Abstract

Temulawak (Curcuma  xanthorrhizaRoxb.) belongs to the family Zingiberaceae, has been empirically used as herbal medicines. The research was aimed to evaluate three promising lines of Temulawak based on their high bioactive contents (xanthorrhizol and curcuminoid) and its in vitro bioactivity (antioxidant and toxicity), and to obtain information on agrobiophysic environmental condition which produced high bioactive compounds. The xanthorrhizol and curcuminoid contents were measured by HPLC. In vitro antioxidant and toxicity were determined by DPPH (1,1-diphenyl-2-picryl-hydrazyl) method and BSLT (Brine Shrimp Lethality Test). The result showed that promising line A produced the highest yield of bioactive and bioactivity, i.e. 0.157 and 0.056 g plant-1of xanthorrizol and curcuminoid respectively. The IC50 of antioxidant activity was 65.09 mg L-1and LC50of toxicity was 69.05 mg L-1. In this study, Cipenjo had the best temulawak performance than two other locations. According to the agrobiophysic parameters, Cipenjo environmental condition was suitable for temulawak cultivation with temperature 28-34 ºC, rainfall ± 223.97 mm year-1 and sandy clay soil. Abstract

The crude ethanol extracts of four Indonesian medicinal plants namely Curcuma xanthorrhiza Roxb.,Phyllanthus niruri Linn., Andrographis paniculata Ness., and Curcuma aeruginosa Roxb. wereexamined for their antioxidant (radical scavenging) activity using 2, 2-diphenyl-2-picrylhydrazyl(DPPH) free radical and cytotoxicity using brine shrimp lethality test (BSLT). The total phenoliccontent was used the Folin-Ciocalteu method. IC50 values for DPPH radical scavenging activityranged from 14.5 to 178.5 μg/ml, with P. niruri having the lowest value and therefore the mostpotent, and C. aeruginosa having the highest value. LC50 values for BSLT ranged from 210.3 to593.2 μg/ml, with C. xanthorrhiza and A. paniculata having the lowest and highest values,respectively. The total phenolic content of the Indonesian plants ranged from 133.0 ±3.7 to863.3±54.7 mg tannic acid equivalent per 1 g extract, with C. aeruginosa and P. niruri having thelowest and highest values, respectively. A positive correlation between free radical scavengingactivity and the content of phenolic compounds was found in the four of Indonesian medicinal plants.

Abstract

Abstract: The Curcuma xanthorrhiza rhizome, Curcuma longa rhizome, and Phylanthus niruri herbs has been used medicinal plant in Indonesia as a single herb for medicine. The purpose of this research was to determine potency of biological and chemical finger printing of extract herbs combination of C. xanthorrhiza, C. longa, and P. niruri. The extract herbs combination were made with equal combination from C. xanthorrhiza, C. longa, and P. niruri. The potency of biological activities of extract herbs combination was investigated using a brine shrimp lethality test (BSLT). The chemical finger printing of extract herbs combination were investigated using Fourier transformation infra red (FTIR) and thin layer chromatography (TLC). The potency of biological of extract herbs combination expressed by LC50 value from all combination herbs were in the range from 5.54 to 324.91 ppm. The combination herbs on 1:1:1 was the best potency of biological showed  TLC profile with thick spot and without C≡C bond for FTIR spectrum. Abstrak: Rimpang Curcuma xanthorrhiza, rimpang Curcuma Longa, dan herbal Phylanthus niruri telah digunakan tanaman obat di Indonesia sebagai ramuan tunggal untuk obat. Tujuan dari penelitian ini adalah untuk menentukan potensi biologis dan kimia dari kombinasi ekstrak herbal C. xanthorrhiza, C. Longa, dan P. niruri. Kombinasi ekstrak herbal dibuat dengan kombinasi yang sama dari C. xanthorrhiza, C. Longa, dan P. niruri. Potensi aktivitas biologis dari kombinasi ekstrak herbal diselidiki menggunakan tes kematian udang laut (brine shrimp lethality test, BSLT). Sifat kimia kombinasi ekstrak herbal diselidiki menggunakan Fourier transformation infra red (FTIR) dan kromatografi lapis tipis (KLT). Potensi biologis dari kombinasi ekstrak herbal diekspresikan oleh LC50 bahwa nilai dari semua herbal kombinasi berada di kisaran 5,54-324,91 ppm. Pada kombinasi herbal 1:01:01 memiliki potensi bioloogi terbaik seperti ditunjukkan pada profil KLT dengan spot yang tebal dan tanpa ikatan C ≡ C untuk spektrum FTIR.

Abstract

Temulawak or Curcuma xanthorrhiza Roxb, is usually used as traditional medicine (herbal medicine) that has antioxidant, anticancer, antihyperglycemic, anti-inflammatory, hepatoprotective and gastroprotective properties. The main components contained in temulawak responsible for its efficacy as a medicine are xantorhizol and curcuminoid. Curcuminoid has drawbacks, which are difficult to absorb and very quickly metabolized by the body, so that limit its bioavailability. The use of solid lipid nanoparticle carrier system (SLN) in form of palmitic acid, is known to improve the bioavailability of curcuminoid. This study aims to find the effective dose of nanocurcuminoid coated with palmitic acid that can be used as an anti-inflammatory agent. The methods used in this study, include the production of nanocurcuminoid with homogenization and ultrasonication methods, determination of particle size, polydispersity index, entrapment efficiency and anti-inflammatory activity test through rat feet edema. Nanocurcuminoid obtained in this study was 561.53 nm in size, with polydispersity index 0.309 and concentrations of curcuminoid absorbed and entrapment efficiency were 0.61±0.031 mg/mL, 58.93±3.021%, respectively. Anti-inflammatory activity of nanocurcuminoid through treated Sprague Dawley rats, showed that there were no significant difference compared with the positive control, curcuminoid extracts and empty nanoparticle. These results indicate that nanocurcuminoid with 175, 200 and 250 mg/kg.bw in doses, has greater anti-inflammatory activity (31.70%) compared to the other treatments.

