Abstract

Hemagglutination activities and phenotypic expressions of fifty-five S. agalactiae isolates consisted of 19 standard and 36 field isolates from subclinical mastitis cattle were observed. Five (eighteen); 3 (4); 2 (7); 1 (2); and 5 (11) isolates (numbers in brackets indicated field isolates) were able to hemagglutinate cattle, horse, sheep, chicken, and human erythrocytes, respectively. The distribution of hemagglutination pattern was discissed in this paper.

Abstract

The ultimate goal of fresh milk formulations are efficacious against-bird flu and diarrhea (against Escherichia coli and Salmonella enteritidis). Outputs generated in this first year of immunoglobulin Y anti-bird flu and anti-diarrhea in various forms of spray dry yolk, dry freeze egg yolks, and pure extracts. lg Y that collected is located in the egg yolks and tested positive for HSNl virus, E. coli and S. enteritidis. Production of Ig Y begins with the treatment in the form of vaccination in laying hens 4 (four) times during the 4 (four) weeks. The vaccines were used in the form of HSNl in the first and third week, while E. coli and S. enteritidis used every week. Serum and yolk was collected and checked for the presence of anti-lg Y was the third agent since the first week after the last vaccination. Inspection method for E. coli and S. Enteritidis is the order Presipitation Gel Test (AGPT) while the HSNl method Haemagglutination Inhibition (HI). The results give a positive result against all three antigens, but in a different week. This situation is common given the immunity of each chicken is different depending on race, age and environment.

Abstract

Coleus scutellarioides [L.] Benth. known as medicinal plant which contains antidiarhea and antimicrobialcompounds. Extraction of bioactive compound from plants is not efficient because it needs a large biomass,therefore the one of appropiate ways is using endophytic bacteria. The purpose of this research was to isolateand test of antibacterial activity of endophytic bacteria from Coleus scutellarioides [L.] Benth. againsttwo pathogenic bacteria i.e Eschericia coli and Staphylococcus aureus. A total of 22 isolates of endophyticbacteria were obtained from Coleus scutellarioides [L.] Benth. Based on antibacterial activity test, 13 isolatesof endophytic bacteria were able to inhibit the growth of E. coli and 15 isolates were able to inhibit S. aureus,whereas endophytic bacteria which can inhibit both types of pathogens are 10 isolates.

Abstract

Endophytic bacteria are beneficial microorganisms that interact with host plants without causingdisruption or damage to the host. Some studies suggest that certain endophytic bacteria can produce chemicalcompounds that have an effect on health, especially endophytic bacteria isolated from medicinal plants. Greenbetel (Piper betle L.) is a medicinal plant that has been used for years and has many benefits. The purposesof this study are to isolating and screening of endophytic bacteria from green betel against four pathogenicbacteria (Staphylococcus aureus, Escherichia coli, Bacillus cereus and Salmonella enteritidis). The numberof endophytic bacteria that has been isolated are 14 isolates. Based on the screening results, three isolatesof endophytic bacteria have potential activity (characterized by the formation of inhibition zone) against S.aureus. The inhibition zone may indicate that those isolates produce compounds that have antibacterial effects. Those isolates are AS1 , BS1 and BS2. The biggest inhibition zone showed by BS1, so it can be concluded thatBS1 is the most potential isolate as a novel source of antibacterial compound.

Abstract

Paratuberculosis or Johne’s Disease is a granulomatous enteritis chronic disease of domestic and wild ruminants caused by infection of Mycobacterium avium subspecies paratuberculosis. The disease commonly infects dairy cattle with clinical signs of chronic diarrhea, decreasing body weight, low milk production, oedema, anemia and occasionally infertility. The basic procedure in order to control Paratuberculosis in farms is to do a good and proper handling of animal faecal. Disinfection of animal environments such as pens, faecal, sewerage and sewage are important in prevention of transmission of this disease. The purpose of this research is to determine specific disinfectan and dosage for Mycobacterium avium subspecies paratuberculosis decontamination in cattle feces so  it can be applied as disease control measures. Cows feces were contaminated with MAP 105CFU/ml and treated with ammonium quartener, phenolic and formaldehyde disinfectant doses 10%, 15% and 20%. The effectiveness of the disinfectant was tested based on MAP identification using Löwenstein-Jensen culture medium and nested Polymere Chain Reaction(PCR). The results showed 15% and 20% doses of formaldehyde disinfectants efective to decontaminate Mycobacterium avium subspecies paratuberculosis  in catle feces.

Abstract

This study aims to analyze antimicrobial activity of Allium chinense G. Don extract against Eschericia coli, Salmonella typhi, Staphylococcus aureus, Bacillus subtilis and Candida albicans, and to examine the active compounds. Allium chinense G. Don was extracted using maseration method and treated with ethanol 70% (v/v), ethanol 96% (v/v), ethyl acetate, n-hexane, and aquadest. Antimicrobial activity assay was conducted using agar difusion method and compounds analysis using Gas chromatography-mass­spectrometry (GC-MS). Antimicrobial activities assay showed that all extracts could inhibit microbial growth. Ethyl acetate extract has the highest antimicrobial activity against all the microbial test. Minimum inhibitory concentration (MIC) of ethyl acetate extract against C. albicans, B. subtilis, E. coli, S. aureus, S. typhi was 25, 100, 250, 250, 1000 mg ml-1 respectively. As many as 25 compounds were derived from GC-MS analysis and most of them were known as the antimicrobial compounds. This study revealed that Allium chinense G. Don contains biologically active compunds as antimicrobial agent particularly anti Candida.

Abstract

Endophytic bacteria can produce antibacterial compounds. Their existence in the medicinal plant of Ficus variegata Blume enables the production of bioactive compounds similar to those contained by the host plants. The purpose of this study was to isolate and identify endophytic bacteria from F. variegata which is potential to produce antibacterial compounds. The methods used include isolation of endophytic bacteria from leaves, stem, aerial root, and fruit of F. variegata plants. Antibacterial activity assay was done against four types of bacteria i.e. Staphylococcus aureus, Bacillus subtillis, Escherichia coli, and Psedomonas aeruginosa. Identification of endophytic bacteria was conducted based on morphological analysis, biochemical test, and molecular analysis of 16S rRNA. Endophytic bacterial culture was extracted by ethyl acetate and analyzed by GC-MS. A total of 29 isolates of endophytic bacteria were obtained from F. variegata. The BH2 isolate was found to have potential activity. Analysis of 16S rRNA showed that BH2 isolate was related to Pseudomonas aeruginosa strain SV1 with 99 % identity. The result of GC-MS analysis showed that the antibacterial compound was Nonanoic acid ethyl ester

Abstract

This experiment aims to produce immunoglobulin Y (Ig-Y) Staphylococcus aureus isolates origin staphylococcosis case in rabbits. Vaccination is done several times using Staphylococcus aureus active in 109 cfu/ml. Injecting the chicken is done four weeks in a row, the first week is done by intravenous injection of S. aureus antigens, the second week of the injection sub cutan S. aureus emulsified with Freunds complete adjuvant, followed by the third and fourth week of the injection of S. aureus antigen emulsified with incomplete Freunds adjuvant. Egg samples were taken after four weeks of the last injection for identification, purification, and determination of specific IgY against S. aureus in egg yolk. IgY specificity qualitatively tests performed by the AGP test (To Gel Precipitation). IgY extraction is done using PEG - Ammonium sulfate, purified IgY concentration calculated by the method of Bradford. The results showed that IgY began to be detected in egg yolk at week six after immunization, with the average levels of IgY is 1.7 mg/ml.
