Abstract

The objectives of this research were to determine the genetic gain value of Tilapia Pandu F5 fry and to compare the growth between the fry of best 10 % (Top 10) of  Tilapia Pandu F5and the average Tilapia Pandu F5 (without selection). This study was done at Satker PBIAT (Centre of Fresh Water Fish Culture Working Unit) Janti Klaten. The  fries was kept in a happa (net cage) at the size of 4x2x1 m3 from nursery I-III. The stocking density was 500 fries/cage. The results of this investigation shows that the weight genetic gain of Tilapian Pandu F5 at nursery I-III was at the range of 60,12 % - 88,58 %. It was also shown that the best 10 %  selection (Top 10) Tilapia Pandu F5 fries growth better than the average Tilapia Pandu F5 fries without selection.

Abstract

Ikan nila di Indonesia merupakan salah satu ikan air tawar yang memiliki nilai ekonomis penting. Berbagai upaya terus dilakukan dalam meningkatkan produksi ikan nila, salah satunya adalah dengan pemuliaan seperti perbaikan genetik. Selective breeding adalah riset genetik yang dominan untuk memperbaiki pertumbuhan sebagai tujuan utama baik dari seleksi famili maupun individu. Harapannya adalah generasi F5 ikan nila kunti memiliki peningkatan performa dari ikan nila kunti F4. Tujuan dari penelitian ini adalah untuk membandingkan karakter reproduksi yang dihasilkan dari ikan nila kunti F4 dan F5 sehingga diketahui terjadinya  peningkatan performa karakter reproduksi dari generasi 4 ke generasi 5. Karakter reproduksi diamati heritabilitasnya, heritabilitas merupakan nilai dugaan yang mampu menunjukkan laju perubahan yang dapat dicapai dengan seleksi untuk suatu sifat di dalam populasi. Penelitian ini dilakukan dengan 2 perlakuan dan masing-masing terdiri dari 30 ulangan. Karakter reproduksi meliputi fekunditas, HR, diameter telur, bobot telur, panjang dan bobot larva menetas, panjang dan bobot larva lepas telur dan jumlah larva dimulut. Data tersebut diuji peningkatan performa reproduksi dengan analisis heritabilitas. Hasil penelitian menunjukkan adanya peningkatan performa karakter reproduksi dari induk F4 ke F5 dilihat dari meningkatnya hasil fekunditas; HR; diameter dan bobot telur; panjang dan bobot larva menetas; panjang dan bobot larva lepas telur dan jumlah larva dimulut serta didukung nilai heritabilitas fekunditas dan HR dari induk F4 ke F5 yang dikategorikan sebagai nilai kisaran yang tinggi. Kualitas air selama penelitian tidak dilakukan upaya untuk mempertahankan namun masih berada dalam kisaran yang layak untuk kehidupan ikan nila kunti.

