washing spermatozoa Brahman cattle. The addition of vitamin E 0.2gr/100ml better than vitamin E 0.1gr/100ml and 0.3gr/100ml in maintaining the percentage of motility and live spermatozoa Brahman cattle.

Abstract

ABSTRAKPenelitian ini bertujuan mengetahui kepekaan ekstrak daun mengkudu (Morinda citrifolia L.) terhadap pertumbuhan Salmonella sp. dan Escherichia coli Esktrak daun mengkudu dibagi menjadi 3 konsentrasi yang berbeda yaitu 25%, 50%, 75%. Uji daya hambat dilakukan sesuai dengan metode Kirby-Bauer. Hasil rata-rata zona hambat ekstrak daun mengkudu terhadap bakteri Salmonella sp. pada konsentrasi 25% yaitu 6,2 mm, pada konsentrasi 50% yaitu 7,1 mm, dan pada konsentrasi 75% diperoleh rata-rata 6,6 mm. Zona hambat ekstrak daun pandan wangi terhadap Escherichia coli pada konsentrasi 25% rata-rata 7,3 mm, pada konsentrasi 50% 7,2 mm dan pada konsentrasi 75% diperoleh rata-rata 7,5 mm. Kontrol positif antibiotik amphicilin terhadap bakteri Salmonella sp. didapatkan rata-rata 6,2 mm, sedangkan terhadap bakteri Escherichia coli dengan rata-rata 19,6 mm. Berdasarkan penelitian ini dapat disimpulkan bahwa ekstrak daun mengkudu memiliki daya yang lemah dalam menghambat pertumbuhan Salmonella sp. dan Escherichia coli.Kata kunci: daya hambat, ekstrak daun mengkudu, Salmonella sp. dan Escherichia coli. ABSTRACTThe aims of this research is to know the sensitivity of noni leaf extract (Morinda citrifolia L.) against Salmonella sp. and Escherichia coli. Noni leaf extract was divided into 3 different concentrations 25%, 50%, 75%. The inhibition effect test was performed in accordance to Kirby-Bauer method. The average of inhibition zone of noni leaf extract on Salmonella sp. at 25% concentration was 6.2 mm, at concentration of 50% was 7.1 mm, and at concentration 75% obtained average 6.6 mm. The average inhibition zone of noni leaf extract againts Escherichia coli at 25% concentration was 7.3 mm, at concentrations of 50% was 7.2 mm and at concentration of 75% obtained an average of 7.5 mm. The averaged positive control of ampicillin antibiotics against Salmonella sp. was 6.2 mm, while against Escherichia coli was 19.6 mm. Base on this research it can be concluded that the noni leaf extract had weak activity to inhibit the growth of Salmonella sp. end Escherichia coli.Keyword : inhibition effect, noni leaf extract, Salmonella sp. and Escherichia coli.

Abstract

This study aimed to compare the progesterone concentrations in Waringin sheep estrus cycle which induced by PGF2α or the combination of PGF2α and GnRH. This study used six Waringin sheep those were divided into twice groups, K1 and K2. K1(n=3) were injected by 7.5 mg PGF2α intramuscularly for 10 days, while K2 (n = 3) were injected by PGF2α and GnRH. On the first day K2 were injected by 7.5 mg PGF2α, then followed by injection 50 µg of GnRH on day 8th and re-injected by 7.5 mg PGF2α on day 15th. On day 18th, sample was re-injected using 50 µg GnRH. Waringin sheep those showed estrus symptoms were detected visually and with signs showed by sheep stud. Blood samples were taken on the 7th, 14th, and 21st day after the peak heat. The measurement of progesterone concentration was conducted by enzyme linked immunosorbent assay (ELISA). The results showed those the concentrations of progesterone on day 7th, 14th, and 21st on K1 vs K2 were 1.324±1.079, 7.607±8.922, and 5.220±1.653  vs 4.705±3.369, 4.184±5.512, and 1.797±0.898 ng/mL (P>0,05), respectively. In conclusion, the concentration of progesterone of Waringin sheep after inducing with PGF2α or combination PGF2α and GnRH at different cycle periode did not show differences. Keywords: Estrus Cycle, GnRH, PGF2α, Progesterone, Waringin Sheep
