Abstract

Antifungal Activity of Tegeran Wood (Cudrania javanensis Trécul) Extracts against Fungus Peniophora sp.  ABSTRACTThe fungus Peniophora sp. can degrade natural dyes, causing discoloration on batik cloth. This study was aimed to evaluate the antifungal activity of tegeran wood (Cudrania javanensis) extracts against Peniophora sp. isolated from the aqueous extract of mahogany (Swietenia mahagoni) bark. Aqueous and methanol extraction procedures gave tegeran wood extracts whose concentrations were then varied into 100, 250 and 500 ppm. Phytochemical analysis of the extracts were carried out, and the results showed that the average values of the total polysaccharides and terpenoids of both aqueous and methanol extracts did not differ significantly (p<0.05), whereas those of polyphenols did (p<0.05). The total polyphenols of the methanol extract (484.723 mg GAE/g) was higher than that of the aqueous extract (389.903 mg GAE/g). Results showed that the methanol extract of tegeran wood showed higher antifungal activity against Peniophora sp. than the aqueous extract, and was also higher than ketoconazole control (100 ppm). The minimum inhibitory concentration of the methanol extract against Peniophora sp. growth was of 500 ppm with the antifungal activity value (AFA) of 76.30%.Keywords: antifungal activity, Cudrania javanensis, Peniophora sp., Swietenia mahagoni, wood extract ABSTRAKJamur Peniophora sp. dapat mendegradasi zat warna alam sehingga warna yang dihasilkan pada kain batik lebih pudar. Penelitian ini dilakukan untuk mempelajari aktivitas anti jamur dari ekstrak kayu tegeran (Cudrania javanensis) terhadap Peniophora sp. yang diisolasi dari ekstrak air kulit mahoni (Swietenia mahagoni). Ekstraksi menggunakan metanol dan air, menghasilkan ekstrak kayu tegeran, yang kemudian konsentrasinya divariasi menjadi 100, 250 dan 500 ppm. Analisis fitokimia ekstrak kayu tegeran dilakukan, dan hasilnya menunjukkan bahwa nilai rata-rata total polisakarida dan terpenoid ekstrak metanol dan air kayu tegeran tidak berbeda nyata (p<0,05). Sedangkan total polifenol kedua ekstrak berbeda nyata (p<0,05). Total polifenol ekstrak metanol kayu tegeran (484,723 mg GAE/g) lebih besar dibandingkan dengan ekstrak air (389,903 mg GAE/g). Hasil menunjukkan bahwa aktivitas anti jamur ekstrak metanol lebih baik dibandingkan dengan ekstrak air kayu tegeran, dan juga lebih tinggi dibandingkan dengan kontrol ketoconazole (100 ppm). Konsentrasi minimum ekstrak metanol kayu tegeran yang dapat menghambat pertumbuhan jamur Peniophora sp. adalah 500 ppm dengan nilai penghambatan (AFA) sebesar 76,30%.Kata Kunci: aktivitas anti jamur, Cudrania javanensis, ekstrak kayu, Peniophora sp., Swietenia mahagoni

Abstract

Formulation and in vitro study of propranolol hydrochloride controlled release from carboxymethyl chitosan-based matrix tablets have been conducted. Formulations with various concentrations of carboxymethyl chitosan 2% (F1), 4% (F2), 6% (F3) were done by wet granulation method. Compatibility test was conducted by XRD and FTIR spectroscopy to determine interaction between propranolol hydrochloride and polymer excipients. Dissolution profiles was obtained through in vitro tests release using simulated gastric fluid (without enzymes, pH 1.2) for the first 2 h and followed by simulated intestinal fluid (phosphate buffer solution without enzyme, pH 7.2) for 2 h remaining. The dissolution profile of each formulation was fitted with five kinetics modeling of drug release (zero order, first order, Higuchi, Peppas-Korsmeyer, and Hixson-Crowell). The compatibility test results showed that formulation caused physical interactions between propranolol hydrochloride and polymer excipient but doesnt make crystallinity nature of propranolol hydrochloride disturbed even after formulation. Dissolution profiles of each formulation showed that controlled release of propranolol hydrochloride from the tablet followed Peppas-Korsmeyer model. It is concluded that carboxymethyl chitosan in appropriate proportions is suitable for formulating propranolol hydrochloride controlled release tablets which exhibit Peppas-Korsmeyer release kinetics.

Abstract

A new polyketide derivative, 2-chloro-3,4,7-trihydroxy-9-methoxy-1-methyl-6H-benzo[c]chromen-6-one (1), was isolated from the endophytic fungus Penicillium chermesinum. The structure was established on the basis of UV, IR, HR-ESI MS, and 1D and 2D NMR experiments. The cytotoxicity against four cancer cell lines (HuCCA-1, HepG2, A-549, and MOLT-3) of compound 1 are 14.94-115.71 µM.

Abstract

Labu kuning (C. moschata) merupakan salah satu tanaman yang banyak terdapat di Indonesia. Penelitian ini bertujuan untuk mempelajari aktivitas antioksidan, struktur morfologi, sifat kimia dan fisikokimia dari tiga komponen tepung yakni kulit, biji dan daging buah dari labu kuning. Ekstrak biji, kulit dan daging buah labu kuning diperoleh dengan maserasi menggunakan pelarut etanol. Dua metode pengeringan yang digunakan yakni pengeringan dengan sinar matahari dan oven pada suhu 50oC. Hasil uji aktivitas antioksidan dengan metode DPPH (2,2-diphenyl-1-picrylhydrazyl) menunjukkan bahwa biji labu kuning dengan metode pengeringan oven, memiliki aktivitas antioksidan yang paling tinggi, yakni memiliki aktivitas penangkapan radikal bebas sebesar 22,6% pada konsentrasi ekstrak 400 ppm, diikuti oleh kulit dan daging buah labu kuning masing-masing sebesar 16,7 dan 14,2%. Tepung kulit, biji dan daging buah labu kuning juga sudah dilakukan karakterisasi menggunakan SEM (Scanning Electron Microscopy) untuk mengetahui struktur morfologi permukaan, FTIR (Fourier Transform Infrared Spectroscopy) untuk mengetahui gugus fungsional senyawa dari labu kuning, XRD (X-ray diffraction) dan Uji Warna. Hasil analisis proksimat menunjukkan bahwa kandungan protein dan lemak dari biji labu kuning dengan metode pengeringan oven memiliki kandungan paling tinggi yakni sebesar 33,5% dan 33,3% sedangkan kandungan karbohidrat tertinggi dimiliki oleh daging buah labu kuning dengan pengeringan oven sebesar 75,3%. Hasil yang diperoleh menunjukkan bahwa perbedaan metode pengeringan menghasilkan karakteristik yang berbeda dari tepung labu kuning
