Abstract

The objectives of this experiments were to determine the effects of drought stress initiated at vegetative growth stage on yield of peanut, to evaluate the tolerance of six peanut cultivars against drought stress, and to determine their leaf prolin and total sugar content. One group of peanut plants were grown in plastic pots and subjected to stress condition during the period of 15-45 days (Experiment I) or 15-80 days (Experiment II) by watering them only in every 4-7 days and were harvested at days 75 (Experiment I) or at maturity (Experiment II). The other group was grown optimally in plastic pot up to harvest period. Leaf prolin content was determined at 12 (before stress), 30 (after three periods of stress), 47 (after six periods of stress), and 64 days in Experiment I and only after six periods of drought stress in Experiment II. Results indicated that drought stress that was initiated at vegetative growth stage reduced plant height, branches number, shoot and root dry weight, and peanut yield. Sensitivity index calculated based on peanut yield showed that peanut cv. Jerapah and Singa were a medium tolerance while Gajah, Kelinci, Macan, and Simpai were sensitive against drought stress. Higher increased in leaf proline content after drought stress was observed in peanut cv. Jerapah and Singa, while less increased was observed in peanut cv. Gajah, Kelinci, Macan and Simpai. Total leaf sugar content of peanut cv. Jerapah, Gajah, Macan and Simpai decreased under drought stress while that of Singa and Kelinci were the same as that under optimal condition. The ability to increase leaf proline content and maintain total leaf sugar content under drought stress could be used as indicators of drought tolerance in peanut cultivars.

Abstract

Factors affecting the success of gene delivery into cells/tissues of rice mediated by particle bombardment have been investigated. However, their effects on regeneration capacity of rice explants have not been investigated. Objectives of this experiments were to evaluate the effects of particle bombardment on regeneration capacity of indica rice cv. Bengawan Solo calli and the effects of four regeneration media on the development of bombardment calli. Calli of indica rice were initiated from mature seeds on callus-inducing medium. They were transferred on osmoticum containing medium prior to bombardment and maintained on the medium up to two days. Particle bombardment was conducted at 16 cm bombardment distance, 27 cm Hg helium pressure and using standard procedures for preparation of microprojectile particles. The bombarded calli were cultured on RNB or RMS regeneration medium, with or without spermidine addition. Results of the experiment indicated particle bombardment affected the survival of explants of indica rice but did not affect the development of the explants. The development of explants was rather dependent on the regeneration media than on the particle bombardment treatment. RMS medium was better than RNB medium for shoots inducing from bombarded indica rice explants.

Abstract

Blast disease affected by Pyricularia grisea causes high percentage of yield losses in rice production. The improvement of durable Blast resistance is difficult due to the complexity of the inheritance of this trait. This study was conducted to evaluate the genetic control and inheritance of Blast resistance trait in interspesific population between IR 64 (accepted Indonesian rice type, medium resistant to Indonesian Blast pathogen) and Oryza rufipogon (AA genome; acc. No.IRGC#105491; donor for Blast resistance). Six populations, i.e. P1, P2, F1, F2, BCP1, and BCP2, were inoculated with three Indonesian races of Blast pathogen. The disease intensity was determined on the basis of disease leaf area (DLA). The three types of gene interactions (additive, dominance, and epistatis) were significantly involved in this trait. Among the digenic epistasis interactions, all of the additive x additive [i], additive x dominance (j) and dominance x dominance (l) contributed to the trait. Broad-sense heritability ranged from 50.30 up to 91.20%, while the narrow heritability ranged from 16.98 up to 73.20%. Abstract