Abstract

The species Graptophylum pictum (L.) Griff, also known as “daun ungu” in Indonesia, is a traditional herbaceous plant believed to have antidiabetic potential. The number of people in the world with diabetes has increased dramatically over the recent years. The treatment of type II diabetes is complicated by several factors inherent to the disease. Elevated postprandial hyperglycemia is one of the risk factors and the intestinal digestive enzyme α-glucosidase plays a vital role in carbohydrate metabolism. One of the antidiabetic therapeutic approaches which reduces the postprandial glucose level in blood is by the inhibition of α-glucosidase. In this study, phytochemical analysis, α- glucosidase inhibitory activity and enzyme kinetics of ethyl acetate- and hexane extracts of G. pictum were evaluated with the aim to analyze its antidiabetic potential. Phytochemical analysis revealed the presence of tannins, steroids, and alkaloids. Steroids were present in ethyl acetate extract but absent in hexane extract, while alkaloids were present in hexane extract but absent in ethyl acetate extract. The ethyl acetate and hexane extracts had 30.68 and 49.82 % inhibitory effect on α-glucosidase activity respectively. The kinetics of glucosidase enzyme of ethyl acetate and hexane extracts were determined by Lineweaver Burk plots. These exhibited uncompetitive and noncompetitive inhibition to alpha-glucosidase activity respectively. From the enzyme assay, we infer that ethyl acetate and hexane extracts of G. pictum contain potential α-glucosidase inhibitors that have the potential to be exploited for use in the treatment of diabetes

Abstract

Wungu leaf (Graptophyllum pictum (L.) Griff) is a plant thought to have potential use in alleviating symptoms of diabetes mellitus. The purpose of the present study was to evaluate the activity of wungu leaf extracts in decreasing blood glucose level of alloxan (200 mg/kg BW)-induced hyperglycemic mice. Extracts of wungu leaf were obtained by macerating with ethanol and then partitioning the extract with diethyl ether, ethyl acetate, and butanol. Each extract obtained was used to treat hyperglycemic mice for 28 days. The results showed that wungu leaf extracts have the ability to decrease the blood glucose level of hyperglycemic mice (dose 50 mg/kg BW). The ethyl acetate extract showed the highest activity, bringing about a decrease of blood glucose of 37.6 %. The wungu leaf extract has the potential to be developed as a source of anti-diabetic agents.

Abstract

Temulawak exctract contains curcuminoids which have anticancer potential. However, clinical application of curcuminoid has been limited due to its low bioavailability. One of the efforts that can be developed to solve this problem is incorporated curcuminoids into Solid Lipid Nanoparticles (SLN) carriers system. The objective of this study was to characterize dan evaluate anticancer potential of temulawak ethanolic fraction nanoparticles. HPLC method was used to determined curcuminoids content of temulawak ethanolic fraction. Characterization indicators like polydispersity index, particle size, morpholgy, and entrapment efficiency. HPLC chromatogram has shown of curcumin, demethoxycurcumin, and bisdemethoxycurcumin were found in temulawak ethanolic fraction. The particle size of nanoparticles obtained in this study was 648.4 ± 95 nm with polydispersity index value of 0.216. A uniform size distribution of nanoparticles as observed by Transmission Electron Microscopy (TEM). The entrapment efficiency of curcuminoid in nanoparticles was about 29.8%. Based on results of BSLT obtained temulawak extract Lethal Concentration (LC50) value of 213.24 ppm and 828.78 ppm of nanoparticles.

Abstract

FTIR spectroscopy combined with principal component analysis and discriminant analysis were used for identification and authentication of jahe merah from its related species namely jahe emprit and jahe gajah. FTIR spectra of all samples were recorded in the wavenumber range 4000-400 cm-1 and then subjected for preprocessing signal such as normalization and baseline correction. Combination of FTIR spectra with some chemometrics method such as principal component analysis and discriminant analysis were used to distinguish the three varieties of ginger. By using discriminant analysis, the three varieties of ginger were classified according to its variety. The developed method could be used for identification and authentication of jahe merah.ABSTRAKSpektroskopi FTIR dan kombinasinya dengan analisis komponen utama dan analisis diskriminan telah digunakan untuk identifikasi dan autentikasi jahe merah dari dua spesies yang berkerabat dengannya yaitu jahe emprit dan jahe gajah. Seluruh contoh dibuat spektrum FTIR pada kisaran bilangan gelombang 4000-400 cm-1 dan diberi perlakuan pendahuluan seperti normalisasi dan koreksi garis dasar. Kombinasi spektrum FTIR dengan beberapa metode kemometrik seperti analisis komponen utama dan analisis diskriminan digunakan untuk membedakan ketiga varietas jahe. Analisis diskriminan dapat mengelompokkan ketiga varietas jahe sesuai dengan jenis varietasnya. Metode yang dikembangkan ini dapat digunakan untuk tujuan identifikasi dan autentikasi jahe merah.
