Abstract

ABSTRACT---One of the Indonesian marine natural resources, brown alga of Turbinaria decurrens, used in pharmacy in order of carotenoid pigment as antioxidant. We interested in analysis B-caroten in Turbinaria decurrens and antioxidant and anti-tumor activity. The method that is use to divide carotenoid is the hierarchy maseration method using n-hexane, ethyl acetate  and methanol  and  then using chromatography column. To analyze the qualitative carotenoid uses HPLC  by Crc column and eluenmethanol-acetonitrile  (3:I).  Meanwhile,  the test of bioactivity carotene  uses  radical DPPH  (1Idifenil-2-pibilhidrozil)  to  test  of  antioxidant  qnd  sitotoksik asscy with  MTT  [3,(4,5-dimethylthiazol-2yl)-2,5-diphenyltetrcaolium  bromideJ  for  HeLa tumor cells  to test of anti-tumor. The  result of this research  is  B -caroten  thqt is in qctract  has 0.00387%.  Moreover,  bioacttvity  test  shows  that B -caroten  fraction does  notactive  to neutralize  of DPPHfree radical than  ascorbic  aci{  but  it shows  the  acttvity  to kill HeLa  tumor  cells.Keywords  : Turblnaria decunens,  Carolenoid,  Antioksidan,  Antitumor

Abstract

Marine organisms, in particular nudibranchs (Mollusca: nudibranchia), are known as a rich source of chemically diverse secondary metabolites exhibiting potential as antimalarial, anti-inflammatory, antiviral and anticancer activity. We presented the chemical investigation of an extract of nudibranch Hypselodoris infucata collected from Bali, an unexplored water but rich in nudibranch diversity. The extract contained the known (–)-furodysinin (1), a furanosesquiterpene that for the first time isolated from this species. Metabolite 1 was purified by chromatography and the structure was characterised by comparison of 1H NMR data with that of the reported data. The absolute configuration was determined by comparing the optical rotation values with the known enantiomer. In vitro cytotoxic activity of compound 1 against HeLa cell line displayed an IC50 at 102.7µg/mL. We also report for the first time the development of a method to assay nudibranch extracts for their feeding deterrence activity against local shrimps Penaeus vannamei. The extract show food rejection with highly significant difference in respect to the control (P = 0.0061) at natural concentration of 3.0mg/mL.Keywords: nudibranch, natural product, feeding deterrence, Hypselodoris

Abstract

Fatty acids and carotenoid has been known as an anticancer agent on both preventing and treating cancer disease. This study was conducted to analyze the fatty acid profile, carotenoid and in vitro anticancer activity of 12 sea cucumber harvested from Karimunjawa and Lampung waters. The aim of the study was to determin the potency of sea cucumbers as raw material for nutraceutical products. Fatty acid profile and carotenoid content were characterized by gas chromatography and spectrophotometry techniques, while in vitro anticancer activity was assessed by MTT assay against cervix (HeLa), breast (T47D and MCF-7) and colon (WiDR) cancer cells. Results of the study showed polyunsaturated fatty acid (PUFA) dominated the composition of fatty acids in the samples from both locations. Holothuria sp. was detected to contain the highest amount of carotenoid. Furthermore, the highest in vitro anticancer activity was detected also in the sample of Holothuria sp. The activity of 30 ppm Holothuria sp. extract against HeLa cell was detected to be almost equal to the 5 ppm doxorubicin control. Concentration of 5 ppm Holothuria sp. extract also showed positive result in killing 50% of MCF-7 and T47D, but capable to 100% kill HeLa and WiDR cells. At concentration of 25 ppm, the extract was able to kill all the 4 cells tested. Statistical analysis showed the amount of carotenoid and two particular fatty acid compounds (docosadienoic and eicosapentaenoic acid) significantly (P<0.05) contributed to the cytotoxic activity that was found in the sea cucumber samples. Those compounds were found in highest concentration from Holothuria sp harvested from Lampung waters, thus being the most prospective raw material for nutraceutical or functional food ingredient with anticancer potency.

Abstract

Spons merupakan hewan multiseluler yang paling sederhana sekaligus merupakan sumber senyawa bioaktif yang sangat berguna di dalam bidang farmasi. Senyawa-senyawa baru yang sangat beragam sudah banyak yang diisolasi dari spons. Kesulitan utama dalam memproduksi senyawa obat baru dari spons adalah kurang mencukupinya jumlah senyawa bioaktif yang dihasilkan spons untuk keperluan uji preklinis dan klinis karena rendemen yang dihasilkan sangat rendah. Artikel ini merupakan tinjauan terhadap metode kultur sel spons untuk memproduksi senyawa bioaktif  dalam rangka mengatasi masalah suplai senyawa bioaktif yang diuji.  Produksi senyawa avarol dari kultur primmorph spons Dysidea avara dibahas dalam review ini.  Avarol dilaporkan  memiliki bioaktivitas yang kuat sebagai substansi antitumor, antibakteri, dan antivirus. Model pendekatan melalui kultur primmorph dapat digunakan sebagai cara yang menjanjikan  untuk memproduksi senyawa bioaktif dari spons.

