Abstract

The addition of katuk leaf meal (saurupus androgynus l.merr) in feeds on body weight gain and scrotal circumference  male ettawa grade goatsABSTRACT. This research was aimed to study the effect of addition of katuk leaf meal (Sauropus androgynus L. Merr)  in feed on body weight gain and scrotal circumference of male Ettawa grade goat. The research lasted for eight months, starting Agustus 2012 to Maret 2013, with feeding trial for two months. Twenty male Ettawa grade goats of 18 to 24 months old, were divided into 4 treatment groups. Treatment groups were added with 0% Katuk leaf meal (R0);  5% (R1);  10%  (R2) and 15%  (R3), with 5  goats in each replication. Treatment diets were feed in the morning and afternoon for 60 days. Weighing body weight and measuring scrotal circumference was done every week. Data were analyzed using analysis of variance (ANOVA) and if there was significantly different, the data were further tested using Duncan multiple range test. The result shows that the body weight gain of the treatment were, R0 (2,39 ± 0,43 kg); R1 (2,84 ± 0,36 kg); R2 (2,85 ± 0,20 kg) and R3 (3,42 ± 0,78 kg) respectively. Scrotal circumference were, R0 (0,74 ± 0,10 cm); R1 (1,24 ± 0,26 cm); R2 (1,26 ± 0,40 cm) dan R3 (1,32 ± 0,32 cm) respectively. The result of this research indicated that the addition of katuk leaf meal in feed had significant effects (P<0,05) on body weight gain and scrotal circumference of Ettawa grade goat.

Abstract

(The ratio of birth sex of ettawa crossbreed goat by artificial insemination using spermatozoa swim up)ABSTRACT. The aim of this research was to determine the ratio of birth sex of ettawa crossbreed goat by artificial insemination using spermatozoa swim up. This research was at Reproduction Laboratory Faculty of Veterinary medicine Syiah Kuala university and Livestock Group SMD, Bener Meriah Regency. Fifteen female goats with age 8-24 months where used in this study and divided into 3 treatment groups (P0, P1, P2) and 5 replications of each. The first group (P0) inseminated with sperm without swim up; P1 and P2 inseminated with sperm which swim up for 5 minutes (P1) and 10 minutes (P2). The result of conception rates observation and sex ratio of children born tabulated into tables. The data was analyzed by using analysis of variance (ANOVA) one way pattern followed by the multiple test Duncan. The results showed that the conception rate and birth sex ratio of male born had significantly different (P<0,05). The segregation of sperm which swim up increased the conception rate and birth sex ratio of male born.

Abstract

Effect of Supplementation of ENERVON-C and SANTA-e in Feed on Sperm Motility of Local CocksABSTRACT. This study aimed to observe the effect of supplementation of ENERVON-C and SANTA-e in feed on sperm motility of local cocks. The research was conducted at Reproduction Laboratory, Faculty of Veterinary Medicine, Syiah Kuala University, Banda Aceh. The research used 12 local cocks divided into 4 treatment groups : P0 (feed of 521), P1 (feed of 521 + 200 mg ENERVON-C/kg of feed), P2 (feed of 521 + 200 mg SANTA-e/kg feed) and P3 (feed of 521 + 100 mg ENERVON-C/kg of feed and 100 mg SANTA-e/kg of feed). During the research, all cocks were given 100 g of 521 feed/day. Feed with ENERVON-C and SANTA-e was given for 2 weeks before semen collections. Drink was given ad libitum. Data were analysed with analysis of variance and if there are effects among the treatments, analysis will be continued with Duncan Test. Results shows that supplementation of ENERVON-C or SANTA-e and their combination in feed can increase the percentage of sperm motility. There are significant differences (p<0,05) between control and treatment groups. To sum up, supplementation of ENERVON-C or SANTA-e in feed can improve the quality of sperm (motility) of local cocks. The sperm quality is better after giving of ENERVON-C and SANTA-e together than single supplementation.

