Abstract

Penelitian ini bertujuan untuk mengetahui perkembangan dari siklus hidup Eimeria tenella gambaran histopatologik membran chorioallontois sebagai akibat infeksi sporosista dan oosista Eimeria tenella, untuk mengetahui kemungkinan penggunaan membran chorioallantois sebagai medium k-ultur Eimeria tenella. Penelitian ini menggunakan 50 butir telur ayam berembrio umur 10 hari yang dibagi ke dalam 5 kelompok, masing-masing 10 butir. Kelompok 1, telur diinfeksi dengan 5.000 oosista, kelompok II diinfeksi dengan 10.000 oosista, kelompok 111 diinfeksi dengan 25.000 sporosista, kelompok IV diinfeksi dengan 50.000 sporosista dan kelompok V diperlukan sebagai kontrol. Telur-telur tersebut diinkubasi pada suhu 41°C selama 6 hari. Mulai hari ke 2 setelah infeksi, dua telur dad masing-masing kelompok diperiksa dengan membuka kerabangnya, membran chorioallantois diamati untuk melihat perubahan makroskopik dan histologik. Hasil yang didapat adalah kelompok I dan II tidak ada perbedaan dan oosista tidak berkembang bahkan mengalami nekrosis. Kelompok III dan /V menunjukkan perkembangan di dalam membran chorioallantois, hari ke 2 setelah infeksi sel epitel membesar dan terisi oleh sporozoit, hari ke 3 setelah infeksi sudah ada skizon generasi I, hari ke 4 setelah infeksi sel-sel epitel membesar dan ada stadium makrogamet, skizon generasi II dan zigot, dan had ke 5 setelah infeksi sudah terdapat stadium oosista. Inokulasi sporosista Eimeria tenella pada membran chorioallantois dapat berkembang menjadi stadium berikutnya, sedang inokulasi oosista Eimeria tenella pada membran chorioallantois tidak menunjukkan perkembangan. Membran chorioallantois dapat digunakan sebagai medium kultur Eimeria tenella.Kata kunci : Chorioallantois, Eimeria tenella, Skizon , Makrogamet

Abstract

Study in order to develop molecular techniques using specific primers for the early diagnosis oftoxoplasmosis have been conducted. Detection of Toxoplasma gondii genome was performed usingpolymerase chain reaction (PCR) technique. The primers used in this study were rDNA, P30, and B1. ThePCR products were further run using gel electrophoresis (gel 1.5% – 2.0%) and the band was documented.Toxoplasma was detected at 500 bp and 600 bp using primer P30 and B1, respectively. Whereas usingprimer rDNA no band was observed. It was assumed that primer rDNA was not sensitive since the targetamplification was 88 bp.

Abstract

Coccidiosis is one of the important diseases in poultry industry. In Indonesia the morbidity of thedisease is between 80 to 90%. A rapid and prompt diagnosis would be one of the essential steps ineradication and control of the disease. The objective of this study is to determine the protein profile ofsporozoite of Eimeria tenella isolated in Yogyakarta using sodium dodecyl sulphate polyacrilamide gelelectrophoresis (SDS-PAGE) protein analysis. Protein analysis was performed in 12% polyacrilamide geland further electrophoresis at 100 volts and over-staining with Coomasie brilliant blue. The resultsshowed that the sporozoite of E. tenella isolated in Yogyakarta contained five proteins with molecularweights of 15, 26, 32, 80, and 91 kDa, respectively.

Abstract

The aim of study was, to identify Clinostomum complanatum (Digenea: Clinostomidae) infectingfreshwater fish in Yogyakarta and Riau on the bases of their molecular profiles in the internal transcribedspacer region (ITS1). Samples of climbing gouramy (Anabas testudineus) infected by Clinostomum sp. wereobtained from Kali Progo River, Yogyakarta. Whereas the climbing perch (Trichogaster trichopterus) wereobtained from the Sail River, Riau. Metacercariae of Clinostomum sp. found in the gills and visceralorgans were aseptically removed using needle, preserved in absolute ethanol. Molecular examination wasperformed by Polymerase Chain Reaction method consisted of extraction, amplification, electrophoresisand sequencing of DNA sample. The DNA sewuences of the samples were analysed by maximum parsimonyand neighbour-joining method. Phylogenetic analysis showed that Clinostomum sp. from Yogyakarta wasgenetically idential to Clinostomum complanatum, whereas Clinostomum sp. from Riau was geneticallysuspected as a new species (difference > 2%) which is included in one cluster to Clinostomum phalacrocorasis.

