Abstract

The objective of this study was to identify the type of specific glycoconjugates and its distribution in testicular spermatogenic cells in muntjak (Muntiacus muntjak muntjak) based on lectins histochemistry. An adult male muntjak aged 4-5 years old in hard antler period was used in this study. Testicular tissue was fixed in Bouin solution and processed histologically. Histochemistry method was performed using six types biotinylated lectins such as peanut agglutinin (PNA), soybean agglutinin (SBA), wheat germ agglutinin (WGA), ricinus communis agglutinin (RCA), concanavalin A (Con A), and ulex europaeus agglutinin I (UEA I) with 20 µg/ml of concentration for PNA lectins and 15µg/ml for other type of lectins. The results showed that glycoconjugates were detected by all type of lectins except UEA I in testicular spermatogenic cells with variation in distribution pattern and also the intensity of lectins binding. Glycoconjugates β-galactose, β-glucose, mannose, Nacetylgalactosamine, N-acetylglucosamine and sialic acid were stained intensely by lectins in golgy-cap phase and acrosomal phase of spermatids. Glycoconjugate N-acetylgalactosamine was the sugar residues which distributed abundantly that marked by positive reaction with PNA, SBA, and RCA lectins. In conclusion, glycoconjugates are detected in testicular spermatids cells of muntjak indicated that glycoconjugates have an important role in spermatogenesis particularly in spermiogenesis. Key words: glycoconjugates, lectins, spermatid, spermatozoa, muntjak

Abstract

The aim of this research was to observe the histological of the thymus in tegal duck (Anas javanicus) according to the variation of ages. The animals used in this research were one month, two months and three months old of tegal duck. The thymus of each duck was analyzed by Hematoxylin-Eosin (HE) method and then observed under a light microscope. The result of the research performed differences in thymus structure of these three ages. The cortex had increased at the age of 2 months and decreased at the age of 3 months. The longest diameter of medulla at the age of 3 months was followed by ages 2 months and 1 month of tegal ducks.  The distribution of Hassall's body had increased at every age level. So, the cortex thickness decreased at the age of 2 months to 3 months, while the diameter of the medulla was getting longer.

Abstract

The objective of this study was to investigate the pattern of carbohydrate distribution of accressory sex glands in male muntjac (Muntiacus muntjak muntjak) in the velvet antler stage. In macroanatomy observation, muntjac accessory sex glands consist of the ampulla, vesicular gland, pars diseminate prostat, and bulbourethral gland. The observation of neutral carbohydrate distribution has done by periodic acid Schiff (PAS) staining. In addition, acid carbohydrate was observed by using alcian blue (AB) pH2,5 staining. Result of this research showed that neutral and acid carbohydrate scattering in each accessories sex gland with different pattern. Neutral carbohydrate with medium (++) and high (+++) intensity reaction was found in the ampulla, vesicularis gland, and bulbourethral gland and strong in prostat gland. The acid carbohydrate with low (+) and medium (++) intensity reaction was observed in the ampulla, low in vesicularis gland, and negative in prostat and bulbourethral glands. In conclusion, the neutral carbohydrate was found dominantly on surface of all accessory sex glands of male muntjac in velvet antler stage with highest concentration and intensity of colour in prostat gland.Key words: carbohydrate, accessory sex glands, muntjac (Muntiacus muntjak muntjak)

Abstract

The aim of this research was to determine the anatomy and histology of pancreas of water monitor lizard (Varanus salvator). Two pancreas were used in this research (a male and a female). Water monitor lizard was euthanized using chloroform and subsequently necropsied to collect the pancreas. The pancreas was washed in 0.9 % NaCl solution. Anatomical observations were carried out including location, shape, and color of pancreas, also the length and weight measurements. Then, pancreas was fixed with 4% paraformaldehyde for 7 days to proceed to histological preparation and hematoxylin eosin (HE) staining. The results of the anatomical observations showed that pancreas of water monitor lizard have two lobes, dorsal lobe (splenic lobe) and ventral lobe (duodenal lobe). Dorsal lobe which was in oval form adhered with the spleen, and ventral lobe was located in the cranial of intestines with the form of long spherical. Both of pancreas lobes were white-yellow color with the length of 1.45±0.64 cm (dorsal lobe) and 7.00±0.42 cm (ventral lobe). Weight of dorsal lobe was 0.15±0.01 g and ventral lobe was 6.35±2.30 g. The histological observations found the acinar cells, ducts, and blood vessels in both of pancreas lobes. Additionally, the similar structure of Langerhans islet only found at the ventral lobe of pancreas. In conclusion, pancreas of water monitor lizard consists of two lobes which located differently. Ventral lobe is larger than the dorsal lobe. Both lobes have similar histological structure except the Langerhans islets are only appeared in the ventral lobe.

Abstract

The objective of this study was to identified the development of the immunoreactive follicle stimulating hormone (ir-FSH) cells in pituitary glands of rats (Rattus norvegicus)in different age using immunohistochemical (IHC) methods. The pituitary organ that used in this study was collected from eight white female rats aged 2, 4, 6, and >12 months. Pituitary glands were processed histologically and stained with IHC methods to detect the ir-FSH cells. The result showed that the ir-FSH cells were observed in pars distalis (PD), pars tuberalis (PT), and pars intermedia (PI) of adenohypophysis with different number of cells. Furthermore, ir-FSH cells were found abundantly (+++) in the PT and a few (+) in PD of rats aged two months. In rats aged 4 months those cells were distributed in slight number (+) in PT and abundant (+++) in PD. In addition, the cells were also found in a great quantities in rats whose ages were 6 months, (+++) in both PT and PD, but these cells decreased (++) in rats aged >12 months in PD and absent (-) in PT. In conclusion, the devalopment and distribution of ir-FSH cells were found in pars tuberalis and pars distalis adenohypophysis in the pituitary glands of rats aged 2 up to >12 months with different number and distribution patterns.

