Abstract

Merbau [Intsia bijuga (Colebr.) O. Kuntze] is one of valuable timber tree in south-east Asia which has been known having ectomycorrhizae (EcM) though being ignored. Identification of the ectomycorrizae is prime important for being basis of further studies. This study investigated the EcM fungi associated with Merbau by using both sporocarp morphology and EcM morphotypes. Morphological characters of sporocarps and basidiocarps of the fungi and EcM morphotypes obtained from seedlings and trees from natural and plantation stands of merbau, as well as from nurseries were compared to the description of those resulted from baiting method. Only one species of ectomycorrhizal fungus was found associated with merbau [Intsia bijuga (Colebr.) Kuntze] which has not been described yet. The fungus formed mycorrhizae with monopodial pinnate branching. The fungus was identified belonging to the genus of Scleroderma. The fungus was more common to occur beneath merbau seedlings than trees. The sporocarps may be yielded under greenhouse condition and could be cultured in MMN agar media, thus it facilitates to the production of inoculums used for further studies.

Abstract

Shallot bulb generally stored for several month before planted in the field. Since explant age is one of important factors in tissue culture development, storage period of shallot bulb might alter the explant growth in vitro. Shoots of shallot formed in the in vitro culture should form bulbs before can be use as seedling, and temperature may affect micro bulb induction. Two experiments had been conducted to evaluate 1) the effect of storage period in the field on the growth of shallot explant in vitro and 2) the effect of culture room temperature in microbulbs induction of shallot. In the first experiment, shallot bulb had been stored for 1, 2, 3 and 4 months before used as explants. Storage period significantly influenced the explant growth in vitro. Bulb with 2 months storage gave the best performance on number of micro shoot, number of leaves and roots, and less of vitrification. Micro shoots on three weeks after planting (WAP) was feasible to use as propagule for shallot micro bulb induction. In the second experiment, shoots from propagation medium was transplanted to bulb induction medium and  grown in growth chamber with different temperatures (day/night) 20/17 oC and 30/27 oC, respectively.  Micro bulb induction was influenced by temperature. Lower temperature showed good results for number of leaves, length of leaves, number of roots, and length of roots. However, temperature of 30/27 oC gave the best result on number of micro bulb, diameter of bulb and bulb width:bulb disk diameter ratio.   Keywords: Allium, explants age, micro bulb,  temperature, 2ip

Abstract

Callus induction was studied in five genotypes of Dianthus chinensis using 2.4 D and NAA. Calluses can be obtainedfrom unfertilized ovule culture and ovary culture. The aim of the research was to study gynogenic potential and responseof Dianthus chinensis through ovule slice and ovary slice culture for obtaining haploid plants. Five genotypes of Dianthuschinensis and five media were used in ovule slice culture and two genotypes and three medium were used in ovary culture.Flower buds in the 7th stage were incubated for the purpose of dark pre-treatment at 4 oC for one day. Ovules and ovaries wereisolated and cultured in induction medium. Cultures were incubated for the purpose of dark pre-treatment at 4 oC for seven days, followed by 25 oC light incubation. The result showed that 2.4D was better than NAA in inducing callus. Percentage of regenerated calluses were produced in V11, V13 and V15 genotypes in M7 medium (MS + 2 mg L-1 2.4D + 1 mg L-1 BAP + 30 g L-1 sucrose and M10 medium (MS + 1 mg L-1 2.4D + 1 mg L-1 BAP + 20 g L-1 sucrose). All calluses originated from ovule and ovary cultures flowered prematurely. Double haploid (V11-34) were obtained from ovule slice culture based on PER (peroksidase) and EST (esterase) isoenzym marker.Keywords: ovule slice culture, ovary slice culture, callus, Dianthus sp., haploid

Abstract

Mutants of Spathoglottis plicataBlume were obtained from plants treated with Gamma Irradiation at 30-100 Gray. The mutants showed variations in the flower morphology. The objective of this experiment was to identify genetic variations of orchids S. plicataand its mutants using inter-simple sequent repeat (ISSR) markers. The amplified product of 10 ISSR primers produced 360 bands and 71 ISSR of the loci (90.14%) were polymorphic. The coefficient of similarity and principal component analysis produced five major groups with similarity coefficient of 0.68. The goodness of fit correlation matrix value reached 0.91. Therefore the ISSR isa good marker for identification of S. plicatamutants.Keywords: gamma irradiation, ISSR marker, mutant, orchid

