Abstract

A research on the synthesis of sodium pentagamavunonat (Na-PGV-0) salt as an analgesic compound from pentagamavunon-0 (PGV-0) were carried out. The synthesis of Na-PGV-0 carried out by reacting PGV-0 in tetrahydrofuran solvent refluxed with sodium ethoxide in mol comparison (1:2)for two hours. Structure of compound was identified using UV-Visible, IR, 1H-NMR and 13C-NMR Spectroscopy methods.The synthesis yields 107,21% and 100,52% products. Structure identification results indicated that the formed compound was a polar salt where the –OH phenolic groups replaced by sodium ions.Key-words: sodium pentagamavunonat, pentagamavunon-0, analgesics.

Abstract

This research was aimed to know chemical constituents or screening phytochemistry and the antiasthma activity test from water, ethanol and boiled water extracts the leaves of sirih duduk (Piper sarmentosum Roxb. ex Hunter) with in vitro against Pseudomonas aeruginosa. Antibacterial test have been done with agar diffusion method used paper dish. Screening phytochemistry showed the leaves of sirih duduk has chemical constituents alkaloid, steroid, polyphenol and saponin. Antibacterial testshowed ethanol extract has more height activity than boiled water extract. Thus the water extract did not show antibacterial activity until concentration 5,0 mg/mL. It was found that the optimum concentration 5,0 mg/mL the zone diameter of inhibitory of ethanol and boiled water extract were 19,5and 17,0 mm respectively.Key words: Piper sarmentosum, sirih duduk, antiasthma, antibacterial, phytochemistry

Abstract

An acetone extract of Curcuma xanthorrhiza rhizomes and the n-hexane and chloroform fractions obtained from it were tested on eight pathogenic bacteria. The results showed that the acetone extract and the nhexane fraction exhibited significant activities against Bacillus subtilis, Pseudomonas aeruginosa, and Staphylococcus aureus, and weak activities against Shigella dysenteriae and Vibrio cholerae. They were inactive against Escherichia coli, Enterobacter aerogenes and Salmonella thypi, while the chloroform fraction was devoid of activities. NMR analysis disclosed the presence of α-curcumene, xanthorrhizol and an unknown monoterpene in the nhexane fraction. In the chloroform fraction, curcumin was found to be the main compound, together with xanthorrhizol as a minor compound. These results suggest that the antibacterial potency of acetone extract of C. xanthorrhiza is contained in the n-hexane fraction, in which the active constituents are terpenoid compounds. This is the first report of the use of NMR analysis for compound identification contained in an extract or fractions of C. xanthorrhiza.

Abstract

Piper sarmentosum Roxb. ex Hunter (“sirih duduk”) has long been used as a traditional medicine against fungoid diseases, especially at vagina and foot-sole. The investigation of the bioactive compounds of P. sarmentosum roots has not infections been carried yet. This research was then aimed to fractinate and to examine the antifungus activity of the extract of P. sarmentosum roots.The extraction was performed by using organic solvents such as methanol, ethyl acetate, chloroform, benzene, and n-hexane. The bioactivity tests were performed against C. albicans. The active extracts were then fractionated by coloumn chromatography.The result showed that the ethyl acetate fraction of methanol extract of P. sarmentosum roots have an antifungus activity against C. albicans. Phytochemical study showed that the active fractions of the methanol extracts of P. sarmentosum roots contained alkaloid.Key words : Piper sarmentosum, activity test, Candida albicans.

Abstract

Previous  reasearch  reported  that  chloroform  extract  that  was  partitioned from etanol exctracts  of  R. mucronata  stem bark had sitotoxic against Myeloma cells  (Diastuti,  2008).  This  research  was  aimed  to  fractinate  the  chloroform extract  of  R.  mucronata  stem  bark  and  cytotoxicity  test  againts  Myeloma  cells, then  identification  the  cytotoxic  compounds.  The  ethanol  extracts  of R.  mucronata  steam  bark  was  partitioned  with  chloroform.  The  chloroform extracts was fractinated by coloum chromatography with gradient eluent were nhexane, chloroform, etilacetate  and methanol respectivelly. Cytotoxicity test the fraction  of  chloroform  extracts  were  conducted  on  Myeloma  cells  by  MTT methods.  Identification   the  compoud  of  fraction  that  had  a  highest  cytotoxic againts  Myeloma  cells  conducted  using  IR  spectrometer,  and  GCMS.  The  result showed  that  2b  fraction  of   R.  mucronata  stem  bark  had  a  highest  cytotoxic againts  Myeloma  cells,  having  IC50  equal  to  5.122   µg/mL.  Identification  of chemical compounds  showed that 2b fraction  contained two steroid compounds were 4 methyl- colest-24-en-3-ol and 4-methyl-stigmast-22-en-3-ol.Key words : R. Abstract

