Development of New CAPS/dCAPS and SNAP Markers for Rice Eating Quality

Abstract

 Rice eating quality traits are very complex and essential to be evaluated not only through physicochemical analysis and sensory test but also by PCR-based marker approach. To date, simple markers based on single nucleotide polymorphism (SNP) discovery to evaluate eating quality of cooked rice are still limited. Thus, the aims of this study were to develop PCR-based markers, called SNAP (single nucleotide amplified polymorphism) as alternative markers of cleaved amplified polymorphic sequence/derived cleaved amplified polymorphic sequence (CAPS/dCAPS). Four primer pairs specific to targeted alleles (CAPS/dCAPS and SNAP) of four loci were successfully designed based on the discovered SNPs according to the eating quality-QTL and searching genomic database. The primer pairs were able to identify alleles corresponding loci among indica and japonica varieties with diverse palatability (overall eating quality). There was consistent allele pattern produced by SNAP and CAPS/dCAPS for the same base mutation. The SNAP marker for rice eating quality trait could be easily assayed by standard agarose gel electrophoresis, allowing to increase the advantage of genotyping methods. Moreover, the SNAP markers together with our previous developed markers which were recommended as applicable marker set for evaluation of rice eating quality, will facilitate as marker-assisted selection for rice breeding program.

Abstract

The low of presentation toddler’s mother to the posyandu is probably caused by some reasons, they are: mothers are too busy/cannot manage their times on housework, the less of information spreading about weighing advantage so that toddler’s mother have less or no understanding about the purpose and the advantage of weighing, the less of family support and also the poor of economic condition. Measurement of antropometri has been used most because of its more practicality, it is accurate enough, and it is easy to be done by everyone by simple practices that are common used in posyandu. The less of nutrition is not because of the lack of food but also a disease. Indirectly motive are family food endurance, children care pattern, and health services.The study which is done is explanatory research study in society nutrition field with cross sectional study approach which studies the relation of toddlers presentation to posyandu with toddler antropometri measurement result. Population which is taken in this study are toddlers who registered in “Balitaku Sayang” posyandu Rw 04, Jangli district, Tembalang subdistrict which are 160 toddlers. While samples which are taken are 62 toddlers registered to “Balitaku Sayang” Posyandu. This study uses random sampling method as the technique.The result shows that 90,30% of toddlers are not active in posyandu, antropometri of index of measurement is BB/U 80,60% good nutrition, antropometri of index of measurement is TB/U 69,40% normal. Antropometri of indeks of measurement is BB/TB 85,50% is normal.The study result showed that there is no relation between toddler’s presentation in Posyandu and the result of antropometri measurement of toddlers index of BB/U. There is a relation between toddlers presentation in Posyandu and toddler antropometri measurement of TB/U index. For last, there is no relation between toddlers presentation in posyandu and the toddlers antropometri measurement result of BB/TB index.Kata Kunci : Posyandu, Antropometri, under five years old

Abstract

ABSTRACTFactors influencing elderly visit to elderly community health care (posyandu lansia): Case study in Tamantirto Village, Kasihan, Bantul, DIYBackground: Elderly community health care (ECHC) is the base of holistic-continuous care for the elderly. The number of elderly at Tamantirto Village is 11.3% of the total residents, which has not been accompanied by a high scope of ECHC visit, where 6 of 9 the ECHCs still have the coverage lower than 50%. Purpose of this study is to prove that age, education, occupation, socio economic status, knowledge, attitude, facilities, access, cadre and health workers services, social role and family role can affect elderly people’s visitations to the ECHC.Methods: Study design: observational analytic, case-control approach. The case: elderly as the ECHC members who actively visited, the control: elderly as ECHC members were who did not actively visit ECHC. The sample’s are 52 cases and 52 controls. The quantitative data were gathered by interviews, while the qualitative data by using focus group disscussion.Results: The factors influencing elderly people’s visitations to the ECHC are age of  71 years old (OR:4.6), not working (OR:8.1), good attitude (OR:3), good facilities (OR:5.4), good service of cadres and health workers (OR:6.5) and a good family role (OR:3.2). The factors that do not affect elderly people’s visitations are educational level, socio economic, knowledge, access, and social role ofthe elderly.Conclusion: Characteristics of elderly activity visit ECHC are age of 71 years old, not working, good attitude, good facilities, good service of cadres and health workers and a good family role.Keywords: Elderly community health care, actively visit, influencing factorsABSTRAKLatar belakang: Posyandu lansia merupakan dasar pelayanan yang holistik dan berkesinambungan pada lansia. Jumlah lansia yang cukup banyak di Desa Tamantirto yaitu sebanyak 11,3% dari jumlah penduduk, belum diiringi dengan cakupan kunjungan yang tinggi, dimana 6 dari 9 posyandu masih mempunyai cakupan di bawah 50%. Tujuan penelitian adalah membuktikan bahwa umur, pendidikan, pekerjaan, kondisi sosial ekonomi, pengetahuan, sikap, fasilitas posyandu, akses terhadap posyandu, pelayanan kader dan petugas kesehatan, peran sosial lansia, serta peran keluarga berpengaruh terhadap kunjungan lansia ke posyandu.Metode: Desain penelitian: analitik observasional, pendekatan kasus kontrol. Kasus adalah lansia anggota posyandu (umur 60 tahun) yang aktif melakukan kunjungan ke posyandu. Kontrol adalah lansia anggota posyandu (umur 60 tahun) yang tidak aktifmelakukan kunjungan ke posyandu. Sampel penelitian 52 kasus dan 52 kontrol. Abstract

