Abstract

The purpose of this research was find out the effect of age and ejaculation frequency on quality of Aceh bull spermatozoa.. This research was conducted at Frozen Semen Prossesing Laboratorium, Saree, Aceh Besar from Januari to April 2013. Six healthy aceh bulls were divided into two groups of age, those were 3.0-4.0 years old; and 4.0-5.0 years old. Three numbers of bull at each group were assumed as the replication. Semen was collected once a week using artificial vagina and collected until third semen ejaculation. After collecting, volume, pH, color, consistency, concentration, motility, mortality, and abnormality of spermatozoa were immediately measured. This data was analyzed by analysis of variance (ANOVA) two-way pattern, followed by Duncant test. The result showed that the evaluation of bull semen at 3.0-4.0 years old and 4.0-5.0 years old affect significantly (P<0.05) on semen volume and spermatozoa concentration and had no contribute significantly on motility, mortality and abnormality of spermatozoa,while ejaculation frequency effect significantly (P<0.05) on semen volume, concentration, and abnormality spermatozoa, but had no effect significantly on motility and mortality (P>0.05). It is concluded that aged and ejaculation frequency effect significantly on quality of aceh bull fresh spermatozoa.Key words: ejaculation frequency, quality of spermatozoa, aceh bull, age

Abstract

This research aimed to determine the effect of spermatozoa X and Y separation using swim-up method on the quality of etawah crossbreed goat’s spermatozoa. This research used 6 male etawah crossbreed goats aged between 18-24 months, which were collected the semen once a week using electroejaculator. Immediately after semen collection, the quality of semen was examined, and then grouped into three treatment groups. Group 1 was control group (P0) refers to the semen was not separated by swim-up method. Group 2 (P1) was the semen separated by the swim-up method for 5 minutes. Group 3 (P2) was the semen separated by swim-up method for 10 minutes. Each treatment was repeated six times. Parameters for semen quality examined were the percentage of spermatozoa motility, live spermatozoa, and abnormal spermatozoa. Data were analyzed by analysis of variance (ANOVA) one way pattern, followed by Duncans multiple test. The results showed that the mean ± SD percentage of sperm motility on each treatment groups (P0; P1; P2) was 81.33±3.44, 89.67±3.21, and 90.00±3.10 %, respectively. The percentage of live spermatozoa on each group was 86.50 ± 2.07, 92.33 ± 2.08, and 91.83 ± 1.72%, respectively. The percentage of abnormal spermatozoa on each group was 13.00±2.53, 7.33±2.52, and 7.67±2.16%, respectively. Results of this research showed that the percentages of motility, live spermatozoa, and abnormal spermatozoa after separation by swim-up method were significantly different (P<0.05) compared to without separation. It can be concluded that the separation of spermatozoa using swim-up method can improve significantly (P<0.05) the percentage of live and motility of spermatozoa and decrease the percentage of abnormal spermatozoa of etawah crossbreed goats.Key words: Etawah crossbreed, spermatozoa quality, swim-up method

Abstract

The aim of this research was to find out anti-cholesterol activity intomato extract (Lycopersicon esculentum) on blood lipid profile and its potential application in the prevention of atherosclerosis in rats fed with high cholesterol diet. This research employed a complete randomized design with unidirectional pattern by using white rats (Rattus norvegicus) divided into five groups. Negative control group was fed with standard diet, while positive control groups were fed with high cholesterol (hypercholesterolemia) diet, group K were fedwith tomato extract at a dose of 10, 20, and 40 mg/kg together with high cholesterol diet. The result of this researchshowed that high cholesterol diet fed decreased simultaneously with tomato extract at a dose of 10, 20, and 40 mg /kg of rats after 60 days of treatment. Tomato extract was able prevent the formation of plaque in the coronary arteries of rats fed with hypercholesterolemia diet. Effective dose oftomatoextractused in this study was 20-40mg/kg.

