Abstract

Erythrina plants (Leguminosae) were used as traditional antifertility agents. In the continuing research for novel plant with antifertility capacity from Indonesian plants, the methanol extract of Erythrina plants showed significant antifertility activity in an in vitro assay of spermatozoa of white rat (R. norvegicus). The methanol extract was separated using bioassay-guide n-hexane, ethylacetate, and n-buthanol fractionation. The ethylacetate fraction was tested in vitro on spermatozoa of white rat (R. norvegicus) at a concentration of 0.25 x 10-3 μg/μL. It showed a decrease of their motilities and viabilities, as well as an increasing abnormality of the spermatozoa.

Abstract

Traditional medicines originating from plants have been widely used as an alternative therapy and extensive research have identified several bioactive compounds of the plants. In Indonesia, Erythrina variegata (Leguminosae) plants have been used for medication to many diseases including anthelmintic, anticancer,antimalaria, and antifertility.Objective: In the course of our continuing search for novel bioactive compounds from Indonesian Erythrina plants, we isolated and described several bioactive compounds such as anticancer, antimalaria, and antifertility from E. variegata. Method: Activity assay in vitro against breast cancer cell T47D using the Sulforhodamine B (SRB) method, activity assay in vitro against antiplasmodium using lactatedehydrogenase (LDH) method, and antifertility activity on spermatozoa Rattus novergicus. Their chemical structures were determined based on spectroscopic evidences and comparison with related compound previously reported. Result: Extract and compounds of E. variegata showed anticancer in vitro against breast cancer cell T47D, antiplasmodial in vitro K1 and 3D7 strain parasites, and antifertility on spermatozoa R. norvegicus with midle and high activity. Besides that, the toxicity assay of it was also conducted using white mice to show the safety grade of the plant. Conclusion: This results strongly suggested that E. variegata is a promising sources as herbal medicine of anticancer, antimalaria, and antifertility agents.

Abstract

Malaria merupakan salah satu masalah serius yang dialami oleh beberapa negara tropis karena meningkatnya parasit malaria (Plasmodium) yang resisten terhadap obat-obat antimalaria. Oleh sebab itu perlu dicari obat antimalari baru, salah satunya tanaman cocor bebek (Kalanchoe blossfeldiana Poelln.) digunakan secara luas oleh masyarakat indonesia sebagai tanaman obat dan tanaman hias. Penelitian ini diawali dengan proses pembuatan ekstrak etanol daun cocor bebek menggunakan alat maserator dan etanol 96% sebagai pelarut. Plasmodium falciparum 3D7 yang akan digunakan dalam uji, terlebih dahulu dilakukan kultur sinambung sesuai metoda Trager and Jansen. P. falciparum  ditempatkan ke dalam lempeng sumur 24 masing-masing berisi 1 mL dengan tingkat  parasitemia ± 1% dalam medium RPHS. Diseluruh sumur, medium RPHS diganti dengan medium RPHS yang mengandung ekstrak etanol daun cocor bebek berbagai konsentrasi (1 sampai1x10-7 µg/mL).  Kultur diinkubasi selama 48 jam, setelah inkubasi parasit dipanen dan dibuat sediaan apusan darah tipis yang diberi pewarnaan Giemsa. Uji aktivitas antimalaria ditentukan dengan parasitemia, persen pertumbuhan dan hambatan parasit. Data dianalisis secara statistika menggunakan metode analisis probit untuk menghitung hambatan parasit sebesar 50% (IC50). Hasil penelitian menunjukan bahwa ekstrak etanol daun cocor bebek memiliki efek antimalaria dengan nilai IC50 sebesar 0,022 µg/mL.

Abstract

Dysoxylum merupakan salah satu genus yang memiliki metabolit sekunder dan aktivitas yang beragam, salah satu aktivitas yang menarik adalah mampu bertindak sebagai agen P-gp pada sel kanker payudara MCF-7. Kulit batang D.alliaceum dimaserasi berturut–turut dengan n-heksana, etil asetat dan metanol. Ekstrak n-heksan dipisahkan dan dimurnikan dengan berbagai teknik kromatografi dipandu dengan analisis kromatografi lapis tipis diperoleh dua senyawa stigmast-5-en-3β-ol (1), stigmasterol (2). Struktur kimia senyawa tersebut telah ditetapkan berdasarkan intrepretasi data-data spektroskopi dan dibandingkan dengan data spektra dari penelitian sebelumnya. Semua isolat senyawa dievaluasi aktivitas sitotoksiknya terhadap sel kanker payudara MCF-7. Senyawa 1 dan 2 dirujuk dari literatur menunjukkan aktivitas kuat dengan IC50 8,65 μg/mL.