Abstract

ABSTRAK Penelitian ini bertujuan untuk mengetahui pengaruh penambahan air kelapa dan gliserol terhadap fertilitas spermatozoa ikan mas (Cyprinus carpio L.) dan kondisi motilitasnya serta perlakuan yang terbaik pada penyimpanan sperma ikan mas strain Rajadanu. Percobaan ini menggunakan Rancangan Acak Lengkap (RAL) dengan 3 perlakuan dan 3 kali ulangan, yaitu Perlakuan A : 0,5 ml sperma ditambah air kelapa muda 75% + gliserol 25%, Perlakuan B : 0,5 ml sperma ditambah air kelapa muda 50% + gliserol 50%, Perlakuan C : 0,5 ml sperma ditambah air kelapa muda 25% + gliserol 75%. Hasil penelitian menunjukkan bahwa penambahan air kelapa dan gliserol pada penyimpanan sperma berpengaruh nyata terhadap fertilitas spermatozoa ikan mas dan spermatozoa masih bertahan hidup selama penyimpanan 4 hari, namun semakin lama disimpan motilitas akan semakin menurun. Nilai fertilitas pada hari ke- 0 tidak berpengaruh, namun berpengaruh setelah dilakukan penyimpanan. Hari ke- 1 nilai fertilitas tertinggi pada perlakuan B 53,1%. Hari ke- 2 perlakuan B memiliki nilai tertinggi sebesar 39,3%. Hari ke- 3 perlakuan B didapatkan nilai tertinggi sebesar 15,9%. Hari ke- 4 fertilitas tertinggi dihasilkan pada perlakuan B, yaitu 6,5%. Berdasarkan hasil penelitian dapat disimpulkan bahwa penambahan air kelapa dan gliserol pada penyimpanan sperma berpengaruh nyata terhadap fertilitas spermatozoa ikan mas dan spermatozoa masih bertahan hidup selama penyimpanan 4 hari, namun semakin lama disimpan motilitas akan semakin menurun, serta perlakuan yang terbaik adalah perlakuan B dengan dosis air kelapa muda 50% dan gliserol 50%. ABSTRACT This study aimed to determine the effect of the addition of coconut water and glycerol on fertility carp (Cyprinus carpio L.) spermatozoa and motility conditions as well as the best treatment on sperm storage carp strains Rajadanu. This experiment used Completely Randomized Design (CRD) with 3 treatments and 3 replications, namely Treatment A: 0.5 ml sperm plus coconut water 75% + glycerol 25%, Treatment B: 0.5 ml sperm plus coconut water 50 % + glycerol 50%, Treatment C: 0.5 ml sperm plus coconut water 25% + glycerol 75%. The results showed that the addition of coconut water and glycerol on sperm storage significantly affect sperm fertility carp and surviving spermatozoa during 4 days of storage, but the longer it is stored motility decrease. Fertility values ​​at day-0 has no effect, but the effect after storage. Day-1 value of the highest fertility treatment B 53.1%. Day-2 treatment B had the highest score of 39.3%. Day-3 treatment B earned the highest score of 15.9%. Day-4 produced the highest fertility treatment B, which is 6.5%. Based on the results of this study concluded that the addition of coconut water and glycerol on sperm storage significantly affect fertility carp spermatozoa and spermatozoa were still alive during 4 days of storage, but the longer it is stored motility will decrease, and the best treatment is the treatment of B at a dose of coconut water 50% and glycerol 50%.

Abstract

Tilapia Pandu (Oreochromis niloticus) is the result repair genetic conducted activities cross breeding between females nila gift and male nila singapore called pandu. This research intent to know and hematology tilapia pandu infected by bacteria Streptococcus agalactiae.Method is used a experimental laboratories. A completely randomized design was applied to the research with 3 treatments and 3 replicates, treatment A (density bactery 105), B (density bactery 107) and C (density bactery 109). Variables tested include the number of erythrocyte, leuchocyte, haemoglobin, hematocrite, blood glucose, and survival rate on days 7 and 14. The results obtained by the infection bacteria S. agalactiae are not significant against hematology and survival rate tilapia pandu. The number of erythrocytes was highest in treatment C for 1.78±0,07x 106 sel/mm3, the highest leucocyte in treatment C for 120,57±4,82 x 103 sel/mm3, the highest haemoglobin in treatment C for 7.70±0.36 g/dL, the highest hematocryte in treatment C for 25.80±1.73%, the highest trombocyte in treatment A for 72,00±61,05 x 103 sel/mm3, the highest blood glucose in the treatment B for 74.53±34,10 g/dL and the highest survival rate in the treatment B and C each for 89.78±0.12%. The results obtained that bacterial infection of S. agalactiae gives no real influence on the blood profile and survival rate tilapia pandu.

