Abstract

Tarsius is an endemic species in Indonesia but is is getting threatened. In-situ and ex-situ conservationof this species would yield better results when its genetic information and diversity determined. Theobjective of this ressearch was to study the genetic diversity of ND3 gene of Tarsius sp. Based on sequencingof PCR products using primer ND3F and ND3R resulted in 437 nts base sequence. Results of ND3 fragmentssequencing were put on multiple alignment with other primates from Genbank using of software ClustalW, and then were analyzed using MEGA program version 4.1. A different nucleotide site was found(nucleotide no. 24). The genetic distance based on nucleotide ND3 was calculated using Kimura 2-parametermodel. The genetic distance showed the smallest genetic distance 0%, the biggest 0,03% and average0,01%. The phylogenetic tree using neighbor joining method based on the sequence of nucleotide ND3 genecould not be used to differentiate among T. Dianae (from Central Sulawesi), T. Spectrum (from NorthSulawesi) and T. bancanus (from Lampung, South Sumatera).

Abstract

Insertion and deletion are information that reflect evolutional potency of species or population, that nowadays, are central topics in conservation genetics. This research aims to reveal the proportion of insertion and deletion on RDP1of species Megapodes in Indonesia. Whole blood DNA was extracted according to the protocol of Qiamp DNA Blood Mini Kit (Qiagen), and used as template for PCR (Polymerase Chain Reaction) which was conducted according the protocol of Takara Ex TaqTM PCR Kit in a Perkin-Elmer Thermal Cycler 9600. PCR products were purified using a Qiaquick PCR Purification Kit (Qiagen, USA). The sequencing reaction for RDP1 was performed using BigDye v.3.1. Cycle Sequencing Kit (Applied Biosystems,USA) followed the protocol of ABI Prism 3100-Avant Genetic Analyzer (Applied Biosystems, USA). Alignment of RDP1 nucleotide sequences of megapodes and that are obtained from international DNA data base (DDBJ-DNA Data Bank of Japan) with accession number AF 394649 (RDP1) were conducted by using Clustal-X. Characteristics of RDP1 were analyzed by employing the DNA Sequence Polymorphism (DnaSP) version 3.51 and Program MEGA version 4. The analysis result demonstrated there were insertion and deletion on RDP1 of species of Megapodes in Indonesia, which resulting in a gap of 1-10 bp and was concentrated in nucleotide 407-568. Alignments between species of Megapodes  and two species of Galliformes showed more gaps, which were concentrated on nucleotides 480-708.

Abstract

Abstract: Balinese language is divided into two dialects, namely Bali Dataran(DBD), and Bali Aga (DBA) dialect. The DBD variation happens vertically, but not in DBA. Some vocabularies in DBD that are classified into low(Tak-Alus/TA) are classified as common variants in DBA. This research aims to explore the Balinese personal pronoun variations in DBD  which  DBA in  Sembiran(DS), and Seraya Timur(DST). The results, by applying the dialectology theory and correlation method or metode padan, showed that the personal pronouns of DS and DST varied lexically and phonologically. Lexically, the personal pronoun of first person singular in DBD /(ti)tiyaŋ/(A), /(i)–caŋ/ and /yaŋ/(TA) was realized as /oke/ and /kaka/ in DS also /(b)–iba/, /uke/, and /wane/ in DST. The second person singular /ragane/(A), /cai/ /ɲai/, and /ibə/(TA) in DBD, were realized as /cai/, /ɲai/, and /ŋko/ in DS also /cai/ and /ɲai/ in DST; and the third person singular /idə/, /dane/(A) and /(i) –yə/(TA) in DBD, were realized as /ya/, in DS and DST. Abstract

Abalon (Haliotis asinina) adalah salah satu bahan makanan yang lezat dan memiliki nilai ekonomis tinggi serta merupakan sumber protein. Penelitian ini bertujuan untuk menentukan kadar protein abalon (Haliotis asinina) asal Kecamatan Dako Pemean Kabupaten Tolitoli dan pemanfaatannya sebagai sumber  belajar. Penelitian yang dilakukan merupakan jenis penelitian deskriptif eksploratif.Data kadar protein diperoleh melalui analisis Kjedhal mikro dengan tiga tahapan yaitu destruksi, destilasi dan titrasi pada lima sampel.Hasil penelitian  menunjukkan nilai  rata-rata kadar protein abalon (Haliotis asinina) pada bahan basah sebesar 60,79%.Setelahmelalui validasi tim ahli yaitu ahli isi, desain dan media serta 30 mahasiswa penguji, layak dimanfaatkan sebagai sumber belajar berupa poster dengan persentase kelayakan sebesar 76,30% Kata Kunci: Kadar Protein,Haliotis asinina, Sumber Belajar.

Abstract

Biji kakao adalah salah satu komoditas utama di dunia yang hingga saat ini masih memiliki masalah penurunan produksi yang mayoritas disebabkan oleh serangan hama dan penyakit. Mengembangkan kultivar kakao yang secara genetik tahan terhadap serangan hama dan penyakit adalah salah satu solusi yang ideal dalam mengatasi permasalahan tersebut. Penelitian ini bertujuan untuk mengetahui gen-gen Theobroma cacao pada database genom kakao yang homolog dengan gen yang terlibat dalam sistem ketahanan terhadap hama dan penyakit pada tumbuhan lain melalui analisis in silico. Survei informasi dasar dan identifikasi gen yang terlibat dalam sistem ketahanan tumbuhan model Arabidopsis thaliana dan Oryza sativa dilakukan pada GenBank dan The Arabidopsis Information Resource. Analisa in silico yang dilakukan berupa BLAST urutan protein, uji homologi protein kandidat masing-masing gen, dan identifikasi gen homolog pada Cacao Genome Database menggunakan sumber data genom “Theobroma cacao cv. Matina 1-6 v1.1”. Berdasarkan hasil identifikasi, gen Thecc1EG006332t1 (PR-1), Thecc1EG024509t1 (PR-2), Thecc1EG024514t1 (PR-2), Thecc1EG026942t1 (CTL1), Thecc1EG021326t1 (CTL1), dan Thecc1EG033984t1 (PR-5) pada Cacao Genome Database (CGD) adalah gen Theobroma cacao yang homolog dengan gen yang terlibat dalam sistem ketahanan tumbuhan terhadap hama dan penyakit, serta kemungkinan besar memiliki fungsi yang sama dengan gen homolognya masing-masing.

