Abstract

High blood cholesterol in human might cause atheroskelerosis, a disease that eventually leads to the occurrence of coronary heart attack. This research was aimed to investigate level of sago waste or shrimp waste on plasma cholesterol and growth in rats. The experiment was conducted on 28 males Spraque dowley rat of one month old and was set up in a completely randomized design with seven treatments and four replications. The treatments were R0 (0 % sago waste and srimp meal) as a control, ASA 10 (10% of sago waste), ASA 20% (20% of sago waste), ASAF 10% (10% fermentation of sago waste), ASAF 20% (20% fermentation of sago waste), LU 10% (10% of srimp waste), LU 20% (20% of srimp waste). Variables measured included feed intake, body weight gain, plasma cholesterol, high density lipoprotein, low density lipoprotein, and triglyceride. Abstract

This research was undertaken to evaluate energy utilization and blood  metabolites status of male Indonesian local sheep involving nutrient balances and using urea space techniques for measurement body composition. Factorial randomized completely design was used in this study using 16 growing male local sheep. Factor A was breed which consisted of 8 Garut sheep (av.BW 14.90 kg) and 8 Jonggol sheep (av.BW 13.60 kg). Factor B was different ration which received of diet containing Indigoferasp. and sproutbean waste. The ration offered were pellets containing CP around 16% and energy 15.6 MJ GE/kg. Energy balance and blood metabolites studies were conducted during three-months trial. Intake, digestibility, urinary and metabolism variables were measured based on total collection method.  Methane energy was measured by rusitec technique and methane gas production multiplied by calory equivalent value of methane. Energy retention (RE) was measured by urea space  technique. Heat production was calculated from ME-RE. Plasma glucose, cholesterol and urea-N were measured by using spectrophotometry. Results showed that there were signiϐicant differences of energy intakes, digestibilities and metabolism among the rations. Animals fed with sproutbean waste ration has higher energy utilization compared to those fed Indigofera sp ration. There were no signiϐicant different for all blood metabolite variables among breed and its interaction between combination treatments. It is concluded that sproutbean waste in local sheep ration has higher energy utilization and daily gain than Indigofera sp.

Abstract

Local beef cattle is still the major source of domestic beef consumption, yet their productivity and quality is relatively low. This study was aimed to investigate productivity and meat quality traits of local beef cattle through fattening using cereals based concentrate containing different energy level. Nine Ongole cattle with average initial liveweight of 254.67 ? 20.7 kg and age between I1-I2 permanent incisor teeth were used in the study. They were fattened for four months and alloted into three different energy rations; they were low (ER) 57.88% TDN, medium (ES) 63.72% TDN, and high (ET) 69.67% TDN. The three rations had simmilar protein contents, between 12.42-12.96%. Observed parameters included cattle performance (ration and nutrition dry mater intake, daily gain, and feed conversion), carcass traits (carcass weight and percentage, fat thickness, and loin eye area at the 12th rib), and meat quality traits (pH, water holding capacity, tenderness, cooking loss, marbling score, and meat color). The results showed that cattle fed with high energy (ET) ration had the highest body weight and marbling score, and more efficient in feed conversion than medium (ES) and low energy (ER) rations. Local beef cattle productivity and there meat quality could be improve through fattening using high energy cereals based concentrate.

Abstract

Morphometric measurements using digital image technique give a lot of benefits i.e., a safer method both for the researchers and the experimental animals, produce a more accurate morphometric data, reduce the stress in the experimental animals and produce a more detail and complete morphometric data. This experiment used 20 Ongole cross cattle and 20 Bali cattle with the average age of 3 years. Digital image data were collected by using DSLR camera with 18Mp resolution. The collected digital image data were analyzed by using Image J program. Parameters measured in this observation were body length, body height, hip height, pelvic height, loin height, the length of ossa vertebrae cervicales, the length of ossa vertebrae thoracicae, the length of ossa vertebrae lumbales, the length of os scapula, the length of os humerus, the length of ossa radius-ulna, the length of os metacarpale III, the length of os femoris, the length of ossa tibia-fibula, and the length of os metatarsale III. Digital image technique could replace the manual technique for morphometric measurement with a lower level of variance (<10%). Application of digital image technique for measurement of morphometric parameters in Ongole Cross and Bali cattle showed that Bali cattle has a potential for muscle deposition in body frame and body axis while Ongole Cross cattle has a potential for muscle deposition in fore and hind legs.

Abstract

Meat quality represents meat properties known by consumers and affects their acceptance.  Feed is one of the factors affecting meat quality.  Utilization of pineapple waste as a fiber source in cattle feeding is potential to reduce feed cost.  Pineapple waste can be used as feed and as meat tenderizer.  However, many believes that feeding pineapple waste to cattle distract beef quality.  This study was aimed at assessing meat sensory quality of Brahman Cross cattle fed pineapple waste as fiber source.  Six Brahman Cross (BX) cattle aged 2.5-3 years were used.  The cattle were fattened for 120 days.  Three cattle was fed rations containing concentrate and pineapple waste silage and the remaining cattle was fed rations containing concentrate and corn leaf silage. In the end of the study period, the animals were slaughtered and meat was cut based on the commercial slaughtering standard.  Meat samples of Longissimus dorsi muscles were taken.  Beef sample aging was conducted at 0ºC for 3 months.  Measurements of beef characteristics were taken monthly.  A completely randomized design was used.  Data were subjected to a Kruskal Wallis test and meat physical characteristics were analysed by repetaed measurement analysis of variance.  The parameters observed comprised pH values, tenderness, cooking loss, water holding capacity, meat colors (L*, a*, b* value), organoleptic test hedonic and hedonic quality (aroma, tenderness, color, and mucus).  The results showed that the pineapple waste did not have significant influences on meat physical characteristics observed, except meat color a* values at two month aging, meat color L* value at three month aging. It was concluded that pineapple was could be used as a fiber source in beef cattle fattening with no negative effect on meat quality.

