Abstract

This research has purposes to isolating and identifying the lactic acid bacteria genus Pediococcus in sumatran orangutan (Pongo abelii) feaces. Feaces are isolated from one orangutan at Kandi Zoo and two orangutans at Kinantan Zoo. The isolating of the lactic acid bacteria was done by using streak plate method with the MRSA (de Man Rogosa Sharpe Agar) specific media. Identification was done by the morphological observing, Gram stain, catalase test, oxidase test and biochemical test. The result showed that one isolate feaces from Kandi Zoo has cocci shaped with the isolate prediction approaches genus Pediococcus sp. The characteristic features of isolate are cocci shaped, Gram positive, tetrad cell structure, nonmotile, catalase negative, and oxidase negative. Based on these characteristics, it can be concluded that there is a lactic acid bacteria coloni which grouped into genus Pediococcus in male orangutan feaces from Kandi Zoo

Abstract

Seroprevalence of avian influenza H5N1 in birds in north aceh districtABSTRACT. Avian influenza virus H5N1 infections are an important cause of diseases in humans and several animal species, including birds. The present study conducted to investigate the seroprevalence Avian Influenza H5N1 in native birds from 15 sub-districts of North Aceh.  This study utilized 1108 serum samples collected from the axilaris vein (left or right) of birds. The standard Hemaglutination Inhibition (HI) assay was conducted at Microbiology Laboratory Faculty of Veterinary Medicine of Syiah Kuala University to determined serum antibody possitive or negative reaction against Avian influenza H5N1. The result showed that seroprevalence Avian influenza H5N1 virus was 4,7 % in North Aceh District. There were nine sub-districts were tested positively by HI test. However, the serum collected from six sub-districts did not react (negative) against Avian influenza H5N1. Based on the results we obtained, a conclusion that natural infection by Avian influenza virus in native birds occured in part of North Aceh District.

Abstract

Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi bakteri Escherichia coli dan Salmonella sp pada telur ayam kampung yang gagal menetas di Laboratorium Lapangan Peternakan Universitas Syiah Kuala. Sebanyak 20 butir telur ayam kampung yang gagal menetas diperiksa di Laboratorium Mikrobiologi Fakultas Kedokteran Hewan, Universitas Syiah Kuala Banda Aceh. Sampel diisolasi berdasarkan metode Carter yang dimodifikasikan. Pada media Brilliance E.coli/ Coliform Selective Agar koloni bakteri tumbuh berbentuk bulat berwarna ungu, pada pewarnan Gram bakteri bewarna pink berbentuk batang pendek. Pada media SSA koloni bakteri berbentuk bulat bewarna hitam dan pada pewarnaan Gram bakteri bewarna pink berbentuk batang panjang. Hasil uji IMViC dan gula-gula sesuai dengan karakteristik Escherichia coli dan Salmonella sp. Hasil penelitian dari 20 sampel telur ayam kampung yang diteliti 3 sampel positif terinfeksi Escherichia coli dan 12 sampel positif Salmonella sp. Maka dapat disimpulkan Escherichia coli dan Salmonella sp merupakan salah satu penyebabab kegagalan menetas pada telur ayam kampung yang gagal menetas di Laboratorium Lapangan Peternakan Universitas Syiah Kuala.Kata kunci : telur ayam kampung, gagal menetas, Escherichia coli, Salmonella spThis study aims to isolat and identify bacteria Escherichia coli and Salmonella sp from failed to hatch domestic chicken in Field Laboratory of Animal Husbandry Syiah Kuala University. Twenty unhatched domestic chicken eggs were examined in microbiology laboratorium of Veterinary Faculty of Syiah Kuala University, Banda Aceh. Modified Carter methode was used to isolate bacterial from the samples. On media Brilliance E.coli/ Coliform Selective Agar colonies bacterial were purple and circular, on Gram staining it formed pink color cocobacil shape. On media SSA the colonies were black and circular, on Gram staining it formed pink color bacil shape. The result of Biochemistry test this showed that 3 of 20 domestic chicken eggs were positive infected by Escherichia coli and 12 of 20 sample were infected by Salmonella sp. It can conclude that Escherichia coli and Salmonella were one of the causes of failed to hatch of domestic chickens  egg’s.Keywords : domestic chicken eggs, fail to hatch, Escherichia coli, Salmonella sp