Abstract

ABSTRAK. Penelitian ini merupakan penelitian observasional yang dengan  rancangan acak lengkap (RAL) dengan 3 kelompok perlakuan jenis semen beku sapi aceh (S1), semen beku sapi bali (S2) dan semen beku sapi simmental (S3).  Masing-masing kelompok diulang sebanyak 10 kali. Data kualitas spermatozoa dan angka kebuntingan yang diperoleh dianalisis dengan analisis of variance (ANOVA) dan dilanjutkan dengan uji Duncan dan hubungan kualitas spermatozoa semen beku dengan tingkat kebuntingan diuji dengan regresi berganda.  Persentase motilitas, spermatozoas hidup dan TAU sapi aceh dan sapi bali tidak memperlihatkan perbedaan yang nyata (P>0,05), namun keduanya berbeda secara nyata (P<0,05) dengan sapi simmental. Hasil uji regresi menunjukan ada hubungan yang nyata (P<0,05) antara kualitas spermatozoa (motilitas, spermatozoa hidup dan TAU) semen beku dengan tingkat kebuntingan, dengan persamaan regresi adalah  Y=  - 2,586 + 0,017 X1 - 0,001 X2 + 0,043 X3, nilai koefisien korelasi r = 0,695 dan nilai koefisien determinasi (r2) sebesar 0,483. Keutuhan TAU memiliki hubungan yang lebih kuat (r = 0,695) dibanding dengan motilitas spermatozoa (r = 0,505) dan spermatozoa hidup (r = 0,195) terhadap angka kebuntingan. Disimpulkan kualitas semen beku sapi unggul berpengaruh terhadap tingkat kebuntingan setelah inseminasi pada induk aseptor sapi aceh betina.(Comparison of frozen semen quality of aceh cattle,  bali cattle,  brahman cattle and simmental cattle and the relationship with the success level of artificial insemination in female aceh cattle)ABSTRACT. This study is an observational study with a completely randomized design (CRD) with 3 groups of frozen semen aceh cattle (S1), bali cattle frozen semen (S2) and simmental cattle frozen semen (S3). Each treatments groups was repeated 10 times. Spermatozoa quality data (motility, live spermatozoa, and intact acrosome) of frozen semen and pregnancy rates obtained were analyzed by analysis of variance (ANOVA) and continued with Duncant test. Percentage motility, live sperm and intact acrosome of aceh cattle, bali cattle and simmental cattle showed no significant differences (P> 0.05), but they differ significantly (P <0.05) compared with brahman cattle. Results of regression analysis showed no significant relationship (P <0.05) between spermatozoa quality (motility, live sperm and intact acrosome) with a pregnancy rate of frozen semen, the regression equation is Y = - 2,586 + 0,017 X1 - 0,001 X2 + 0.043 X3, Correlation coefficient value r = 0,695 and coefficient of determination (r2) equal to 0,483. intact acrosome of spermatozoa have a stronger relationship (r = 0.695) compared with sperm motility (r = 0.505) and live spermatozoa (r = 0.195) on the pregnancy rate. It was concluded that the quality of frozen semen had an effect on the pregnancy rate after insemination on female aceh cow.

Abstract

Effect of vitamin E addition (alfa-tokoferol) into sperm washing medium by centrifuge on the quality of Brahman cattle spermatozoa ABSTRACT. The aims of study to determine the effectiveness of the addition of vitamin E in the washing medium by centrifugation on sperm quality Brahman cattle. frozen semen of Brahman cattle, divided into 4 treatment groups addition of vitamin E in the washing medium: 0.0gr/100 ml medium (K0), 0.1gr /100 ml medium (K1); 0.2gr/100 ml medium (K2) and 0.3 g / 100 ml medium (K4), each group was repeated 5 times. Examination of motility, viability and integrity of sperm membrane performed according to WHO standards. The data obtained were analyzed with one-way ANOVA and Duncan test. The average percentage of motility, viability and membrane integrity of spermatozoa in the addition of vitamin E were significantly different (P <0.05) compared to the control. Percentage of motility, viability and membrane integrity of spermatozoa in the group K2 significantly higher (P <0.05) compared with the group K3: K1 and K0. Percentage of motility, viability and sperm capacitation and sperm live on group K3 significantly higher (P <0.05) compared with the K1 and K0. While the percentage motility of spermatozoa in the group K1 higher were not significant (P> 0.05) compared with the group K0. The addition of vitamin E in the medium on the process of washing spermatozoa Brahman cattle. The addition of vitamin E 0.2gr/100ml better than vitamin E 0.1gr/100ml and 0.3gr/100ml in maintaining the percentage of motility and live spermatozoa Brahman cattle.