Abstract

The aim of the  study was to determine the histological structure of tegal duck (Anas javanicus) spleen according to ages variation. One month, 2 month and 3 months of tegal duck were used as the sample of this study. The spleen of each duck was analyzed by Hematoxylin-Eosin (HE) method, then observed under light microscope. The result of this study performed there were the differences of histological structure of the ducks spleen. The capsule thickness had increased along with increasing age, which was 16.0 ± 1.0 µm; 25.3 ± 2.5 µm; and 29.0 ± 1.0 µm. The boundary between the two pulp in the tegal duck 3 months  old more obvious than 1 month or 2 months old. The diameter of white pulp had increased in the oldest duck. It can be concluded that the development of spleen histological structure was more perfect with the  increasing age of tegal duck.

Abstract

The aim of this research was to find out the effect of water temperature increase on leucocyte count of nile tilapia. The blood sample from 9 tilapia with weigh of 40-50 g were used in this study. The research was conducted using completely randomized design (CRD) with 3 treatments and 3 replications. Treatment P1 as a control with aquarium temperature was maintained at 29±1 C. Water temperature in treatment P2 and P3 was maintained at 32±1 C and 35±1 C respectively. The aquarium size in each treatment group was 80x60x40 cm with height of water 30 cm. The treatment was done for 15 days and on day 16th, the blood were collected from caudal vein. Data were analyzed with analysis of variance (Anova) and followed with Duncans multiple range test. The result showed that total leucocyte count (x 105/mm3) of nile tilapia in group P1, P2, and P3 were 3.08±0.88, 1.70±0.52, and 1.69±0.20 respectively. lts showed that an increase in temperature significantly affect (P<0.05) the leucocyte count of nile tilapia. The conclusion of this research is the increase in water temperature in aquarium decrease the leucocyte count of nile tilapia.

Abstract

This research aimed to calculate the number of phsycotrophic bacteria of nile tilapia reared in increased water temperature. For this purpose, 12 nile tilapias with body weight of 35-40 g were randomly allotted into 2 treatments groups with 3 replications. Fish in group 1 (P1) was reared in normal temperature (29±1° C) and group II (P2) was reared in high water temperature (35±1 C). All fish were fed commercial diet 3 times a day for 21 consecutive days. T-test analysis revealed a significant decrease in number of psychotropic bacteria (P<0.01) in group P2. Total plate count (TPC) resulted in fish gills, intestinal, and muscle were 1.5×10³, 7.7x10³, and 9.7x10³ cfu/g respectively. In conclusion, increasing of water temperature reduce the number of phsycotrophic bacteria of nile tilapia.

Abstract

This research aimed to determine the effect of giving Sipatah-patah extract toward the histopathological and femur bone growth of white rat that was ovariectomized. The experimental animals that were used were 12 white rats divided into 4 treatment group with 3 repetitions. K0 was the ovariectomized rat without giving Sipatah-patah extract (ESP); K1, K2 and K3 were ovariectomized rats which were given Sipatah-patah extract with multilevel doses of 500 mg/kg BW, 700 mg/kg BW and 900 mg/kg BW for 30 days. On the 31st day, rats were euthanized using chloroform and os femur that was taken to being made into histological preparation.  There was the decrease in the bones density of the K0 group which is characterized by thinning of trabecular structure, there were lots of osteoclast cells on the edge of the trabecular and lower density of active osteoblasts and passive osteoblasts. The rat of group K1 and K2 showed an improvement on the trabecular structure and lower osteoclast than group K0. The rat of group K3 had a visible improvement of the most congested trabecular structures, cohesive with the most density cell of active osteoblasts than the other groups. The result of this research concluded that the giving of Sipatah-patah extract doses 900 mg/kg BW showed a higher density of trabecular and active osteoblasts than the control group, K1 and K2 on the white rat bones that were ovariectomized.

Abstract

This research aimed to observe and identify the development of estrogen receptor alpha (ERα) in the ovary and uterus of rats (Rattus norvegicus) at various age levels using immunohistochemistry (IHC). The ovary and uterus were collected from eight female rats aged 2, 4, 6, and > 12 months and processed histologically and stained with IHC staining. This research showed, the intensity of ERα expression was found in high abundance (+++) in stroma and the corpus luteum and was weakly found (+) in the theca cells, blood vessels, oocytes and germinal epithelium of rats aged 2 months. Additionally, ERα expression in rats aged 4 months had similarities with rats aged 2 months, but the intensity of ERα had increased (++) in ovarian theca cells, blood vessels, and oocytes. In ovaries of rats aged 6 and > 12 months, ERα expression had decreased (++) in the stroma and the corpus luteum and had a low expression (+) in the blood vessel and oocytes. The uterus of rats aged 2 months had low expression of ERα (+) in the stroma and luminal epithelium, moderate (++) in the myometrium, but was not found in the uterine glands. Furthermore, the expression in the uterus of rats at the age of 4 months had increased in the stroma and luminal epithelium (++) and uterine glands (+).  At the age of 6 months, it found excessively (+++) in the stroma, myometrium, and luminal epithelium, moderate expression (++) in the uterine glands.  The expression of ERα in uterus aged > 12 months was almost equal to the uterus of rats aged 6 months, but the expression in the myometrium and epithelial luminal had decreased (++). It can be concluded that the development and distribution pattern of ERα in the ovaries and uterus of rats are different at each age levels, it may be related to the growth and development of reproductive organs and also closely related to the estrus cycles of rats.