Abstract

ABSTRAKPenelitian ini bertujuan untuk mengisolasi dan mengidentifikasi bakteri Pseudomonas sp pada ikan asin di TPI Labuhanhaji Aceh Selatan. Penelitian ini menggunakan 15 sampel ikan asin dari 3 pedagang. Setiap pedagang diambil sejumlah 5 macam ikan asin, diantaranya ikan asin tongkol, dencis, layur, kembung, dan ikan kepala batu lalu diperiksa di Laboratorium Mikrobiologi Fakultas Kedokteran Hewan, Universitas Syiah Kuala Banda Aceh. Ikan asin di stomacher lalu di swab steril dan dimasukkan ke media Nutrien Broth  dan diinkubasikan pada suhu 37oC selama 24 jam, selanjutnya dilakukan penanaman pada media Pseudomonas Agar Base lalu diinkubasikan kembali pada suhu 37o C selama 24 jam. Koloni bakteri yang tumbuh pada media PAB dilakukan pengamatan bentuk bakteri, pigmentasi, permukaan, pinggiran, elevasi dan aspek koloni. Pewarnaan Gram dilakukan untuk memastikan bakteri yang didapat adalah kelompok Gram negatif. Terakhir dilakukan uji IMVIC,uji biokimia Triple Sugar Iron Agar dan uji gula-gula. Hasil penelitian ini di analisis secara deskriptif. Hasil pemeriksaan dari 15 sampel ternyata di temukan adanya pertumbuhan koloni bakteri Pseudomonas sp pada media PAB dan hasil pewarnaan Gram menunjukan Gram negatif. Oleh karena itu dapat di simpulkan bahwa ditemukan  bakteri Pseudomonas sp pada ikan asin di TPI Labuhanhaji Aceh Selatan. ABSTRACTThis study was aimed to isolate and identify Pseudomonas sp bacteria in salted fish in Fishing Auction Station (TPI) in Labuhanhaji Aceh Selatan. This study used 15 salted fish as the samples from 3 different traders. From each trader taken a number of 5 kinds of salted fish,  including Euthynnus affinis, Sardina piichardus, Trichiurus lepturus, Rastrelliger, and Osmeridae then they were checked in Microbiology Laboratory Faculty of Veterinary Medicine, Syiah Kuala University Banda Aceh. The salted fishs were placed in stomacher continued by putting them in sterile swab then they were put into Nutrient Broth medium incubated at 37oC for 24 hours, then planted on medium Pseudomonas Agar Base followed by incubating them again at 37oC for another 24 hours. Coloni of bacteria that grow on PAB were conducted to observe its bacterial form, pigmentation, surfaces, edges and elevation and the aspects of the colony. Gram staining was underdone to ensure the bacteria are the Gram-negative group. In the last treatment, IMVIC, Triple Sugar Iron Agar biochemical and sugar test were conducted. The result of this research is analyzed descriptively. The Results of examining 15 samples showed that there are the growth of Pseudomonas sp on PAB media and Gram staining results showed Gram negative. Therefore, it can be concluded that the Pseudomonas sp bacteria on salted fish in TPI Labuhanhaji Aceh Selatan were found.

