Abstract

The  research  has  been  conducted  to  determine  hepatoprotective  effect  of  ethanolic  extract  of Acalypha  indica  L.  leaves  on  rats  (Rattus  novergicus)  induced  with  paracetamol.  The  study  was performed  following  a  completely  randomized  design, using  45 male  rats  (Rattus  novergicus)  divided into  5  groups.  Rats  in  group  1  (K1)  were  given  CMC  1  %  for  7  days  and  followed  by  aquadest administration 8 hours post given CMC on day 7. Group 2 was given CMC 1 % for 7 days and treated with paracetamol at dose 2,5 g/kg BW 8 hours post given CMC on day 7. Groups 3 (K3), 4 (K4), and 5 (K5) were pretreated with ethanolic extract of Acalypha indica L.  leaves at the dose of 50, 100, and 200 mg/kg BW for 7 days respectively, followed by paracetamol administration at the dose of 2.5 g/kg BW in the  next  8  hours  on  day  7. Paracetamol  hepatotoxicity  and  extracts  activity were measured  based  on SGPT and SGOT activity level on day 0, 8 hours after  last administration of  extract on day 7, and 24 hours post paracetamol administration. Data were analyzed using one way ANOVA. The result showed that  the  administration  of  ethanolic  extract  of Acalypha  indica  leaves  at  the  dose  of  50,  100,  and  200 mg/kg BW was  significantly  reduced SGPT  and SGOT  level  (P<0,05)  in  rats  that were  induced with paracetamol. Based on the results of SGPT and SGOT serum activity, it can be concluded that ethanolic extract of Acalypha indica L. potential as hepatoprotective agent.

Abstract

A research was conducted to find out an alternative diagnose in detecting toxoplasmosis in livestock/animal using intradermal test  from protein membrane of  T. gondii bradizoite stage. Local isolate ofmembrane T. gondii bradizoite stage  was used in the research. Ten of domestic sheep with the age of ± 1year and 10 mice strain Balb/c with the age ± 2 month were used in this research. The reaction ofhypersensitivity on the skin post protein membrane bradyzoite injection was indicated by the process ofskin thickening. The diameter skin thickening was measured using cutimeter, in which diameter e” 10 mmindicated positive diagnose. The result showed that optimal dosage of membran protein bradyzoite thatcould be applied to detect toxoplasmosis in livestock and animal using intradermal test were 0,6 ml and0,2 ml for sheep and mice respectively. The sensitivity and specificity level of antigen use (protein membrane)of T. gondii bradizoite stage from local isolate to diagnose toxoplasmosis in mice using intradermal testwere: 85.0 %  and 66.6 % respectively, while in sheep the sensitivity and specificity level were 85.0 % and66.6 % respectively. It can be concluded that  intradermal  test was appropriate to be implemented fordetecting toxoplasmosis in sheep and mice induced with tachyzoite T. gondii.

Abstract

The purpose of this research was to know the influence of dipping time in natural methyl testosterone hormone (MT) on successful male forming process of tilapia (Oreochromis niloticus) larvae. This research used 360 7 days old tilapia larvae, purchased from cultivation of brackish water, Ujung Batee, Banda Aceh. The samples were divided into 4 treatment groups with three repetitions, they were P0 (control group without treatment), PII (Tilapia larvae were dipped in natural MT for 10 hours), PII (Tilapia larvae were dipped in natural MT for 20 hours), and PIII (Tilapia larvae were dipped in natural MT for 30 hours). All tilapia larvae were maintained until the age of 60 days, before the sex of the fish examined. The average percentage of male tilapia in group P0, PI, PII, and PIII were 50.28, 87.77, 90.06, and 95.05%, respectively. Statistical analysis showed that there were significant differences (P<0.05) among P0, PI, PII, and PIII. Based on result of this research, it can be concluded that the longest dipping time the highest the percentage of male tilapia.Key words: larvae, tilapia, natural hormone methyl testosterone, the male sex percentage

