Increasing of Plasma Cholecystokinin Level and Jejunum Histological Changes After Treatment with Soybean Extracts Protein

Abstract

It is well known that soybean has beneficial health effects. There are lot of active compounds in soybean, like protein and anti nutrition factors (ANF). Trypsin inhibitor and lectin, two kinds of ANF have an adverse effect on the morphology and function of digestive tract in animals. b-conglycinin in soybean protein, has been proven  has reducing body weight effect through increasing cholecystokinin (CCK) level. The aim of this study was to measure plasma CCK level and the histological changes of jejunum in Wistar rats after treatment with protein extract of Willis raw soybean (PEWS), protein extract of Detam 1 raw soybean (PEDS) and protein extract of Detam 1 tempeh (PEDT) for 14 days. This study was also to ascertain whether b-conglycinin and ANF contribute  to  reducing body weight by giving PEWS, PEDS, and PEDT to 4 groups of 6 rats for 14 days. We observed food intake, body weight, CCK level, and histological profile of jejunum. As a conclusion, PEWS, PEDS, and PEDT treatment to Wistar Rats  for 14 days caused increasing CCK plasma level and jejunum villi atrophy. The reducing body weight is caused not only by b-conglycinin but probably by ANF as well.

Abstract

Soybean is a good source of protein. It has two major fractions, b-conglycinin (7S) and glycinin (11S). b-conglycinin’s function was known to suppress food intake, and this effect may be due to stimulating endogenous cholecystokinin (CCK) release. The aims of this study were to determine the highest content of total b-conglycinin and b-conglycinin sub unit-b level obtained from two varieties of soybean i.e. Wilis and Detam 1 varieties using different preparation and extraction methods. These two soybean varieties were prepared into tempeh. Then the seed and tempeh were extracted using Deak and Panthee methods. There were six extracts analysed using sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS PAGE) and coomassie brilliant blue (CBB) staining. The result was shown that Detam variety and raw seed contained the highest total b-conglycinin level. And Panthee method was the best method for extraction of total b-conglycinin, while Deak method was the best method for extraction of b-conglycinin subunit-b.

Abstract

In the last decade, the number of overweight children and adults has increased all over the world. In Indonesia,  obesity  needs  serious  management  because  it  is  multi-factorial  etiology.  One  of  the predominant  factors  of  obesity  is  excessive-appetite.  This  paper  aims  to  discuss  a  gut  hormone  called cholecystokinin  (CCK),  which  has  been  proven  to  be  able  to  decrease  appetite.  CCK  has  a  short  term anorectic effect, which becomes the reason for the hormone to be one of therapeutic targets of obesity in the future.  CCK  is  produced  in  the  enteroendocrine  cell  along  the  small  gut  mucous  and  the  secretion  is stimulated  by  protein  and  fat  through  signal  transduction  mechanisms  called  G  Protein  Coupling Receptor  (GPCR),  GPR  93.  Then  they  activate  Extracellular  Signal-Related  Kinase  (ERK)  1/2  through Gαq and Gαi signal transduction. Soybean contains a bioactive protein, β-conglycinin (BconP) subunit–β, which  has  been  proven  to  be  the  best  inducer  for  CCK  secretion.  The  well-known  signal  transduction pathway is Gαq-coupled  GPCR,  which  influences  the  calcium  signal  and  activates  Protein  Kinase  C (PKC). However, the downstream signaling to complete the pathway remains unclear and further study will be needed to explore the whole process.   Keywords: signal transduction, cholecystokinin, fat, protein, β-conglycinin

Abstract

Diarrhea in Indonesia is potential to be epidemic; its morbidity is high and it may cause death and malnutrition in infants and toddlers. In 2006, 41 districts of 16 provinces in Indonesia reported the incidents of diarrhea epidemic in their area. At Cikampek Saraswati General Hospital, diarrhea was the fourth highest disease in 2007 and children of 1-4 years old were the highest risk group. The aim of this research is to describe the respondents’ education, economic level, knowledge, attitudes, behavior, toddler’s nutrition, personal hygiene, as well as the provision of clean water, environmental sanitation, and health facilities that have affected the incidents of diarrhea in Cikampek Saraswati General Hospital. This study uses descriptive methods, the cross-sectional research design and 46 questionnaire items as the research instruments. The participants of this research were mothers or guardian mothers with children aged between 1-5 years old suffering from diarrhea and being registered as patients in Cikampek Saraswati General Hospital in July 2008. The incidental samples were 104 people. The findings showed that the education, economic level, knowledge, behavior, toddler’s nutrition, personal hygiene, and environmental sanitation of the respondents were still poor, while the attitudes of the respondents, the provision of clean water, health facilities in the residence of respondents were already good.