Abstract

The addition of katuk leaf meal (saurupus androgynus l.merr) in feeds on body weight gain and scrotal circumference  male ettawa grade goatsABSTRACT. This research was aimed to study the effect of addition of katuk leaf meal (Sauropus androgynus L. Merr)  in feed on body weight gain and scrotal circumference of male Ettawa grade goat. The research lasted for eight months, starting Agustus 2012 to Maret 2013, with feeding trial for two months. Twenty male Ettawa grade goats of 18 to 24 months old, were divided into 4 treatment groups. Treatment groups were added with 0% Katuk leaf meal (R0);  5% (R1);  10%  (R2) and 15%  (R3), with 5  goats in each replication. Treatment diets were feed in the morning and afternoon for 60 days. Weighing body weight and measuring scrotal circumference was done every week. Data were analyzed using analysis of variance (ANOVA) and if there was significantly different, the data were further tested using Duncan multiple range test. The result shows that the body weight gain of the treatment were, R0 (2,39 ± 0,43 kg); R1 (2,84 ± 0,36 kg); R2 (2,85 ± 0,20 kg) and R3 (3,42 ± 0,78 kg) respectively. Scrotal circumference were, R0 (0,74 ± 0,10 cm); R1 (1,24 ± 0,26 cm); R2 (1,26 ± 0,40 cm) dan R3 (1,32 ± 0,32 cm) respectively. The result of this research indicated that the addition of katuk leaf meal in feed had significant effects (P<0,05) on body weight gain and scrotal circumference of Ettawa grade goat.

Abstract

This study aimed to determine the effect of vitamin C addition in andromed® dilution on procentage of motility and intact plasma membrane post thawing of aceh bull spermatozoa. This research used fresh semen aceh bull were collected using artificial vagina and divided into four treatments.Control treatment (P0) semen without the addition of vitamin C, first treatment (P1) semen plus vitamin C 0.5 g/100 ml of dilution, second treatment (P2) semen plus vitamin C 1.0 g/100 ml of dilution, third treatment (P3) semen plus vitamin C 2.0 g/100 ml of dilution. The experiment design used was completely randomized design (CRD). Data was analyzed using ANOVA (Analysis of Variant) and followed by Duncan’s multiple range (Duncan’s Multiple Range Test) 5% confidence interval. The result showed that the addition of vitamin C influence the motility and intact plasma membrane percentage of aceh bull spermatozoa after thawing. The addition of vitamin C 0.5 g/100 ml and 1.0 g/100 ml increase motility and intact plasma membrane percentage, while vitamin C 2.0 g/100 ml decrease motility and intact membrane plasma percentage after thawing.Key words: intact plasma membrane, motility, aceh bull, spermatozoa, vitamin C

Abstract

This study aimed to evaluate the genetic potential of local meat chickens (LMC) to get the candidate of parent stock through the development of the selection method applied, as well as economical and practical approach and adjusment of nutritional requirement, especially protein and energy balance. Research was divided into 2 groups, where the female and male parent was reared for 2 months and the offspring (F1) was reared for 4 months and selected. The parent was mated by the method of artificial insemination (AI). Method of selection was used on the orientation of the meat chicken production parameters with a positive correlation with the gene expression Diet treatments were formulated by optimal nutrition standards in order to drive maximum primary production of local meat chicken, contained 17, 19, and 21% of protein. The result showed that feeding diet with 19% protein can stimulate optimum growth and food conversion of ALPU male, whereas female growth LMC in line with the increasing availability of protein in the diet, wherein feeding 21% of protein result in optimal growth until age 4 months.

Abstract

ABSTRAK Penelitian ini bertujuan untuk mengetahui apakah terdapat perbedaan keberhasilan Inseminasi Buatan pada sapi aceh menggunakan semen beku sapi bali, sapi simental, dan sapi limosin di Kecamatan Mesjid Raya  Kabupaten Aceh Besar. Materi penelitian terdiri dari 30 orang responden dan 61 ekor induk sapi aceh yang dipelihara masyarakat di Kecamatan Mesjid Raya Kabupaten Aceh Besar. Metode yang dipakai dalam penelitian ini adalah metode survey, dengan pengambilan sampel secara purposive sampling. Data diperoleh dari hasil IB sapi aceh oleh petugas inseminator, dan hasil wawancara berupa kuesioner dengan peternak. Data dianalisis menggunakan analisis diskriptif. Hasil dari penelitian ini menunjukkan tingkat Conseption Rate (CR) sapi aceh menggunakan semen beku sapi Bali adalah 63,15%, lebih tinggi dari semen beku sapi Simental sebesar 54,54% dan sapi Limosin sebesar 60,00%. Nilai Service Per Conception menggunakan semen beku sapi Bali adalah 1,42 kali lebih rendah dari semen beku sapi simental dan semen beku sapi Limosin yaitu  1,54 dan 1,55. Kata Kunci :  Inseminasi Buatan (IB), sapi aceh, sapi bali, simental dan limosin. ABSTRACT            This study aims to determine whether there are differences in the success of Artificial Insemination on Aceh cows using frozen semen of  Bali cows cattle, simental cows and limousine cows in the District Mesjid Raya Aceh Besar regional. The research material consisted of 30 respondents and 61 cows of Aceh cattle that are herded by society in District Mesjid Raya Regency of Aceh Besar. The method used in this study is the survey method, with sampling by using purposive sampling. The data obtained from AI results of aceh cows by the inseminator officer, and interview  in the form of quisioners with farmer. Data were analyzed using using purposive sampling. The data obtained from AI results of aceh cows by the inseminator officer, and interview  in the form of quisioners with farmer. Data were analyzed using descriptive analysis. The result of this research shows that the level of Conseption Rate (CR) of Aceh cows using frozen semen of Bali cows is 63,15%, higher than Siment cattle semen 54,54% and Limousine cows is 60,00%. The value of Service Per Conception using frozen semen of Bali cattle was 1.42 times lower than semen frozen simental cows and frozen  Limosin cows of 1.54 and 1.55.  Keyword : Artificial Insemination (AI), Aceh cow, Bali cow, simental and limosin.