Abstract

Penelitian ini bertujuan mengetahui gambaran histopatologis hepatopankreas udang windu yang terinfeksi hepatopancreatica parvovirus (HPV). Sebanyak 10 ekor udang windu yang berasal dari tambak rakyat digunakan sebagai sampel penelitian terdiri atas 5 sampel positif terinfeksi HPV dan 5 sampel normal. Semua udang dinekropsi untuk diambil hepatopankreas dan difiksasi dengan larutan Davidson. Selanjutnya hepatopankreas dibuat preparat histopatologis sesuai dengan prosedur teknik yang biasa dilakukan di Laboratorium Patologi Fakultas Kedokteran Hewan Universitas Syiah Kuala Darussalam Banda Aceh dan diwarnai dengan hematoksilin eosin (HE) serta diamati di bawah mikroskop. Hasil penelitian menunjukkan bahwa pada hepatopankreas udang windu yang terinfeksi HPV ditemukan adanya hipertrofi basofilik badan inklusi intranuklear, degenerasi, dan lisis sel.

Abstract

 The aim of this research was to observe behavior and gill profile of nile tilaphia (Oreochromis niloticus) induced by density stress. A total of 50 nile tilapia fish with the age of 3-8 months and 80-100 bwwere divided into 4 groups. Group I was control group contained 5 fish, group II,III, and IV contained 10, 15, and 20 fish, respectively. Treatment was performed for 24 hours, and fish behavior was observed every 3 hours. Gillsample was examined by anatomical pathology then tissue samples were collected and fixed in 10% Davidson solution, routinely processed,embedded in paraffin, sectioned at 5 microns, stained with hematoxylin and eosin (H&E), and examined by light microscopy. The data wasanalyzed descriptively. Behavior changessuch as swim up to water surface and rapid movement of operculum occur earlier than passivemovement and decreased reflex of fish on group II, III, and IV. The anatomical pathology of nile tilapia gills observed were darker colour ofgills, gill rott, and over mucus.Histopathological result showed that haematoma, necrosis, hyperplasia of primary and secondary lamella, andfusion lamella were observed in gills of nile tilapia induced by density stress.____________________________________________________________________________________________________________________Key words: histopathology, nile tilapia, gill, density stress

Abstract

This research was aimed to study histopathology of rat kidney infected with Trypanosoma evansi after administration of sernai leaves extract (Wedelia bifolora). The samples used were kidneys of 25 male rats strain Wistar which were equally divided into 5 treatment groups. Group P1 was uninfected and untreated control, group P2 was infected with 1x103 T. evansi but not sernai leaves extract, rats from groups P3, P4, and P5 were infected with 1x103 T. evansi and given sernai leaves extract with the dose of 30 mg/kg bw, 45 mg/kg bw, and 60 mg/kg bw respectively. The sernai extract was administered orally using a stomach tube for 3 consecutive days. The next day after last treatment, all rats were sacrificed and necropsied. Kidneys were collected for histopathological examination such as degeneration and necrosis of tubules cells, convoluted contortus duct lumen, and adhesion and atrophy of glomerulus. Sernai leaves extract with the dose of 45 mg/kg bw was the most effective dose to prevent kidney damage due to T. evansi infection.

Abstract

The objective of this research is to know Aceh cattle follicle dynamic under environmental heat stress condition. This research was conducted on December 2012 until August 2013. Animals used in this research were 20 Aceh cattle aged 5-8 years old, weighing of 150-250 kg, and had at least 2 regular cycles. All cattle used were clinically in good body condition score. The cattle were divided into two groups of 10, used in two different period of times (December-January and July-August) to examine the effect of environmental heat to their follicle dynamic. Each group is separated into two different keeping management; one group was kept in pens while the others in the pasture, 5 cattle in each management group. Research timing was based on information acquired from Indrapuri Agency for Meteorology, Climatology, and Geophysics (BMKG) which predicted that extreme weather would last from July to August while December to January would be relatively normal in Aceh region. All cattle’s estrous cycle were synchronized by 5 mg/mL of PGF2α intramuscularly (Lutalyse™, Pharmacia & Upjohn Company, Pfizer Inc.). Ultrasonography examination was performed to monitor ovary follicle’s growth and dynamic during one cycle. Days during ovulation marked by standing heat was regarded as Day 0 of estrous cycle. Follicular dynamic examination during estrous cycle on December-January and also on July-August, both penned or pastured cattle showed the follicle waves that was 3 follicle growth waves. The size of follicle growth on first wave (1st DF) on cattle kept in pen on July-August and December-January were relatively similar. The size of follicle growth on first wave for cattle kept in pasture on July-August and December-January were relatively similar. To conclude, Aceh cattle’s follicle dynamic is not change during environment heat stress condition and is not affected by different cattle-keeping management.

