Abstract

The research aims to find out the  incidence of gastrointestinal parasite in wild long tail macaque in wildlife  reserve  park, Weh  Island, Sabang. For  that purpose,  fecal  specimens  from 25 wild  long  tail macaques  in Weh  Island-  Sabang were  collected.  The  fresh  faeces  samples were  then  preserved  using AFA  (alcohol-formal-acetic)  liquid  prior  to  be  examined  in  Parasitology  Laboratorium,  Faculty  of Veterinary Medicine, Syiah Kuala University.   The  presence of gastrointestinal  parasite was  observed using centrifuge method as well as Boray  sedimentation method. The obtained data were tabulated and analyzed  descriptively.  The  result  revealed  that  two  parasite  species  were  found  in  gastrointestine; Eimeria  spp.    (12%)  and  Strongloides  spp.  (8%).  It  could  be  concluded  that  the  prevalence  of gastrointestinal  parasite  in wild  long  tail macaque  in wildlife  reserve  park, Weh  Island-  Sabang was considered mild.

Abstract

The study has been done to examine Leucocytozoonosis case using organ mashed method in broiler and ducks slaughtered in chickens slaughter house in Banda Aceh Nanggroe Aceh Darussalam. From each of broiler and ducks fifty organs were collected for sample. Before those animals were killed blood smear were taken to find merozoites and gametosites stadium while smashed organs were made to find schizont stadium. The number of schizont of Leucocytozoonosis were analyzed descriptively. Result of research indicate that was higher positiveLeucocytozoonosis with mashed method better than blood smear in broiler and duck. The prevalence of Leucocylozoonosis with blood smear respectively are 30 % and 24 % from broiler while by using organ smashed methods are 58 % and 54 % respectively.

Abstract

Telah dilakukan penelitian tentang intensitas cemaran larva nematoda dan jenis larva yang dominan pada rumput pengembalaan ternak domba di Aceh Besar. Sampel rumput diambil dari 4 desa, yaitu Meunasah Papeun, Lamreung, Leung Ie dan Cot Leupan. Pengambilan sampel dilakukan dengan membuat plot berbentuk huruf W pada sampel rumput yang akan diperiksa.

Abstract

A study was conducted to isolate the membrane protein of Toxoplasma gondii at bradyzoid stage forthe development of intradermal diagnostic test in livestock infected by the parasite. Toxoplasma wasinitially collected from meat of goat serologically positive to the parasite. The infected meat was then fedinto uninfected cat to obtain oocyst. The oocyst was inoculated into the stomach of mice to produce tachyzoitwhich in turn produce cyst in tissue known as bradyzoit . The membrane protein was then isolated from thebradyzoit. The protein was analysed by sodium dodecyl sulfate electrophoresis (SDS-PAGE). The dataobtained were presented descriptively. The protein concentration isolated from each mouse infected at thedose of 1x107 oocysts was 11.91 mg. Two protein bands specific for bradyzoit were identified at 97.72 kDaand 67.60 kDa.

Abstract

A research was conducted to find out an alternative diagnose in detecting toxoplasmosis in livestock/animal using intradermal test  from protein membrane of  T. gondii bradizoite stage. Local isolate ofmembrane T. gondii bradizoite stage  was used in the research. Ten of domestic sheep with the age of ± 1year and 10 mice strain Balb/c with the age ± 2 month were used in this research. The reaction ofhypersensitivity on the skin post protein membrane bradyzoite injection was indicated by the process ofskin thickening. The diameter skin thickening was measured using cutimeter, in which diameter e” 10 mmindicated positive diagnose. The result showed that optimal dosage of membran protein bradyzoite thatcould be applied to detect toxoplasmosis in livestock and animal using intradermal test were 0,6 ml and0,2 ml for sheep and mice respectively. The sensitivity and specificity level of antigen use (protein membrane)of T. gondii bradizoite stage from local isolate to diagnose toxoplasmosis in mice using intradermal testwere: 85.0 %  and 66.6 % respectively, while in sheep the sensitivity and specificity level were 85.0 % and66.6 % respectively. It can be concluded that  intradermal  test was appropriate to be implemented fordetecting toxoplasmosis in sheep and mice induced with tachyzoite T. gondii.

