Abstract

Background. Factors that affect the effectiveness of flotation method in Soil-transmitted helminths diagnostic are flotation solution and optional period. Further research is required to determine the optional period that gives the best effectiveness from three flotation solutions, which are NaCl, MgSO4, and ZnSO4.Aim. To compare the effectiveness of NaCl, MgSO4 and ZnSO4 as flotation solution in flotation method and the best optional period of each floating solution determined by counting number of eggs.Methods. This research was quasi-experimental analytic with two independent variable. Independent variables 1 were flotation solutions (saturated NaCl, MgSO4 and ZnSO4) . Independent variables 2 were optional period (15,30,45,60 and 70 minutes). Each combination of treatments performed on 8 samples.Results. There were significant differences between different of optional periods in one solution .The same optional period for each solution ( saturated NaCl, saturated MgSO4 and saturated ZnSO4) were not gives siginificant differences. The difference considered significant when the p value < 0.05. The most number of eggs found in NaCl was at 60 minutes, ZnSO4 at 45 minutes, and MgSO4 at 45 minutes).Conclusion. The optimum of optional period for each solution were: saturated NaCl (60 minutes), ZnSO4 (45 minutes), and MgSO4 (45 minutes). Flotation method using ZnSO4 as solution with 45 minutes as optional period considered as the most effective method based on number eggs found and time. In other hand, the effectiveness of the flotation method obtained either with NaCl, MgSO4 and ZnSO4 in all level of optional period were same.

Abstract

Background: Consumption of high-fat diet (HFD) is increasing blood total cholesterol, LpPLA2 enzyme and MDA level which is a cause of atherosclerosis that should be controlled by antioxidants. Paddy straw mushroom (Volvariella v.) extract has an antioxidant compounds. This research proves the effect of Volvariella v. extract on blood total cholesterol (TC), LpPLA2 enzyme and MDA level, and proves correlation between TC with LpPLA2 enzyme and MDA level.Methods: A randomized post test only control group design to twenty four of 8-10 weeks old  male Rattus novergicus Wistar strain with 180-200 gram were divided into four group; K1 with standard diet, K2 with HFD, each X1 and X2 with HFD+Volvariella v. extract 500 mg/kg.bwt/day and 1000 mg/kg.bwt/day. All treatments for 56 days. TC levels assesed by enzymatic colorimetric test method, LpPLA2 enzyme with ELISA method and MDA with TBARs method. Data were analyzed by one way Anova, LSD post hoc and Pearson’s correlation.Results: TC mean of K1 (91,28±1,55 mg/dL), K2 (181,96±4,19 mg/dL), X1 (122,94±2,75 mg/dL) and X2 (105,86±2,25 mg/dL); p=0,001. Enzyme LpPLA2 mean of K1 (2,65±3,19 ng/mL), K2 (1,29±1,35 ng/mL), X1 (3,92±2,18 ng/mL),  X2 (2,05±3,29 ng/mL); p=0,385 and blood MDA mean of K1 (1,17±0,12 nmol/mL), K2 (5,75±0,21 nmol/mL), X1 (2,96±0,91 nmol/mL) and X2 (1,77±0,92 nmol/mL); p=0,001. Correlation between TC and LpPLA2 enzyme was shown by X1 group (p=0,042; r = - 0,827).Conclusion:Volvariella v. extract decreased blood total cholesterol and MDA level significantly, did not significant to LpPLA2 enzyme. There wss a correlation between TC and LpPLA2 enzyme but was not to TC and blood MDA.

Abstract

BACKGROUNDFormalin is a xenobiotic that is now commonly used as a preservative in the food industry. The liver is an organ that has the highest metabolic capacity as compared to other organs. Mangosteen or Garcinia mangostana Linn (GML) peel contains xanthones, which are a source of natural antioxidants. The purpose of this study was to evaluate the effect of mangosteen peel extract on formalin- induced liver cell mortality rate and p53 protein expression in Wistar rats.METHODSEighteen rats received formalin orally for 2 weeks, and were subsequently divided into 3 groups, consisting of the formalin-control group receiving a placebo and treatment groups 1 and 2, which were treated with mangosteen peel extract at doses of 200 and 400 mg/kgBW/day, respectively. The treatment was carried out for 1 week, and finally the rats were terminated. The differences in liver cell mortality rate and p53 protein expression were analyzed.RESULTSOne-way ANOVA analysis showed significant differences in liver cell mortality rate among the three groups (p=0.004). The liver cell mortality rate in the treatment group receiving 400 mg/kgBW/day extract was lower than that in the formalin- control group. There was no p53 expression in all groups.CONCLUSIONSGarcinia mangostana Linn peel extract reduced the mortality rate of liver cells in rats receiving oral formalin. Involvement of p53 expression in liver cell mortality in rats exposed to oral formalin is presumably negligible.

Abstract

Latar Belakang: Malaria masih menjadi masalah kesehatan masyarakat di Indonesia. Beberapa wacana penggunaan terapi adjuvant sebagai pendamping obat anti malaria standard (ACT) diperlukan untuk memperbaiki disfungsi vaskuler pada malaria. Bukti bahwa ekstrak daun A.muricata dapat menurunkan sebukan sel leukosit otak mencit swiss yang diinokulasi PbA belum ada dan perlu diteliti.Tujuan: Membuktikan pengaruh pemberian Annona Muricata terhadap sebukan leukosit otak mencit malaria yang diterapi Artemisinin Based Combination therapy Metode: Penelitian ini adalah penelitian Experimental dengan desain Post Test Only Control Group Design. Sampel  terdiri dari 20 ekor mencit swiss yang dibagi menjadi 4 kelompok. Kelompok K diberi air. Kelompok P1 diberi A.muricata 3.12 mg/KgBB/hari sebagai dosis pencegahan dan 6.24 mg/KgBB/hari sebagai dosis pengobatan. Kelompok P2 diberi ACT 0.546 mg/KgBB/hari. Kelompok P3 diberi A.muricata 3.12 mg/KgBB/hari sebagai dosis pencegahan dan 6.24 mg/KgBB/hari sebagai dosis pengobatan dan ACT 0.546 mg/KgBB/hari. Perlakuan selama 14 hari diakhiri dengan terminasi mencit dan isolasi organ otak untuk sediaan histopatologi dan koleksi darah untuk pengamatan tingkat parasitemia. Uji statistik menggunakan uji Chi-square untuk leukosit otak serta menggunakan uji Kruskal-Wallis dan uji Mann Whitney untuk persentase parasitemia.Hasil: Uji Chi-square limfosit otak didapatkan hasil tidak bermakna p>0,05 dan syarat uji Chi-square tidak terpenuhi. Penggabungan dilakukan pada kelompok 1 dan 2 kemudian diuji kembali menggunakan Chi-square didapatkan hasil tidak bermakna p>0,05 namun syarat uji Chi-square terpenuhi. Parasitemia didapatkan perbedaan bermakna antara kelompok K-P2 (p=0,008), K-P3 (p=0,009), P1-P2 (p=0,008), P1-P3 (p=0,009) maupun P2-P3 (p=0,011). Tidak didapatkan perbedaan bermakna antara kelompok K-P1 (p=0,249).Simpulan: Pengaruh pemberian A.muricata maupun A.muricata dengan ACT terhadap leukosit otak mencit swiss yang diinfeksi PbA didapatkan tidak bermakna.
