Abstract

Tempe is an Indonesian fermented soy bean, produced by using mainly Rhizopus oligosporus. Diversification of tempe raw materials is required for better food security, e.g., using lupin. The potency of lupin bean has been studied as a substitute of soy in tempe production. Nutritionally, lupin bean offers additional advantages over soy bean, i.e., it has a lower level of many anti-nutritional factors (ANFs) but contains higher protein (32-36%) than most pulses. The objectives of this study were to select the most suitable lupin bean from different particle sizes for tempe production and to characterize the quality of lupin tempe. In this study, the lupin tempe samples were analyzed for moisture content, firmness, aroma, and the total count of colony after 1 and 2 days of fermentation. Sensory evaluation was conducted by 17 Indonesian untrained panelists (hedonic) in three different days. The data were evaluated statistically using General Linear Model (GLM) and Tukey’s Honest Significant Difference (HSD). Results showed that the moisture content of lupin tempe ranged from 60 to 66%, higher than that of soy tempe (58.90%). Meanwhile, the firmness of lupin tempe was between 21.38-30.89 N. Mostly, tempes which were produced from ≤50% of particles sized ≥5 mm (L3, L5 and L6) exhibited poor quality in terms of firmness and aroma. The mean of total count of colony in lupin tempe were 3.0x104 CFU/g, lower than that of soy tempe (218x103 CFU/g), indicating a slower growth of mycelia on lupin beans. However, the overall acceptability of lupin tempe which was prepared from whole beans (L4 and L7) was not significantly different from that of soy tempe. Based on these quality and sensory tests, lupin tempe using whole lupin beans was recommended for substitute of soy bean in tempe production.

Abstract

Cytotoxic assay of secondary metabolite endophytic fungus 1.2.11 from Brucea javanica (L) Merr has been carried out. Brucea javanica fruit collected from Cianjur was used in this experiment. Cytotoxic assay was done on Raji, NS-1, HeLa and Vero cells. The observation was done for 24 hours and also for 48 hours. IC50 was calculated using the Rich and Muench theory. To observe the working mechanism of cytotoxic process, DNA staining with etidium bromide and acridine orange was conducted. The cytotoxic assay of endophytic fungi 1.2.11 showed an IC50 of 58.35 μg/ml, 88.39 μg/ml on Raji cell,; 162.09 μg/ml, 66.24 μg/ml on NS cell; 361.21 μg/ml, 219.97 μg/ml on HeLa cell; and lastly 1075.18 μg/ml, 656.82 μg/ml on Vero cell after 24 and 48 hour incubation respectively. The results of this study showed that secondary metabolite of endophytic fungus 1.2.11 has selective cytotoxic effect towards cancer cell and also showed that it might cause apoptosis in NS-1cell. Abstract

Colletotrichum species were reported producing various bioactive compounds that may potentially be used in the development of anti-microbial, antioxidant, anticancer and anti-diabetes drugs. Our previous studies showed that the methanol extracts of mycelia from Colletotrichum sp. TSC13, an endophytic fungus isolated from Taxus sumatrana (Miq.) de Laub., were found to have α-glucosidase inhibitory activities. The aim of present study was to examine the effects of initial inoculum on growth and the content of fatty acids as an α-glucosidase inhibitor in Colletotrichum sp. TSC13 that grew under shake and static condition. The results showed that shake and static conditions had more effect compared to the initial inocculum on growth and glucose consumption in growth and glucose consumption. However in terms of fatty acids contents and α-glucosidase inhibitor activities the initial inoculum showed more effect than shake and static conditions

Abstract

Macaranga known locally as mahang-mahangan has uniquely ecological function, and also became a part of traditional medicine in Indonesia. Macaranga genus also known as a sources of terpenoid and phenolic (flavonoid) compounds which have biological activity as antioxidant and anticancer (cytotoxicity).There are few phytochemical investigations have been done on M. gigantifolia species. As a part of our continuing research of isolation anticancer compound from natural product, a geranylated flavonoid compound (macarangin) has been isolated from ethyl acetate fraction of Macaranga gigantifolia leaves using chromatography methods. The isolated compund (isolat MG) was elucidated to gain the chemical structure based on spectroscopic data (LC-MS and FT-NMR). Cytotoxicity test of this compound was tested against MCF 7 cell lines, showed that macarangin has a potential activity with IC50 value 119.12μg/mL. Key words: Macarangin, geranylated  flavonoid, Macaranga gigantifolia, MCF 7 cell lines.

