Abstract

Analisis logam berat Cu, Zn, dan Pb pada lamun Halodule uninervis dari pulau Barrang Caddi Kotamadya Makassar telah dilakukan dengan menggunakan spektrofotometer serapan atom tanpa nyala. Hasil analisis menunjukkan bahwa konsentrasi logam Cu lebih tinggi jika dibandingkan dengan konsentrasi logam Zn dan Pb. Konsentrasi logam Cu, Pb, dan Zn pada setiap bagian morpologi lamun masing-masing adalah, 1,97 – 3,20; 0,01 – 0,13; 0,01 – 0,16 ppm. Konsentrasi Cu tertinggi pada batang (2,89 -3,20 ppm), dan pada daun 1,97 – 2,47 ppm. Konsentrasi logam Zn pada umumnya lebih rendah dibagian batang daripada pada akar dan daun. Sedangkan untuk konsentrasi logam Pb tertinggi pada bagian batang dibanding konsentrasi Pb pada akar dan daun.   Kata kunci : lamun, logam berat , analis

Abstract

Chitin from crab shell (Scylla serrata) can be converted into chitosan using the enzyme of chitin deacetylase isolated from thermophillic microbe. Conversion process  initiated with microbe rejuvenation, grown in solid medium at temperature 55 oC and pH 7,0. Next optimum time determination producing the enzyme, fractionation of enzyme and producing the chitosan and finally analysed Infra Red spectrophotometry. Thermophillic microbe can yield the enzyme with optimum activity in  three to four days and fractionated with the amonium sulfate 40 % (w/v). Infra red spectrosgram data, chitosan was obtained with the deacetylated degree between 66.53 – 78.9 %, water content 5.75 – 7.56 % and ash content between 33.26 – 34.25 %.   Keywords :  Thermofilik microbe,  crab shell, chitin and  chitin deacetylase

Abstract

An investigation on the content and the type of aromatic hydrocarbon found in the sediment of the coastal area has been carried out using microgravimetric and spectrometric methods. From microgravimetric result, it was found that there are 131.7 – 1738.7 mg/kg dry sediment of Extractable Organic Matter (EOM), 49.85 – 533.72 mg/kg dry sediment of Total Hydrocarbon (TH), 20.68 – 508.67 mg/kg dry sediment n-Alkana Fraction (F1), and 13.23 – 320.94 mg/kg  dry sediment Aromatic Fraction (F2). From synchronized spectro-fluoroscence analysis, it was estimated that the types of aromatic hydrocarbons found in the sediment were benzene, fluorene, fluoranthene, naphthalene, phenyl -1- naphthalene, chrysene, phenantrene, anthracene, and 3,4-benzopyrene. The hydrocarbon in station A, B, and C are anthropogenic hydrocarbon while in station D,E, and F are natural source. A modelling of aromatic sources in sediment is also proposed.

Abstract

An investigation to convert chitin into its deacetylated product, chitosan, has been conducted in this research. Prior those deacetylation work, the chitin was isolated from waste of crab (Portunus pelagicus, L) undergoes 3 (three) main steps : deproteination, demineralisation, and decolourisation. It was found that the optimum condition of deacetylation process which gave 86 % yields were at the ratio of chitin- NaOH solvent 1 : 20, 2 molar of NaOH concentration, and temperature of 80 ºC respectively. These resulted chitosan was also verified with an FTIR spectroscopy that gave the identical spectrum with the chitosan FTIR standard spectrum. Abstract

Enzymatic bioconversion of chitosan from crab shell chitin using thermophillic bacterium has been carried out. With chitinolytic optimum activity, called M-1 isolator, at 550C and pH 7.0, the microbe could grow best at fourth day of fermentation with O.D. of 0.62 as well as the enzyme at 7.03 UA/mL. With the same optimum condition, deacetylated chitin enzyme, utilised for converting chitin into chitosan, was also produced at 7.95 UA/mL. After further purification with ammonium sulfate 40%, chitosan was produced, examined, and confirmed accordingly. Keywords : Bioconversion, chitin, shell, chitosan, fermentation

Abstract

A study towards the influence of solvent concentration and temperature variations in extraction pocess of chitin from crab shell as well as its qualitative examination has been investigated in this research. The result showed that the concentrations for the optimum process of deproteinations using sodium hydroxide, demineralisation with hydrochloric acid and decolorisation by hydrogen peroxide were 3.5%, 1.5 N and 0.35% respectively. their best extraction temperatures were 85oC for deproteinations, 35oC for both demineralisation and decolorisation processes. These optimum extraction conditions represent the need for a suitable reactor design allowing sequential acid-base treatments. futher upscaling effort may be required.Keywords : Chitin, extraction, crab, qualitative

