Abstract

ABSTRAK   Banyak mahasiswa yang  mengalami kesalahan konsep pada saat memprogram skripsi tentang keanekaragaman hayati terutama mengaplikasikan materi hubungan kekerabatan.  Pengertian konsep dan penerapan konsep dalam perkuliahan Taksonomi Tumbuhan Tinggi/TTT  tidak berbekas. Hal ini menunjukkan bahwa mahasiswa masih tidak dapat menerapkan pengetahuan yang diperoleh.  Dengan munculnya permasalahan ini tampak bahwa kompetensi yang diharapkan dimiliki mahasiswa tidak muncul dalam jangka panjang (longterm memory). Dari  fakta di atas, pertanyaan yang muncul adalah “Bagaimanakah sebaiknya proses pembelajaran TTT terutama materi hubungan kekerabatan  agar pengetahuan yang telah diperolah pada mata kuliah TTT dapat bertahan lama dalam benak mahasiswa?” Oleh karena itu perlu dirancang  proses pembelajaran TTT yang dapat menjawab pertanyaan tersebut. Salah satu cara agar pengetahuan dapat bertahan lama dalam benak mahasiswa yaitu memberikan tugas yang menantang kemampuan mahasiswa dan memberi kesempatan  bekerjasama. Pembelajaran yang tepat untuk memfasilitasi hal tersebut  adalah Pembelajaran Berbasis Proyek (Project Based Learning). Kajian ini  bertujuan mendeskripsikan pelaksanan  penerapan pembelajaran berbasis proyek yang dianalisis secara deskriptif dengan jenis penelitian  eksperimen semu. Subyek penelitian ini adalah mahasiswa pendidikan biologi angkatan 2010 yang berjumlah 41 mahasiswa.  Hasil penerapan  menunjukkan bahwa nilai tugas proyek yang terdiri dari tahap perencanaan, persiapan, koleksi, pembuatan laporan dan seminar hasil memiliki nilai yang baik dan mahasiswa mempunyai respon positif terhadap penerapan pembelajaran berbasis proyek.   Kata Kunci : pembelajaran, berbasis proyek, hubungan kekerabatan, TTT.

Abstract

Mutation sub Topic is one of an abstract learning material. Learning object in this sub topic cannot be observed using light microscope, thus it needs learning media that able to illustrated the material itself. So, this material would be simply understand by students. The objectives of this  research is to produce and to described the theoritical feasibility of student worksheet. This research conducted based on Four-D (4-D) development methods. The result of this research indicates that developed student worksheets based on guided discovery on the mutation material as learning media could be declared feasible with worksheets validation and material validation score namely 86% and 90%   Keywords: student worksheet, guided discovery, mutation.

Abstract

Genetics substance sub topic is abstract, so an appropriate media needed to motivate and help students to understand it. The aim of this study is to produce a web-based learning media by utilizing the LMS Moodle on Genetics substance sub topic and describe media feasibility theoretically based on media experts, Genetics experts, and Biology’s teacher validation. Empirically based on media legibility. The results showed that the web-based learning media on Genetics substance sub topic were feasible for use in learning process with a very decent categories based on media format aspect (93.65%); suitability of the material (100%), and legibility (95.7%).   Keywords: LMS Moodle, web-based learning, genetic substance

Abstract

Shigella flexneri is one of pathogenic bacteria that cause dysentery. Disease caused by Shigella sp. can be cured using antibiotics, but the excessive use of antibiotics causes bacterial resistance. One of antibiotics alternate is secondary metabolites of binahong (Anredera cordifolia) leaves. This research aimed to determine the growth inhibitory of S. flexneri strain BW 1201 and the optimal concentration of binahong leaf extracts to inhibit the growth of S. flexneri strain BW 1201. Binahong leaves extracts was prepared by maceration method, using 96% ethanol. The filtrate of binahong leaves was evaporated by vacuum rotary evaporator to obtain a viscous extract. Binahong leaf extracts tested concentration were 0% (distilled water/control), 40%, 60%, 80% dan 100% with 4 repetitions. Data obtained were the diameter of clear zone (mm) formed in the culture medium of S. flexneri strain BW 1201. Data were analyzed using one way variant analysis test and Duncan´s test. Results showed that the binahong leaves extracts had inhibitory effects on the growth of S. flexneri strain BW 1201 and optimal clear zone of 27,2 mm against the test bacterial at the concentration of 100%.   Key words: Shigella flexneri; binahong leaf extracts; growth inhibitory of bacterial; well difussion method

Abstract

Escherichia coli is commonly known as nonpathogenic bacteria, but some strains are pathogenic and cause diarrhea, urinary tract infections, and meningitis. Escherichia coli is initially sensitive to antibiotics, but lately began resistant. Alternative antibacterial can be obtained from extracts of certain plants, such as mahogany (Swietenia mahogany). This research aimed to determine the effect of various concentrations of ethanolic extract of mahogany seeds to growth inhibition of  Escherichia coli FNCC 0091 and determine the concentration that could optimally inhibited the growth of Escherichia coli FNCC 0091. Mahogany seeds extract was prepared by maceration method with 96% ethanol. Antibacterial activity was done by disc diffusion method. Data obtained were diameter of clear zone. Data were analyzed using one-way analysis of variance followed by Duncan´s test. Results showed that the ethanolic extract mahogany seeds could inhibited the growth of Escherichia coli FNCC 0091. The best treatment was ethanolic extract of mahagony seeds at concentration 100% and 80% with clear zone of 2.33 mm and 2.13 mm.     Key words: Escherichia coli FNCC 0091; ethanolic extract of mahogany seed; discs diffusion method