Abstract

ABSTRAK Gen FTO berfungsi sebagai regulasi homeostasis, deposisi lemak dan pengaturan obesitas. Penelitian ini bertujuan untuk mengidentifikasi polimorfisme SNP g.125550A>T di ekson 3 gen FTO pada bangsa sapi potong Indonesia. Sampel darah diperoleh dari 209 ekor sapi, terdiri atas sapi bali (44), madura (20), Pesisir (20), katingan (20), Peranakan ongole (PO) (22), Pasundan (20), Sumba Ongole (SO) (11), brahman (20), simental (15), dan limousin (18). Polimorfisme gen FTO dianalisis menggunakan metode PCR-RFLP (HpyCH4III) dan direct sequencing. Hasil genotiping SNP g.125550A>T adalah polimorfik (genotipe AA, AT, dan TT) pada sapi madura, pesisir, katingan, PO, pasundan, SO, brahman, simental, dan limousin. Frekuensi alel A dan T masing-masing adalah 0,70, 0,68, 0,84, 0,89, 0,70, 0,86, 0,90, 0,73, 0,69 dan 0,30, 0,33, 0,16, 0,11, 0,30, 0,14, 0,10, 0,27, 0,31. Nilai Ho dan He masing-masing adalah 0,60-0,14 dan 0,44-0,18 serta dalam keseimbangan Hardy-Weinberg (P>0.05). Sementara pada sapi bali bersifat monomorfik hanya bergenotipe AA. Hasil sekuensing SNP g.125550A>T ditemukan mutasi tranvesi A menjadi T pada posisi nukleotida  g.125550. Berdasarkan hasil penelitian ini, dapat disimpulkan bahwa SNP 125550A>T gen FTO beragam dan berpotensi dijadikan marka genetik untuk kualitas daging pada bangsa sapi potong Indonesia.Kata Kunci: gen FTO, PCR-RFLP, Sapi, SNP g.125550A>TABSTRACTThe FTO gene functions as regulation of homeostasis, fat deposition and regulation of obesity. This study aimed to identify the polymorphism of SNP g.125550A>T in exon 3 of FTO gene in Indonesian beef cattle. Blood samples were collected from 209 cattle, including bali (44), madura (20), pesisir (20), katingan (20), PO (22), pasundan (20), SO (11), brahman (20), simental (15), and limousin (18). Polymorphism of the FTO gene was analyzed using PCR-RFLP (HpyCH4III) and direct sequencing methods. The results of genotyping SNP g.125550A>T was polymorphic (AA, AT and TT genotypes) in madura, pesisir, katingan, PO, pasundan, SO, brahman, simental, and limousin cattle. The frequency of A and T alleles were 0,70, 0,68, 0,84, 0,89, 0,70, 0,86, 0,90, 0,73, 0,69 and 0,30, 0,33, 0,16, 0,11, 0,30, 0,14, 0,10, 0,27, 0,31 respectively. The values of Ho and He were 0,60-0,14 and 0,44-0,18 respectively and in Hardy-Weinberg equilibrium (P>0,05). While in Bali cattle was monomorphic (AA genotype). Results of sequencing SNP g.125550A>T of the FTO gene found a transverse mutation A to T at the nucleotide position g.125550. As a result of this study, it can be concluded that SNP 125550A>T of the FTO gene was diverse and potentially used as genetic markers for meat quality in Indonesian beef cattle.Keywords: cattle, FTO gene, PCR-RFLP, SNP g.125550A>T.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh penambahan antidenaturan dan natrium tripolifosfat terhadap sifat fisik ( pH, daya mengikat air, kekuatan gel) dan kimia (kadar air, kadar protein, kadar lemak) nikumi daging kuda.  Pada penelitian ini  diamati penambahan antidenaturan berupa sukrosa dan sorbitol (1:1) dengan level 0% : 0%, 1% : 1%, dan 2% : 2% dan natrium tripolifosfat dengan level 0%, 2.5% dan 5% dari berat daging kuda yang sudah dileaching.  Rancangan penelitian yang digunakan adalah Rancangan acak lengkap pola faktorial dimana antidenaturan sebagai faktor pertama dan Natrium Tripolifosfat sebagai faktor kedua masing-masing diulang sebanyak 3 kali untuk peubah yang diukur. Hasil penelitian menunjukkan penambahan antidenaturan dan natrium tripolifosfat berpengaruh terhadap pH, daya mengikat air, kekuatan gel, kadar air, kadar protein dan kadar lemak pada nikumi kuda.  Peningkatan persentase antidenaturan dan natrium tripolifosfat akan meningkatkan nilai pH, daya mengikat air, kekuatan gel, kadar air  dan kadar protein sedangkan kadar lemak mengalami penurunan Kata Kunci : Nikumi Daging Kuda, Antidenaturan, Natrium Tripolifosfat
