Abstract

In the year 2009-2010, the Department of Agriculture, Malaysia introduced Abandoned Land Development Project (ACDA). Under this project, abandoned lands are replanted with crops of economic value. The cultivator of this case study was one of the participants of ACDA project. With the subsidies provided by the Government of Malaysia, the cultivator established a banana farm. Conventionally, the cultivator’s main source of income should be generated from selling the banana fruit. However, we found this cultivator cum entrepreneur diversified his land productivity. The monthly income generated from selling banana fruits, suckers and rubber seedlings were 30.2 %, 39.9% and 29.9% of his total farm income, respectively. The cultivator provide a novel insight in managing banana farm by introducing new techniques of planting, fertilization regime and diversification of income in his banana farm intercropped with rubber seedlings.

Abstract

The aim of this study was to determine the effect of added betel leaves towards fat oxidation on smoked seasoned catfish fillets during storage. This study consisted of seven treatments, there were control (without added of spices, liquid smoke, and betel leaves powder), the added spices, liquid smoke, and also the combination of added spices, liquid smoke, betel leaves powder at various concentration of betel leaves powder (0.05, 1, 5, and 10%). The spices and soy sauce were dissolved in water and used to dissolve betel leaves powder. The order of smoked seasoned catfish fillets were soaked in a spice solution and betel leaves powder for 2 hours and then drained for 5 hours at 5 °C, and dipped in liquid smoke 10% (v/v) for 1 minute, then cooked in oven at 75 °C for 6 hours, cooled and packed (0.8 mm polypropylene plastic). The result showed that the betel leaves powder and spices interacted synergistically to inhibit fat oxidation and increased shelf life of smoked seasoned catfish fillets until the six days of storage. Added betel leaves powder could inhibit fat oxidation, peroxide value content varied 12.16 to 14.14 mgeq/1000 g, lower than control (13.53 to 14.09 mgeq/1000 g). Added 1% betel leaves powder was the most preferred smoked seasoned catfish fillets by consumers test, however it induced darker color, harder texture, and could covered up the smell of smoked on product. Abstract

Telah dilakukan penelitian untuk mengetahui bagian-bagian lamun Cymodocea sp. yang berpotensi sebagai antioksidan dan uji toksisitasnya. Ekstrak daun dan batang Cymodocea sp. dibuat dengan cara maserasi menggunakan tiga macam pelarut, yaitu metanol, etil asetat, dan n-heksan. Pengujian kadar total fenol menggunakan metode Follin-Ciocalteu. Aktivitas antioksidan dilakukan dengan metode spektrofotometri menggunakan DPPH (1,1-diphenyl-2-picrylhydrazyl) sedangkan kadar toksisitas dilakukan dengan uji BSLT (Brine Shrimp Lethality Test). Hasil penelitian menunjukkan bahwa ekstrak metanol daun Cymodocea sp. mempunyai kandungan total fenol  paling tinggi (26,73 mg GAE/gram ekstrak) dengan aktivitas antioksidan (IC50) sebesar 518,57 ppm. Berdasarkan nilai IC50, sampel tersebut tergolong antioksidan lemah. Hasil uji toksisitas menunjukkan semua sampel Cymodocea sp. bersifat toksik terhadap larva udang Artemia salina Leach. Ekstrak etil asetat daun Cymodocea sp. mempunyai toksisitas terbesar dengan nilai LC50 sebesar 67,14 ppm. Uji fitokimia menunjukkan bahwa ekstrak Cymodocea sp. mengandung senyawa golongan saponin, steroid, flavanoid, fenol hidrokuinon, dan terpenoid.