Abstract

ABSTRAK. Penelitian bertujuan mengetahui pengaruh konsentrasi pengencer AdroMed® terhadap kualitas semen sapi aceh setelah proses pembekuan. Penelitian ini menggunakan rancangan acak lengkap (RAL) dengan 4 kelompok perlakuan pengenceran AndroMed®. Kelompok A1: AndroMed® 10% (5 ml AndroMed® + 45 ml Aquadestilata), A2: AndroMed® 15% (7,5 ml AndroMed® + 42,5 ml Aquadestilata), A3: AndroMed® 20%(10 ml AndroMed® + 40 ml Aquadestilata) dan A4: AndroMed® 25% (12,5 ml AndroMed® + 37,5 ml Aquadestilata). Masing-masing kelompok diulangi sebanyak 6 kali. Variabel yang di amati pada penelitian ini adalah Motilitas spermatozoa yang diamati tiap kelompok setelah pembekuan yang selanjutnya dianalisis dengan analysis of variance (ANOVA) pola satu arah yang dilanjutkan dengan uji Duncant. Rata-rata persentase motilitas spermatozoa pada kelompok A1, A2, A3 dan A4 secara berturut-turut adalah 30,69±3,68%, 39,79±2,44%, 45,22±3,17% dan 42,42±4,24%. Hasil penelitian menunjukkan bahwa konsentrasi pengencer AndroMed® berpengaruh secara nyata (P<0,05) terhadap persentase motilitas spermatozoa sapi aceh. Persentase motilitas spermatozoa pada kelompok A1 tidak berbeda secara nyata dengan A2, dan keduanya berbeda secara nyata (P<0,05) dibandingkan dengan kelompok A3dan A4. Konsentrasi AndroMed® 20% lebih baik dari pada 10%, 15% dan 25% dalam mempertahankan motilitas spermatozoa sapi aceh setelah pembekuan. (Analysis of Aceh Cattle spermatozoa motility after freezing using Andromed® with different concentration)ABSTRACT. The study aims to determine the effect of diluent concentration of AndroMed® against the motility of aceh cattle spermatozoa following freezing process. This study uses a completely randomized design (CRD) with 4 treatment groups. Group 1 used diluent 15%; AndroMed®, Grroup 2 diluent 15%; Group 3 with AndroMed® 20%; and, Group 4 with AndroMed® 25%. Each group was repeated 6 times. Motility of spermatozoa assessed which each group observed after freezing.The motility data obtained were analyzed by one way analysis of variance (ANOVA) followed by Duncant test. The average percentage of motility after freezing were found in the group A1, A2, A3 and A4 respectively are 30,69 ± 3,68%, 39,79± 2,44%, 45,22± 3,17%, and 42,42± 4,24%. Statistical analysis showed that the concentration of diluent AndroMed® significantly affected (P<0.05) the motility of aceh cattle spermatozoa. There is no significant difference of sperm motility percentage, between A2 with A1 treatment but  both are significantly different (P<0.05) compared to group A3. AndroMed® concentration affects the motility of aceh cattle spermatozoa after freezing. The treatment of 20% AndroMed® concentration were improved the quality of aceh cattle spermatozoa following freezing compared to those with AndroMed® 10%, 15%, and 25%.