Abstract

Plumbago zeylanica (Ki Encok) merupakan salah satu tumbuhan obat multifungsi yang banyak ditemukan di Indonesia. Daun dan akarnya berkhasiat sebagai obat pada berbagai penyakit termasuk anti jamur dan anti bakteri. Penelitian ini dilakukan untuk mengetahui senyawa bioaktif antibakteri yang dapat digunakan untuk mengobati penyakit gigi dan mulut. Dari hasil pemisahan dan pemurnann fraksi aktif, tiga senyawa telah berhasil diisolasi dari ekstrak n-heksana dan etilasetat daun Ki Encok (Plumbago zeylamica), yaitu 3β-galaktosasitosterol (1), epi-isoshinanolon (2), dan metil 2,4-dihidroksibenzoat (3). Ketiga senyawa tersebut telah diuji aktivitasnya terhadap bakteri untuk penyakit mulut dan gigi yaitu Streptococcus mutan dan Streptococcus sanguinis. Hasilnya, diameter zona hambat dari senyawa 1, 2, dan 3 terhadap bakteri S. Sanguinis berturut-turut sebesar 12,0; 10,3; dan 12,0 mm, sedangkan terhadap bakteri S. Mutans berturut-turut sebesar 13,3; 11,7; dan 13,5 mm.

Abstract

Spons pencuci piring 200.000 kali lebih kotor dibanding dudukan toilet. Berbagai bakteri penyebab penyakit seperti Eschericia coli, Pseudomonas dan Staphylococcus, berkembang biak di permukaan yang basah. Selain itu, 500 ribu bakteri hidup di dalam saluran pembuangan bak cuci piring. Jika spons dalam kondisi tidak kering (lembab karena direndam), maka akan menjadi markas semua bakteri. Penelitian ini bertujuan untuk menentukan populasi bakteri yang terdapat pada spons cuci piring yang basah. Identifikasi dilakukan dengan metoda molekular berbasis DNA yaitu teknik PCR-RFLP gen 16s rDNA. Metoda PCR-RFLP merupakan genetik fingerprinting yang akurat, cepat dan sederhana. Koloni bakteri yang ditumbuhkan dari spons pencuci piring di lisis dan digunakan sebagai templat untuk PCR. Hasil PCR gen 16s rDNA yang berukuran 1400 pb, dipotong dengan enzim restriksi, Hinf1. Hasil penelitian menunjukkan terdapat empat pola pemotongan yang berbeda-beda. Pola yang berbeda ini menunjukkan terdapat empat populasi yang berbeda hidup pada spons basah. Kami mengidentifikasi bahwa salah satu mikroba yang tumbuh adalah E. coli yang merupakan bakteri patogen.

Abstract

Akway (Drimys beccariana Gibs) is an endemic plant of Papua. Ethnobotany is used as a medicinal plant, one of which is used as a malaria drug. This study aims to isolate, determine the chemical structure and evaluate the antimalarial activity of D. beccariana stem bark. Powder of D. beccariana bark is macerated with methanol and fractionated using n-hexane and ethyl acetate. Ethyl acetate extract was fractionated using gradient liquid chromatography (KCV) (n-hexane: ethyl acetate: methanol). The KCV fraction was separated by repeated gravity column chromatography and preparative thin layer chromatography (TLC-P) to obtain Isolate 1. Isolate 1 was determined by spectroscopic methods including UV-Vis, IR, 1D NMR, and 2D. Isolate 1 is determined by its compound structure which is 5,3'-dihydroxy-7,4'-dimethoxyflavone. 5,3'-dihydroxy-7,4'-dimethoxyflavone compounds evaluated antimalarial activity against Plasmodium falciparum in vitro with antimalarial activity better than artemisinin.

Abstract

Various species of Kalanchoe plants have been widely used as raw materials in traditional medicines. This study was the continuation of the researches on secondary metabolites from Indonesia Kalanchoe plants, focused on Kalanchoe blossfeldiana. Fresh K. blossfeldiana leaf was extracted with methanol at room temperature to obtain the concentrated extract. The concentrated methanol extract was dissolved in water and then partitioned successively with n-hexane and ethyl acetate. The methanol, n-hexane, ethyl acetate extracts were tested using antiplasmodial assay against Plasmodium falciparum 3D7. The IC50 of methanol, n-hexane, and ethyl acetate extract were 13.002; 2.807, and 11 nM, respectively. Ethyl acetate extract was separated by the combination of chromatography on silica and ODS. This process produced the yellow solid. The chemical structure of the compound was determined based on UV, IR, MS, 1H-NMR, and 13C-NMR analyses and the comparison of data obtained from the literature and identified as phenolic compound, namely 3,3,4,5,7-pentahydroxyflavone or quercetin (1), and displayed antiplasmodial activity with IC50 3.97x10-2 nM.

Abstract

Flavonol compounds, quercetin (1) and quercetin-3-O-?-D-glucopyranoside (2) have been isolated from the ethyl acetate extract of Lygodium microphyllum leaves. The chemical structures of flavonol compounds were identified based on spectroscopic data and by comparison of spectral data obtained previously. The discovery of flavonol compounds in Lygodium microphyllum was shown in this study for the first time.
