Performance of Nile tilapia juvenile fed diet containing different dosages ofrecombinant common carpgrowth hormone

Abstract

The studywas conductedtodetermine thedose of recombinant common carp growth hormone (rCcGH) supplemented in the commercial diet that generates the best performanceonbodyweight, biomass, andfeed conversion rate of juvenile tilapia.The dose of rCcGH administered was10, 20, and30mg/kg of commercial feed, andnorGH supplementation as control. Eachtreatment was designed in triplicates. Juveniles at average bodylengthof about2cm(average body weight of 0.7 g) were reared in a density of25fish/m2, for three weeks in hapa net at a size of 2×2×1 m3. Fish were fed diet containing rCcGH twicea week with an interval ofthreedays. The juvenile were fed with the rCcGH supplemented diet three times daily at satiation. The results showedthat average body weight andbiomass of fish treated by rCcGH with the doses of 20 and 30 mg/kg feed was higher (p<0.05) than that of 10 mg/kg and control. In addition, feed conversionrate of 20 and 30 mg/kg feed treatments was lower (p<0.05) than that of 10 mg/kg and control. RT-PCR analysisshowedthat expression level of IGF-1 gene in juvenile liver was increased in parallel with the increasing of rCcGH dosages supplemented in diet.This suggestedthatIGF-1 plays an important rolein the induction ofgrowth,andfeed conversionefficiencyof tilapiafed diet containing rCcGH. Aromatase gene expression and masculinization of Nile tilapia immersed in water 36 °C containing 17α-methyltestosterone

Abstract

ABSTRACT  Immersion of undifferentiated larval tilapia in high temperature and 17α-methyltestosterone (MT) cab increase the male ratio. However, the effectiveness of immersion in high temperature of water containing MT remains to be evaluated. The purposes of this study were: 1) evaluate the male ratio, growth, and survival of tilapia, and 2) analyze the aromatase brain-type gene expression level in tilapia after immersing in high temperature (36 °C) containing MT at 2 mg/L for four hour with single and double immersion. Aromatase gene expression was analyzed by semi-quantitative RT-PCR (sqRT-PCR) method. The result showed that higher monosex male ratio was obtained by single immersion of MT at 36 °C at room temperature. Gene expression level of aromatase brain-type was lower on single immersion and increased significantly at second immersion compared to control (immersion at room temperature without MT). Immersion using MT and high temperature had no significant effect on fish survival. However the specific growth rate and fish biomass were higher than control. Thus, monosex male tilapia can be produced by single immersion of undifferentiated larvae at 36 °C temperature containing MT. Keywords: male ratio, aromatase, Oreochromis niloticus, temperature, 17α-methyltestosterone  ABSTRAK  Perendaman larva ikan nila yang belum terdeferensiasi kelaminnya dengan suhu tinggi dan hormon 17α-metiltestosteron (MT) dapat meningkatkan nisbah kelamin jantan. Tetapi, efektivitas perendaman menggunakan MT pada suhu tinggi belum diteliti. Tujuan dari penelitian ini adalah 1) mengevaluasi nisbah kelamin jantan, pertumbuhan, dan kelangsungan hidup ikan nila, dan 2) menganalisis ekspresi gen aromatase tipe-otak pada ikan direndam menggunakan MT dengan dosis 2 mg/L selama empat jam sebanyak satu dan dua kali perendaman pada suhu 36 °C. Ekspresi gen aromatase dianalisis menggunakan metode RT-PCR semi-kuantitatif (sqRT-PCR). Hasil penelitian menunjukkan bahwa kombinasi perendaman MT satu kali pada suhu 36 °C lebih tinggi menghasilkan ikan nila jantan monoseks dibandingkan perendaman MT satu kali pada suhu ruang. Tingkat ekspresi gen aromatase tipe otak pada perendaman satu kali lebih rendah, dan meningkat secara signifikan pada perendaman kedua dibandingkan dengan kontrol (perendaman pada suhu ruang tanpa MT). Perendaman larva menggunakan MT dan suhu 36 °C tidak berpengaruh nyata terhadap kelangsungan hidup, tetapi laju pertumbuhan spesifik dan biomassa ikan perlakuan tersebut lebih tinggi daripada kontrol. Dengan demikian, ikan nila jantan monoseks dapat diproduksi dengan perendaman satu kali pada larva yang belum terdeferensiasi jenis kelaminnya menggunakan MT pada suhu 36 °C. Kata kunci: rasio jantan, aromatase, Oreochromis niloticus, suhu, 17α-metiltestosteron

Identification and expression analysis of c-type and g-type lysozymes genes after Aeromonas hydrophila infection in African catfish

Abstract

ABSTRACTLysozymes play an important role in the first line of defense in fish and potentially used as an immunity status biomarker and immune responses evaluation in fish, which often found in two types, i.e. chicken-type and goose-type (c- and g-type, respectively). To recent, the information related to the sequences and the expression analysis of the c- and g-type lysozyme genes in African catfish is still limited. In the present study, we report a partial cloning and mRNA expression analysis of c-type and g-type lysozymes in African catfish Clarias gariepinus. We have successfully cloned and partially identify the c-type, and g-type lysozyme genes of C. gariepinus, which consist of 594 and 560 of coding sequences, respectively. Catalytic and other conserved residues were identified by multiple sequences alignment and they showed high similarity with other teleost fish species. mRNA levels of the genes were analyzed by using qPCR method and their expressions in the spleen, liver, and head kidney were rapidly modulated after Aeromonas hydrophila injection, with different patterns were observed in each organ. These results confirmed that c- and g-type lysozymes played an important role in non-specific immunity against A. hydrophila infection. This study provided valuable information that can be used to understand the African catfish immune systems for better disease and stress management in C. gariepinus culture.Keywords: lysozymes, gene identification, gene expression, bacterial infection, African catfish ABSTRAKLisozim berperan dalam sistem pertahanan dini pada ikan dan sangat potensial digunakan sebagai marka status imunitas dalam evaluasi respons imun. Lisozim umum ditemukan dalam dua tipe pada ikan: tipe-ayam (tipe-c) dan tipe-angsa (tipe-g). Informasi terkait sekuens gen dan ekspresi gen kedua tipe lisozim pada ikan lele dumbo sangat terbatas. Pada penelitian ini, kami melaporkan kloning gen secara parsial, dan analisis ekspresi gen dari kedua tipe lisozim pada ikan lele dumbo C. gariepinus. Sekuens parsial gen lisozim tipe-c dan tipe-g yang berhasil diidentifikasi adalah sepanjang 594 dan 560 pasang basa. Situs katalitik dan residu khas memiliki tingkat kesamaan yang tinggi dengan spesies ikan yang lain. Analisis mRNA dilakukan dengan metode quantitative PCR (qPCR). Ekspresi kedua gen di jaringan ginjal depan, limpa, dan hati dengan cepat terinduksi pasca infeksi bakteri A. hydrophila dengan pola yang berbeda. Hasil ini menunjukkan bahwa lisozim tipe-c dan tipe-g memiliki peran yang sangat penting dalam sistem imun ikan lele dumbo terhadap infeksi A. hydrophila. Penelitian ini menghasilkan informasi penting yang dapat digunakan untuk mempelajari sistem imun ikan lele dumbo dan sebagai acuan dalam penanganan dan manajemen penyakit pada budidaya ikan lele dumbo.Kata kunci: lisozim, identifikasi gen, ekspresi gen, infeksi bakteri, ikan lele dumbo