Abstract

Telah dilakukan formulasi tablet salut selaput yang mengandung kombinasi ekstrak daun sirsak, kulit buah manggis dan jamur Ling Zhi yang diharapkan mempunyai aktivitas antioksidan dan imunomodulator yang tinggi. Penelitian meliputi formulasi tablet salut selaput F1 mengandung ekstrak daun sirsak-kulit buah manggis-jamur Ling Zhi (100:100:25 mg) dan F2 (125:75:125 mg) dalam 840 mg tablet. Uji mutu menunjukkan tablet salut F1 dan F2 memenuhi persyaratan Farmakope Indonesia IV, kecuali uji disolusi. Penetapan aktivitas antioksidan metode DPPH menunjukkan IC50 tablet F1 215,93 μg/mL, tablet F2 363,86 μg/mL, sedangkan IC50 ekstrak daun sirsak 602,64 μg/mL, IC50 ekstrak kulit buah manggis 111,80 μg/mL dan IC50 ekstrak jamur Ling Zhi 2199,50 μg/mL. Aktivitas imunomodulator dengan metode in vivo bersihan karbon menunjukkan indeks fagositik sedang, yaitu tablet F1 sebesar 1,385 tablet F2 sebesar 1,469 sedangkan ekstrak Echinacea sebagai kontrol positif sebesar < 1. Uji stabilitas tablet berdasarkan kadar zat aktif α-mangostin menunjukkan masa pakai tablet F1 1,865 tahun dan tablet F2 1,842 tahun. Penelitian menyimpulkan bahwa tablet F1 maupun tablet F2 mempunyai aktivitas imunomodulator maupun aktivitas antioksidan yang sinergis dibandingkan ekstrak tunggalnya. Disimpulkan pula bahwa aktivitas antioksidan dan stabilitas tablet F1 lebih baik dibandingkan tablet F2, tetapi aktivitas imunomodulator tablet F2 lebih baik dibandingkan tablet F1.

Abstract

Telah dilakukan fomiulasi tablet hisap dengan bahan aktif utama: A) ekstrak teh hijau; B) ekstrak teh hijau dan natrium askorbat; C) ekstrak teh hijau dan ekstrak biji anggur, Aktivitas antioksidan tablet hisap dibandingkan dengan aktivitas bahan baku aktif. Tablet hisap diuji karakteristik Hsik, uji kesukaan (nilai hedonik), uji disolusi, dan uji stabilitas dalam penyiinpanan. Uji karakteristik fisik menunjukkan bahwa tablet hisap memenuhi ketentuan-ketentuan Farmakope Indonesia IV Penetapan aktivitas antioksidan tablet hisap dengan metode DPPH memberikan nilai IC50 formula A, B, dan C masingmasing adalah 844,04 µg/mL, 221,80 µg/ml., dan C 57,4 µg/mL. sementara nilai ICM) ekstrak teh hijau, natrium askorbat, dan ekstrak biji anggur masing-masing adalah 4,29 µg/ml., 12,95 µg/mL, dan 1.96 µg/mL. Pengujian stabilitas dipercepat dilakulcan berdasar kadar EGCG dengan KCKT yang kemudian dihitung menggunakan persamaan Arrhenius menunjukkan bahwa waktu simpan formula A, B, dan C masing-masing adalah 2,11 tahun; 1,87 tahun dan 2,73 tahun_ Hasil menunjukkan bahvva tablet hisap mengandung ekstrak teh hijau dan ekstrak biji anggur (formula C) aclalah tablet hisap yang terbaik.