Abstract

Penelitian ini bertujuan untuk menentukan pengaruh penggunaan ekstrak etanolik Padina sp. terhadap daya simpan filet ikan kembung selama penyimpanan pada suhu kamar. Filet ikan kembung direndam selama 30 menit dalam larutan ekstrak etanolik Padina sp. dengan konsentrasi 0; 0,5; 1; 1,5; dan 2 g/ 100 mL akuades. Filet ikan kembung kemudian disimpan pada kotak styrofoam pada suhu kamar selama 24 jam dengan selang waktu pengamatan setiap 6 jam. Parameter yang diamati meliputi: pH, Total Plate Count (TPC), kadar histamin, kandungan bakteri pembentuk histamin (BPH), dan organoleptik (kenampakan, bau, dan tekstur). Hasil penelitian menunjukkan bahwa penggunaan ekstrak etanolik Padina sp. yang berbeda memberikan pengaruh nyata (p<0,05) terhadap nilai TPC, kandungan BPH, dan nilai organoleptik keseluruhan filet ikan kembung selama 24 jam. Filet ikan kembung yang diberi ekstrak etanolik Padina sp. masih layak konsumsi hingga jam ke-6.

Abstract

Industri pengolahan ikan menghasilkan limbah dengan kadar protein tinggi. Penambahan bakteri proteolitik diharapkan dapat membantu mendegradasi limbah berprotein tinggi. Pada penelitian ini, isolasi bakteri proteolitik dilakukan untuk mendapatkan isolat dengan aktivitas proteolitik tinggi. Isolat terbaik diuji kemampuan hidup dan aktivitasnya pada berbagai konsentrasi NaCl dan pH. Kemampuan isolat meningkatkan kualitas limbah juga diuji pada limbah pengolahan bandeng presto. Hasil penelitian diperoleh 36 isolat bakteri proteolitik dari limbah cair pencucian ikan Pasar Kranggan Yogyakarta. Dari ke-36 isolat tersebut, isolat D61 merupakan isolat dengan aktivitas proteolitik tertinggi dengan zone diameter 20,5 mm. Isolat tersebut mampu hidup pada NaCl 0-10% dan kisaran pH 5-10, namun aktivitas proteolitik tertingginya pada NaCl 0-2% dan kisaran pH 7-8 dan pH 10. Berdasarkan karakteristik morfologi dan biokimianya, isolate D61 memiliki kemiripan 93,93% dengan Bacillus soli. Hasil ujicoba pada limbah cair pengolahan bandeng presto ternyata isolat D61 tidak mampu memperbaiki kualitas limbah tersebut.

Abstract

The aim of the research was to determine effect of Padina sp. extract on shelf life of red nile fi llet during storage at low temperature. Red nile fi llet was soaked up into Padina sp. extract solution at various concentration of 0; 0.5%; 1%; 1.5%; and 2% for 30 minutes, then was stored in chilling room for 10 days. The observation was carried out every 2 days. The parameters observed were pH, Total Plate Count (TPC), Total Volatile Base-Nitrogen (TVB-N), and organoleptic still be tests. The results showed that the different concentration of Padina sp. extract yielded the signifi cant effect (P<0,05) on pH, TPC, TVB-N and organoleptic of the red nile fi llet. The treatment of Padina sp. extract on red nile fi llet can still be consumed up to 8 days of storage based on TPC, TVB, and organoleptic namely odor and texture. Penggunaan ekstrak Padina sp. 1% merupakan perlakuan terbaik dalam mempertahankan kesegaran fi let nila merah yang disimpan pada suhu dingin.