Abstract

Genetics is one of topics in Biology with abstract concepts. The abstract genetics concepts could be understood if there are appropriate models/media/practical activities. An alternate to help students understanding genetics concept, especially DNA, is by integrated basic process skill in the learning process. This research aimed to produce students worksheets based on basic process skill on the topic of DNA extraction which is feasible empirically. This research was designed using the 4D development model. The empirical feasibility of student worksheets obtained from student responses, with the percentage 94.67%, so it could be concluded that the student worksheets was very feasible.   Key words: Student worksheet, basic process skill, DNA extraction

Abstract

The structure DNA is one of the biological topic that is abstract, so that the delivery of topic required learning media. One of the learning media that can be used is digital puzzle media. Digital puzzle media is the development of puzzle media. The development of digital puzzle aims to produce a learning media for structure of DNA topic and described its theoretical feasibility. This research is development research by ASSURE method. Research instrument used is the digital media puzzle validation sheets. The data was analyzed by quantitative descriptive. The result showed that developed media was very feasible with percentage 93.75%. Keywords: digital puzzle, DNA structure, theoritical feasibility.

Abstract

Implementation of curriculum 2013 had a lot constraints, namely some material requires learning media to reach the goal of basic competencies, for example chromosome mutation material. Biology teacher of SMAN Kesamben stated that they facing difficulty in presenting the chromosome mutation material. Observation indicated that 84.3% of students said it was difficult to learn complex concepts and 100% of students required the learning media to help concepts understanding. Karyotype magnetic board is kind of learning media that able to solve that problem. The purpose of this study was to determine the feasibility of karyotype magnetic board media theoretically based on the results of the media validation and empirically based on student responses and student learning outcomes. Research and Development (R&D) method was used in this study. The media development was started on November 2013 to April 2014 in the Department of Biology, FMIPA, State University of Surabaya. Media trial was conducted in SMAN Kesamben Jombang on March 18, 2014. The results showed the quality aspect of media got a percentage of 98.25%, the effectiveness aspect of the media got a percentage of 93.25%, the material aspect of the concept got a percentage of 97.5%, and aspect of language got a percentage of 100%. All of them included into very well criteria. Overall the karyotype magnetic board media is good based on theoretical feasibilty with a percentage of 97.8%. Empirical feasibility got percentage of 95.48% and 100% based on student responses and learning outcomes, respectively. Classical completeness average score is 89.25. Abstract

Ubi jalar varietas Papua Patippi merupakan tanaman yang mampu hidup pada tanah yang kurang subur. Kemampuan bertahan hidup tersebut dikarenakan tanaman ubi jalar berasosiasi dengan bakteri endofit melalui berbagai mekanisme di antaranya menghasilkan hormon IAA. Penelitian ini bertujuan untuk memperoleh bakteri endofit penghasil hormon IAA, mengetahui karakteristik isolat bakteri dan melihat similaritas antarisolat bakteri. Sampel berupa akar tanaman ubi jalar diperoleh dari kebun Balai Penelitian Tanaman Kacang-Kacangan dan Umbi-Umbian. Bakteri endofit, diisolasi dan diuji untuk mengetahui kadar hormon IAA yang dihasilkan. Empat isolat yang menghasilkan hormon IAA tertinggi dikarakterisasi morfologi koloni, morfologi sel dan fisiologi biokimia. Data yang diperoleh dianalisis secara deskriptif dan numerik menggunakan Clad97. Hasil penelitian menunjukkan terdapat 4 isolat bakteri endofit penghasil hormon IAA tertinggi. Isolat-isolat tersebut yaitu isolat A1, B1, B2 dan B3. Isolat bakteri endofit tersebut mempunyai karakteristik morfologi koloni dengan bentuk irregular; elevasi umbonate atau raised; tepi serrate, lobate atau entire; warna putih sampai putih kekuningan; karakter optik opaque; permukaan halus, tidak rata dan kasar. Karakteristik morfologi sel adalah Gram negatif, bentuk sel batang pendek, susunan sel monobasil, diplobasil atau streptobasil dan tidak terdapat endospora. Karakteristik fisiologi biokimia isolat bakteri endofit beragam di antaranya motil atau nonmotil, memproduksi katalase, mereduksi gula, memfermentasi asam campuran, memproduksi 2,3 butanadiol, mampu mereduksi nitrat dan memproduksi indol. Berdasarkan karakteristik tersebut isolat B1, B2 dan B3 mempunyai kemiripan yang tinggi dengan tingkat similaritas mencapai 1 sedangkan untuk isolat A1 mempunyai similaritas terendah yaitu 0.73 dibandingkan dengan isolat B1, B2 dan B3.   Kata kunci: karakterisasi; akar ubi jalar; bakteri endofit; hormon IAA