Abstract

Telah dilakukan suatu penelitian untuk mengidentifikasi jenis-jenis parasit nematoda gastrointestinal yang menyerang kambing dan domba jantan lokal yang dipotong di Rumah Potong Hewan (RPH) Banda Aceh. Penelitian ini juga bertujuan untuk mengetahui jumlah masing-masing genus cacing yang menyerang kedua ternak tersebut. Masing-masing sebanyak 50 gastrointestinal kambing dan domba yang ada di RPH Banda Aceh ditentukan secara acak akan digunakan sebagai objek penelitian. Pemeriksaan terhadap cacing untuk keperluan identifikasi cacing nematoda gastrointestinal dilakukan setelah cacingcacing yang didapat dimasukkan ke dalam laktofenol selama 6 jam. Penentuan genus cacing nematoda gastrointestinal dilakukan sesuai dengan metode Soulsby (1982). Hasil identifikasi ternak kambing dan domba yang dipotong di RPH Banda Aceh menunjukkan bahwa kedua ternak tersebut sudah terinfestasi oleh cacing-cacing nematoda gastrointestinal. Adapun masingmasing genus untuk kambing adalah : Trichostrongylus spp, Oesophagustontum spp, Bunostomum spp, Chabertia spp, Trichuris spp dan Haemonchus spp. Sedangkan untuk domba cacing nematoda gastrointestinal dari genus: Gaigeria sp, Strongyloides spp, Bunostomum spp, Oesophagustomum spp, Haemonchus spp, Chabertia spp, dan Trichuris spp. Jumlah cacing nematoda gastrointestinal yang menyerang temak kambing sebanyak 514 ekor cacing sedangkan yang menyerang domba sebanyak 543 ekor cacing. Prilaku atau kebiasaan makan tidak signifikan (P > 0,05) terhadap jumlah cacing.nematoda gastrointestinal.

Abstract

A research has been done to identify the distribution of Culicoides species in Banda Aceh. Culicoides samples were collected by using modified Center for Disease Control (CDC) Miniatur Light Trap. All Culicoides samples has choice a small measured, were identified morfologically. The result showed that the number species Culicoides has caught in Ulee Kareng are: C. huffi, C. arakawae and C. shultzei, whereas in Alue Naga are C. huffi and C. shultzei. The distribution of species Culicoides in Ulee Kareng are: C. huffi has 22, C. arakawae 13 and C. shultzei 11. The average distribution of Culicoides in Alue Naga are C. huffi has 11 and C. shultzei has 4 .

Abstract

Penelitian ini bertujuan mengetahui efektivitas larvasida ekstrak etil asetat daun kemuning terhadap larva Ae. aegypti. Materi yang digunakan dalam penelitian ini adalah ekstrak etil asetat daun kemuning (EEADK). Rancangan penelitian ini menggunakan rancangan acak lengkap (RAL) yang terdiri atas enam perlakuan yaitu P1 (100 ml aquades), P2 (abate 10%), P3 (EEADK 10 ppm), P4 (EEADK 50 ppm), P5 (EEADK 100 ppm), dan P6 (EEADK 1000 ppm). Pada masing-masing perlakuan ditambahkan 25 larva nyamuk Ae. Aegypti dan ragi sebagai makanan. Masing-masing perlakuan diulang sebanyak 3 kali ulangan. Hasil yang diperoleh dianalisis dengan menggunakan analisis varian (ANAVA). Rata-rata jumlah larva yang mati 24 jam setelah pemberian ekstrak pada kelompok P1; P2; P3; P4; P5; dan P6 masing-masing adalah 25,00±0,00; 0,00±0,00; 0,33±0,39; 1,00±0,00 ; 2,33±0,94; dan 3,33±0,47. Berdasarkan analisis uji Puncan menunjukkan bahwa terdapat perbedaan yang nyata (P<0,05) antara kelompok perlakuan dengan kelompok kontrol. Pemberian ekstrak etil asetat daun kemuning dapat menurunkan jumlah larva Ae. Aegypti.

Abstract

The research has been done to know prevalence of toxoplasmosis in livestock, to search the source of infection that have potency to cause toxoplasmosis in human and to make a map of toxoplasmosis infection area in Banda Aceh. The method used in this research was serologic test, Card Aglutination Test (CATT). Data of fertile women in this study are collected through questioner. The result of this research showed that the toxoplasmosis prevalency number of society in Banda Aceh was 3.15%, while at livestock were varied respectively: goats 40%, chickens 25%, cattles 23%, duck 20%, cats 16%, buffalos 15%, and sheeps 10%. The potencial sources of to coures toxoplasmosis in human are: goat, chicken, cattle, duck, cat, buffalo, and sheep. The area found toxoplasmosis in humans and animal were: Baiturrahman, Kuta Raja, Ulee Kareng, Kuta Alam, and Syiah Kuala. Abstract

The purpose of this research was to compare the use of PCR method and histological examination to diagnose toxoplasmosis in tissues of Balb/c mice infected with sporulated oocysts through drinking water. A total of 20 male Balb/c mice aged approximately 2 months were used in this experiment. Each mouse was infected with 1x103 Toxoplasma gondii tachyzoites intraperitoneally. Tissue samples (liver, lung, heart, kidney, and brain) were collected from 5 mice on day 1, day 5, day 7, and day 9 after infection. Samples were then examined by PCR and histological methods. The data collected were analyzed descriptively. The results showed that PCR method was more sensitive than histological examination. PCR examination using primer invitrogen gen can amplify DNA T. gondii at 436 bp of the samples from liver, lung, heart and brain on Day 7 and Day 9 after infection. The histological examination showed that the cyst of toxoplasma was found in the brain while mononuclear cells infiltration was found in other internal organs.