Abstract

Indonesia has a lot of seaweed that have high potential as a producer of alginate, but the method of extraction has not been as expected. The objective of this study to develop a method of extraction of sodium alginate from seaweed Sargassum through the calcium alginate pathway. This study used different variations of the concentration of calcium chloride. The concentration of calcium chloride used varied 0.5, 0.75 and 1 M. As a control, the extraction of alginate performed through alginic acid pathway which was developed at the Center for Research Product Processing and Biotechnology of Marine and Fisheries, Jakarta. Quality parameters were observed including alginate yield, product appearance, viscosity and gel strength. The result showed that the yield of alginate produced successively for 32.67; 44.67 and 53.33 % and 31.67 % for controls. In appearance, the concentration of calcium chloride did not significantly affect the alginate product appearance, but darker when compared with the product extracted through the alginic acid. Viscosity alginate produced successively 149, 131 and 144 cP, while 304 cP for control. In general, the alginate gel strength extracted through of calcium alginate pathway is lower than the alginic acid pathway.ABSTRAKIndonesia  mempunyai  banyak  rumput  laut  yang  berpotensi  tinggi  sebagai penghasil  alginate,  namun  metode ekstraksinya belum sesuai yang diharapkan. Penelitian ini bertujuan untuk mengembangkan metode ekstraksi natrium alginat dari rumput laut Sargassum melalui jalur kalsium alginat. Dalam penelitian ini digunakan berbagai variasi konsentrasi kalsium klorida yang digunakan pada pemisahan alginat dari filtrat hasil ekstraksinya. Konsentrasi kalsium klorida yang digunakan divariasi 0,5; 0,75 dan 1 M. Sebagai kontrol dilakukan ekstraksi alginat melalui jalur asam alginat yang dikembangkan di Balai Besar Riset Pengolahan Produk dan Bioteknologi Kelautan dan Perikanan, Jakarta. Parameter kualitas alginat yang diamati meliputi rendemen alginat, kenampakan produk, viskositas dan kekuatan gel yang dihasilkan. Hasil pengamatan menunjukkan bahwa rendemen alginat yang dihasilkan berturut-turut sebesar 32,67; 44,67 dan 53,33 % dan untuk kontrol 31,67 %. Secara kenampakan, konsentrasi kalsium klorida tidak terlalu mempengaruhi kenampakan produk alginat yang dihasilkan, tetapi lebih gelap jika dibandingkan dengan produk hasil ekstraksi melalui jalur asam alginat. Viskositas alginat yang dihasilkan berturut-turut 149, 131 dan 144 cP, sementara untuk kontrol 304 cP. Secara umum kekuatan gel alginat yang dihasilkan dari jalur kalsium alginat lebih rendah dibandingkan dengan yang diekstrak melalui jalur asam alginat.

Abstract

This paper aims to discuss the history of Acehnese wars and the progress of peace in Aceh after the signing of the Memorandum of Understanding between the Aceh Freedom Movement, or Gerakan Aceh Merdeka (GAM), and the Republic of Indonesia in Helsinki on August 15th, 2005. Prior to this, Aceh was a tense region and home to the longest armed conflict in Southeast Asia – underway since 1982. The people of Aceh were fighting to realize the concept of self-independence. However, the movement came to a stop when the devastating tsunami hit Aceh on December 26th, 2004. The purpose of this paper is to discuss the root of Acehnese wars. The paper will provide the current political, social, economic and cultural achievements since Aceh entered into a new chapter of peace. Using qualitative approach, the theory of inequality and conflict and Fanon’s ideas, this paper offers a comprehensive perspective on learning from Aceh wars and conflicts. This study found that the motive of wars in Aceh can be divided from group motive, private motivation, failure of social contract and environment scarcity. Moreover, the research confirms that social and economic progress in Aceh has not been as successful as its political achievements. Furthermore, in terms of culture, progress has been ambiguous. This paper aims to provide a better understanding of how to maintain peace in Aceh by addressing social, political, economic and cultural issues with the goal of attaining prosperity and well-being for the people of Aceh.

