Abstract

length in North Sulawesi province. Data on animal live weight (LW), body length (BL) and chest girth(CG) were collected from all cows (n=363) kept by traditional household farmers. Regression analysiswas carried out to estimate LW from all linear body measurements. Data were classified based on age ofanimals consisted of five groups with the first age group of two and half years old, to the fifth age groupof seven and half years old. The results showed that age significantly (P<0.05) influenced all bodymeasurements. Correlations between all pairs of measurements were highly significant (P<0.001) for allage groups. Regression analysis showed that live weight could be predicted accurately from chest girthand body length (R2=0.97). Multiple regression model can be recommended to predict live weight ofOngole grade cows based on those variables with their age groups ranging from 2.5 to ≥7.5 years old asfollows: Live weight (kg) = -806.410 + 4.79835 CG (cm) + 2.83500 BL (cm).

Abstract

The objectives of this study were to detect the Mendelian mode inheritance of growth hormone(GH) and to establish genotype frequency of GH gene in Ongole-crossbred cattle mated by the artificialinsemination (AI) technique. Total of 76 blood samples were collected from Ongole-crossbred cows andbulls (G0), and their progenies (G1) at the Tumaratas AI service center in North Sulawesi province,Indonesia. All blood samples were screened for the presence of GH locus using a PCR-RFLP methodinvolving restricted enzyme Msp1 on 1.2 % of agarose gel. Data were analyzed using statistical programfunction in Excel XP. The results showed that GH locus using alleles of Msp1+ and Msp1- enzymerestriction in Ongole-crossbred cows and bulls was inherited to their Ongole-crossbred progeniesfollowing the Mendelian mode inheritance. This Mendelian inheritance generated by AI technique wasnot under genetic equilibrium for the Msp1 genotype frequencies in groups of G0 and G1. The breedingprogram using genotypes of bulls and cows (G0) for generating the genotype of GH Msp1 enzymerestriction by AI technique should be maintained to increase these various allele dispersion rates forbreeding under genetic equilibrium of the Ongole-crossbred cattle population.

Abstract

The aim of the research was to evaluate the genetic improving of production traits selected of Balicattle. Four hundred and twenty eight data of weaning weight, yearling weight and body weight gainwere used to estimate genetic improvement for those traits. One hundred and seven data of bodydimension (body weight, chest circumference, body length and withers height) at 24 months old wereused to estimate genetic improving for those traits. The estimation of genetic and environmentalvariance and co-variance component, and heritability were found by animal model. The fix effect ofweaning weight, yearling weight and body weight gain was rainfall, age of measurement and year ofbirth, whereas the fix effect of body dimension depend on each trait. The result showed that heritabilityof weaning weight, yearling weight, body weight gain, body weight, chest circumference, body lengthand withers height was 0.090.15, 0.270.13, 0.470.15, 0.070.19, 0.500.19, 0.340.28, 0.600.21,respectively. Genetic improvement of all traits have had a different pattern in selection responses. Alltraits did not show genetic improvement due to selection for.

Abstract

Genotypic performance in term of heritability as the crucial factor of animal economical traits for body size inheritance had not been fully studied in Ongole-grade cattle. The objectives of this research were to define the heritability values of live weight, chest girth and body length in Ongole-crossbred cattle. Total of 37 blood samples were collected from parental cows and 2 blood samples from parental Ongole breed bulls. All blood samples were screened for the presence of growth hormone (GH) locus using PCR-RFLP method involving restricted enzyme Msp1 on agarose-gel (1.2%). Data were analyzed using statistical program in Excel XP. Results showed that the phenotypic estimation average of Ongole grade cattle population of live weight, chest girth and body length were 445.41 ± 45.95 kg, 175.35 ± 4.11 cm, and 139.70 ± 5.73cm, respectively. The heritability values of animal live weight, chest girth and body length in this study were 0.24, 0.003, and 0.41, respectively. These heritability values of animal live weight and body length would be categorized as moderate to high genotypic performance values, while the heritability of animal chest girth was included in low heritability standard of the animal economical trait performance.

Abstract

The fruits of Piper cubeba (kemukus;Javanese)have been used in traditional medicine to treat illnesses such as asthma. Extracts were prepared by gradual maceration of the dried fruits with n-hexane, and by extraction of the residue with ethanol, and were tested for their tracheospasmolytic effects on isolated guinea-pig trachea contracted with metacholine at a dose of 0.25 mg/ml. Both extracts demonstrated spasm inhibition but the effect of the ethanol extract was lower than that of the n-hexane extract. However, TLC analysis did not distinguish between the extracts. The ethanol extract was then partitioned with n-hexane, and the first and second n-hexane extracts were combined and dried before further fractionation using vacuum liquid chromatography. The fractions were tested for tracheospasmolytic effects. Two active fractions were obtained and separated by column chromatography and preparative TLC. Four pure compounds were obtained. One of these is a lignan compound and identified as dihydrocubebin, had tracheospasmolytic activities.Key words: Piper cubeba; lignan, dihydrocubebin; tracheospasmolytic effect.