Abstract

S. agalactiae is a bacteria that often attack tilapia. The purpose of this study was to examine the blood profile of Tilapia Kunti that infected by bacterium S. agalactiae with use variable concentration of erythrocytes, leukocytes, haemoglobin, hematocrit, trombocyte, and blood glucose. The method used was a completely randomized design (CRD) with 3 treatments and 3 replications. Treatment I (Tilapia Kunti injected using S. agalactiae with density 105), treatment II (tilapia kunti injected using S. agalactiae with density107), and treatment III (tilapia kunti injected using S. agalactiae with density 109). The results showed that the tilapia kunti injected using S. agalactiae with different densities have significant differences on the hematocrit and no significant differences on erythrocytes, leukocytes, haemoglobin, trombocyte, blood glucose and survival rate. The highest concentration of erythrocytes was observed in treatment II, in the amount of 1.933 ± 0.162 x106 cells/mm3, the highest concentration of leukocytes was observed in treatment II, in the amount of 122.267 ± 3.265 x103 cells/mm3, the highest concentration of hemoglobin was observed in treatment II, in the amount of of 9.333 ± 0.404 g / dL, the highest concentration of hematocrit was observed in treatment II, in the amount of is 30.933 ± 1.168%, the highest trombocyte concentration was observed in treatment I, in the amount of 30.333 ± 14.742 x103/μL, the highest blood glucose concentrations was observed in treatment I, in the amount of 73.933 ± 47.446 mg / dL, and the highest survival rate was observed in treatment II, in the percentage of 94.443 ± 9.624%.

Abstract

The purpose of study is to observe the effect of different tilapia strains in Rice-fish culture systems to the growth (individual absolute weight growth, RGR), feed utilization eficiency (EPP) and survival rate (SR) of tilapia. The method used in this research was experimental method carried out in the field. The material that used was tilapia seed in size 7 -10 cm with average individual weight 12- 24 gr. The trial design in this research used completely randomized design (CMD) with 3 treatment and 3 replications. The treatment tested A (Larasati), B ( Gift), and C (Gesit). The feed was given 2 times a day in the morning and afternoon. The variable measurable were individual absolute weight growth, RGR, EPP and SR. The result shows that the giving growth performance of different tilapia strains has highly significant (P> 0.01) to the individual absolute weight growth (gr), relatif growth rate (RGR), feed utization eficiency (EPP) and not significant influence (P ˃ 0.05) to survival rate (SR) of tilapia. Treatment A gives high relative growth rate and individual absolute weigth growth is 8.81% / day and 125.47 gr. Treatment A provide the best feed utilization eficiency of 84.08% and survival rate ranged from 86.46 to 94.94%. Range of water quality is still in decent condition for tilapia cultivation media. Based on the study it can be concluded that larasati tilapia strain provides the best growth in Rice-fish culture systems.

Abstract

Tilapia (Oreochromis niloticus) is one kind of freshwater fish which are economical important and widely cultivated by people as export commodities. The efforts to improve the quality of tilapia using selection program are needed in order to increase production and profitability of tilapia growers. High quality parents and seeds are absolutely necessary in tilapia farming activities because the superior parent seed quality is expected to be obtained as well. This study aims to compare the growth between Kunti Tilapia F4 males with F5 male and F4 female with F5 female and also to know the value of genetic gain Kunti Tilapia at age 5 months. Tested fish used in the study were Kunti Tilapia F4 and F5 at the age of 4 months. The rearing treatment lasted in 30 days. The variables measured were survival rate, relative growth rate, weight, total length, thickness, food convertion ratio, and genetic gain. This study used two treatments and 3 replications for each gender (F4 and F5 ♂ and F4 and F5 ♀). The research was conducted in October to November in SATKER PBIAT Janti, Klaten.The genetic gain value of Kunti tilapia from F4 to F5 generation at the age of 5 months are as follows, SR genetic gain : male (♂)2,72 % and female (♀)3,02 %; RGR genetic gain : male (♂) 11,37 % and female (♀) 10,78 %; weight genetic gain : male (♂)25,91 % and female (♀)24,9 %; total length genetic gain : male (♂) 8,08 % and female (♀) 1,51 %; thickness genetic gain : male (♂) 12,09 % and females (♀) 11,79 %; food convertion ratio (FCR) genetic gain : male (♂) 0,95 % and females (♀) 0,83 %. The Growth of Kunti Tilapia F5 generation is better than the F4 generation.