Abstract

In Indonesia most people starts their days with eating breakfast.  However, there are still many who do not realize the importance of starting a day with breakfast. Many research suggest that having breakfast can improve short term memory. After breakfast, blood glucose level will raise. It is assumed that glucose plays an important role in improving memory. The purpose of this research is to  find the effect of raised blood glucose level in improving short term memory. In this research Real Prospective Experimental Method and Random Complete Design were used. 30 FK UKM third year female student have been studied, their age range between 19-23 years old, and they were given porridge for breakfast. Blood glucose level was measured before breakfast and 30, 60, 90 minutes afterwards. Short term memory was tested with memory test before breakfast and three times after breakfast. Data was analyzed to analyze the correlation between blood glucose and memory. There are significant increase of blood glucose and memory after breakfast and there is a significant corelation between raised blood glucose and memory. The conclusion is breakfast may improve memory and the raised of blood glucose level matched with the improvement of memory.  

Peran Zn dalam Terjadinya Acne vulgaris

Abstract

Zinc deficiency may lead to many diseases and symptoms. One of them is Acne vulgaris. Many researchers said that plasma zinc level patients with severe acne is lower than normal. The purpose of this research is to measure the plasma zinc level and the zinc intake daily amount in Acne patients. The aim of this research is  to know the role of plasma zinc level in Acne vulgaris. And also the correlation between plasma zinc level and nutrition intake. The subjects are 74 people, consist of 42 sample and  32 control,  who came to skin health center  in RSHS, Bandung, skin health center in RS Imanuel, Bandung and students of Maranatha Christian University who have Acne in their face and also who have normal skin. All subjects have met the selection criteria. Data is taken by private interview, private questionnaire and laboratory plasma zinc test. Data is analysed by Kruskal-Wallis Chi square,  Mann-Whitney test, t-test and Rank Spearman correlation. The result shows there is a  tendency that  if Zn plasma level is lower so the Acne disease  is worse, and adversely if Zn plasma level becomes higher, the disease becomes better. But the Zinc intake does not show  significant correlation with plasma zinc level, either in the subject group nor in the control normal group. Conclusion, plasma zinc level has an  important role  in  Acne vulgaris, but  has no correlation with nutrition intake.

Abstract

Autisme adalah gangguan perkembangan pervasif pada anak yang umumnya muncul sebelum usia 3 tahun. Autisme bukan gangguan perilaku semata, tapi ada sebab-sebab fisik yang mendasarinya. Jadi pengobatannya selain untuk terapi mental, juga diperlukan intervensi biomedis untuk terapi fisik sesuai penyebab gejalanya. Dipaparkan kasus Alan yang didiagnosis sebagai ADD (Attention Deficit Disorder) / ADHD (Attention Deficit Disorder + Hiperactivity) dan Taylor yang didiagnosis sebagai Late onset infantile Autism, keduanya adalah putra Pamela Scott. Mereka berhasil pulih setelah melalui serangkaian intervensi seperti: Pemeriksaan urin, pemeriksaan alergi, Home Based Program 40 jam seminggu untuk terapi bicara, kerja, komunikasi dan perilaku. Serta Diet Anti Candida, pemberian Suplemen, Diet Gluten Free Casein Free, Pemberian Probiotik, dan injeksi EPD (Enzyme Potentiated Desensitization) untuk terapi fisiknya. Penemuan kasus dini sangat penting, karena Diagnosis dan intervensi dini sangat menentukan prognosis.