Phosphorus (P) deficiency is one of major problems in rice cultivation. Development of a tolerant variety to P deficient soil is expected to reduce the needs of P fertilizer. The aim of this research was to evaluate the agronomic performance of Pup1-introgression rice lines. This research consisted of two separate experiments, an evaluation on rice grown in nutrient solution in the greenhouse of ICABIOGRAD, Bogor; and field evaluation at upland rice screening site Taman Bogo, Lampung. The rice lines used were BC2F3 derived from crossing of Situ Bagendit with Kasalath and NIL-C443, and from crossing of Batur with Kasalath and NIL-C443. Molecular analysis was conducted to verifiy the Pup1 introgression in the lines used and successfully confirmed the occurance of homozygote Pup1 segment in those lines. Evaluation of rice lines grown in nutrient solution was performed in a split plot factorial design, using the dose of Al (0 and 45 ppm Al3+) as main plots and dose of P (0.5 and 10 ppm P) as subplots. Field evaluation was performed in split plot design, with P fertilization (0 and 500 kg SP-18 ha-1) as main plots and BC2F3 lines as the subplots. Evaluation using Yoshida nutrient solution showed that the BC2F3 lines had greater total dry weight under low-P condition (37.5-112.5%), especially under Al-toxicity, compared to the respective parental varieties (Situ Bagendit and Batur). Field evaluation showed that the Pup1-introgression lines had greater shoot dry weight than the respective parental lines (10.5-74.82%). Abstract

Aromatic new plant type (NPT) rice lines were selected to obtain high yielding and aromatic lines. The objectives of the research were to study the yield stability and adaptability of 35 NPT rice lines across different environment, with Ciherang and Sintanur as check varieties. The lines planted at two locations, Bogor and Pusakanagara in two seasons (2009 dry and wet seasons, DS-WS). The experiment used  randomized complete block design (RCBD), with three replications. The 21-day-old seedlings were planted with spacing of 20 cm x 20 cm, with a plot size of 2 m x 5 m. Yield stability and adaptability were estimated by using coefficient regression (bi) and general mean of yield by Finlay-Wilkinson method. Combined analysis of variance showed that lines (G), environment (E), and the G x E interaction were significantly different. Lines showed different stability and adaptability. Several lines yielded higher than Ciherang variety. Nine lines were classified as stable and widely adapted at the marginal environment, i.e IPB 116-F-3-1, IPB 117-F-4-1, IPB-117-F-14-2, IPB-117-F-15-2, IPB-117-F-17-5, IPB 140-F-1-1, 140-F-IPB, 2-1, IPB 140-F-3, and IPB 149-F-2. Lines IPB 113-F-2, IPB 140-F-4, IPB 140-F-6, IPB 140-F-7, and B11738-MR-Si-1-2-1-2 were not stable and adapted only in optimum environmental condition (bi > 1) while IPB 116-F-46-1, IPB-117-F 17-4, IPB-117-F 18-3 and B11955-MR-84-1-4 has the value of bi < 1 or adaptable to marginal environments.   Keywords:  NPT rice, aromatic rice, yield stability

Abstract

water deficiency. Genetic engineering at the level of transcription factors (TF) is particulary a promising strategy in developing drought tolerant rice cultivar. HD-Zip genes are TF that function in plant adaptation to some environmental stresses including water deficit. The recombinant plasmid pC1301H Oshox6 which contained HD-Zip Oshox6 gene was placed under a drought inducible promoter called LEA promoter, gusA and hpt genes were driven with CaMV promoter. The aim of research was to obtain indica rice transgenic plants of Batutegi and Kasalath cultivars using pC1301H Oshox6 plasmid. Recombinant plasmid was transformed into immature rice embryos using Agrobacterium tumefaciens. Kasalath cultivar showed a better capacity to form embryogenic calli compared to Batutegi. Transformation efficiency of Batutegi is lower (1.5 - 0.3%) than Kasalath (2.2-28.3%). Regeneration efficiency is 25-83.3% and 7.7-100% for Batutegi and Kasalath, respectively. Number of putative transformant plantlets of Batutegi and Kasalath are 63 and 48 plantlets, respectively. Southern blot analysis (using hpt probe) on 12 independent lines of each Batutegi and Kasalath cultivars showed different gene copy number, ranging from one to four copies of gene.   Keywords: agrobacterium tumefaciens, LEA promoter, HD-Zip Oshox6, rice