Abstract

The aim of this study was to investigate the effect of rootstock and shoot types on in vitro mangosteen micrografting.The experiment was arranged in completely randomized design (CRD) with two factors. The first factor was the rootstocktype, i.e. rooted planlet from the germination of quartered seed, and rooted planlet from the germination of undivided seeds.The second factor was the developmental phase of scion, i.e. dormant buds, and flush (had new leaf more than 2-4 mm). Theresults showed that rootstock derived from the germination of undivided seed had a higher success rate than other treatmentson all variables, except for number of new leaves. The use of flush as scion was better than dormant buds; flush resulted in ahigher percentage of successful micrograft and longer shoots. In vitro micrografting had a better growth rate than grafting at the same age. The results of anatomical observation conducted at four months after micrografting demonstrated that there was a good graft union, indicated by excellent fusion between rootstock and scion xylem tissues.Keywords: flush, in vitro, micrografting, rootstock, scion

Abstract

The objective of this research was to study shallot bulb formation on few concentrations of growth retardant succinic acid daminozid hydrazide (SAOH). Completely Randomized Design with 2 factors were used in this experiment. The first factor was four concentrations of SAOH (0, 30, 60 and 90 ppm) and second was two cultivars of shallot (Bima Juna and Kuning Tablet). The cultivars did not give significant effect to total number of leaf, shoot, root, number and weight of bulb, diameter of bulb, and height of plantlets. While SADH concentrations gave very significant effect to number of leaf, but not significant to number of root, number and weight of bulb, diameter of bulb and height of plantlets. Combinations of the two factors only gave significant effect to number of leaf and shoot but not significant to number of root, number and weight of bulb and height of plantlets.   Keywords: bulb formation, SAOH, shallot

Abstract

Cultivars Granola dan Atlantic are the best cultivars so far for farmers, due to it adaptation in Indonesia. Cv Granola is resistant to several important disease, but the quality is low, thus only produce for vegetable purpose.

Abstract

Improvement of selected induction culture media on callus induction in anther culture of anthurium and a histologicalstudy on its callus formation were studied at the tissue culture laboratory of the Indonesian Ornamental CropsResearch Institute from February to October 2008. The objectives of the study were to optimize selected media forcallus formation, reveal cell origin of callus derived from anther culture and shoot formation process. Selectedmedia improved in the study were 1) MMS-TBN containing 0,5 mg/l TDZ, 1,0 mg/l BAP and 0,01 mg/l NAA (Winartomedium, WM) and 2) MMS III supplemented with 1,5 mg/l TDZ, 0,75 mg/l BAP and 0,02 mg/l NAA (Winarto andRachmawati medium, WRM). Improvement treatments were carried out by omission and application of 2,4-D in 0.5mg/l and reduction of medium strength of full, half, quarter, one eighth, one sixteenth, and zero strength. Afactorial experiment was arranged using a randomized complete block design with four replications. Results ofthis study indicated that the highest callus induction was clearly established in WRM. The medium stimulatedpotential growth of anther (PGA) up to 81% with 49% of percentage of anther regeneration (PAR) and 2.7 number ofcallus formed per replication (NCF). Significant improvement in callus formation was also recorded by reduction ofmedium strength of WRM to one eighth compared to others. The reduction induced PGA up to 58% with 29% of PARand 1.8 NCF. From histological studies it was well recognized that regenerated callus on half anthers cultured wasoriginated from middle layer cells of anther wall. The morphogenic response of anther wall cells caused primarilyon no androgenesis effect in microspore cells.

Abstract

Several bacterial pathogens can cause diseases of potato. Ralstonia solanacearum and Erwinia carotovora are those of the worlds most important diseases of potato, especially in tropical climates. This experiment was aimed to obtain putative potato cultivars having good tolerance to bacterial wilt caused by R. solanacearum and soft rot caused by E. carotovora. Two adopted potato cultivars in Indonesia, cv. Atlantic (2n=4X=48) and cv. Granola (2n=4X=48), were used as parents.  The experiment was arranged in a Randomized Complete Design.  Test for tolerance was perfomed in vitro using BF15 as susceptible control and Solanum stenotonum as tolerance control. The result showed that there was high diversity of tolerance level to both R. solanacearum and E. carotovora.  Some clones showed good tolerance level as compared to cv. Atlantic and cv. Granola, but none showed that as compared to S. stenotonum.   Key words:  in vitro testing,  potato, R. solanacearum, E. carotovora

Abstract

At least ten years needed to abtain new potato cultivar through sexual hybridization, somatic hybridization or through genetic transformation, To short cut this process, Laboratory of Biotechnology, Department of Agronomy, IPB employed a strategy so called single seed in vitro clonal decent ( SSICD) by usings elected parental lines for TPS (True Potato Seed) production. This breeding consist of in vitro pre evaluation for resistance wilt, fusarium wilt, black leg, rot knot nematode and maturity. Using the same number of bacterial cell / (109 cell/ml), there were positive correlation between in vitro test for disease resistance through dripping test or dripping test with greenhouse test through direct inoculation of Ralstonia solanancearum. Resistant clones to fusarium wilt and verticillium were also resistant to bacterial wilt. In vitro tuberization could be use to evaluate maturity of potato cultivar.   Key words: Potato, SSICD