Abstract

One of the isolated active compound of the tracheospasmolytic from kemukus fruits (Piper cubeba L.f) is cubebin. The problem occurred when cubebin (C20H20O6) mixed with its epimer because of the difficultly to identify the structure by 1D-NMR spectroscopy. Structure identification then was conducted by 2D-NMR spectroscopy, so the structure of cubebin and epicubebin can be clear identified. Key words : Cubebin, epicubebin, identification, 2D-NMR

Abstract

Sapi Bali merupakan ternak lokal asli Indonesia sangat berpotensi untuk dikembang-biakkan dan sebagai plasma nutfah harus tetap dilestarikan agar tidak tercemar dengan “darah” bangsa sapi lain. Disinyalir performans sapi Bali cenderung menurun disebabkan aplikasi program pemuliaan yang kurang ketat, sehingga mutu genetiknya rendah. Dalam upaya memperbaiki mutu genetik, Pola Peternakan Inti Terbuka (Open Nucleus Breeding) tampaknya sesuai untuk diterapkan dalam pengembangan sapi Bali yang banyak melibatkan peternakan rakyat sebagai populasi dasar dengan dukungan kegiatan seleksi pada berbagai lapisan peternakan dan harus ditunjang dengan rekording performans produksi dan reproduksi secara tertib, benar dan akurat, serta berkesinambungan. Rekording merupakan metode untuk menunjang keberhasilan program perbaikan mutu genetik ternak yang sangat bermanfaat dalam program seleksi berdasarkan performans individu, dan dapat membantu manajemen beternak yang baik. Dari rekording dapat diketahui silsilah ternak sehingga sangat bermanfaat untuk melakukan analisis komponen ragam dan menduga nilai pemuliaan (breeding value) ternak. Tujuan penelitian untuk mengembangkan manajemen rekording bagi pola perbibitan sapi Bali dan perangkat lunak (software) sistem rekording dan pengolahan data. Luaran berupa laporan Kegiatan Model Rekording dan Pengolahan Data Untuk Program Seleksi Sapi Bali; model rekording  sederhana untuk data primer di lapang dan model tabulasi data secara manual; dan perangkat lunak program rekording (sistem rekording data produksi dan reproduksi, rekapitulasi data, faktor koreksi, dan nilai pemuliaan ternak). Kesimpulan adalah perbaikan mutu genetic untuk memperbaiki performans produksi pada keturunan secara bertahap dan kontinyu perlu dilakukan sehingga perlu perencanaan dan implementasi program breeding yang tepat dan berkelanjutan. Diperlukan format recording dan pengolahan data yang sederhana dan standar sehingga mudah diaplikasikan. Program recording di lapangan yaitu recording langsung di lapangan dengan menggunakan lembar isian recording oleh peternak dan komputerisasi data pada petugas atau di pusat pembimbitan, dinas atau instansi terkait. Diperlukan pelatihan mengenai manfaat dan teknik pengisian paket recording baik di tingkat peternak maupun petugas/dinas. Penggunaan perangkat lunak (software) dalam program recording akan sangat membantu dalam pengarsipan, pengolahan dan pencarian data yang diperlukan dalam rangka mendukung program breeding. Kata Kunci : sapi Bali, data, model rekording, program seleksi

Abstract

Penelitian tentang seleksi pejantan sapi PO berdasarkan nilai pemuliaannyatelah dilaksanakan di kandang percobaan Loka Penelitian Sapi Potong Grati Pasuruan,pada bulan Juni sampai November 2011. Tujuan penelitian untuk memperolehinformasi peringkat keunggulan pejantan sapi PO berdasarkan nilai pemuliaan bobotbadan terkoreksi umur 205 dan 365 hari. Materi yang digunakan adalah data rekordingproduksi sapi PO umur 205 dan 365 hari dari 310 ekor, keturunan dari 27 ekorpejantan dan 163 ekor induk. Metode yang digunakan adalah studi kasus denganmenganalisis data rekording sapi PO tersebut di lokasi penelitian (tahun 2004-2010).Variabel yang diamati adalah bobot badan umur 205 dan 365 hari; yang selanjutnyadilakukan koreksi untuk keperluan estimasi nilai pemuliaannya berdasarkan saudaratiri (half-sib). Hasil penelitian menunjukkan bahwa rataan bobot badan sapi PO umur205 hari sebesar 109,10+18,35 kg dan umur 365 hari sebesar 132,70+19,93 kg.Pejantan nomor 5 mempunyai nilai pemulian bobot badan umur 205 hari yangtertinggi; sedangkan nilai pemuliaan bobot badan umur 365 hari dimiliki pejantannomor 12. Dari hasil uji peringkat terdapat korelasi yang nyata (P<0,01) antaraperingkat pejantan berdasarkan nilai pemuliaan bobot badan umur 205 dan 365 hari.Disimpulkan bahwa pejantan nomor 5 menduduki peringkat pertama dari semuapejantan yang diuji, dengan nilai pemuliaan bobot badan umur 205 dan 365 harimasing-masing pada peringkat pertama dan ketiga.Kata kunci: nilai pemuliaan, pejantan, sapi Peranakan Ongole (PO)SIRE SELECTION BASED ON BREEDING VALUE IN PO CATTLE ATCATTLE RESEARCH CENTRE GRATI - PASURUANABSTRAKResearch on sire selection based on breeding value in PO cattle was conductedfrom June to November 2011 in Cattle Research Station, Grati, Pasuruan. Theobjective of this research was to find out the ranking of sire superiority based onbreeding value of 205 and 365 days corrected body weight. Materials used were 310production records of PO cattle at 205 and 365 days of age, which resulted from 27sires and 163 dams. Method used was case study with analyzing the data collectedfrom 2004-2010. Variables measured were body weight at 205 and 365 days of age,which then were corrected for estimating breeding value based on half-sib relationship.Results showed that mean of body weight of PO cattle at 205 and 365 days of age were109.10+18.35 kg and 132.70+19.93 kg, respectively. The best sire for body weight at205 days of age was sire number 5 and that at 365 days of age was sire number 12.There was significant rank correlation (P<0.01) between breeding value of bodyweight at 205 and that at 365 days of age. The conclusion was that sire number 5 wasthe best sire, with breeding value of body weight at 205 and 365 days of age at the firstand third rank, respectively.Key words: breeding value, sire, PO cattle
