Abstract

Erythrina plants (Leguminosae) were used as traditional antifertility agents. In the continuing research for novel plant with antifertility capacity from Indonesian plants, the methanol extract of Erythrina plants showed significant antifertility activity in an in vitro assay of spermatozoa of white rat (R. norvegicus). The methanol extract was separated using bioassay-guide n-hexane, ethylacetate, and n-buthanol fractionation. The ethylacetate fraction was tested in vitro on spermatozoa of white rat (R. norvegicus) at a concentration of 0.25 x 10-3 μg/μL. It showed a decrease of their motilities and viabilities, as well as an increasing abnormality of the spermatozoa.

Abstract

Traditional medicines originating from plants have been widely used as an alternative therapy and extensive research have identified several bioactive compounds of the plants. In Indonesia, Erythrina variegata (Leguminosae) plants have been used for medication to many diseases including anthelmintic, anticancer,antimalaria, and antifertility.Objective: In the course of our continuing search for novel bioactive compounds from Indonesian Erythrina plants, we isolated and described several bioactive compounds such as anticancer, antimalaria, and antifertility from E. variegata. Method: Activity assay in vitro against breast cancer cell T47D using the Sulforhodamine B (SRB) method, activity assay in vitro against antiplasmodium using lactatedehydrogenase (LDH) method, and antifertility activity on spermatozoa Rattus novergicus. Their chemical structures were determined based on spectroscopic evidences and comparison with related compound previously reported. Result: Extract and compounds of E. variegata showed anticancer in vitro against breast cancer cell T47D, antiplasmodial in vitro K1 and 3D7 strain parasites, and antifertility on spermatozoa R. norvegicus with midle and high activity. Besides that, the toxicity assay of it was also conducted using white mice to show the safety grade of the plant. Conclusion: This results strongly suggested that E. variegata is a promising sources as herbal medicine of anticancer, antimalaria, and antifertility agents.

Abstract

In the course of our continuing research for novel antimalarial agent from Indonesian plants, the methanol extract of the Erythrina variegata leaves showed significant antimalarial activity against Plasmodium falciparum strain K1 in vitro with IC50 6.8 μg/mL. The methanol extract was separated by using variety of chromatography techniques. The chemical structure of an antimalarial compound was determined on the basis of spectroscopic evidence and compared to previous data then this compound is identified as a pentacyclic triterpenoid oleane derivative, namely 3,22,23-trihydroxy-oleane-12-ene. The pentacyclic triterpenoid, 3,22,23-trihydroxy-oleane- 12-ene showed antimalarial activity against 3D7 and K1 strains with IC50 4.3 μg/mL and 24 μg/mL, respectively. Abstract

In the course of our continuing search for novel paralytic compound from Indonesian plants, the methanol extract of the stem bark of Erythrina variegata (Leguminosae) showed significant paralytic activity against the third instar larvae of silkworm (Bombyx mori). The purposes of this research were isolation and structural elucidation of paralytic compound from the stem bark of E. variegata. Using the paralytic activity following the separation, the methanol extract was separated by combination of column chromatography to yield prenylisoflavone, warangalone. The chemical structure of warangalone was identified based on spectroscopic evidence and comparison with the previous reported. The paralytic activity of warangalone showed weak activity against the third instar larvae of silkworm (B. mori).Keywords: Erythrina variegata, Leguminosae, isoflavonoid, warangalone, paralytic activity