Gardenia augusta, Merr is one of the family Rubiaceae species used as traditional medicine because it has medicinal properties, such as drug diabetes mellitus. These research have been done on this plant, among others, is the ability of the extract  and  the active fraction ethyl acetate extract of Gardenia augusta, Merr leaves to lowering blood sugar levels of rats.  The purpose of this research was to purification of the active fraction of the ethyl acetate extract and identify the secondary metabolites of Gardenia augusta, Merr leaves as well as test that can be used antidiabetic. Purification was done by column chromatography which was eluted with n-hexane: ethyl acetate (7:1) and obtained four groups of fractions.  The fractions of the TLC to obtain pure isolates F2 fraction with melting point 158-160 ° C. The antidiabetic activity of the isolate F2 fractions were tested into white rats by glucose tolerance methods. Isolate F2 fraction was identified by using UV and  IR spectrophotometer.  The results showed that isolate F2 fraction of ethyl acetate extract of Gardenia augusta, Merr leaves has a hypoglycemic effect of 21.05%. The secondary metabolites test showed a positive result for steroids. Analysis using UV spectrophotometer showed the electronic transition n → л * and transition л → л *. Analysis using IR spectrophotometric showed functional groups OH, C = O, CO, aliphatic C-H, C = C, C-C, CH3  symmetric and C-H outside the field.   Key word: Gardenia augusta, Merr,  diabetes mellitus,  steroids

Antibacterial Activity of Germacrone Sesquiterpene from Curcuma Xanthorrhiza Rhizomes

Abstract

The aim of this research was to isolate and indentify the terpenoid compound from Curcuma xanthorrhiza rhizomes and its antibacterial activity. Isolation was carried out by using vacuum liquid chromatography and centrifugal chromatography. The structure was determined by NMR spectroscopy (1H-NMR, 13C-NMR 1D and 2D), then compare with data from literatures. Antibacterial test was carried out by using  microdillution methods and evaluated against eight bacteria. They are Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Enterobacter aerogenes, Pseudomonas aeruginosa, Salmonella typhi, Shigella dysentriae and Vibrio cholerae. The result showed that the isolate was a white crystal which was indetified as germacron-type sesquiterpene. Germacron have highest activity againts P. aeruginosa, MIC 15.6 µg/mL and MBC 31.2 µg/mL.

Antibacterial Activity of Germacrane Type Sesquiterpenes from Curcuma heyneana Rhizomes

Abstract

The isolation of terpenoids from C. heyneana rhizomes and their antibacterial activity have been conducted. The terpenoids were isolated by using vacuum liquid chromatography and radial chromatography. The structures of the compounds were determined based on spectroscopic data (1H-NMR, 13C-NMR (1D and 2D)). The antibacterial activity was carried out by using microdilution method and evaluated against eight bacteria. Three germacrane type sesquiterpenes have been isolated from C. heyneana rhizhomes and were identified as germacrone, dehydrocurdione, and 1(10),4(5)-diepoxygermacrone. Germacrone showed highest antibacterial activity against P. aeruginosa with MIC values of 15.6 µg/mL and MBC values 31.2 µg/mL. Dehydrocurdione showed highest antibacterial activity against B. subtilis with MIC values of 31.2 µg/mL and MBC values of 31.2 µg/mL. However, 1(10),4(5)-diepoxygermacrone showed a weak antibacterial activity.

Abstract

Piper sarmentosum Roxb. Ex Hunter or “Sirih duduk” has long been used for traditional medicine to cure various diseases, such as fungus infections. The investigation of the bioactive compounds of P. sarmentosum roots has not been carried out. This research was aimed to isolate the bioactive compounds from P. sarmentosum roots. The results showed that methanol extracts of P. sarmentosum roots have an activity on Candida albicans. The separation a bioactive compounds from methanol extracts of P. sarmentosum roots was performed by column chromatography, thin layer chromatography and recrystalizations. The identifications of the bioactive compounds were carried out using ultra violet spectrometry, infrared spectrometry, gas chromatography-mass spectrometry and nuclear magnetic resonance spectrometry. The results indicated that from methanol extracts, an alkaloid compound of piperoylpyrol derivative was 5-hydroxy-5- (3,4-methylenedioxyphenyl)-2-pentenoyl pyrol, could be purely isolated. Examination of bioactivity at concentration 0.10-2.50 mg/ml showed that this compound had an activity on C. albicans.

Abstract

ABSTRACT             IPTEKS-based community service has been done with the Entrepreneurship and Workshop unit of MAN Purbalingga. The objectives of the activities were (1) Transferring of soap making technology to produce natural soap-base; (2) Transferring of cold-process and hot-process methods for the creation of soap-souvenirs with various shapes and colors; and (3) Transferring the packaging knowledge for souvenir soap products. The methods applied include student education, training, and the diffusion of science and technology to produce souvenir soap. The post-test results showed that there was an increased knowledge of the partners on saponification reactions (0% to 50%), soap making principles (0% to 54.2%), as well as the understanding of raw materials, additives, and soap-making equipment. The three main factors that could encourage the partners to make soap were an eagerness on how to make soap (70.8%); business opportunity (62.5%), and wanted to make soap creations (29.1%). This community service activity was expected to provide benefits in the form of partnership ability to produce souvenir soap made from natural ingredients with various shapes and colors. Keywords: cold-process, hot-process, souvenir soap