Abstract

ABSTRAK Penelitian ini bertujuan mengetahui pengaruh pemberian formalin melalui intraperitoneal terhadap kadar hemoglobin dan jumlah eritrosit pada tikus putih (Rattus norvegicus) strain Wistar. Hewan yang digunakan dalam penilitian ini adalah 24 ekor tikus putih jantan. Rancangan yang digunakan adalah rancangan acak lengkap (RAL) pola searah dengan 4 kelompok perlakuan. Masing-masing kelompok perlakuan terdiri atas enam ekor tikus. Kelompok kontrol tikus hanya diberi pakan dan air minum tanpa perlakuan apapun, kelompok 1 tikus diinjeksi formalin dengan dosis 1 mg/Kg bb, kelompok 2 diinjeksi formalin dengan dosis 2,5 mg/Kg bb, kelompok 3 diinjeksi formalin dengan dosis 5 mg/Kg bb. Masing-masing kelompok diberi perlakuan selama 14 hari berturut-turut secara intraperitoneal. Selama penelitian tikus diberi pakan dan air minum secara adlibitum. Pada hari ke 15, sampel darah diambil melalui sinus orbitalis menggunakan pipet kapiler. Hasil analisis statistik menunjukkan rata-rata (±SD) kadar hemoglobin (g/dl) K0 12,00±0,72, K1 9,63±1,67, K2 9,83±1,30, dan K3 9,76±1,41. Rata-rata (±SD) jumlah eritrosit (106/mm3) K0 7,06±0,78, K1 5,31±1,46, K2  5,35±0,97, dan K3 4,98±0,59. Hasil uji ANOVA menunjukkan kelompok K0 berpengaruh nyata (P<0,05) terhadap kelompok K1, K2, dan K3, sementara kelompok K1, K2, dan K3 tidak berpengaruh nyata (P>0,05) antara kelompok perlakuan. Disimpulkan bahwa pemberian formalin secara intraperitoneal dapat menurunkan kadar hemoglobin dan jumlah eritrosit tikus putih strain Wistar.Kata kunci: formalin, kadar hemoglobin, jumlah eritrosit, Rattus norvegicusABSTRACTThe study aims to determine the effect of formalin administration via intraperitoneal on levels of haemoglobin and the number of erythrocyte in rats (Rattus norvegicus) Wistar strain. Animals used in this study were 24 male rat. The research design was used completely randomized design (CRD) with 4 unidirectional pattern groups treatments. Each group treatment consisted of six rat. Group control rat was given only fed and drinking without any treatment, group 1 rat injected formalin with dosage of  1 mg/Kg bw, group 2 rat  injected formalin with dosage of 2,5 mg/Kg bw, and group 3 rat injected formalin with dosage of 5 mg/Kg bw. Each group was given the specific treatment for 14 consecutive days intraperitoneally. During the study, rat were fed and drinking water in a adlibtum. On day 15, blood sample was taken through orbital sinus using a pipette capillary. The statistical analysis that showed mean (±SD) levels of haemoglobin (g/dl) K0 12,00±0,72, K1 9,63±1,67, K2 9,83±1,30, and K3 9,76±1,4. Mean (±SD) number of erythrocyte (106/mm3) K0 7,06±0,78, K1 5,31±1,46, K2  5,35±0,97, and K3 4,98±0,59. The results of ANOVA showed significant group K0 (P<0,05) against groups K1, K2, and K3, while groups K1, K2, and K3 no significan effect (P>0,05) in between group. It was concluded that the administration of formalin intraperitoneal can lower haemoglobin levels and the number of erythrocyte rats Wistar strain.Keywords:  formalin, haemoglobin levels, number of erythrocyte, Rattus norvegicus

