Abstract

Chitin from crab shell (Scylla serrata) can be converted into chitosan using the enzyme of chitin deacetylase isolated from thermophillic microbe. Conversion process  initiated with microbe rejuvenation, grown in solid medium at temperature 55 oC and pH 7,0. Next optimum time determination producing the enzyme, fractionation of enzyme and producing the chitosan and finally analysed Infra Red spectrophotometry. Thermophillic microbe can yield the enzyme with optimum activity in  three to four days and fractionated with the amonium sulfate 40 % (w/v). Infra red spectrosgram data, chitosan was obtained with the deacetylated degree between 66.53 – 78.9 %, water content 5.75 – 7.56 % and ash content between 33.26 – 34.25 %.   Keywords :  Thermofilik microbe,  crab shell, chitin and  chitin deacetylase

Abstract

An investigation of acidophillic microorganism, producing enzym that can cause chitin degradation, has been carried out. Microbial exploration from soil and water samples, found in several regions in West Java, was carried out by spreading out samples in the acidic medium contained 1 % of colloidal chitin and samples were incubated at various temperature. From isolation result, there were 36 isolates that  have chitinase activity. The isolates were purified and 3 isolates were pure which can grow at pH of 4 – 5 for 4 days with the chitonolitic indeks of 1.6, 1.5 and 1.4 for samples from K-2,  K-22 and K-24, respectively. The three potential isolates were identified and it was found that the three isolates had characteristic of gram positive with a form of bacillus, had spores as well as anerobic and motillity properties. The activities of the three isolates were then  investigated. Results showed that  the optimum pH and temperature for the isolates were 5.0 and 37 0C. Key words: microorganism, acidophilic, chitinase

Abstract

A research on the ability of protein fraction from red algae Gelidium amansii to inhibit the growth of Salmonella thypi and Staphylococcus aureus has been conducted. Proteins was fractionated from the crude extract by salting out method with  0 – 20 %, 20 – 40 %, 40 – 60 % and 60 – 80 % ammonium sulfat saturation. Proteins was purified by a dialysis method using a selophan membrane. The protein level was determined by Lowry’s method, the highest protein concentration, 5.6 mg/mL was found in the 40 – 60 % fraction  of the red algae. Antibacterial activity test was performed using a layered jelly diffusion method on the  MHA medium. Results indicated that crude extract and the protein fraction of the red algae had antibacterial effect to Salmonella thypi and Staphylococcus aureus. The highest antibacterial activity to Staphylococcus aureus was found in the 0-20% fraction with the inhibition diameter of 14.43 mm after incubated for 24 hours. These research might provide a significant results for the following research on the antibacterial action in molecular  level of protein compounds from marine algae.

Abstract

Enzymatic bioconversion of chitosan from crab shell chitin using thermophillic bacterium has been carried out. With chitinolytic optimum activity, called M-1 isolator, at 550C and pH 7.0, the microbe could grow best at fourth day of fermentation with O.D. of 0.62 as well as the enzyme at 7.03 UA/mL. With the same optimum condition, deacetylated chitin enzyme, utilised for converting chitin into chitosan, was also produced at 7.95 UA/mL. After further purification with ammonium sulfate 40%, chitosan was produced, examined, and confirmed accordingly. Keywords : Bioconversion, chitin, shell, chitosan, fermentation

Abstract

Bacillus licheniformis HSA3-1a is able to produce extracellular protease. The aim of this research was to find out its production optimum time, consentration of MnCl2, temperature and optimum pH, also the cofactor influence on activity protease from B. licheniformis HSA3-1a. The protease activity was tested with a modified method of Walter. Protease production time was optimum for 36 hours with an activity value of 0,394 U/mL, protein content was 132,35 mg/mL. The Optimum condition of protease enzyme were pH 7,0 and temperature 45 ⁰C with an activity value of 0,0463 U/mL. Protease activity could be inhibited by MnCl2 concentration of 0,01 - 0,03 M.

Abstract

Red algae Eucheuma cottonii and Eucheuma spinosum which one alternative source of antioxidant and anticancer because contains polysaccharide compound. This research aims to isolate and examine the activity of antioxidant and toxicity polysaccharide extract from red algae Eucheuma cottonii and Eucheuma spinosum . The study was carried out by isolating the polysaccharide extract using water and methanol-ethanol precipitation. Antioxidant activity of crude extract was examined using 1,1-Difenil-2-pikrilhidrazil (DPPH) method, and toxicity test was carried out using Brine Shrimp Lethality Test (BSLT). The research results showed that the crude extract polysaccharide for Eucheuma cottonii and Eucheuma spinosum have strong antioxidant activity with IC50 value of 72,49 ppm and 75,98 ppm. The result of BSLT assay showed that the crude extract polysaccharide has a highest toxicity with LC50 value of 165,88 ppm and 337,21 ppm there are classified as toxic. The crude extract polysaccharide Eucheuma cottonii and Eucheuma spinosum has a potential to be developed as an alternative antioxidant and anticancer agent.

Abstract

The research aimed to find out the effect of Fe2+ and Mn2+ towards β-carotene productivity as result of oxidative stress from photosystem (PS II). The phytoplanton Isocrysis aff galbana (T-iso) were the microalgae species which had the high lipid content primarily and had potentiality to produce β-carotene as the β-carotene supplement. Analysis method was carried out by the sonication extraction for short and cheap in the lysis cell of phytoplankton biomassa, infra red (IR) to find out the interaction metal ion and UV/VIS spectrophotometer to determine β-carotene concentration. The research result indicateds that Fe2+ has higher impact than Mn2+ on the β-carotene productivity. The interaction of ions metal indicates as   M-N, -O-M and M←OH-C complex in the phytoplankton amino acid. The β-carotene concentration is 4,97 µg/g DW in addition Fe2+ 0,30 ppm and 1,95 μg/g DW in Mn2+  0,20 ppm. The dry weight concentrations of β-carotene indicates that  I. aff galbana with Fe2+ addition has potential as β-carotene supplement.

Abstract

Determination of antibacterial activity of each extract of M. umbellata (Houtt.) Stapf var. K. degrabrata leaves has been conducted against three strains bacteria Escherichia coli, Staphylococcus aureus, and Shigella dysenteriae by using amoxicillin as a positive control and DMSO as a negative control. Agar diffusion method has been used with MHA medium (Muller Hinton Agar) on variation concentrations of each extract were 2500 ppm, 5000 ppm, 10,000 ppm, and 20,000 ppm N-hexane extract was the most active extract against S. aureus with zone of inhibitions were 10.50 mm, 10.80 mm, 11.00 mm and 11.45 mm, respectively. The most active extract against S. dysenteriae was ethyl acetate extracts with zone of inhibitions were 9.36 mm, 11.55 mm, 11.58 mm and 17.70 mm, respectively. Isolated compound from n-hexane extract was first compound as white powder with a melting point of 75-77 °C which was identified as 1-Tetracosanol and second compound was obtained in the form of a yellow paste which was identified as steroid compound that had long conjugated substituent. These compounds were elucidated based on the data of IR, UV-Vis, 1H-NMR, and 13C-NMR.