Protease has been used in large application industrial process such as detergent, leather, textil, softdrink, andmedicine. In order to find unique protease, many substances were explored as proteases of bacteria sources. Inthis study, tofu liquid waste was used as a source of bacteria producing proteases. Waste sample was growth inskim milk agar medium showing proteases activity, it was used to produce extracellular protease. The microbialcolonies were identified as Staphyllococcus sp. Protease was extracted with 5000 g centrifugation at 4 0C, andpurificated with ammonium sulphate precipitation continued with dialisis. Optimum production time, pH, metal ion,EDTA, specific activity, KM, and Vmaks were studied for enzyme characterization. Volume of crude enzyme was 300ml, with spesific activity of 3.55 U/mg. Protease obtained from 60% ammonium sulphate fraction had the highestspecific activity of 68.22 U/mg. Study on the protease characterization revealed that optimum temperature of thisenzyme was 400C. The optimum pH of the enzyme was found to be 8.0. The kinetic parameters K M dan Vmaks withcasein as substrate were 0.31% and 51.55 U/ml. Some inhibitory effect was observed in the presence of EDTA, Cu +2,Co+2, Zn+2, and enzyme activity was stimulated by Mg+2, indicating that this ion had a functional role in the molecularstructure of the enzyme.

Abstract

Cultivation of thermophilic bacteria isolate TVII-6 for a.amylase production. Thermophilic bacteria TVIJ-6 was isolatedand selected from the soil sample taken from Dieng vulcanic. The specific activity of ainylase produced by TVfl-6 was 1624.37U/mg protein. Cassava starch was used as carbon source on enrichment culture and the optimum concentration was 1% withspecific activity 2092.23 U/mg protein. Cultivation in a 2-I bioreactor at pH 7.0 and temperature of 50°C, combine with 250 rpmagitation and 1.0 vvm aeration produced the highest a.amylase. The maximum specific growth rate obtained was 0.147/hours.The relationship between product and substrate was linear Y=0.129 X + 0.024. Efficiency of the substrate to produce amylaseY, was 0.129 g proteinlg substrate. Relationship between the bacterial growth and substrate was Y = 03115 X + 0.013.Efficiency of the substrate for growing the bacteria Y,, was 0.315 g cell/g substrate. Based on the bacterial growth and amylaseproduction, amylase could be considered growth associated, with kinetic parameter of product ratep was 0.37gfgfhr.

Purification of alfa-Amylase from Bacillus stearothermophilus by Ultrafiltration Membrane

Abstract

The objective of this experiment was to evaluate the optimum conditions of cc-amylase of Bacillus slearotherinophiluspurification by ultrafiltration and the step of a-amylase purification from indigenous isolate. Culture filtrate frombioreactor was separated by microfiltration membrane with a pore sIze of 0.2 tm. The aupernatant was purifie again byultrafiltration system membrane with cut off 30 000 Dalton. Treatments of cz-amylase purified by ultrafiltration werecarried out at flow rate of 30,45 and 60 mI/minute, and concentrated by about 5, 10 and 15 tImes. The crude enzymesresulted From ultrafiltration were precipitated with acetone. The results showed that the optimum condition ofultrafiltration was using flow rate of 30 mI/minute and concentrated by about 10 times. At the optimum condition ofultrafiltration, the specific activity of ci-amylase was of 6 686.6 U/mg with 2.3 fold purification factor. The effect of flow ratedecreased the total enzyme activity, specific activity and yield. The concentration disposal could decrease total activity andprotein, but not always reduced specific activity of the enzyme. Purification of crude enzyme by ultrafiltration and acetonereduced the total activity, total protein and yield, but specific activity and purification factor increased. Ultrafiltrationfollowed by acetone precipitation, gave enzyme specific activity of 18155.4 U/mg, purification factor of 6.3 and yield of20%, respectively. Zymogram analysis using Native-Polyacrylamide Agarose Gel Electrophoresis indicated´ ci-amylase ofapproximately 192 932.8 Da.