Abstract

(The ratio of birth sex of ettawa crossbreed goat by artificial insemination using spermatozoa swim up)ABSTRACT. The aim of this research was to determine the ratio of birth sex of ettawa crossbreed goat by artificial insemination using spermatozoa swim up. This research was at Reproduction Laboratory Faculty of Veterinary medicine Syiah Kuala university and Livestock Group SMD, Bener Meriah Regency. Fifteen female goats with age 8-24 months where used in this study and divided into 3 treatment groups (P0, P1, P2) and 5 replications of each. The first group (P0) inseminated with sperm without swim up; P1 and P2 inseminated with sperm which swim up for 5 minutes (P1) and 10 minutes (P2). The result of conception rates observation and sex ratio of children born tabulated into tables. The data was analyzed by using analysis of variance (ANOVA) one way pattern followed by the multiple test Duncan. The results showed that the conception rate and birth sex ratio of male born had significantly different (P<0,05). The segregation of sperm which swim up increased the conception rate and birth sex ratio of male born.

Abstract

Penelitian ini bertujuan mengetahui pengaruh penambahan ekstrak kulit buah naga merah dalam media sitrat kuning telur terhadap daya tahan hidup spermatozoa sapi aceh setelah pendinginan. Semen ditampung dari 1 ekor pejantan umur 3 tahun sehat, menggunakan vagina buatan. Semen yang berkualitas baik dibagi ke dalam 5 kelompok perlakuan penambahan ekstrak kulit buah naga merah; 0,00 g/100 ml (P0); 0,2 g/100 ml (P1); 0,4 g/100 ml (P2); 0,6 g/100 ml (P3); dan 0,8 g/100 ml (P4) dalam media sitrat kuning telur dan disimpan dalam suhu 4. Pengamatan daya tahan hidup spermatozoa dilakukan setiap hari selama 5 hari. Data yang diperoleh dianalisa dengan analysis of varian(ANOVA) pola satu arah yang dilanjutkan dengan uji berganda Duncan. Hasil pengamatan daya tahan hidup spermatozoa setelah pendinginan pada perlakuan P0; P1; P2; P3; dan P4 secara berturut-turut 3,8; 4,6; 3,6; 1,0; dan 0,0 hari. Hasil penelitian ini menunjukkan bahwa penambahan ekstrak kulit buah naga merah dalam media sitrat kuning telur berpengaruh secara nyata (P<0,05) terhadap daya tahan hidup spermatozoa. Penambahan ekstrak kulit buah naga merah 0,2 g/100 ml dalam media sitrat kuning telur menghasilkan daya tahan hidup spermatozoa sapi aceh lebih baik selama penyimpanan pada suhu 4. (This study aims to determine the effect of peels red dragon fruit extract addition into citrate-yolk medium on viability of aceh cattle sperm after cooling. Semen was collected from 1 healthy male aged 3 years, using artificial vagina. Good quality semen was divided into 5 groups; addition of peels red dragon fruit extract 0.0 g/100 ml (P0); 0.2 g/100 ml (P1); 0.4 g/100 ml (P2); 0.6 g/100 ml (P3); and 0.8 g/100 ml (P4) into citrate-yolk medium, and stored in a temperature of 4. Observation of sperm viability were done every day up to day 5. The data obtained was analyzed with the one-way analysis of variance (ANOVA), followed by multiple test Duncan. Results show sperm viability after cooling in the groups P0; P1; P2; P3; and P4 respectively was 3.8 days, 3.6 days, 4.6 days, 1.0 day, and 0.0 day. The results show that the addition of peels red dragon fruit extract in citrate-yolk medium affects significantly (P<0.05) sperm viability. The addition 0.2 g/100 ml peels red dragon fruit extract into citrate-yolk medium during storage in a temperature of 4 results greater sperm viability.)