Abstract

The research on developing diagnostic method, vaccine, and drugs for coccidiosis has been focused onthe finding of the immunogenic molecule in Eimeria. The identification of this agent will need the antibodywhich can recognize the biomolecule in the antigen. Antibody that has been developed for this purposeshould be analyzed first, and one of the simple methods for analyzing this antibody is through dot blotanalysis. The objective of this research was to analyze the polyclonal antibody which developed from theoocyst, sporocyst, and sporozoite of  E. tenella using dot blot analysis. The antigen for this polyclonalantibody was made from each of the E. tenella stadium by sonication. Fifteen mice, divided into 3 groups,were then injected subcutaneously with each antigen. The sera from these mice were then collected, analyzedby using ELISA and then it will be used for the dot blot analysis. The research result showed that thepolyclonal antibody which has been developed in mice from each antigen can react with the antigen itself.From this result it can be concluded that the developing of this antibody is successful and it can be used forfurther research in immunoproteomic.

Abstract

Telah dilakukan penelitian tentang infeksi nematoda gastrointestinal pada orangutan (Pongo pygmaeus) di Kebun Binatang Gembira Loka Yogyakarta. Penelitian ini bertujuan untuk mengetahui garnbaran infeksi nematoda gastrointestinal dengan melakukan inventarisasi cacing tersebut di Kebun Binatang Gembira Loka Yogyakarta. Penelitian ini dilakukan dengan mengambil sampel tinja dan 13 ekor orangutan seminggu sekali selama 8 minggu dan diperiksä dengan metode natif, sentrifus dan Mc. Master. Telur cacing diidentifikasi berdasar bentuk dan ukuran, serta dihitung jumlahnya setiap gram tinja. Gejala klinis yang diarnati adalah kondisi tubuh seperti berat badan, aktivitas, nafsu makan dan bentuk tinja. Basil penelitian menunjukkan bahwa semua orangutan di Kebun Binatang Gembira Loka Yogyakarta terinfeksi cacing Strongyloides fitelleborni dan 2 ekor orangutan terinfeksi cacing Trichuris trichiura. Dengan analisis t-test dibuktikan bahwa tidak ada penganih yang nyata antara jenis kelamin dan tipe kandang dengan kejadian nematodiasis pada orangutan. Infeksi yang ditimbulkan oleh cacing Strongyloides fuellehorni cukup berat hingga menimbulkan gejala klinis diare, nafsu makan turun, lemah dan lesu, sedangkan infeksi oleh Trichuris trichiura tergolong ringan. Abstract

This study is conducted to identify species of trematode and its pathological features in pigeons’kidney. Twenty five of Yogyakarta pigeons were examined for trematode infection in kidneys. One ofkidneys was mashed in mortar with a little water, the other was examined for histopathological featuresstained with hematoxyline-eosin.Trematodes found were stained with Schmison ´s Carmine. Seven pigeons(28%) were infected by trematode with non significant clinical features. Identification trematodescharacterized by oral sucker, pharynx, testes are slighty diagonal in position, irregular in shape and intracaecal. Ovary pretesticular and vitellaria widespread from anterior ovarium to the posterior body.Histopathological examination showed segment of trematode in the medullary collecting ducts lumen,dilatation, flattening and emptyness of ducts epithelial cells, emptyness, giant cells and dominationmononuclear cell in interstitial tissue, characterizing a granulomatous nephritis. Trematodes foundidentified as Paratanaisia bragai. The trematodes were found in very dilated medullary collecting ductsand caused inflammation in adjacent tissues. Further studies are needed to find out vector of trematodein pigeons .