Abstract

Separation of spermatozoa with x and y chromosome at boer goat and its application by artificial insemination for kid sex purposeABSTRACT. The purposes of this experiment are to investigate the separation of X and Y spermatozoa by measuring the spermatozoa quality, sex ratio between X and Y, capacity of fertility indicated by conception rate and sex ratio of goat boar kids. Samples, used in this experiment, are fresh semen from Boer goat with high quality consists of 4 group treatments with 6 replications 1) group of spermatozoa without separation (control), 2) group of spermatozoa separated by percoll gradient density centrifugation 3 levels (P1), 5 levels (P2) and swine up (P3). The observed parameters are spermatozoa quality, X and Y spermatozoa ratio, fertility’s capacity and sex ratio on the birth. Quality examination of spermatozoa and identify X and Y spermatozoa is based on the standard method of WHO. The conception rate was based on the ratio of pregnant goat after the first insemination. Data of spermatozoa quality and spermatozoa ratio were analyzed by using analisis of variance (ANOVA) and further analysis by LSD if there were differences between treatments. The results of this experiment showed that spermatozoa quality Boer goat significantly reduced (p<0,05) after separation with percoll gradient density centrifugation  and swim up. Percentage spermatozoa X after percoll gradient density centrifugation was significantly higher (P<0,05) compared to control and swim up. Meanwhile, the Y spermatozoa population was significantly higher (P<0,05) after swim up treatment compared to percoll gradient density centrifugation and control. The percentage of sex ratio (male: female) after insemination from percoll gradient density centrifugation produced more female than male. On the other hand, insemination from swim up produced more male than female. Sex ratio produced from separation of percoll gradient density centrifugation, swim up was difference from control semen (P<0,05). From this experiment, it was concluded that spermatozoa separation by percoll gradient density centrifugation and swim up can be used as one of the methods to separate   X and Y spermatozoa and further can be applied to get preferred sex animals.

Abstract

ABSTRAK. Penelitian ini bertujuan untuk menentukan karakteristik dan komposisi plasma semen sapi aceh. Sampel semen dikoleksi dari 2 ekor sapi aceh jantan sehat berumur 3,0-3,5 tahun menggunakan vagina buatan. Koleksi semen dilakukan satu kali dalam seminggu selama enam minggu. Evaluasi karakteristik semen meliputi volume, warna, konsistensi, pH, konsentrasi spermatozoa, motilitas, viabilitas, abnormalitas spermatozoa dan komposisi kimia plasma semen. Data kualitas dan komposisi kimia plasma semen yang diperoleh dianalisa secara diskriptif. Hasil penelitian menunjukkan semen segar sapi aceh memiliki volume 3,82 ± 0,47 ml, warna krem keputihan, konsistensi kental, pH 6,84 ± 0,17, konsentrasi 1194 ± 4,80 × 106sp/ml, persentase motilitas 77,28 ± 3,17 % dan abnormalitas 5,98 ± 1,77%. Plasma seminalis semen segar sapi aceh mengandung protein total sebanyak 1,51 ± 0,21 g/100ml, Kalium 75,80 ± 22,63 mg/100 ml dan Magnesium 32,30 ± 17,64 mg/100 ml lebih tinggi, tetapi Natrium 170,00 ± 81,60 mg/100 ml; kalsium 40,00 ± 12,76 mg/100 ml, Fosfor 16,70 ± 6,17 mg/100 ml, dan Mangan 7,08 ± 2,48 mg/100 ml lebih rendah dibandingkan dengan sapi potong. Secara umum kuantitas dan komposisi plasma seminalis semen sapi aceh yang dihasilkan cukup baik, dan memenuhi syarat untuk diproses sebagai semen beku. (The characteristics of semen and chemical composition of the seminal plasma of aceh cattle maintaned in BIBD Saree Aceh Besar) ABSTRACT. This study aim were to determine characteristics and composition of semen plasma Aceh cattle. Semen samples were collected from two healthy Aceh bulls that aged from 3.0 to 3.5 year. Samples were yielded by using an artificial vagina. Samples collecting were conducted once a week for six consecutive weeks. The evaluation of semen characteristics includes volume, color, consistency, pH, sperm concentration, motility, viability, abnormal spermatozoa and seminal plasma chemical composition. Data of semen quality and chemical composition of seminal plasma were analyzed descriptively. The evaluation showed that Aceh Cattle semen had 3.82 ± 0.47 ml volume, beige whitish color, thick consistency, 6.84 ± 0.17 pH, 1194 ± 4.80×106 sp/ml sperm concentration, 77.28 ± 3.17% motility and 5.98 ± 1.77% abnormal spermatozoa. Plasma seminalis fresh semen of Aceh Cattle contained 1,51 ± 0,21 g/100ml total protein; 75,80 ± 22,63 mg/100ml potassium and 32,30 ± 17,64 mg/100ml magnesium higher than the heifer, but 170,00 ± 81,60 mg/100ml sodium; 40,00 ± 12,76 mg/100ml calcium, 16,70 ± 6,17 mg/100ml fosfor, and 7,08 ± 2,48 mg/100ml mangan lower. It was concluded that quality and composition of seminal plasma Aceh cattle semen could be made as frozen semen.