Abstract

Dalam penelitian berkelanjutan untuk pencarian senyawa metabolit sekunder baru dari tumbuhan Kalanchoe Indonesia, telah dilakukan kajian fitokimia terhadap Kalanchoe serrata. Daun segar K. serrata sebanyak 14,8 kg diekstraksi dengan metanol pada temperatur kamar. Ekstrak metanol (292 g) dipartisi berturut-turut dengan n-heksana dan metilenklorida. Ekstrak metilenklorida (0,80 g) selanjutnya dipisahkan pada kromatografi cair vakum pada silika gel G60 dengan eluen kloroform-aseton yang meningkat kepolarannya sehingga dihasilkan 10 fraksi yang dikelompokan berdasarkan analisis KLT. Padatan yang diperoleh pada fraksi yang terelusi dengan 20% aseton selanjutnya dipisahkan pada kromatografi kolom pada silika gel (230-400 mesh) dengan eluen kloroform dan dimurnikan lebih lanjut dengan kristalisasi pada aseton sehingga dihasilkan isolat berbentuk kristal jarum tak-berwarna sebanyak 23 mg. Isolat menunjukkan titik leleh 115-118oC dan memberikan warna hijau-kebiruan pada uji Liebermann-Burchard menunjukkan adanya kerangka steroid.  Hasil analisis spektroskopi yang meliputi UV, IR, 1D-NMR dan 2D-NMR menunjukan bahwa isolat merupakan turunan sterol dan diidentifikasi sebagai poriferasta-5,22E,25-trien-3β-ol,22-dehidrokolesterol.

Abstract

Erythrina plants, locally known as “dadap ayam”, are higher plant species and have been used as a folk medicine for treatment of cancer. To prove the effectiveness of the leaves and stem bark of E. variegata as an anti-cancer agent, the assay in this research was focused on in vitro  test  towards  breast  cancer  cell  T47D.  In  the course  of  our  continuing  search  for novel anti-cancer agent from Erythrina plants, the methanol extract of the leaves and stem bark of  E. variegata  showed  significant  anti-cancer  activity  against  breast  cancer  cell  T47D  in  vitro  using the Sulphorhodamine B (SRB) assay. By using the anti-cancer activity to follow the separations, the methanol extract was separated by combination of column chromatography. The chemical structure  of  an  anti-cancer  compounds  were  determined  on  the  basis  of  spectroscopic evidence  and  comparison  with  the  previously  reported  and  identified  as  an  erythrina  alkaloid (1)  and  isoflavonoid  (2).  Compounds  (1-2)  showed  anti-cancer  activity  against  breast  cancer cell  T47D  used  with  IC50  of    1.0  and  3.3  µg/mL,  respectively.  This  results  strongly  suggested that E. variegata is promising sources for anti-cancer agents.Keywords: Anti-cancer, Erythrina variegata, Leguminoseae

Abstract

Dalam penelitian yang berkelanjutan ini bertujuan untuk menemukan senyawa bioaktif alami dari tanaman Indonesia, diperoleh hasil bahwa ekstrak metanol dari biji Erythrina variegata (Leguminosae) menunjukkan aktivitas paralitik dan daun E. variegata menunjukkan aktivitas antimalaria. Ekstrak metanol dipisahkan komponen-komponennya dengan kombinasi kolom kromatografi diperoleh dua senyawa aktif (1 dan 2). Struktur kimia senyawa aktif (1 dan 2) ditetapkan berdasarkan data-data spektroskopi dan perbandingan data dari senyawa yang berhubungan dari penelitian sebelumnya, dan diidentifikasikan sebagai alkaloid eritrina (1) dan turunan triterpenoid pentasiklik (2). Senyawa (1) memperlihatkan aktivitas paralitik terhadap instar ketiga ulat sutera (Bombyx mori) dengan ED50 15 μg/g diet. Senyawa (2) memperlihatkan aktivitas antimalaria dengan IC50 0,243 μg/mL terhadap pertumbuhan P. falciparum secara in vitro. Abstract

