Abstract

The research of microflora and amino acids component in climbing perch wadi had been done.  Aims of this research are to know amino acids composition in fresh fish and climbing perch wadi; and to know kind of halofilic bacterial (until genus level) that have a role during climbing perch wadi fermentation process.  Wadi is tradisional fermentation  product of fish that have fish  fermentation specific aroma with salty taste.  Was the processes by dry pickling in closed chamber with hight concentration salt (≥ 25% w/w). The result, arranged component of amino acids in fresh climbing perch is the same  of climbing perch wadi, but the compositions are decreased during fermentation process. Isolation, characterization and identification of bacterial were done, obtain three kinds of  bacterial from  Acinetobacter, Enterobacteriaceae, and Brucella genus, and the dominant genus group is Acinetobacter.   Key words: microflora, amino acid component, wadi, climbing perch

Abstract

Marine environment comprises of many organism which are known to posses bioactive compound as a common means of self-defense or for the protection of eggs and embryos. Class Cephalopods (such as squidand cuttlefish) are notable for their defences, such as jetting escape movements, changes in colouration, toxic venom and inking.This study aims to compare the antibacterial activity of melanin from cuttlefish ink (Sepia sp.) with squid ink (Loligo sp.) against E. coli. Extraction and purification studies were carried out on Sepia and Loligo melanin using a hydrochloric acid 0,5M treatment under mechanical.The melanins were obtained and further evaluated their activity by direct contact methods between melanin and E. coli in nutrient broth.Total microbes was counted by total plate count.Both inks also was tested their activity against E. coli. The results showed that melanin from cuttlefish and squid inks had inhibitory activity at concentrations of 10 mg / ml and 20 mg / mL, respectively reaching 99.99% against E. coli.The inks of both Cephalopods at the same concentration as melanin, did not show any inhibitory activity against E. coli.  The melanin of Sepia sp. have a higher antibacterial activity than the melanin of Loligo sp.

Abstract

Fishery product diversification activities can be used to overcome problems that are often experienced by fish cage entrepreneurs. If there is an excess harvest of fresh fish or the yield is forced to do due to sudden changes in water conditions (such as water pollution, excess capacity of the buyer, the death of fresh fish), the problem has not resolved so far. The purpose of this activity is to provide knowledge and skills for housewife of “karamba” businesspeople in the form of making presto of soft thorn carp and nugget of tilapia in Bangkal Village, Cempaka District, Banjarbaru City, South of Kalimantan. The method used in this activity is the survey, demonstration, and evaluation of the implementation of activities. The results of this activity were in the form of nuggets of tilapia and presto of soft thorn carp produced by housewives of businesspeople from Banyu Hirang Village in Bangkal Village as participants. In addition to its delicious taste, has a high protein nutritional value, these processed products can also sell as a source of household income.