Abstract

The objective of the research was to determine the roles of fertilizers application in improving rice ratoons. Statistical design used was a factorial randomized complete block design with three replications. The first factor was three levels of fertilizer rate (kg ha-1) i.e. 5 N + 27 P2O5; 45 N + 30 K2O; and 27 P2O5+30 K2O. The second factor was rice genotypes, i.e. Cimelati, Hipa-5 and Rokan varieties, and two lines of rice i.e. IPB106-7-47-DJ-1 and IPB106-F-8-1. The growth and production characters of main crops and ratoon were observed. The results showed that both genotypes and rates of fertilizer affected yield of ratoon. Following application of (kg ha-1) 45 N + 27 P2O5 three genotypes produced the highest ratoon, i.e. IPB106-7-47-DJ-1 and IPB106-F-8-1 lines and Rokan variety. Cimelati and Hipa-5 varieties produced higher number of ratoon with fertilizer rate (kg ha-1) 45 N + 30 K2O. Ratoon production of all genotypes ranged 38.1-56.6% of the main crop. This study clearly demonstrated that N, P and K fertilizer had important roles in increasing yield and yield components of rice ratoon. Keywords: fertilizer, productivity, rice genotypes, rice ratoon

Abstract

Breeding programs to improve rice varieties for tidal swamp areas have successfully developed a number of promisingbreeding lines. The objectives of this study were to evaluate the performance of ten advance breeding lines in multilocationyield trials and to analyze grain quality of the lines. The lines and two check rice varieties, IR42 and Batanghari, wereevaluated in replicated yield trials in six different tidal swamp environments. Grain quality of these lines were analyzed todetermine physical and chemical properties of the milled and cooked rice. Result from multilocation yield trials showedthat the breeding lines had higher yield potential compared to popular variety IR42 and their yields were comparable tothe control variety Batanghari. The lines showed different adaptability against different environments; some of the linesdemonstrated wide adaptability while the others showed specific adaptation ability. All of the lines had good grain qualitywhile they had different shape and texture which made them potential to be accepted in different regions. The result fromthis study indicated that all of the lines have potential to be commercially cultivated in tidal swamp areas. In addition, dataobtained from this study have been used in the registration of three lines as new varieties for swampy area namely IPB 1RDadahup, IPB 2R Bakumpai and Inpara 6.Keywords: adaptation, grain quality, multilocation trials

Abstract

The objectives of the study was to determine the effect of cutting height of main crop on the yield of rice ratoon, and to select genotypes with high productivity on tidal swamp land. The method used was a factorial randomized block design with three replications. The first factor was cutting height of 10 cm, 20 cm and 30 cm from soil surface. The second factor was five genotypes selected from previous study, namely Cimelati, Hipa-5, Rokan, IPB106-7-47-DJ-1 and IPB106-F-8-1. The results showed that combination of genotypes and cutting height affected ratoon yield.  Cutting height of 20 cm increased the number of filled grain in almost all genotypes, increased growth rate of ratoon to 1.5 cm day-1  and increased growth uniformity of ratoon of Cimelati, Hipa-5 and Rokan geontypes, with production 3.0 ton ha-1, 2.8 ton ha-1 and 2.2 ton ha-1, equal with 57.2%, 60.1%, 52.7%, of the main crop respectively.  Cutting height of 10 cm above soil surface showed the best results for IPB106-7-47-DJ-1 and IPB106-F-8-1 genotypes, with the highest yield of ratoon 2.1 ton ha-1 and 1.5 ton ha-1, equal with 45.8% and 35.4%  of the main crop respectively. Abstract

Twenty seven land races of tropical japonica rice were test-crossed with a &acute;WA&acute; type cytoplasmic male sterility (CMS) line IR58025A to evaluate their fertility restoration ability.  Based on pollen fertility and spikelet fertility of their F1 hybrids, genotypes are classified into maintainer or restorer.  The result showed frequency of maintainers among genotypes was higher than restorers.  Four genotypes, i.e., Ase Lapan, Ase Mandi, Hawara Bunar and Lampung Kuning were designated as maintainers.  These maintainers possess a number of desirable traits such as pest resistance and abiotic stresses tolerance; thus they will be useful for improving parental lines of hybrid rice.     Key words: Land race, test cross, fertility restoration
