Abstract

Introduction: Curcumin and its analog pentagamavuon-0/PGV-0 can inhibit steroidogenesis of luteal cell culture. However, the action site of the curcumin and PGV-0 in luteal cells steroidogenesis is still unknown.Objectives: To analyze the action site of curcumin and PGV-0 in luteal cells steroidogenesis through its influences on the expression of cytochrome P450scc enzyme.Methods: PMSG-induced corpus luteums of Sprague Dawley rat were used as the subjects. Curcumin and PGV-0 with a dosage of 100 µM was given shortly after the stimulation of LH and/or PGF2a. Immunohistochemistry staining was done to assess the expression of cytochrome P450scc.Results: LH stimulation (50 ng/ml) increased the expression of cytochrome P450scc significantly (p<0.05) compared with the control group. In the other hand, PGF2a (0.56 µM) inhibited the expression of cytochrome P450scc significantly. Curcumin (100 µM) and PGV-0 (100 µM) inhibited the expression of cytochrome P450scc in the control group and groups stimulated by LH and/or PGF2a.Conclusion: Curcumin and PGV-0 inhibit the upstream transduction signal of cytochrome P450scc in the steroidogenesis of luteal cell culture. Abstract

Introduction: Pentagamavunon-0 (PGV-0) compound, a curcumin analog, can inhibit steroidogenesis of luteal cell culture by inhibiting the production of progesterone. The action site of PGV- 0 on steroidogenesis of luteal cells is still unknown. The objective of this study is to analyze the effect of PGV-0 on the Extracellular Signal Regulated Kinase (ERK) phosphorylation in steroidogenesis of luteal cell culture.Methods: This study used corpus luteum of rat Sprague Dawley strain that was induced withPregnant Mares Serum Gonadotropine (PMSG). Pentagamavunon-0 was given shortly after the stimulation of LH and or PGF2a with or without forskoline. The phosphorylation of ERK was assessed by IHC method.Results: The result showed that LH stimulation increased ERK phosphorylation significantly.However, PGF2a decreased ERK phosphorylation significantly. Forskoline increased ERKphosphotylation significantly but no significant effect was found with LH stimulation. PGV-0inhibited ERK by either LH or forskoline stimulation.Conclusion: Pentagamavunon-0/PGV-0 can inhibits signal transduction of steroidogenesis lutealcells by acting on the up stream to ERK. Abstract

Trophoblast participates in preventing allorecognition and controlling pathogens that compromise fetal wellbeing. Toll-like receptors recognize conserved sequences on the pathogens surface and trigger effector cell functions. Porphyromonas gingivalis is thought to spread to the umbilical cord and cause fetal growth restriction. Objective: To characterize expression and function of TLR-2 and TLR-4 in trophoblast cells from Porphyromonas gingivalisinfected pregnant rats. Methods: Live Porphyromonas gingivalis were challenged into the maxillary first molar subgingival sulcus of female rats before and/or during pregnancy and sacrified on gestational day (GD) 14 and 20. Porphyromonas gingivalis was detected by API-ZYM system in the maternal blood of the retro-orbital venous plexus and the umbilical cord. TLR-2 and TLR-4 expressions in trophoblast cells was detected by immunohistochemistry. Results: Porphyromonas gingivalis was first detected in the maternal blood and finally spread to the umbilical cord. Syncytiotrophoblast, spongitrophoblast and trophoblastic giant cell in treated groups had significantly higher expression of TLR-2 and TLR-4 than control group (p<0.05). Conclusion: Syncytiotrophoblast, spongitrophoblast and trophoblastic giant cell are able to recognize Porphyromonas gingivalis through TLR-2 and TLR-4 expression. The ligation of TLR-2 and TLR-4 promoted cytokine production and induced trophoblast cell death. These findings strengthen links between periodontal disease and fetal growth restriction.DOI: 10.14693/jdi.v20i2.150

Abstract

Fetal growth restriction remains a major cause of neonatal morbidity and mortality. Porphyromonas gingivaliscan induce placental inflammatory response resulting in fetal growth restriction. Objective: This study aimed to evaluate the potential utility of the pro-inflammatory cytokine TNF-α and anti-inflammatory cytokine IL-10 in rat placental tissues to understand whether these events were causally related. Methods: Female rats were infected with live-Porphyromonas gingivalis at concentration of 2x109 cells/ml into subgingival sulcus area of the maxillary first molar before and/or during pregnancy. They were sacrificed on gestational day (GD)-14 and GD20. The expression of TNF-α and IL-10 in macrophages and trophoblast cells were detected by immunohistochemistry. Results: A higher expression of TNF-α was found in spongiotrophoblast of the Pg-BD group on GD14 (6.30±1.16), and in trophoblastic giant cells of Pg-D group on GD20 (5.50±1.35). Furthermore, a higher expression of IL-10 was found in trophoblastic giant cells of the Pg-BD group on GD14 (4.50±1.51) and in syncytiotrophoblasts of Pg-BD group on GD20 (8.70±2.67). Conclusion: The expression of TNF-α on GD14 and GD20 were accompanied by increased expression of IL-10. The placental pathologic conditions induced by Porphyromonas gingivalis can be inhibited by elevated expression of IL-10 in macrophages and trophoblast cells.DOI: 10.14693/jdi.v20i3.199