Abstract

This current paper brings into discussion cultural meaningfulness on intercultural perspective of English learning materials for elementary school. Students might be perceived to master English, but there has been no guarantee that they can properly make use of their acquired language skills upon direct interaction with native speakers of the target language. This is reasonable as English instruction in elementary school is projecting grammatical aspects, not touching cultural aspects of the target language yet. In fact, those cultural aspects are playing pivotal roles in achieving interactional comprehension between speakers and interlocutors. In regards to cultural studies, foreign language students are to be apt to target language’s cultures. Departing for the mentioned concerns, English is to be taught by incorporating cultural aspects of native speakers, and thus intercultural perspective. This current paper pinpoints the development of intercultural perspective in English learning materials for elementary school students aging from 6-12 years old. Cultural meaningfulness on intercultural perspective is set to be the target that encapsulates various topics for teaching English in elemenatry school. The so called perspective not merely comprises social skills, but training sensitivity and comprehension on values, point of view, proper way of life and thinking as well. A number of materials containing intercultural perspective and cultural meaningfulness are described in this paper. Those materials are presented within six intercultural topics in the forms of culture-related vocabularies, expressions, and meanings.

Abstract

Penelitian ini bertujuan untuk meningkatkan hasil belajar dengan tema lingkungan pada kelas I SD Negeri 018 Letawa Kecamatan Sarjo Kabupaten Mamuju Utara. Penelitian dilaksanakan dengan dua siklus, dan setiap siklus terdiri dari empat tahap yaitu : Perencanaan, Pelaksanaan, Observsi dan refleksi. Subjek penelitian ini adalah semua siswa kelas I SD Negeri 018 Letawa Kecamatan Sarjo Kabupaten Mamuju Utara Tahun Pelajaran 2012/2013 dengan jumlah siswa 32 orang. Data penelitian ini diperoleh dari observasi kegiatan guru dan kegiatan belajar siswa. Berdasarkan hasil analisa data, pada siklus I diperoleh Daya Serap Individu 69% dan Ketuntasan Belajar Klasikal 75%. Sedangkan Pada siklus II meningkat dengan Daya Serap Individu 75% dan Ketuntasan Belajar Klasikal 81%, serta hasil belajar siswa mengalami peningkatan secara klasikal dari siklus I ke sisklus II yaitu 75,0% ke 81,25%. Abstract

Javanese is a well known for its speech levels called ngoko ‘low’ and krama ‘high’ which enable its speakers to show intimacy, deference, and hierarchy among the society members. This research applied critically Brown and Gilman (1960)’s theory of terms of address to analyze the asymmetrical, factors which influence, and politeness of the use of speech levels in Javanese.                                                                                                   Method of observation, in depth interview, and document study were applied to collect the data. Recorded conversation was then transcribed into written form, classified and codified according to the speech levels, and analyzed using politeness system (Scollon and Scollon, 2001) and status scale (Homes, 2001).                                                                       The use of speech levels shows asymmetric communication: two speakers use two different codes, i.e. ngoko and krama because of power (+P) and with/without distance (+/-D), and it is the reflection of hierarchical politeness. The asymmetrical use of ngoko and krama by God and His Angel, God and human beings strongly explicated the asymmetrical communication between superiors and inferiors. The finding of the research shows that the use of ngoko and krama could present the phenomena of code-switching, code-mixing, and the fundamental phenomenon is ‘code-crossing’. It is concluded that hierarchical politeness in Javanese is ‘social contract’ i.e. the acknowledgment of the existence of high class (superior) and low class (inferior)  implemented in ‘communications contract’  using speech levels of the Javanese language  in line with status scale. Abstract

Penelitian ini bertujuan untuk mengetahui peningkatkan hasil belajar siswa kelas V tentang energi alternatif dan cara penggunaannya menggunakan Model Pembelajaran Kooperatif Tipe STAD. Penelitian ini adalah penelitian tindakan kelas yang dilaksanakan dalam dua siklus. Tahapan pada penelitian ini meliputi tahapan perencanaan, pelaksanaan tindakan, observasi dan hasil refleksi. Data yang diperoleh dalam penelitian meliputi hasil belajar siswa, hasil observasi aktivitas guru dan siswa. Hasil penelitian menunjukkan bahwa Penerapan Model Pembelajaran Kooperatif Tipe STAD dapat meningkatkan hasil belajar siswa kelas V tentang energi alternatif dan cara penggunaannya di SDN 01 Pasang Kayu. Hasil penelitian menunjukkan pada tindakan siklus I nilai ketuntasan klasikal diperoleh 41,67%, sedangkan nilai ketuntasan klasikal pada siklus II meningkat menjadi 83,33%. Pada hasil analisa terhadap daya serap individu pada siklus I sebesar 69,44%, sedangkan pada siklus II sebesar 83,33%.
