Abstract

The eradication of Helicobacter pylori is important for the treatment ofpatients with infection of gastrointestinal system. However, there were only a few reports on anti-Helicobacter pylori activity of traditional medicines. The anti-H. pylori activity of n-hexane extract, chloroform extract, etanol extract and fraction of n-hexana of Lempuyang wangi (Zingiber aromaticum) was carried out by agar dilution method and chloramphenicol was used as positive control. Prior to assessment, identification of H. pylori by urease, oxidase and catalase tests was done. Only n-hexana extract assayed that inhibit the growth of H. from Malang with Minimal Inhibitory Concentration (MIC) 1.25 mg/ml and no extract or fraction of Lempuyang wangi could inhibited the growth of H. pylori from Mataram.

Abstract

The objective of this research was to evaluate the activity of andrographolide extracted from sambiloto (Andrographis paniculata Nees)   in induction of apoptosis in HeLa cells culture in vitro. This research consisted of two phases, the first phase was isolation and identification of andrographolide. The HeLa cells were incubated in RPMI media and Fetal Bovine Serum 10%. The concentration of   andrographolide  were  0 ;  0.05 ; 0.10 ; 0.25 ; 0.50 ; 1.00 ; 2.50 ; 5 ; 10 ; 25 ; 50 ; 100 ; 250 µg/ml dissolved in DMSO. Each andrographolide concentrations was added into HeLa cells culture and then incubated for 24 hours at 37oC in CO2 incubator. Apoptosis induction was determined by ethidium bromide-acridine orange exclusion and analyzed by flourecent microscope. The result of this research showed the andrografolide extracted from sambiloto (Andrographis paniculata Nees) induced apoptosis activity in HeLa cells culture in vitro with IC50 of 109.90µg/ml. The result of this experiment suggested to study the effect of andrografolide on expression of gene responsible for apoptosis mechanism in cancer cells in vitro. Key words: Andrographis paniculata Nees, andrographolide, apoptosis

Abstract

Plasmodium falciparum, the agent of malignant malaria, is one of mankinds most severe scourges, mainly in the tropic world. Efforts to develop preventive vaccines or remedial drugs are handicapped by the parasites rapid evolution of drug resistance. Here, we presented an advance work on examination of antimalarial component from marine life of Xestospongia sp., the study is based on hexane extraction method. The premier result, we obtained five fractions. Among these five fractions, the fourth has the most potent inhibitory against the growth of P. falciparum 3D7 with an IC50: 7.13 µg/mL. A compiled spectrum analysis, FTIR, 1H-NMR and GC-MS, revealed that the fourth fraction consisted abundantly of two secondary metabolites such as flavonoids and triterpenoids. Finally, our results suggest a plausible structure rooted to the base of ibuprofen.

Abstract

Native people or ethnic societies that live in endemic malaria islands such as in Bangka Island and Belitung Island have used many medicinal plants to cure malaria. Leaves of kesembung (Scaevola taccada (Gaertn Roxb), roots of kebentak (Wikstroemia androsaemofolia Decne), and roots of medang mencena (Dapniphyllum laurinum (Benth) are the examples. This research was aimed to investigate the present of some biochemical compound and evaluate the antimalarial activity of ethanol extract of the plants against Plasmodium falciparum 3D7 in vitro. The IC50 level was determined through visual observation under microscope over 5000 of giemsa-stained erythrocytes then analyzed by probit analysis. Results showed that kebentak root ethanol extract was effective to inhibit P. falciparum 3D7 with level 0.485 g/mL. Furthermore, the IC50 level of kesembung leaves and medang root were 44.352 g/mL and 1486.678 g/mL respectively. Phytochemical test result showed that kebentak leaf ethanol crude extract contained triterpenoid, kesembung root contained phenol and tannins; moreover, medang root contained alkaloid, saponin, and triterpenoid. How to CiteHelmi, H., Afriyansyah, B. & Ekasari, W. Abstract

Background: Antimalarial screening against nine species of the genus Cassia showed that the methanol extract of leaves Cassia spectabilis have the highest activity. Since it will be used as a traditional medicine, hence it is needed further studies of antimalarial activity of these plants by choosing a safer solvent, namely ethanol. Objective: In vitro anti-malarial activity against Plasmodium falciparum was conducted using the method of Trager and Jensen. Methods: The serial solution tested were: 100, 10, 1,  0.1 and 0.01 µg/ mL, while the in vivo test was performed based on Peter’s test (The days suppressive test) that using P. berghei (strain ANKA) infected mice. Results: The results showed that ethanolic extract of C. spectabilis leaves has inhibitory activity against P. falciparum with IC50 value of 12.52 µg/ mL and against P. berghei with ED50 value of 131.5 mg/kg body weight. Conclusions: A further study to see the potential of ethanol extract from C. Spectabilis leaves as anti-malaria is warranted.