Abstract

Seed quality can be increased by fish genetic amelioration. Good quality seed is characterized by its immunity from diseases. Nila Larasaty (O. niloticus) Hybrid F5 is ameliorated genetic product of cross breeding between Nila Gift F5 female and Nila Singapore F5 male. This research aims to know Nila Larasati F5 Hybrid’s immunity system infected by S. agalactiae. Completely randomized design (RAL) is applicated on the research with 3 treatments and 3 repetitions, which treatments are: A (105 CFU/ml bacteria); B (107 CFU/ml bacteria) and C (109 CFU/ml bacteria). This research done in 35 days started from September until October 2012 at 2nd Class of BKIPM, Semarang Examined variabels are erythrocyte, leucocyte, hemoglobin, trombocyte, hematocrite, blood glucose, and survival rate. The result shows that erythrocyte highest on treatment C (1,87±0,19x106 sel/mm3); leucocyte highest on B (126,53±13,36 x103sel/mm3); hemoglobin on C (8,43±0,40 g/dl); trombocyte on B (50,33±45,72x103sel/mm3); hematocrite on C (9,20±1,71%); blood glucose on A (57,07±23,10 mg/dl); and survival rate highest on A (97,22±4,81%). Water quality parameters checked i.e DO (Dissolved Oxigen) 0,8 – 1,4 mg/l; pH 7,7 – 7,9; tempeature 28 – 28,3°C and ammonia 0,173 – 0,215 mg/l. Also the result shows that infection by S. agalactiae with different concentration has no significant effect (P>0,05) on seed of Nila Larasati F5 hybrid’s immunity system.

Abstract

Tilapia (Oreochormis niloticus) is one type of potential to cultured intensive in floating net cage because tilapia (Oreochormis niloticus) have biological characteristic benefit. Cultured tilapia (Oreochormis niloticus) in floating net cage is a aquaculture technic the most of productive. The case that happen is densities to applied for cultured in floating net cage on Wadaslintang, Wonosobo not optimal.The purpose of study is to observe the effect of different stocking densities and to observe the best of stocking densities towards feed utilization eficiency (EPP), growt h (W, RGR) and Survival rate( SR) of tilapia cultivated in floating net cage. The method used in this research was experimental method. The material that used was tilapia seed size 10 ± 20 cm with average individual weight 40± 60 g. The research design used completely randomized design (CMD) with 3 treatment and 3replications. The treatment tested A ( stocking densities of 50 fish/m³), B (stocking densities of 75 fish/m³), and C (stocking densities of 100 fish/m³). The feed was given 3 times a day. The result shows that the giving of different densities tilapia gives significant effect (P< 0,05) to the feed utilization eficiency (EPP),growth(W, RGR) and not gives significant (P>0,05) efect to Survival rate(SR).

Abstract

This research aimed to compare the heterosis effect of Larasati Tilapia from F5 generation the growth variable weight, length, thickness, survival rate and food convertion ratio, for ♂ Larasati F5, ♂ Pandu F5, ♂ Kunti F5; and ♀ Larasati F5, ♀ Pandu F5, ♀ Kunti F5 at the age of 5 month. This research was conducted in Freshwater Hatchery and Aquaculture Unit Janti, Klaten. Larasati Tilapia F5 generation, Pandu Tilapia F5 generation, and Kunti Tilapia F5 generation at the age of 4 months. This research were used 3 treatments and 3 replications for each sex. The treatment used was Larasati Tilapia F5, Pandu Tilapia F5, and Kunti Tilapia F5 generation. The result show that growth of Tilapia Larasati F5 generation was better than the parent generation Tilapia Pandu F5 generations and Tilapia Kunti F5 generations. Heterosis value of Larasati Tilapia F5 generation at the age of 5 months were, weight of (♂) 31.45% and (♀) 26.94% . Length of (♂) 22.90% and (♀) 19.96%. Thick of (♂) 22.84% and (♀) 19.88%. Survival rate of (♂) 1.33% and (♀) 1.48%. Food convertion ratio of (♂) -0.76% and (♀) -0.68%.