Abstract

The aim of this research was to observe inhibitory activity of LAB isolated from tape waterlily seed to enteric pathogenic bacteria (Vibrio cholera, Salmonella typhi, Shigella disentri, E.coli ATCC 25922) and it’s susceptibility to antibiotic, in bile salt and under acidic condition. Microbia in the tape ( a fermented product) of waterlily seed to showed were Streptococcus thermophilus (IKH-1), Pediococcus pentosaceus (IKH-2) and Leuconostoc mesentroides (IKH-8). Streptococcus thermophillus showed inhibition against the growth of Shigella disentri with inhibition zones 16,28 mm, but did not against the growth of V. Cholera, S. typhi, E.coli. Pediococcus pentosaceus inhibit Vibrio cholera, dan Salmonella thypi with inhibition zones 18,59 mm dan 7,91 mm. So that, Leuconostoc mesenteroides inhibit Salmonella thypi with zones inhibits average 8,25 mm. Chloramfenicol at 0.05 mg concentrations did not show inhibition against the growth of isolated Streptococcus thermophillus, Pediococcus pentosaceus and Leuconostoc mesentroides. These isolates could survive too in bile salt (2%) and acidified media (pH 3).ABSTRAKPenelitian ini bertujuan untuk menguji kemampuan penghambatan bakteri asam laktat yang diisolasi dari tape biji teratai terhadap patogenik enterik (Vibrio cholera, Salmonella thypi, Shigella disentri, E. Coli ATCC 25922), antibiotik, bile salt dan asam. Jenis bakteri yang diketahui tumbuh selama fermentasi tape biji teratai adalah Streptococcus thermopilus (IKH-1), Pediococcus pentosaceus(IKH-2), dan Leuconostoc mesentroides (IKH-8). Pengamatan terhadap uji penghambatan patogenik enterik (Vibrio cholera, Salmonella thypi, Shigella disentri, dan E. Coli ATCC 25922), menunjukkan bahwa Streptococcus thermophillus mampu menghambat Shigella disentri dengan luas rata-rata zona hambat sebesar 16,28 mm tetapi tidak mampu menghambat V. cholera, S. typhi dan E. coli. Pediococcus pentosaceus mampu menghambat Vibrio cholera, dan Salmonella thypi dengan luas rata-rata zona hambat 18,59 mm dan 7,91 mm. Sedangkan Leuconostoc mesenteroides mampu menghambat Salmonella thypi dengan luas rata-rata zona penghambatan 8,25 mm. Penghambatan terhadap antibiotik, bile salt dan asam menunjukkan bahwa Streptococcus thermophillus, Pediococcus pentosaceus, dan Leuconostoc mesenteroides tahan terhadap klorampenikol pada konsentrasi 0,05mg, tahan terhadap bile salt 2% dan  asam (pH 3).

Abstract

The purpose of this research are develop to usage of lotus seed as base medium tape and study of functionally lotusseed as probiotik agentia. The potency of lotus seed tape as probiotic agentia was done tape suplemented by Lactoba- cillus acidophillus SNP-2. Observation of product belong one month for volunter that consumption. Result of research show that lactic acid bakteria, Lactobacillus, and Enterobacter on feses volunter that consumption lotus seed tape belong observation not significancy. There is no difference values of microflora on feses volunter for before and after consumption of lotus seed tape with suplemented biomass. Range of means LAB between log 8.73 CFU/g – Log 10.12CFU/g, Lactobacillus Log 7.37 CFU/g – Log 10.38 CFU/g end Enterobacter Log 9.19 CFU/g – Log 10.49 CFU/g. ThepH values of feces volunter during observation is significant different with range on 6.11–6.68.ABSTRAKPenelitian ini bertujuan untuk mengembangkan pemanfaatan biji teratai sebagai bahan dasar pembuatan tape danmempelajari fungsionaliti tape biji teratai sebagai agensia probiotik. Potensi tape biji teratai sebagai agensia pro- biotik dilakukan dengan suplementasi tape dengan Laobacillus acidophilus SNP-2 dan diamati pada relawan yang mengonsumsi tape biji teratai selama 1 bulan. Hasilnya menunjukkan Total Bakteri Asam Laktat, Lactobacillus dan Enterobacter pada feses relawan yang mengonsumsi tape biji teratai selama pengamatan tidak menunjukkan pen- garuh yang nyata. Tidak terdapat perbedaan nilai mikroflora feses relawan sebelum dan sesudah mengonsumsi tape biji teratai yang disuplementasi biomassa. Rata-rata kisaran BAL adalah Log 8,73 CFU/g – Log 10,12 CFU/g, Lacto- bacillus Log 7,37 CFU/g – 10,38 CFU/g dan Enterobacter Log 9,19 CFU/g- Log 10,49 CFU/g. Nilai pH feses relawan selama pengamatan berbeda nyata dengan kisaran berada pada nilai 6,11–6,68.

Abstract

The objective of this study was to evaluate microbiota in mice consuming fermented lotus seeds. There were two groups of mice studied, namely a group of mice given a standard diet (K), and a group of mice given standard ration added with fermented lotus seeds (TP). The parameters measured in the study were body weight of mice, total microbes, number of BAL, and number of E. coli, (14- and 28- day experiments). Each observation was repeated three times. The results showed that the consumption of fermented lotus seeds did not have any effect on microbiota (the number of microbes, BAL, and E. coli) in mice within the 28- day experiment, but the mice lost weight as much as 2.13%.