Abstract

The purpose of this study was to determine antioxidant activity of polyphenol and phlorotannin extracts from brown algae Sargassum polycystum. Polyphenol was extracted with ethanol 96% then tested total phenolic content, while phlorotannin was extracted with methanol, chloroform, aquabidest, and ethyl acetate then tested total phlorotannin content. After crude polyphenol and crude phlorotannin extracts were obtained, the studied compounds were extracted using column chromatography. The extracts were then analyzed with HPLC (High Performance Liquid Chromatograpys). Antioxidant activity was analyzed by radical scavenging activity (RSA) and ferrous ion chelating ability (FIC) for four samples, crude polyphenol, crude phlorotannin, polyphenol, and phlorotannin. The result of total phenolic content was 1.18±0.67 mg GAE/g dry sample and total phlorotannin content was 0.61±0.27 mg PGE/g dry sample. Based on RSA and FIC analysis, phlorotannin (IC50 1.12±0.02 mg/mL and 1.34±0.01 mg/mL) showed the highest activity followed by crude phlorotannin (IC50 1.20±0.01 mg/mL and 1.47±0.10 mg/mL), polyphenol (IC50 1.23±0.01 and 1.55±0.02 mg/mL), and crude polyphenol (IC50 1.27±0.01 mg/mL and 1.63±0.02 mg/mL). The results of chromatographic analysis showed that S. polycystum extract contains phenolic compounds similar to the phloroglucinol.ABSTRAKPenelitian ini bertujuan untuk menentukan aktivitas antioksidan ekstrak polifenol dan florotanin dari rumput laut cokelat Sargassum polycystum. Polifenol diekstrak menggunakan pelarut etanol 96% kemudian diuji kandungan total fenolik, sedangkan florotanin diekstrak dengan menggunakan pelarut metanol, kloroform, akuades, dan etil asetat kemudian diuji kandungan total florotanin. Setelah memperoleh ekstrak polifenol dan florotanin, kedua senyawa tersebut dipurifikasi dengan kromatografi kolom. Hasil kromatografi kolom dianalisis menggunakan HPLC (High Performance Liquid Chromatography). Uji aktivitas antioksidan dilakukan dengan metode radical scavenging activity (RSA) dan ferrous ion chelating ability (FIC) terhadap empat sampel, yaitu ekstrak polifenol, ekstrak florotanin, polifenol, dan florotanin. Kandungan total fenolik yang diperoleh adalah 1,18±0,67 mg GAE/g ekstrak kering, sedangkan kandungan total florotanin diperoleh 0,61±0,27 mg PGE/g ekstrak kering. Berdasarkan uji RSA dan FIC, florotanin (IC50 1,12±0,02 mg/ mL dan 1,34±0,01 mg/mL) memiliki aktivitas tertinggi diikuti ekstrak florotanin (IC50 1,20±0,01 mg/mL dan 1,47±0,10 mg/mL), polifenol (IC50 1,23±0,01 dan 1,55±0,02 mg/mL), dan ekstrak polifenol (IC50 1,27±0,01 mg/mL dan 1,63±0,02 mg/mL). Hasil analisis kromatografi menunjukkan ekstrak S. polycystum mengandung senyawa fenolik relatif seperti senyawa floroglusinol.

Abstract

Fresh fishery products are highly perishable and susceptible to spoilage caused by rapid growth and activity of bacteria. Psychotropic bacteria are the major microorganisms responsible for spoilage when these products stored in ice or under refrigeration. The purposes of this study were to isolate psychrotrophic bacteria from fresh fishery products and to evaluate its resistance to bacteriocins. Fresh fishery products (kembung fish, shrimp, shellfish) samples were obtained from Demangan and Kranggan traditional market. Colony count methods were used to enumerate bacteria, whereas isolation and identification of bacteria was performed by the identification method of Lahallec and Colin (1995). The identification conducted were covering Gram staining and cell morphological examintaion (cell form), motilitym presence or absence of an oxidise, mode of glucose utilization, and capacity for glucose or lactose fermentation, H2S production, and gas production. The resistance assyas by bacteriocin were carried out using agar diffusion method. Seventeen isolates were found abd they were classified as Pseudomonas, Xanthomonas, and Flavobacterium/ Cytophaga. The results show that psychrotropic bacteria resistance to supernatant of lactic acid bacteria and nisin.