Abstract

ABSTRAKPenelitian ini bertujuan untuk mengetahui daya hambat ekstrak biji buah pala (Myristicafragrans Houtt) terhadap pertumbuhan bakteri Escherichia coli. Biji pala yang digunakan dalam penelitian ini diperoleh dari pasar tradisional Darussalam, Banda Aceh. Proses ekstrasi biji buah pala menggunakan pelarut Etanol 96% dengan tingkat konsentrasi yang berbeda yaitu 1%, 10%,20%.  Menggunakan kertas cakram kosong (Blank disk) yang telah dicelupkan ke dalam ekstrak biji buah pala masing-masing pengulangan dan antibiotik ampisilin yang diletakkan pada media MHA. Pengujian daya hambat dilakukan dengan metode difusi Kirby-Bauer menggunakan kertas cakram pada media Mueller hinton agar (MHA) dengan 5 perlakuan dan 3 pengulangan. Media agar diinkubasi pada suhu 36-37ºC selama 24 jam. Hasil penelitian ini menunjukkan bahwa bahwa ektsrak  biji  buah  pala  memiliki  daya  hambat  yang  lemah  terhadap  pertumbuhan  bakteri Escherichia coli.  Ekstrak biji buah pala pada konsentrasi 1% memiliki daya hambat sebesar 9,73 mm, konsentrasi 10% sebesar 8,5 mm, konsentrasi 20% sebesar 8 mm dan kontrol positif sebesar20,83 mm.Kata kunci : antibakteri, ekstrak biji buah pala, Escherichia coli.. ABSTRACTThis study aims to determine the inhibitory power of nutmeg extract (Myristica fragransHoutt) on the growth of Escherichia coli bacteria. The nutmeg seed used in this study was obtained from the traditional market of Darussalam, Banda Aceh. Extraction process of nutmeg seed using96% ethanol solvent with different concentration level that is 1%, 10%, 20%. Using blank disc paper (Blank disk) which has been dipped into nutmeg extract of each repeating and ampicillin antibiotic placed on MHA media. Inhibitory testing was performed by the Kirby-Bauer diffusion method using paper discs on Mueller hinton agar medium (MHA) with 5 treatments and 3 repetitions. Media to be incubated at 36-37ºC for 24 hours. The results of this study indicate that the nutmeg nut extraction has a weak inhibitory effect on the growth of Escherichia coli bacteria. The extract  of nutmeg  seed  at concentration  of 1%  has inhibitory power  of 9.73  mm,  10%concentration of 8.5 mm, 20% concentration of 8 mm and positive control of 20.83 mm.Keywords : antibacterial, nutmeg ekstact, Escherichia coli.

Abstract

Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi bakteri Escherichia coli dan Staphylococcus aureus pada keumamah yang dijual di pasar tradisional Lambaro, Aceh Besar. Penelitian ini menggunakan 10 sampel keumamah yang di jual di pasar tradisional Lambaro, Aceh Besar. Isolasi Escherichia coli dan Staphylococcus aureus berdasarkan metode Carter. Data hasil penelitian ini dianalisis secara deskriptif, berdasarkan keberadaan Escherichia coli dan Staphylococcus aureus yang terdapat pada Keumamah. Hasil penelitian ini menunjukkan bahwa satu sampel positif tercemar Escherichia coli, sedangkan sembilan sampel negatif  sementara pada Staphylococcus aureus didapat hasil positif untuk semua keumamah yang di jual di pasar tradisional Lambaro, Aceh Besar. Disimpulkan bahwa keumamah yang dijual di pasar tradisional Lambaro, Aceh Besar telah tercemar Escherichia coli dan Staphylococcus aureus.This study aimed to isolate and identify Escherichia coli and Staphylococcus aureus on keumamah sold at Lambaro traditional market,  Aceh Besar. Ten keumamah were used for sample. Escherichia coli and Staphylococcus aureus isolate based on the Carter methode. The result of this study were analyzed descriptively,  based on the presence of Escherichia coli and Staphylococcus aureus in keumamah.  The result showed that one sample is positive contaminated with Escherichia coli, and nine samples are negative. However for Staphylococcus aureus all those ten samples of keumamah  sold at Lambaro traditional market Aceh Besar are positively contaminated. It can be concluded that keumamah sold at Lambaro traditional market,  Aceh Besar were contaminated by Escherichia coli (10%) and Staphylococcus aureus (100%).

Abstract

ABSTRAK                Penelitian ini bertujuan mengetahui jumlah total bakteri pada telur ayam yang dijual diwarung kopi kawasan Darussalam Kecamatan Syiah Kuala Banda Aceh. Penelitian ini menggunakan 12 butir telur ayam yang diperoleh dari enam warung kopi kawasan Darussalam. Jumlah total bakteri dilakukan dengan menggunakan metode Total Plate Count dengan pengenceran berseri 10-1 – 10-4 untuk isi telur. Hasil penelitian menunjukkan jumlah total mikroba tertinggi dari warung kopi B 2,0 x 105 CFU/ml, sedangkan terendah dari warung kopi E 3,0 x 103 CFU/ml. Kesimpulan dari penelitian ini sebanyak 0,16% warung kopi kawasan Darussalam, Kecamatan Syiah Kuala, Banda Aceh terdapat telur yang batas maksimum cemaran mikroba melebihi standar SNI 3926:2008 (1 x 105 CFU/ml).    Kata kunci :Jumlah Total Bakteri, Warung Kopi, telur ayam.ABSTRACT This study aimed to detetermined total plate count of bacteria in eggs that were  sold in coffee stalls at Syiah Kuala, Banda Aceh. This study used 12 eggs from each stall. Assessment were done in total plate count method with diluent of 10-1 – 10-4 to each egg content. The result showed highest count was from ‘B’ Coffee stall at 2,0 x 105 CFU/ml, while the lowest was found in ‘E’ 3,0 x 103 CFU/ml. This study concluded that 0,16% coffee stalls in region of Darussalam subdistrict in Banda Aceh exceed national standard of Indonesia which set at SNI  3926:2008 (1 x 105 CFU/ml).Keywords :Bacteria Total Plate Count, Coffee Stall, Chiken Egg.

