Abstract

The aim of this study was to evaluate acute toxicity of methanol extract of Sargassum echinocarpum on male and female of mice (Mus musculus) strain BALB/c two month age. S. echinocarpum was obtained from Talango Island in April 2008. The powder of S. echinocarpum was macerated by methanol (1:3) at 4 °C for 3 x 24 hours, then concentrated, dried, and extracted. The extract was screened to observe the phytochemical compounds. For acute toxicity extracts was given orally and once on one hour before feeding. The dose treatments were 0; 625; 1250; 2500; and 5000 mg/kg body weight (BW). Parameters measured were phytochemical contain, body weight during the study, 50 percent of lethal doses, and histology score of liver and kidney. The phytochemical study showed that methanol extract of S. echinocarpum contained tannins, polyphenols, saponins, glycosides, and steroids. On 1250 mg/kg BW or more, methanol extract of S. echinocarpum inhibited the increasing of body weight on mice, but, until 5000 mg/kg BW, it did not cause mortality on mice. On 1250 mg/kg BW or more, methanol extracts caused necrosis on hepatocyt and tubulus.Key words: acute toxicity, methanol extract, Sargassum echinocarpum

Abstract

Buah mengkudu sering digunakan masyarakat sebagai obat antihipertensi, namun belum diketahui dimana tempat kerjanya. Penelitian ini bertujuan membuktikan efek buah mengkudu terhadap otot polos pembuluh darah. Digunakanmodel percobaan aorta terpisah marmut yang endotelnya telah dihilangkan.  Variabel yang diukur adalah besar perubahan kontraksi otot polos aorta tanpa endotel yang distimulasi fenilefrin [10-5M] terlebih dulu kemudian ditambahkan ekstrak buahmengkudu dosis 0,2%, 0,6%, dan 1,0%. Hasil penelitian menunjukkan bahwa ekstrak buah mengkudu dosis 0,2% menyebabkan peningkatan kontraksi,sedangkan dosis 0,6% dan 1,0% menyebabkan relaksasi. Efek relaksasiini terbukti bermakna (p < 0,05) dan terdapat hubungan yang kuat dan bermakana antara dosis dan efek relaksasi (R = 0,894; p < 0,05). Dari hasil penelitian di atas dapat disimpulkan bahwa ekstrak buah mengkudu dapat menyebabkan relaksasi otot polos aorta terpisah marmut tanpa endotel pada dosis > 0,2%,sedang pada dosis 0,2% terjadi peningkatan kontraksi. Disarankan untuk melakukan penelitian lebih lanjut untuk mengetahui dosis efektif mengkudu sebagai terapi antihipertensi. Kata kunci:mengkudu, relaksasi, aorta, endotel

Abstract

The cananga flower (Canangium odoratum) is one of the herbal medicine used for asthma bronchiale. Cananga extract have been proved to relax the isolated guinea pig’s tracheal  and bronchus smooth muscle. The relaxation of the tracheal smooth muscle could be inhibited by propanolol 10-9M, however propanolol 10-9M could not inhibit the relaxation of the bronchialsmooth muscle. Therefore the objective of this study is to investigate the effect of cananga extract on relaxation of isolated  guinea pig’s lung strip through  β-adrenergic receptor. Three different dose of propranolol was used as  β-adrenergic receptor antagonist. This study was an experimental research using guinea pig’s isolated lung strip. The control group was given histamine and cananga extract. The first group of treatment was administrated histamine, propanolol 10-9M and cananga extract. The second group of treatment was given histamine, propanolol 10-8M and cananga extract. The third group of treatmentwas given histamine, propanolol 10-7M and cananga extract. The results of this study showed that cananga extract caused relaxation of lung strip and there was a reduction in the relaxation of lung strip after administration of propanolol. The reduction in relaxation lung strip caused by propranolol inhibition was found to be significant using regression testing. In conclusion, cananga extract works as a partial agonist to β-adrenergic receptors in bronchial smooth muscle of guinea pigs since the dosage response curve showed shift to the right but it was not paralel. Keyword :cananga extract, relaxation of lung strip, β-adrenergic receptor.

Abstract

Methicillin-resistant Staphylococcuc aureus (MRSA) merupakan bakteri Staphylococcus aureus yang multi resisten terhadap antimikroba. Tumbuhan ceplukan (Physalis angulata L) dapat dijadikan sebuah alternatif pengobatan infeksi MRSA karena pada umumnya memiliki senyawa aktif yang berperan sebagai senyawa antimikroba. Penelitian ini bertujuan untuk mengetahui efektivitas ekstrak daun ceplukan sebagai antimikroba terhadap MRSA in vitro. Metode yang digunakan adalah dilusi tabung untuk menentukan Kadar Hambat Minimum (KHM) dan Kadar Bunuh Minimum (KBM). Konsentrasi ekstrak daun ceplukan yang digunakan pada penelitian adalah 50%; 55%; 60%; 65%, 70%, serta 2 kelompok kontrol yaitu kelompok kontrol bakteri tanpa diberi ekstrak daun ceplukan (0%) dan kelompok kontrol bahan (100%). Hasil dari penelitian ini didapatkan pertumbuhan koloni bakteri yang lebih rendah pada pemberian ekstrak daun ceplukan dibandingkan kontrol dan jumlah koloni semakin rendah dengan meningkatnya konsentrasi ang (r=-0,770). Dapat disimpulkan bahwa ekstrak daun ceplukan memiliki efektivitas sebagai antimikroba terhadap bakteri MRSA secara in vitro.