Abstract

Endometriosis is a gynecological disease associated with inherited genetic traits. HOXA10 gene whichis expressed in uterine plays an important role in the pathogenesis of endometriosis. The protein affects thedevelopment of pinopodes as a biomarker of endometrial receptivity in endometriosis.The aim of this study isto examine if there is a mutation or polymorphism within HOXA10 gene among patients with endometriosis.Thirty twopatients and 32 healthy women were recruited as subjects of this study. The exon 2 of HOXA10which covers most of coding region was amplifi ed using PCR. The presence of a mutation or polymorphismwas detected by direct seguencing. The distribution of genotype and allele was analyzed using Chi square test with p<0.05 is considered as signifi cantly different. A novel heterozygous variant within exon 2 of HOXA10 which substitute an adenine into thymine was detected at base position 55. This missense alteration changed amino acid serine to cystein (Ser19Cys). Interestingly, this variant was detected in 12 endometriosis cases (38%) but none in control. Patients carry HOXA10 Ser19Cys variant were associated with dismenorea and more frequent in stage I endometriosis. The role of this variant in the function of HOXA10 protein and frequency among Indonesians need to be clarifi ed. We found a novel heterozygous HOXA10 gene variant, Ser19Cys.The genotype frequency is 38% among endometriosis patients but none in control. Abstract

Low birth weight (LBW) babies remains a major problem world wide as it causes a high rate ofneonatal morbidity and mortality. One effort to reduce the prevalence of LBW babies is byproviding iron supplementation to the pregnant women. The aim of this study was to examinethe relationship between iron supplementation program and prevalence of LBW babies in Palu,Central Sulawesi. This was an observational study with case control design. Subjects were allLBW babies from mothers that had accepted iron supplementation during pregnancy. Samplesize in this study was 87 for case and 87 for control. Bivariate analysis showed that ironsupplementation that not comply the program had a significant correlation with prevalence ofLBW babies (p=0.01). Other significant factors include abnormal hemoglobin level (p=0.01),body mass index (BMI) (p=0.02), educational level (p=0.02), multiparity (p=0.03), andgestational age (prematurity) (p=0.03). Multivariate analysis revealed that the strongest riskfactor for the occurrence of LBW babies was iron supplementation that not comply the program(OR= 3.82; 95% CI: 1.77- 8.22). Other risk factors were hemoglobin level (OR= 3.45; 95% CI:1.59-7.49), BMI (OR= 2.27; 95% CI: 1.05-4.91), gestational age (OR= 3.11; 95% CI: 1.45-6.67), multiparity (OR= 2.98, 95% CI: 1.36-6.51), and educational level (OR= 2.38, 95% CI:1.12-5.03). Based on the analysis, the strongest risk factors that affected the prevalence of LBWwas iron supplementation, abnormal hemoglobin level, gestational age, multiparity and educationlevel. In conclusion, iron supplementation during pregnancy that not comply with the programwas the strongest risk factor of LBW babies. The prevalence of LBW babies can be reduced bycontrolling of iron supplementation, hemoglobin level, BMI, gestational age, parity and education.