Abstract

This research was aimed to isolate marine derived-fungi which associated with macro algae and sponge from Indrayanti Beach, Jogjakarta and to screen the antitumor (T47D and HeLa) bioactivity of the fungi extracts. Three solid media of MEA (malt extract agar), GPY (glucose peptone yeast) and MFM (minimal fungi medium) were used as isolation medium. Each of the pure fungi isolates was then cultivated in 100 ml of liquid medium for 4 weeks at room temperature (27-28°C) in static conditions. The antitumor activity of the fungi extracts were tested against breast tumor cells (T47D) and cervical cancer cells (HeLa) using Thiazolyl Blue Tetrazolium Blue (MTT) assay method. A number of 21 isolates of fungi were isolated from 4 macro algae and 1 sponge samples. The identification of fungi isolate was conducted using combination of molecular approach (ITS1-5.8S-ITS4 DNA regions) and macro-micro morphological characteristics. Among those 21 marine fungi species isolated, MFGK-21 extract showed the best anti-servical tumor (HeLa) with an IC50 value of 240.1 µg/ml and MFGK-27 extract showed the best anti-breast tumor (T47D) with an IC50 value of 59.6 µg/ml. The MFGK-21 fungi isolate was identified as Penicillium steckii, while the MFGK-27 fungi isolate was identified as Aspergillus sydowii.

Abstract

Sea cucumbers are one of Indonesia’s marine organism with the potential to be developed as possible herbal medicines. In a preliminary study of cytotoxic activity of ethanol extracts of 14Indonesian sea cucumber species, the most active extract came from the Holothuria sp. Thecurrent research aimed to identify the major cytotoxic component in the sea cucumber Holothuriasp., that yielded the most active extract. The samples were collected from South Lampung beach.Isolation of the cytotoxic component was done employing liquid flash and preparative reversedphase (C 18) chromatography. Cytotoxic ity evalu atio n of fractio ns collected durin g thechromatography was conducted using the MCF-7 tumor cell line. Elucidation of the structure ofmost active isolate was done by NMR (Nuclear Magnetic Resonance) spectroscopy and GC-FID(Gas-Chromatography-Flame Ionisation Detector) analysis. The results of these analyses showedthe most active compound to be stearic acid; IC 50towards MCF-7 cells 10.32 ppm.

Abstract

Marine algae contains bioactive secondary metabolites that  have potential to be developed as antioxidant. The aims of  this research were to investigate antioxidant activity and total phenolic compound of marine algae collected from Binuangeun Beach, Banten and to characterize antioxidant compounds from selected algae species. Antioxidant activity was determined by 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) assay and Ferric Ion Reducing Antioxidant Power (FRAP) Assay, while total phenolic content was estimated by Folin-Ciocalteau method. Isolation of bioactive antioxidant compound was performed using SiO2 column chromatography and preparative high performance liquid chromatography (HPLC). The antioxidan activity evaluation was conducted to methanol extracts  from 20 species (7 Chlorophyta, 9 Phaeophyta and 4 Rhodophyta). Results showed that Padina australis extract was found to be the most active. The major bioactive antioxidant compound was identified as fucoxanthin and a polar compound that was suspected as phenolic compound. The extracts of P. australis presented the highest phenolic content (58.59 mg GAE/g). A significant correlation between antioxidant capacity and total phenolic content was found, indicating that phenolic compounds are the major contributors to the antioxidant properties of P. australis.

Abstract

Skrining senyawa bioaktif ekstrak metanol dari karang lunak familia Alcyoniidae telah ditakukan. Uji hayati dilakukan terhadap ekstrak metanol dari 4 jenis karang lunak yaitu sinularia leptoclados, Lobophytum pauciflotum, Lobophytum A dan Lobophytum B Skrining dilakukan dengan menggunakan 3 jenis uji hayati yaitu uji toksisitas metode Brine Shrimp Lethatity Test (BSLT); uji sitotoksisitas menggunakan ser kanker HeLa dengan metode uji 3-[4,5-dimethyrthiaz or-2yry-2,idiphenyltekazolium bromide (r.4TT assay), dan uji antibakteri menggunakan metode agar difusi.

Abstract

Penelitian yang bertujuan untuk mengetahui hubungan antara keberadaan duri dengan toksisitas ekstrak metanol karang lunak dan sponge telah dilakukan. Sebanyak 28 jenis ekstrak karang funak dan sponge diuji toksisilasnya menggunakan uji Brine Shrimp Lethality Iest (BSLT). Perbedaan mortalitas Artemia salina akibat pemberian ekstrak diuji dengan uji t dua sampel independen. Korelasi antara keberadaan duri dengan tingkat toksisitas dianalisis dengan korelasi nonparametrik Spearman.

Abstract

Penelitian ini bertujuan untuk mengetahui aktivitas sitotoksik dua jenis algae coklat (Sargassum cinerum dan Sargassum polycystum) dan kandungan fukosantin kedua jenis rumput laut tersebut. Rumput laut diperoleh dari pantai Binuangeun, Lebak, Banten, Indonesia. Ekstrak aseton yang diperoleh dipekatkan dengan menggunakan konsentrator vakum. Aktivitas sitotoksik terhadap sel HeLa dan T47D dievaluasi dengan menggunakan uji MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide). Kandungan fukosantin pada ekstrak aseton S. cinerum dan S. polycystum dianalisis dengan menggunakan Kromatografi Cair Kinerja Tinggi (KCKT). Hasil uji sitotoksik memperlihatkan bahwa ekstrak S. cinerum dan S. polycystum memiliki aktivitas sitotoksik yang lebih tinggi terhadap sel T47D dibanding terhadap sel HeLa. Nilai IC50 S. cinerum dan S. polycystum terhadap sel T47D masing-masing sebesar 79,2 dan 52,2 µg/ml. Analisis KCKT memperlihatkan kadar fukosantin pada ekstrak aseton S. cinerum dan S. polycystum masing-masing sebesar 0,179 dan 0,318 mg/g ekstrak.