Abstract

Penelitian ini bertujuan untuk mengisolasi Aspergillus sp pada paru-paru ayam kampung. Sampel yang digunakan dalam penelitian ini adalah 12 paru-paru ayam kampung yang diambil secara acak dari rumah potong unggas (RPU) Peunayong Banda Aceh. Isolasi Aspergillus sp dilakukan sesuai dengan metode Thompson (1969). Sampel dicuci dengan aquades steril yang berisi antibiotik selanjutnya ditanamkan pada media Sabouraud’s Dextrose Agar (SDA) kemudian diinkubasikan pada suhu kamar selama 2-7 hari. Pertumbuhan morfologi Aspergillus diamati secara makroskopis. Koloni yang diduga Aspergillus sp diperiksa secara mikroskopis. Data yang diperoleh dianalisis secara deskriptif. Hasil pemeriksaan menunjukkan bahwa Aspergillus sp dapat diisolasi pada 7 dari 12 sampel paru-paru. Dapat disimpulkan bahwa 58,33 % sampel paru-paru yang diperiksa positif terinfeksi Aspergillus sp dan 41,67 % bebas Aspergillus sp. This study aimed to isolate Aspergillus sp from the lungs of native chicken. Samples used in this study were 12 chicken lungs taken randomly from the poultry slaughterhouse Peunayong Banda Aceh. Isolation of Aspergillus sp were done based on Thompson method (1969). The samples were washed with sterile aquadest containing antibiotics before implanted on Sabouraud’s Dextrose Agar (SDA), then incubated at room temperature for 2-7 day. The plate was observed from Aspergillus sp colony macroscopically and microscopically. Data were analyzed descriptive. The result of examination showed that Aspergillus sp can be isolated 7 out of 12 lungs. It can be concluded that 58,33% of the lungs samples infected with Aspergillus sp and 41,67% were free from Aspergillus sp.

Abstract

Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak daun kelor (Moringa oleifera L.) terhadap pertumbuhan bakteri Staphylococcus  aureus. Ekstrak daun kelor diekstraksi dengan cara maserasi yaitu menggunakan pelarut etanol  96%. Pengujian aktivitas antibakteri dilakukan dengan metode difusi agar  menggunakan kertas cakram yang  telah direndam dalam ekstrak etanol daun kelor dengan konsentrasi 20%, 40%, 60% dan 80% pada media Mueller Hinton Agar (MHA). Parameter yang diukur adalah besarnya diameter zona hambat yang  terbentuk disekitar kertas cakram. Hasil uji aktivitas antibakteri dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa ekstrak etanol  daun kelor memiliki daya hambat terhadap bakteri Staphylococcus  aureus yaitu pada konsentrasi 80% sebesar 14,02 mm (kategori kuat), 60% sebesar 12,03 mm (kategori kuat), 40% sebesar 9,00 mm  (kategori sedang), 20% sebesar 7,98 mm (kategori sedang), kontrol positif 28,63 mm  (kategori sangat kuat) dan kontrol negatif tidak menunjukkan efek antibakteri. Dapat disimpulkan bahwa ekstrak daun kelor memiliki antibakteri terhadap pertumbuhan bakteri Staphylococcus aureus.                The aim of this research is to observe the antibacterial activity of Moringa oleifera L. extract on Staphylococcus aureus. It can be concluded that Moringa oleifera L. extract has an antibacterial effect on Staphylococcus aureus.
