Abstract

The hemiparasite Santalum album Linn. (cendana) grows very slow, in nature the rare and difficult seeds need stimulation to germinate. Gibberellins (including GA4) are growth regulators, usually used to increase growth as well as to break seed dormancy. The objectives of this research were to investigate the influence of gibberellins on germination percentage and height of cendana growth. Experiment was laid out in a Completely Randomized Design with 3 replicates of 300 seeds for germination percentage and 3 replicates of 10 seeds for seedling growth. Gibberellin was applied as treatment with 100, 300, and 500 ppm. The seed germination was recorded until 9 weeks, and height of plants measured until 8 months at the greenhouse. The results showed that the addition of gibberellins at all treatment increased the percentage of germination and caused the seeds germinated four weeks earlier than the control. In the first 4 months, 500 ppm gibberellins gave the highest acceleration of germination, afterward all treatments have relatively the same influences. Gibberellins gave positive effect on height of cendana growth. After 7 months, the growth decreased although all gibberellin treatments gave higher growth than the control.Keywords : Santalum album Linn., Gibberellins (GA4), germination, height growth.

Berbagai Rencana Penelitian Integratif (RPI) telah ditetapkan termasuk RPI Adaptasi Bioekologi dan Sosial Ekonomi Budaya terhadap Perubahan Iklim yang diinisiasi pada tahun 2010. Dalam RPI ini salah satu penelitian dilakukan untuk mengidentifikasi berbagai spesies pohon dari berbagai provenans yang potensial untuk mengantisipasi perubahan iklim dengan penekanan untuk tujuan pengujian pada daerah kering. Abstract

Penyakit karat tumor menyerang secara luas pada sengon (F. moluccana Miq.), membentuk tumor (neoplasmik) yang disebabkan oleh patogen obligat parasit Uromycladium tepperianum (Sacc.) McAlpine. Senyawa fenol mempunyai kemampuan berfungsi sebagai co-factor penentu patogenisitas dari hasil perkembangan patogen dan pertahanan kimia tanaman. Tujuan penelitian ini adalah untuk mengetahui kandungan senyawa fenol pada toleransi sengon terhadap karat tumor dengan melakukan pengamatan tinggi tanaman, analisis kuantitatif senyawa fenol total, analisis anatomi kayu sengon toleran dan uji toleransi kalus sengon hasil budidaya jaringan dengan filtrat karat tumor sebagai agen kimia. Hasil penelitian ini adalah (1) sengon yang diinokulasi spora karat tumor mempunyai senyawa fenol total yang lebih rendah dibandingkan kontrol (tanpa inokulasi), (2) sengon toleran karat tumor mempunyai kandungan senyawa fenol lebih rendah dibandingkan sengon yang sensitif, (3) hasil mikroskopis anatomi kayu menunjukkan adanya kandungan substrat yang lebih gelap pada sengon tidak toleran dan (4) pada media filtrat karat tumor dengan konsentrasiyang sama, sengon toleran menunjukkan sel-sel kalus hidup yang lebih tinggi. Konsentrasi tertinggi sel kalus dapat hidup adalah25% (v/v).

Abstract

This study aims to observe acid tolerance of Vitex pubescens culture growth. Planlet cultures were established from seeds on Murashige & Skoog medium supplemented with 1 µM benzylaminopurin; 0,1 µM kinetin and 0,01 µM naphtalenaceticacid. The pH was adjusted to 5,7;4,0 and 3,0. The mean height of planet were 4,44 cm ± 0,32 for pH 5,7; 2,88 cm ± 0,54 for pH 4 and 1,10 cm ± 1,38 for pH 3. The mean length of root were 13,72 cm ± 0,47 for pH 5,7; 18,88 cm ± 0,60 for pH  and 19,27 cm ± 0,73 pH 3. The mean number of branch roots per cm were 1,65 ± 0,058 for pH 5,7;3,88 ± 0,18 for pH 4 and 0,57 ± 0,30 for pH. After 12 weeks acclimatization in Yellow Red Podzolic (YRP) soil, the mean height of plant with pH 5,7 culture were 29,86 cm ± 0,64 and 26,72 cm ± 0,40 for pH culture. In vitro acid tolerance of Vitex pubescens were at pH 4 and can grow in ex vitro condition in YRP soil with pH 4,48.