Abstract

ABSTRAK. Penelitian ini bertujuan mempelajari pengaruh pemberian kombinasi pakan fermentasi dengan multi enzim pencernaan dan vitamin E dalam ransum komersial terhadap peningkatan kualitas semen ayam arab. Penelitian ini menggunakan rancangan acak kelompok (RAK) yang terdiri dari 4 kelompok perlakuan dengan 5 kali ulangan. 1) kontrol (P0) ayam diberikan ransum komersial 100 %, 2) perlakuan 1 (P1) ayam diberi 89,83% ransum komersial + 10% pakan fermentasi + 0,15% multi enzim + 0,02% vitamin E, (3) perlakuan 2 (P2) ayam diberi 79,83% ransum komersial + 20% pakan fermentasi + 0,15% multi enzim + 0,02% vitamin E, dan (4) perlakuan 3 (P3) ayam diberi 69,83% ransum komersial + 30% pakan fermentasi + 0,15% multi enzim + 0,02% vitamin E. Koleksi semen dilakukan pada hari ke 36 dengan metode masase. Data kualitas semen yang diperoleh dianalisis dengan menggunakan analysis of variance (ANOVA) satu arah yang dilanjutkan dengan uji Duncan. Hasil penelitian menunjukkan pemberian kombinasi pakan fermentasi dengan multi enzim dan vitamin E dalam ransum komersial berpengaruh secara nyata (P<0,05) terhadap peningkatan volume, pH semen, konsentrasi spermatozoa, motilitas, spermatozoa hidup, dan penurunan abnormalitas ayam arab, namun tidak berbeda nyata (P>0,05) terhadap warna, konsistensi dan gerakan massa spermatozoa. Dapat disimpulkan pemberian kombinasi pakan fermentasi dengan multi enzim dan vitamin E dalam ransum komersial dapat meningkatkan kualitas semen ayam arab. (Effect addition of fermented feed with the multi-enzyme digestion and vitamin E in commercial feed on improvement the quality of semen arab chicken)ABSTRACT. The study aimed to determine the effect of fermented feed combination with the multi enzymes digestion and vitamin E addition on the quality of semen arab’s chicken. This research used a randomized block design (RAK) consisting four treatments with five replications : P0: 100% Commercial feed (524), P1: 89.83% commercial feed + 10% fermented feed + 0.02% viamint E + 0.15% multi enzyme, P2: 79.83% commercial feed + 20% fermented feed + 0.02% vitamin E + 0.15% multi enzyme, P3: 69.83% commercial feed + 30% fermented feed + 0.02% vitamin E + 0.05% multi enzyme. Semen sample was collected at 36th  day using a massage method. Data obtained were analyzed using analysis of variance (ANOVA) and continued by Duncan test. The results showed that addition of combination of fermented feed with digestive multi enzymes and vitamin E has significantly effect (P<0,05) on increases of the volume, pH, spermatozoa concentration, percentage of spermatozoa motility, spermatozoa life, and decreased the percentage of spermatozoa abnormalities. This research can be concluded that the addition of  fermented feed combination with digestive multi enzymes and vitamin E able to improve the quality of arab chicken semen.