Tumbuhan Erythrina variegata (Leguminosae) secara tradisional dikenal oleh masyarakat Indonesia sebagai obat antikanker dan antimalaria. Bagian tumbuhan ini yang biasa digunakan sebagai bahan pengobatan adalah daun dan kulit batang, tapi kandungan senyawa kimia aktif biologisnya belum banyak dilaporkan. Tujuan penelitian ini adalah untuk mengungkapkan senyawa aktif antikanker dan antimalaria secara in vitro yang terdapat di dalam daun dan kulit batang E. variegata. Penelitian dilakukan dengan cara ekstraksi metanol dan fraksionasi dari daun dan kulit batang E. variegata yang dipandu dengan uji antikanker secara in vitro terhadap sel kanker payudara T47D menggunakan metode Sulforodamin B (SRB) dan uji antimalaria secara in vitro terhadap Plasmodium falciparum 3D7 (sensitif klorokuin) dan K1 (resisten klorokuin) menggunakan metode laktatdehidrogenase (LDH). Selanjutnya dilakukan pemisahan fraksi etil asetat daun dan kulit batang E. variegata yang dipantau dengan uji antikanker dan antimalaria secara in vitro menggunakan kombinasi kolom kromatografi diperoleh tiga senyawa aktif (1, 2, dan 3). Struktur kimia senyawa aktif (1, 2, dan 3) ditetapkan berdasarkan data-data spektroskopi dan diidentifikasikan sebagai turunan triterpenoid pentasiklik glikosida(1); flavonoid, eristagallin A (2); dan steroid, (22E)-5α,8α-epidioksiergosta-6,22-dien-3β-ol (3). Senyawa (1) menunjukkan aktivitas antimalaria secara in vitro terhadap P. falciparum strain 3D7 (sensitif klorokuin) dengan IC50 1,8 µg/mL dan terhadap strain K1 (resisten klorokuin) dengan IC50 3,3 µg/mL. Senyawa (2) dan (3) menunjukkan aktivitas antikanker secara in vitro terhadap sel kanker payurada T47D dengan IC50 masing-masing 3,0 dan 3,2 µg/mL. Tumbuhan E. variegata mempunyai potensi sebagai bahan dasar obat herbal antikanker payudara dan antimalaria.

Abstract

Dadap ayam (Erythrina variegata) secara tradisional dikenal oleh masyarakat lokal sebagai obat antikanker. Bagian tumbuhan ini yang biasa digunakan sebagai pengobatan adalah daun, tetapi kandungan senyawa kimia aktif biologisnya belum banyak dilaporkan. Pencarian komponen aktif yang terdapat di dalam tumbuhan E. variegata sangat penting sebagai nilai tambah untuk bahan obat herbal antikanker. Tujuan penelitian ini adalah untuk mengetahui senyawa yang beraktivitas antikanker dari bagian daun dan kulit batang E. variegata secara in vitro terhadap sel kanker payudara T47D. Penelitian dilakukan dengan cara ekstraksi metanol dan fraksionasi dari daun serta kulit batang E. variegata yang dipandu dengan uji antikanker secara in vitro menggunakan metode Sulforhodamin B (SRB). Selanjutnya, fraksi aktif dipisahkan komponen-komponennya dengan kombinasi kolom kromatografi yang dipandu dengan uji antikanker secara in vitro, diperoleh tiga senyawa aktif (1-3). Struktur kimia senyawa aktif (1-3) ditetapkan berdasarkan data spektroskopi dan perbandingan data dari senyawa yang berhubungan dari penelitian sebelumnya, dan diidentifikasikan sebagai turunan steroid (1-3). Senyawa (1-3) memperlihatkan aktivitas antikanker secara in vitro terhadap kanker payudara T47D dengan IC50 masing-masing 6,5; 5,3; dan 3,2 ?g/ml. Abstract

The leaves of Erythrina variegata (Leguminosae) used tradisional plant of an antimalarial. In the course of our continuing search for novel an antimalarial compound from Erythrina plants, the methanol extract of the leaves ofE. variegata showed significant antimalarial activity in vitro toward Plasmodium falciparum in vitro using the lactate dehydrogenase (LDH) method. The methanol extract of the leaves of E. variegata showed against bothstrains of parasite with IC50of 6.8 μg/ml against K1 and > 60 μg/ml against 3D7, respectively. The methanol extract of the leaves of E. variegata was separated by using bioassay-guide fractionation. The n-buthanol fraction yieldedthe most activity, exhibiting equipotency against both strains of parasite with IC50of 5.1 μg/ml against K1 and 13.5 μg/ml against 3D7, respectively. Furthermore, by using the antimalarial activity to follow separation, the n-buthanol fraction was separated by combination of column chromatography to yield an active compound. The active compound showed antimalarial activity against both strains of parasite used with IC50 of 4.3 μg/ml against K1 and 23.5 μg/ml against 3D7, respectively. Its inhibition of the resistant strain (K1) was also much better compared to its inhibition of the sensitive strain (3D7), indicated that the leaves of E. variegata to be potential as antimalarial agents, but its lower potency compared to artemisinin and chloroquin.