Abstract

Pongamia pinnata is an arboreal legume with high conservation value (Conference of The Parties to The Convention on Biological Diversity at the IX/2 nd  on plants of high conservation volue criteria). Biotechnology breeding program with the selection  of legume crops that have a high nitrogen use efficiency becomes an important issue to improve productivity and quality of P. pinnata.Thus the important symbiotic effectiveness test done to improve the ability of N fixation as the main element forming a plant tissues . The purpose of this study was to obtain Rhizobium strain from different P. pinannta habitats which has the highest effectiveness compared with strain Rhizobium sp. Commercial (USDA 122) and controls to P. pinnata seedlings. The parameters used are the number of nodules, plant height, root length and diameter . From this activity obtained 3 strains of Ambon and 2 strains of Banyuwangi .  Strains selected on the basis of colony growth speed of Rhizobium sp.in vitro.The average number of nodules, plant height,  root length and diameter of P. pinnata that are inoculated with strain  AMI. AM2, AM3, Bal and Ba2 from seram, Ambon higher than from Baluran , Banyuwangi. On average, the highest number of nodules occurred in simbiose of P.  pinnata) Ambon  Rhizobium AM3  (50 ± 0,66) eguels P. pinnata Ambon –Rhizobium USDA 122 (50 ± 0,88); the best plant height occurred in P. pinnata Ambon-Rhizobium Ba2 (72cm ± 1,14) ; the best root length  (25 cm ± 0.33) and the best stem diameter (4.0 mm  ±  0.33) occurred in P. pinnata Ambon-Rhizobium AM2. The highest symbiose effectiveness value of P. pinnata-Rhizobium occureed in P. pinnata origin from Ambon with Rhizobium strain AM3 (32,45).

Abstract

Cendana (Santalum album Linn.) is one of the important hemiparasite species due to its high value essential oil for pharmaceutical industries. However, since1998 this species has been categorized as vulnerable by the IUCN Red List. The propagation of cendana has been hampered by inadequacy in regeneration, either through sexual or vegetative propagation. Regeneration of cendana through in vitro technique is still limited due to the difficulty in rooting and acclimatization. The purpose of this study is to observe the effect of clones, in vitro and ex vitro techniques on the primary and secondary root development. Two clones of cendana: Clone A.III.4.14 and WS28 were tested in Gresshoff & Doy culture media enriched by IBA 20 mg/l; IAA 0.15 mg/l and kinetin 0.15 mg/l. Root development was observed for six months of culture for in vitro and three months after acclimatization in a greenhouse for ex vitro. The results of this study showed that Clone A.III.4.14 formed primary root in lower percentage rate (41.85%) than Clones WS28 (60,44%), on the contrary it grew secondary root in higher percentage rate (58.15%) than Clone WS28 (39,56%). The ex vitro following the acclimatization showed that the root hairs grew only in the plantlets which formed secondary root during in vitro. This result indicates an important of clone’s selection for secondary root development during in vitro to obtain a better root system in the success of acclimatization of cendana.

Abstract

The availability of source material to multifunctional tree Toona sp. have constraints for macro and micro propagation, in this regard, this study aims to:1)observe the formation of buds on T. sinensis and T. sureni stem cuttings in an effort to sustain the availability of explants and 2)analyze the effect of GA4, IBA and BAP growth hormone on regeneration and propagation of the axillary bud initiation and callogenesis through tissue culture. Isolation of stem cuttings was conducted in a greenhouse with number of shoots per nodule and nodule distance as observed.Variables Tissue culture techniques were used to study the effect of exogenous GA4 hormones application on the initiation of axillary buds. The application of IBA hormone on plantlet rooting and callogenesis. The results showed that T. sinensis has 2-7 buds per nodule and nodule distance of 2-5 mm, while T. sureni has one bud per nodule and nodule distance of 5-10 cm. The average length of T. sinensis shoots were 15.8 cm and 17.4 for T. sureni. Enrichment with 1 mg/l GA4 gave the highest axillary shoot length on T. sinensis (8.5 cm ± 0.7228) and (9.8 cm ± 0.1022) for T. sureni. Two mg/l IBA enrichment gave the highest root length of T. sinensis (10.9 cm ± 1.8392) and for T. sureni (7.9 cm ± 0.7633). Three mg/l concentration BAP application gave the best effect in term of callus weight of T. sinensis (1.4 g ± 0.3833). Scoring of callus weight showed moderate response categories to both T. sinensis and T. sureni.

Abstract

The quality of Santalum album seedling is influenced by growing media where their root grow and develop. This research was conducted to confirm that organic growing media is a good characteristics as top soil media and to determine the quality index value as indicator of seedling quality. There types of media; top soil, composted sawdust and pure sawdust were arranged in a completely randomized design with 4 replicates of 5 seeds. The observation periode was 8 month, from January until September 2007. The resuly showed that index value for S, album seedling in organic media is as relatively high as in top soil media; 1,42 for top soil, 1,10 for composted sawdust and 1,08 for pure sawdust.

Abstract

Preliminary shoot groeth on acacia hybrids (Acacia mangium x A. Auriculiformis) was observed to assess their in- vitro shoot initiation. Explants were taken from 4 months old seedlings of A. Mangium, A. Auriculiformis and their hybrid. Murashige & Skoog’s (MS) medium was used supplemented with various concentration of BAP, IBA,NAA, and GA4. Parameters measured were bud breaking, number average number of leaves of acacia hybrid was 7 and 1.9 cm and 5.2 respectively, found in medium Ak2 culture. Bud breaking of acacia hybrids on that medium happened 15 days earlier than pure Acacia. This finding showed that acacia hybrid could be propagated by  micro propagation succesfully.