Abstract

Kurkumin merupakan bahan alam yangmempunyai berbagai aktivitas biologis, dintaranya sebagai antifertilitas. Belumada penelitian tentang efek kurkumin khususnya yang berkaitan dengan stimulasiLH di endometrium. Tujuan penelitian ini untuk mengetahui ekspresi enzim COX-2pada sel epitel luminal dan epitel glandular uterus, akibat pemberiankurkumin  setelah stimulasi LH.          Dilakukanpenelitian dengan rancangan eksperimental post-testcontrol randomized group design terhadap 30 ekor tikus Rattus norvegicus Strain SpragueDawley betina, berusia 28 hari, dibagi menjadi 6 kelompok, diinduksiovulasi dengan PMSG, kemudian diberi perlakuan sebagai berikut : (K1) aquades,(K2) LH 10 IU + aquades, (K3) CMC 1 ml, (K4) LH 10 IU + CMC 1ml , (K5) kurkumin 100mg/kg BB + CMC 1 ml dan K6)LH 10 IU + kurkumin 100 mg/kg BB + CMC 1 ml. Kurkumin diberikan secara oralselama 4 hari dan  LH diberikan denganinjeksi intramuscular satu kali pada hari pertama ovulasi. Hari ke 5 tikus dikorbankanuntuk diambil uterusnya dan dibuat preparat IHC untuk pengukuran ekspresi COX-2Analisis statistik menggunakan Anovasatu jalan, dilanjutkan dengan DMRT (Duncan’sMultiple Range Test). Untuk mengetahui perlakuan yang berpengaruhmenggunakan multiple comparation analysis.Analisis statistik ekspresi COX-2 menunjukkan peningkatan yang bermakna antara kelompok K5 dengan kelompokK6 (p<0,05). Penurunan yang bermakna (p<0,05) ditunjukkan  antara kelompok K4 dengan kelompok K6. Hasilanalisis multiple comparationmenunjukkan yang berpengaruh terhadap ekspresi COX-2 di epitel luminal adalahLH, kurkumin, kurkumin + LH dan di epitel glandular,  LH dan kurkumin. Dari hasil tersebut  disimpulkan bahwa kurkumin 100 mg/kg BBsetelah stimulasi LH 10 IU menurunkan ekspresi COX-2 baik di epitel luminalmaupun epitel glandular uterus  Rattus norvegicus Strain Sprague Dawley.melalui salah satu jalurdari jalur rangsangan LH  Kata kunci : Siklooksigenase, Kurkumin, LuteinizingHormon

Abstract

Perilaku seksual pranikah yang tinggi pada remaja disebabkan oleh faktor personal seperti pengetahuan kesehatan seksual, Infeksi Menular Seksual (IMS) dan HIV / AIDS, sikap terhadap seksualitas, harga diri dan efikasi diri. Penelitian ini bertujuan untuk mengetahui pengaruh faktor personal seperti pengetahuan tentang kesehatan seksual, IMS dan HIV / AIDS, sikap, harga diri dan efikasi diri terhadap perilaku seksual pranikah pada remaja di Kota Kendari yang diukur melalui kuesioner dan wawancara mendalam. Desain penelitian adalah studi potong lintang dengan jumlah sampel 200 remaja yang berasal dari empat sekolah negeri di Kota Kendari mulai dari Agustus sampai dengan Oktober 2014. Analisis regresi logistik menunjukan bahwa remaja yang memiliki pengetahuan yang rendah tentang kesehatan seksual, IMS dan HIV / AIDS berisiko sebesar 4,28 kali, sikap permisif terhadap seksualitas berisiko 5 kali, harga diri rendah berisiko sebesar 3,3 kali dan efikasi diri rendah sebesar 2,5 kali untuk melakukan perilaku seksual pranikah berisiko. Analisis kualitatif menunjukan variabel sikap sebagai faktor yang memberikan risiko terbesar di dalam berperilaku seksual pranikah yang berisiko pada remaja. Influence of Personal Factor to Premarital Sexual Behavior among TeenagersHigh premarital sexual behavior among teenagers are caused by personal factors, such as health sexual knowledge, Sexually Transmitted Infections (STIs) and HIV / AIDS, attitudes towards sexuality, self-esteem and self-efficacy. This study aimed to find out the influence of personal factor to premarital sexual behavior among teenagers in the Kendari City as assessed through questionnaires and in-depth interviews.The study design was cross-sectional study with a sample of 200 adolescents from four public schools in Kendari City from August to October 2015. Logistic regression analysis showed teenagers having lack of knowledge of sexual health, STIs and HIV / AIDS had 4.28 times risk having permissive attitude toward sexuality had 5 times risk, having low self-esteem had 3.3 times risk and having low self-efficacy had 2.5 times to perform premarital sexual behavior. Qualitative analysis showed that attitude variable was the factor giving the biggest risk in risky premarital sexual behavior among teenagers.

Abstract

Background: Porphyromonas gingivalis as a major etiology of periodontal disease can produce virulence factor, lipopolysaccharide/LPS, which is expected to play a role in the intrauterine fetal growth. Trophoblast at the maternal-fetal interface actively participates in response to infection through the expression of a family of natural immune receptors, toll-like receptor (TLR). Purpose: the aims of study were to identify endotoxin concentration in maternal blood serum of Porphyromonas gingivalis-infected pregnant rats, to characterize the TLR-4 expression in trophoblast cells, and to determine its effect on fetal growth. Methods: Female rats were infected with live-Porphyromonas gingivalis at concentration of 2 x 109 cells/ml into subgingival sulcus area of the maxillary first molar before and/or during pregnancy. They were sacrified on 14th and 20th gestational day. Fetuses were evaluated for weight and length. Endotoxin was detected by limulus amebocyte lysate assay in the maternal blood serum.