Abstract

The effcet of storage in liguld or freeze dried condition and calcium ion addition to activity md stability of asmylase from Bacillus stewotkcrmophilhis TlI. have been carried out. The enzyme was stored under several condition:I enzyme was freeze dried, ii enzyme was precipitated by aceton and resoluble in phosphate citrate buffer, iiienzyme was freeze dried after aceton precipitation and reaolibilizstioii in phosphate citrate buffer, Iv enzymeprecipitated by *ceton, resoluble in phosphate Citrate buffer and added by 5 mM calcium ion, and v enzyme wasfreeze dried after aceton precipitation, resolibilization in phosphate citrate buffer and added by 5 mM calcium ion.ReuIts show after nine month storage that the beat enzyme obtained in the addition of calcium ion in the freezedried condition 79.5%.

Abstract

The isolation and selection of xylanase producing bacteria have been done from soil and waste of starch Industry.Strains were cultivated in neutral and alkaline media with xilan as a substrate. Colonies which produced clearingzone were presumed as xylanolytic bacteria and chosen for further screening. Results show that 10 isolates ofbacteria In neutral condition and 25 bacteria in alkaline condition. There are three isolates in neutral condition ON.12, ON-13 and ON-33 have high enzyme activities at 19.93, 23.42 and 19.32 U/ml, and specific activities which were80.37, 71.85 and 114.36 U/mg protein respectively. On the other hand there are two isolates in alkaline condition TAIS and TA-fl15 which have high enzyme activities at 12.43 and 11.18 U/mI, and specific activities which were 36.72 and42.68 U/mg protein respectively.

Abstract

The product of a-amylase hydrolysis was evaluated for their application In sugar syrup Industry. The objectives ofthis experiment were to determine the enzyme production of Bacillus stearothermophilus TH,2 in a five liter bioreactorand to analyze its hydrolysis product on cassava starch using thin layer chromatography and high performance liquidchromatography. The bacterium was cultured in the bloreactor for 48 hours, and then the biomass, enzyme activity,protein and reducing sugar contents in the filtrate were evaluated In the course of cultivation. The strain secreted anextraceiiuiar a-amylase in the optimal condition at pH 6.5, 5O´C, agitation of 300 rpm and aeration of 1.5 vvm for 24hours. The highest activity of a.amylase and reducing sugar content of 1 068.87 U/mI and 4.48 g/l respectively wereobtained after 24 hours incubation. Hydrolysis products by the crude enzyme on cassava starch were evaluated atdifferent Incubation time. In the course of incubation the content of glucose, dextrin, maltose and oligosaccharideswere increasing. After 24 hours the concentration of glucose and maltose reached 51 970 and 10 090 ppm respectively.Based on the enzymatic products, we concluded that thermostabie a.amylsse produced by B. slearothermophilus was aaendo-a-amylase.

Prediction of Physicochemical Properties of Indonesian Indica Rice Using Molecular Markers

Abstract

Physicochemical properties determine the palatability and cooking quality of rice, which must be determined efficiently in order to satisfy consumer demand. To date, little information exists on the use of molecular markers to predict physicochemical properties of the “indica” rice varieties found in Indonesia. The objective of this study was to investigate physicochemical properties and genetic variation of Indonesian rice varieties, and to formulate regression equations to analyze sets of DNA markers which could predict  amylose content (AC), protein content (PC) and pasting properties of the varieties. A total of 24 Indonesian indica rice varieties were chosen based on their genetic background and agricultural characteristics. We then measured selected physicochemical properties, and genotyped the varieties using 30 DNA markers. The chosen varieties showed favorable values for PC, AC, and six rapid viscosity analyzer (RVA) pasting properties, which was supported by molecular data. As demonstrated by principal component analysis (PCA), markers could provide a complementary method for differentiating rice varieties, as an alternative to measuring physicochemical properties. PCA analysis also allowed us to establish marker sets using multiple regression analysis. We formulated eight model regression equations comprising data regarding 15 to 19 markers with high coefficients (R2=0.98-0.99). The formulas provided results that consistently correlated and therefore predicted the physicochemical properties of indica rice. Further validation of these marker sets may provide rapid and efficient means for predicting the physicochemical properties of Indonesian-bred indica rice in the future.
