Abstract

Banana is commonly propagated vegetatively by suckers since most of edible banana are triploid, male sterile and parthenocarpic, use of conventional breeding for banana improvement is difficult.  Mutation induction and in vitro technique are alternative tools for banana improvement. The objectives of this research were (1) to determine radiosensitivity of banana cv. Ampyang against gamma irradiation, and  (2) to evaluate performance of plantlets regenerated from gamma irradiated explants of banana cv. Ampyang.  Explants of in vitro grown shoots were exposed to gamma irradiation at 0, 20, 25, 30, 35, 40, 45, and 50 Gy to determine their radiosensitivity. Growth and development of regenerated plantlets were recorded after 10 months of proliferation and regeneration periods. The CurveExpert ver. 1.4 analysis results indicated that lethal doses of irradiation reducing 20% to 50% of shoot growth (LD20-50) were 51.07 - 64.54 Gy.  All regenerated plantlets from irradiated explants produced less numbers of roots, and some of regenerated plantlets, showed significantly less plantlet fresh weight and height than the control one. Plantlets regenerated from explants irradiated with 25, 40, 50 Gy have longer leaves than the control. The regenerated plantlets from gamma irradiation treatments were successfully transferred into soil and they would be used to evaluate existence of variants among regenerated banana plantlets.   Keywords: induced mutation, gamma irradiation, lethal dose (LD20-50)

Abstract

The purpose of the experiment was to identify  the effect of growing medium and SADH (Succunic Acid Dimethyl Hydrazide) on growth and development of African Violet (Saintoaulia ionanthqJ. The experiment was conducted in Baranang Siang Bogor (240 m above sea level), and arranged in factorial randomized complete block design. The first factor was medium (1 cocopeat : 1 sand : 1 compost ;  1 cocopeat : 2 sand : 1 compost ; 1 cocopeat : 1 sand : 2 compost ;  2 cocopeat  : 1 sand :  1 compost). The second factor was concentration of SADH (0, 15, 30, 45 mg/l). There was not significant effect of medium and SADH qn vegetative growth, except leaf area. SADH (15 mg/l) and medium 4 (2 cocopeat : 1 sand : 1 compost) had largest leaf Growing medium and SADH had significant effect on generative growth especially on time to visible flower buds, flower bud number, flower number, flower diameter and flower senessence. SADH (15 mg/l) applied to medium 4 (2 cocopeat : 1 sand : 1 compost) initiated time to flower. SADH (30 mg/l) applied to medium 4 affected to highest number of flower buds, and SADH (0 mg/l) applied to growing medium 4 affected highest flower diameter.

Abstract

Zoysia matrella (L.) Merr. is a widely used ornamental grass, particularly in home and landscape gardens, because of its soft appearance and high quality. One of main problem is its low growth rate. Therefore, specific cultural techniques are required to accelerate its growth while maintaining its attractive appearance. This experiment was aimed to study the effect of sod size and planting distance of Zoysia mairella. on grass growth and development. The experiment was conducted at the IPB experimental station at Darmaga IV, Bogor, for 8 months and was completed in August 1990. The trials used a split plot design with 2 factors and 3 replications. Treatment consisted of 2 planting distances i.e. (20 x 20) cm and (25 x 25) cm, and 4 sod sizes, i.e. (2 x 2) cm, (5 x 5) cm, (8 x 8) cm, and (10 x 10) cm. The variables observed were percentage of coverage, rate of coverage, quality of leaf color quality of appearance, and uniformity of growth. The results indicated that population density did not significantly affect variables observed except for uniformity of growth. Hence, the larger the sod size, the higher its rate of coverage and the more uniform its appearance, but the lower its quality of appearance. Statistically, the sod size significantly affected the percentage of coverage, rate of coverage, quality of appearance, and uniformity of growth

Abstract

ABSTRACTAn effective and efficient in vitro propagation system has important roles in preparing and producing high quality-seedlings of Dendrobium for commercial scale. The objective of this research was to establish an effective and efficient embryogenic callus (EC) proliferation method using bioreactor system and regeneration EC into plantlet for producing high quality seedlings of Dendrobium Indonesia Raya ?Ina?. Differences in callus densities (5, 10, 15, and 20 g callus in 250 mL medium), aeration levels (2.5, 5.0, and 10.0 O2 volume? per? medium volume per minute; vvm), and regeneration media half-strength MS and 2 g L-1 NPK (32:10:10) combinated by 0.00, 0.05 mg L-1 BA, 150? mL L-1 coconut water and their combinations were tested in this experiment. The experiments were arranged using randomized completely block design (RCBD) with three replications for EC proliferation and randomized completely desaign (RBD) for EC regeneration. The results showed that combination of? aeration at 2.5 vvm and 10 g of EC was the most suitable aeration level and callus density for proliferation of EC in the 500 ml airlift bioreactor with 6.85 multiplication rate, 92.5% EC formation, and malformed callus morphology as low as 6.1%. The highest somatic embryos (SEs) formation was 87.7% with 44.5 SEs per clump and 92.1% SEs germination with 41.0 germinated-SEs per clump, 85.1% normal germinated-SEs, and whereas the best performance of plantlet was obtained from 1/2 MS + 0.05 mg L-1 BA semi solid medium. Plantlets were successfully acclimatized using Cycas rumphii medium with high survival rate (91.6%). Keywords: aerations, callus densities, germination, media, somatic embryos