Abstract

ABSTRAK. Penelitian ini bertujuan mengetahui konsentrasi hormon testosteron kerbau Simeulue dan korelasinya dengan tingkat umur dan lingkar skrotum. Sebanyak 15 ekor kerbau Simeulue jantan dibagi dalam tiga kelompok yaitu umur 2,1-3,0 tahun; 3,1-4,0 tahun, dan 4,1-5,0 tahun. Parameter yang diamati terdiri dari lingkar skrotum dan konsentrasi hormon testosteron. Data yang diperoleh dianalisis dengan analisis varian satu arah dan selanjutnya diuji dengan uji berganda Duncan. Untuk mengetahui hubungan antara umur, lingkar skrotum dengan konsentrasi hormon testosteron digunakan analisis regresi sederhana. Hasil analisis pada lingkar skrotum dan konsentrasi hormon testosteron memperlihatkan perbedaan yang nyata (P<0,05). Lingkar skrotum dan kadar testosteron serum umur 2,1- 3,0 tahun berbeda secara nyata (P<0,05) dengan umur 3,1- 4,0 tahun dan 4,1 - 5,0 tahun. Terdapat hubungan yang nyata (P<0,05) antara konsentrasi hormon testosteron dengan umur dan lingkar skrotum, dengan persamaan regresi Y = -4,925-0,436 X1 + 0,697 X2. dengan nilai koefesien korelasi (r) sebesar 0,675 dan koefisien determinasi (r2) sebesar 0,455. Hasil ini menunjukkan bahwa lingkar skrotum (X2) berpengaruh sebesar 67,10 % terhadap konsentrasi hormon testosteron, lebih tinggi dibanding umur (X1) yang berpengaruh sebesar 46,60%. Kesimpulan, umur berpengaruh terhadap lingkar skrotum dan konsentrasi hormon testosteron kerbau Simeulue. Kerbau Simeulue berumur 4,1 - 5,0 tahun memiliki korelasi yang lebih kuat dari pada kerbau Simeulue umur 2,1 - 3,0 dan 3,1 - 4,0 tahun terhadap lingkar skrotum dan konsentrasi testosteron.  (Testosterone hormone concentration of Simeulue buffalo and its correlation with age level and the scrotum circumference) ABSTRACT . The objective of this study was to determine the testosterone hormone concentration of Simeulue buffalo and its correlation with age level and the scrotum circumference. Fifteen male Simeulue buffalo were divided into three groups: 2.1 - 3.0 years; 3.1-4.0 and 4.1-5.0 years where each group consists of five buffaloes. The parameter observed consisted of age, scrotal circumference and testosterone hormone concentration. The data obtained were analyzed by one-way analysis of variance. The difference is tested with Duncan multiple tests. Meanwhile, simple regression analysis was used to testing the relationship between age and scrotal circumference with testosterone hormone concentration. The results showed that the scrotal circumferences and testosterone hormone concentration have significant differences (P <0.05). Scrotal circumference and testosterone level of 2.1-3.0 years were significantly different (P <0.05) with age 3.1- 4.0 years and 4.1 - 5.0 years. Additionally, there is a relationship (P<0,05) between testosterone hormone concentration with age and scrotal circumference, with correlation coefficient value (r) = 0.675 and determination coefficient(r2) = 0,455, with regression equality Y =- 4,925 - 0,436 X1 + 0,697 X2. In conclusion, age affected the scrotal circumference and testosterone hormone concentration in the Simeulue buffalo. 4.1 - 5.0 years Simeulue buffalo has stronger correlation than 2.1 - 3.0 and 3.1 - 4.0 years of Simeulue Buffalo at scrotal circumference and testosterone concentration.