Abstract

Penelitian ini bertujuan mempelajari gambaran histopatologi hati tikus putih (Rattus norvegicus) akibat mengonsumsi minyak jelantah.  Duapuluh ekor tikus berumur 3 bulan dengan bobot badan ±250 g dibagi atas 4 kelompok perlakuan dan setiap kelompok terdiri at as 5 ekor. Perlakuan K1 (minyak goreng curah), K2, K3, dan K4 (minyak jelantah 3x, 6x, dan 9x pemakaian). Perlakuan dilakukan selama 60 hari, dan pada hari ke 61 dilakukan eutanasia yang dilanjutkan dengan nekropsi. Hati tikus diambil dan difiksasi dalam larutan buffered neutral formaline 10%untuk diproses sediaan histopatologis dan diwarnai dengan hematoksilin dan eosin (HE). Hasil penelitian menunjukkan bahwa hati berwarna coklat kehitaman, bengkak, dan ditemukan nodul multi fokal dengan konsistensi padat. Abstract

The Purpose of  this study were to identify the type of endoparasite and to find out the prevalance of endoparasite infected digestive tract of Colossoma macropomum. The study was conducted at parasitology Laboratory of Veterinary Medicine Faculty, Syiah Kuala University in May - June 2015. The sample used were collected randomly. As many as 10% of total fish in a population. The exa mination of endoparasite were carried out using smear method. Parameter measured were prevalence and physic – chemical of water. The results showed that type of endoparasite found in digestive tract of Colossoma macropomum fish was Capillaria sp. with the prevalence of 15%. Tujuan penelitian ini untuk mengidentifikasi jenis endoparasit yang menginfeksi dan mengetahui prevalensi endoparasit yang menginfeksi saluran pencernaan ikan bawal air tawar. Penelitian dilakukan di Laboratorium Parasitologi Fakultas Kedokteran Hewan, Universitas Syiah Kuala pada Bulan Mei dan Juni 2015. Penelitian  menggunakan metode smear method  dan penyamplingan sampel dilakukan secara acak. Sampel yang digunakan dalam penelitian sebanyak 10% dari total ikan dalam satu populasi. Parameter yang diukur yaitu prevalensi dan parameter fisika – kimia air. Hasil dari penelitian menunjukkan bahwa jenis endoparasit  yang teridentifikasi pada ikan bawal air tawar adalah Capillaria sp. Nilai prevalensi yang diperoleh sebesar 15%.

Abstract

Penelitian ini bertujuan mengetahui gambaran histopatologi insang ikan nila yang dipelihara dalam temperatur air di atas normal. Dalam penelitian ini digunakan 12 sampel ikan nila yang dibagi atas 4 kelompok. Kelompok I adalah perlakuan kontrol dengan temperat ur air 28° C, sedangkan ikan pada kelompok II, III, dan IV dipelihara masing-masing pada temperatur 30, 32, dan 34° C selama 6 jam, mulai dari jam 09.0015.00  WIB. Kemudian insang sampel difiksasi dalam larutan Davidson 10% selanjutnya dilakukan pembuatan sediaan histologi dengan menggunakan pewarnaan humatoksilin dan eosin (HE). Pengamatan histopatologi dilakukan dengan mikroskop cahaya biokuler, kemudian dilakukan pemotretan dengan fotomikrograf. Data yang diperoleh dianalisis secara deskriptif. Hasil pemeriksaan histopatologi terhadap insangikan nila ditemukan adanya epitelium terlepas, hiperplasia lamella primer, hiperplasia lamella sekunder, nekrosis, dan fusi lamella pada insang ikan nila yang  dipelihara dalam temperatur air di atas normal.
