Abstract

Eugenia uniflora Lam. is one of herbal products developed for anticancer. The aim of the present study was to identify the antiproliferative effect of ethanolic extract of Eugenia Uniflora Lam. leaves (EEU) on breast cancer cell line T47D. This Research was initiated by extracting the active contents of Eugenia uniflora Lam. leaves by maceration with ethanol 96%. The extract was then analyzed by thin layer chromatography (TLC). Cytotoxic assay of EEU was carried out by using MTT assay. Apoptosis phenomenon was observed with double staining using acridine orange-ethidium bromide. EEU showed cytotoxic effect on T47D cells with IC50 value of 65 µg/ml. Moreover, EEU 50µg/ml and 100µg/ml induced apoptosis. TLC examination showed that EEU used in this study contain phenolic, flavonoid, and saponin compounds which were suggested to be responsible for antiproliferative effect. Further molecular mechanism underlying EEU antiproliferative effect needs to be done.Keywords: Eugenia uniflora Lam., T47D cells, antiproliferative, apoptosis

Abstract

Breast cancer is the second leading cause of cancer death among women in the world. To overcome the high incidence of cancer, the discovery and development of cancer treatments continues to be pursued. Pandan leaf (Pandanus amaryllifolius Roxb) is one of natural product that had cytotoxic activity. This study investigated the cytotoxic activity on MCF-7 breast cancer cells and to identify the chemical subtance groups of pandan leaves ethanol extract and fractions as chemopreventive agent. Active contents of pandan leaves was extracted by remaceration with 70% ethanol. Variation solvent of the water, chloroform, and ethyl acetate used to fractionation of pandan leaf ethanolic extract. Qualitative test were done to identify the chemical substances of pandan leaves etanolic extracts and fractions. Cytotoxic activity of sample was measured using MTT assay. The result of the phytochemical screening showed that the etanolic extract and water fractions of pandan leaf contained alcaloids, poliphenol and saponin. Chloroform fractions contained alcaloids and poliphenol. Pandan leaves ethanol extracts and chloroform fractions showed cytotoxic activity on MCF-7 cell line with IC50 of 540 ?g/ml and 160 ?g/ml whereas ethyl acetate fractions and water fractions of pandan leaves showed no cytotoxic activity on MCF-7 cell line.Keywords: Pandanus amaryllifolius Roxb, MCF-7 cell line, cytotoxic activity, etanolic extract,fractionation

Abstract

The  prognosis  of  breast  cancer  patients  is  closely  associated  with  the  response  of tumor  cells  to  chemotherapy  agent.  Doxorubicin  is  one  of  the  primary  chemotherapeutic agents  used  for  the  treatment  of  breast  cancer.  Resistance  to  chemotherapy  is  believed  to cause  treatment  failure  in  cancer  patients.  Furthermore,  long  time  exposure  to chemotherapeutic  agent  induces  cancer  cells  resistance.  MCF-7  sensitive  cells  used  as chemoresistance  model  have  overexpression  P-gp  (P-glycoprotein).  Chemoresistance  was established by treating MCF-7 cells with 0.5 µg/ml doxorubicin-contained medium for a week. 50% inhibiting concentration (IC50) doxorubicin on MCF-7 cells/DOX were determined using MTT assay. Western blot assay and immunocytochemistry assay was performed to determine the expression of P-gp. Morphological of MCF-7 cell/DOX was changing to become larger and have  lamellapodia.  IC50  value  of  doxorubicin  was  700  nM  on  MCF-7/DOX  and  400  nM  on sensitive MCF-7 cells. The MCF-7/DOX sensitivity to doxorubicin was decreased, shown by 1.5  fold  higher  IC50  of  doxorubicin  on  MCF-7/DOX  compared  to  MCF-7  sensitive  cells. Treatment doxorubicin to sensitive MCF-7 cells leads to the increasing P-gp expression. The P-gp  level  expression  has  strong  correlation  with  the  low  sensitivity  of  MCF-7/DOX  to doxorubicin.Key words: doxorubicin, resistance cells, sensitive MCF-7 cell

Abstract

Graduates students mostly oriented into a job seeker, not the job creators. There is an assumption that the preparations become entrepreneurs during college, lead to failure in passing study on university. Science and technology-based entrepreneurship education can change those assumption, so it can affect a student's readiness for entrepreneurship on 3 aspect: affective (attitude), cognitive (knowledge), and psychomotor (skills). This research aimed to analyzed the effect of science and technology-based entrepreneurship programs to changes in student readiness.This research was quasy experimental by intervented 20 tenants, the participant of Ipteks bagi Kewirausahaan (Science and Technology for Entrepreneurship) at the Polytechnic of Health Bhakti Setya Indonesia. Students attended a series of science and technology-basedentrepreneurship education program especially by approaching of herbal saintification in various kinds of products that lead to supporting therapy. The intervention consist of a motivational training program, training on creating products, internships in industry partners, and comparative study. Data was collected by questionnaire test in pre and post treatment to determine the student?s readiness changing. Data was analyzed by paired T-test. The result showed that the intervention of science and technology-based entrepreneurship programs can improve student?s readiness in entrepreneurship.Keywords: entrepreneurship, entrepreneurship readiness, student, science and technology

Abstract

Kanker leher Rahim merupakan salah satu kanker yang paling terjadi pada wanita. Angka kematian yang tinggi menunjukkan bahwa kemoterapi tidak memberikan hasil yang baik sehingga perlu dikembangkan strategi untuk mengatasinya. Daun alpukat (Persea americana Mill) adalah salah satu produk alami yang memiliki aktivitas antioksidan dengan menangkap radikal bebas dan memiliki aktivitas sitotoksik. Tujuan dari penelitian ini adalah untuk mengevaluasi aktivitas sitotoksik dari ekstrak etanolik daun alpukat pada sel kanker leher Rahim HeLa dan mengidentifikasi senyawa aktif  yang ada pada daun alpukat. Senyawa aktif dari daun alpukat diekstraksi dengan metode maserasi menggunakan etanol 96%. Uji kromatografi dilakukan untuk mengidentifikasi senyawa aktif. Aktivitas sitotosik ekstrak etanolik daun alpukat dilakukan dengan uji MTT. Hasil skrining fitokimia menunjukkan bahwa daun alpukat mengandung senyawa golongan flavonoid, alkaloid dan saponin. Ekstrak etanol daun alpukat menghambat pertumbuhan sel HeLa dengan nilai IC50 sebesar360 µg/ml.

Abstract

Fraksi kloroform daun pandan (Pandanus amaryllifolius) diketahui memiliki efek sinergis sebagai kokemoterapi doksorubisin pada sel kanker payudara MCF-7 dan T47D. Penelitian ini untuk mengetahui efek toksik subkrosis fraksi kloroform daun pandan dikombinasi dengan doksorubisin. Penelitian dilakukan dengan menggunakan tikus Sprague Dawly 1-2 bulan sebanyak 40 ekor dibagi dalam 8 kelompok yang terdiri dari kelompok sehat, kelompok pelarut, kelompok doksorubisin, dan empat kelompok perlakuan kombinasi doksorubisin dan fraksi kloroform. Setelah 30 hari dilakukan pemeriksaan biokimiawi darah  trigliseride, LDL, HDL dan kolesterol. Hasil penelitian menunjukkan kombinasi doksorubisin dengan fraksi kloroform daun pandan tidak menyebabkan toksisitas terhadap jantung dan mampu menurunkan efek toksik doksorubisin terhadap jantung.

Abstract