Abstract

Telah dilakukan usaha mengisolasi komponen kimia daun Paliasa, Kleinhovia hospita Linn., berdasarkan variasi kelompok polaritas mulai dari tetapan dielektrik 2 sampai 78. Proses pemisahan dimulai dari maserasi, fraksinasi, ekstraksi, dan dilanjutkan dengan identifikasi kromatografik lapis tipis. Telah ditemukan 109 isolat yang cukup murni dan dapat dilanjutkan untuk analisis struktural komponen yang bersangkutan. Hasil analisis empat isolat dengan spektrometri IR, UV dan GC-MS menunjukkan adanya kelompok senyawa terpenoid dan fenolik (serapan puncak UV pada 207, 228, dan 297 nm) sebagaimana yang lazim dijumpai dalam senyawa bahan alam lainnya. Sejauh yang diketahui belum ada rujukan struktural komponen kimia daun Paliasa selain yang telah dilakukan. Perlu penelitian lebih banyak dalam skrining maupun bioassay esktrak yang dihasilkan.   Keywords : Isolasi, identifikasi, paliasa, polaritas

Abstract

A triterpenoid derivative compound, oleane-12-en-2,3,23-triol-28-oat, from chloroform fraction of root wood ofPterospermum subpeltatum C. B. Rob (Sterculiaceae) was identified using IR, 1H dan 13C NMR data. The compoundhas shown toxicity against Artemia salina, LC50 of 46.06 μg/mL, and antibacterial potential upon Shigella boydii andStaphylococcus aureus with inhibition zones of 8.3 and 9.8 mm respectively.

Abstract

ABSTRAK Telah dilakukan identifikasi kuantitatif jenis-jenis asam lemak lamun dari spesies Thalassodendron ciliatum, Cymodocea rotundata, Cymodocea serrulata, Thalassia hemprichii, dan Enhalus acoroides. Analisis lemak contoh yang diambil di perairan kabupaten Donggala ini dilakukan dengan metode soxhlet dan analisis asam lemak menggunakan metode GC. Ternyata diperoleh variasi kandungan lemak antara 0,27 % - 1,01 %, sementara konsentrasi asam lemak jenuh yang ditemukan bervariasi antara 4,39 % - 8,03 %, asam lemak tak jenuh bervariasi antara 4,48 % - 18,39 %, dan omega 3 (EPA) bervariasi antara 0,27 % - 1,24 %. Hasil di atas, jika digabungkan bersama hasil analisis protein, fosfat, dan mineral dapat menjadi dasar untuk memperkirakan status kesuburan lamun. Kata kunci : Lamun, asam lemak, ABATRACT The quantitative analyses of fatty acids various was conducted from the seagrass Thalassodendron ciliatum, Cymodocea rotundata, Cymodocea serrulata, Thalassia hemprichii, and Enhalus acoroides speciess. The sampel takken from Donggala coast is analyses as the soxhlet method and the Gas Chromatography. The result obtained obstetrical variation of  fatty are vary among 0,27 - 1,01 %, for a while concentration of saturated fatty acids are vary among 4,39 - 8,03 %, unsaturated fatty acid are vary among 4,48 - 18,39 %, and omega 3 (EPA) are vary among 0,27 - 1,24 %. The result is it is joined with the protein, phosphate, and mineral analyses result can become basic for estimate the fertility status the sea grass. Keyword: Sea grass, fatty acid

Abstract

An isolation and structure determination of secondary metabolites of sponge of Biemna triraphis has been carried out. Separation techniques used consisted of  maceration (ethanol) and fractionation (mixture of n-hexane & acetone), while structural determination were conducted using GC-MS, H-NMR, and IR. Activity tests to Artemia salina were applied. White crystals were found with Rf value 0.68 on TLC with eluent of n-hexane:acetone 5:2 and m.p 1300C. Instrumental analysis indicated fragmentation values (m/e) were  414, 396, 381, 371, 329, 303, 273, 255, 231, 213, 159, 57, and 43 and chemical shift of  H-NMR (δ) were 0.678, 0.819, 0.854, 0.905, 1.008, 3.525  and 5.345 and infra red  absorptions, n (cm-1) were  3432.67,  2931.26,  2869.55,  1635.33,  1457.92,  1380.78, 1056.79, and 802.24. Apparently, β-sitosterol was the most possible candidate for the structure of the compound. Activity test results also indicated its high bioactivity.   Keywords : isolation, sponge, b- sitosterol