Efektifitas Dosis Tunggal Berulang Mebendazol500 mg Terhadap Trikuriasis pada Anak-Anak Sekolah Dasar Cigadung dan Cicadas, Bandung Timur

Abstract

Untuk penanggulangan infeksi cacing-cacing yang ditularkan melalui tanah pada anak-anak Sekolah Dasar dilakukan program penyuluhan kesehatan dan pengobatan masal. Penelitian ini dilakukan pada 501 anak-anak di Sekolah Dasar Cigadung dan Cicadas, Bandung Timur. Anak-Anak yang positif trikuriasis diberi mebendazol dengan dua cara, kelompok I diberi mebendazol500 mg dosis tunggal, sedangkan kelompok II diberi mebendazol500 mg dosis tumggal dan diulangi setelah satu minggu. Prevalensi infeksi cadng di Sekolah Dasar Cigadung dan Cicadas (sebelum pengobatan) adalah askariasis 5,99 %, trikuriasis 14,57 %, berbeda dengan pola prevalensi tahun 1991 dimana askariasis 83,33 % dan trikuriasis 57,97% .Hal ini mungkin disebabkan perbedaan obat cacing yang digunakan, dimana umumnya efektif terhadap askariasis, tetapi kurang efektif terhadap trikuriasis. Dua minggu setelah pengobatan angka penyembuhan trikuriasis adalah 89,47 % untuk kelompok I dan 91,43 % untuk kelompok II. Angka penurunan jumlah telur 93,53 % untuk kelompok I dan 97,59 % untuk kelompok II.

The Analgetic Effect of Kayu Rapat Bark Infusion (Parameria laevigata (Juss.) Moldenke) on Male Mice Treated with Thermal Induction

Abstract

Introduction: Kayu rapat bark has been empirically used to treat many kinds of disease, and was assumed to have analgetic effect because it contains flavonoid and polyphenol in its bark.Objectives: The aim of this experiment is to discover the analgetic effect of kayu rapat (Parameria laevigata (Juss.) Moldenke) bark.Methods: The method used in this experiment was laboratoric experiment. The analgetic property was examined through heat-induced pain using a heating plate equiped with a thermostat of 550C. The experimental animal used were Swiss -Webster 25 male mice weighing ± 28 grams which were then divided into 5 treatment groups (n=5), each group was given a kayu rapat bark infusion (IKKR) of 0.975 g/kgBW, 1.95 g/kgBW , 3.9 g/kgBW, Aquadest (as control) and Sodium diclofenac 17.86 mg/kgBW (drug for comparison). The datas taken were the reaction times of the earliest visible response marked by lifting up or licking the front paws or even jumping of the mice. Data were analyzed using one way ANOVA, followed by mean difference test of Tukey HSD with α = 0.05.Results: The experimental results showed a significant difference for the IKKR of 0.975 g/kgBW (p<0.05). On the other hand, a highly significant difference was observed for th e IKKR of 1.95 g/kgBW and 3.9 g/kgBW (p<0.01). The experimental group that was given the IKKR of 3.9 g/kgBW showed a similar potency as Sodium diclofenac (p>0.05).Conclusion: It is therefore concluded that kayu rapat bark infusion had analgetic effect possesses an an algetic property.

REPPELENT EFFECT OF LAVENDER, ROSE AND ROSEMARY OIL ON Aedes aegypti MOSQUITOES

Abstract

Introduction :Dengue Haemmorhagic Fever (DHF) was one of arthropode borne diseases which was spread by mosquitoes. To prevent insect bites we could use synthetic repellent like diethyltoluamid (DEET). DEET usages for long time period could cause many side effects so we needed to seek natural repellent which was safe and effective. Objective : to know the potential effect of lavender, rose and rosemary  oil repellent to adult female Aedes aegypti mosquitoes. Methods :The research design was true experimental with Complete Random Sampling, comparatively. The effects of reppelents were examined by using one side method. Seven hundred and fifty  Aedes aegypti mosquitoes which were separated into 5 groups, were given treatment randomly with of lavender, rose, rosemary  oil, control and DEET 12.5%. Data were measured by counting the number  of mosquitoes which migrated from the side that had been given treatment. Data were analyzed by using ANOVA one way, continued with Tukey HSD test, α = 0,05 using computer software. Results :This study showed the amount of mosquitoes which migrated from the side which had been given lavender oil was 89.33%, rose oil  92,67%, rosemary  oil 94,00% highly different significant  (p<0,01) from control 8.67%. Conclusion was lavender, rose, and rosemary  oil had potential effects as repellents. The potential effects of rose, and rosemary  oil were similar  to DEET 12,5 % (p>0,05), but lavender oil was lower than DEET 12,5 % (p<0,05).
