Abstract

First generation (F1) of hatchery produced humpback grouper (Cromileptes altivelis) has been characterized genetically in order to serve the information of their status in related to their breeding strategy. PCR-RFLP method was used to detect the variation of mtDNA D-loop region of F1 population at BBPBL Lampung and BBAP Situbondo. The result of study showed that reducing of haplotype diversity had been arised from broodstock (0.8548) to F1 generation population (0.7473; 0.7273; and 0.6947, respectively).  Genetic divergence that had found between population BBPBL Lampung and BBAP Situbondo make it possible to do outbreeding in order to get its heterosiss effect. Keywords: mtDNA, haplotype diversity, genetic differentiation, Cromileptes altivelis   ABSTRAK Ikan kerapu tikus (Cromileptes altivelis) generasi pertama (F1) hasil domestikasi di hatchery telah dikarakterisasi secara genetik untuk menyediakan informasi status sehubungan dengan program pemuliaannya.  Metode PCR-RFLP digunakan untuk mendeteksi variasi sekuens D-loop mtDNA ikan kerapu tikus F1 yang diproduksi di BBPBL Lampung dan BBAP Situbondo.  Hasil penelitian menunjukkan bahwa telah terjadi penurunan keragaman haplotipe dari induk (0,8548) ke populasi generasi F1 (masing-masing 0,7473; 0,7273; dan 0,6947).  Adanya keragaman genetik antara populasi ikan kerapu tikus di BBPBL dan BBAP Situbondo memungkinkan dilakukannya outbreeding untuk mendapatkan efek heterosis. Abstract

The key success of producing grouper seed depends on the availability of high and complete nutritive live food  (zooplankton) during larvae development stage. Rotiferand Artemia are common zooplankton that has been used in most of grouper hatchery. The cysts of Artemia is expensive because it’s still imported, therefore potential zooplankton from Indonesians waters should be developed to replace or to substitute Artemia. Copepod Oithona sp can be found easily in Indonesian marine waters. Oithona sp can be developed to adult stage within 7 days. One female Oithona can produce 8 to 22 nauplii with average 14,39 ± 3,62 nauplii. The larva ofhumpback grouper (Cromileptes altivelis) fed by Oithona sp showed a higher DHA content (0,30 % b/b) than those fed by copepod + Artemia (0,15 % b/b) and and Artemia only (0,03 % b/b). The EPA content in larva fed by Oithona also showed thehighest, 0,30 % b/b. However, improvement technique of monoculture Oithona should be carried out due to mass mortality and contamination by protozoan and rotifer.

Abstract

An evaluation of the quality of the Black Tiger Broodstock Shrimp (Penaeus monodon) in the southern coastal area of Pangandaran, West Java was conducted during period of Juny-October 2005. The evaluation was focused on the waterquality identification and the quality of the broodstock collected from the farmer sorround the Pangandaran Area. The result of the evaluation showed that the water quality of the coastal area of Pangandaran was good compare to the northern coastal area of West Java in which the organic and inorganic pollutantwas . This situation was affected on the quality of the  broodstock. The identification result on the quality of broodstock using polymerase chain reaction (PCR) method shows that the broodstock shrimp of the Black Tiger Shrimp(Penaeus monodon) in the coastal area of Pangandaran is free from the White Spot Syndrome Virus (WSSV) and Infectious Hypodermal and Hematopoietic Necrosis Virus (IHHNV). From 99 broodstock identified, only 4 indicated by very light contaminated WSSV and 2 by IHHNV. This condition shows that in general the quality of the broodstock shrimp of the Black Tiger Shrimp (Penaeus monodon) in the coastal area of Pangandaran was excellent and these sources are potentialto be applied as broodstock sources for developing the Black Tiger Shrimp (Penaeus monodon) culture in the national level.

Abstract

An evaluation of the environmental status of the northern coastal area of Karawang was conducted in the wet and the dry season to understand their feasibility to support aquaculture development. Theevaluation result showed that in the wet season, the water quality of the coastal and breackishwater ponds was poor due to high concentration of total suspended solid (TSS), high turbidity, ammonia,nitrate, nitrite and sulfide that stimulated phytoplankton bloom which indicated by high concentration of chlorophyll-a. To anticipate the water quality degradation, it is necessary to build a sedimentationfacility along the inlet channel which accomapied by seaweed and mangrove plantation to eliminate and reduce an organic waste that entered to the breackishwater ponds. An application of the“Integrated Multi-Trophic Aquaculture (IMTA)” in the coastal area in the future is the best prospective to develop sustainable aquaculture. This technology is green technology with the biorecycle systemand zero emition. Hasil evalusi menunjukkan bahwa pada musim hujan telah terjadi penurunan kualitas air baik di perairan pantai maupun perairan tambak yang diindikasikan dengan meningkatnyakekeruhan dan konsentrasi ammonia, nitrate, nitrit dan sulfide yang memacu terjadinya ledakan (blooming) fitoplankton (klorofil-a). Untuk mengantisifasi penurunan kualitas air, perlu dikembangkanbak pengendapan disepanjang pantai yang disertai penanaman rumput laut dan mangrove untuk mengurangi kekeruhan dan menetralisir limbah organik yang masuk ke pertambakan sebelumdigunakan untuk kegiatan budidaya perikanan. Penerapan Teknologi Budidaya Perikanan terintegrasi “Integrated Multi-Trophic Aquaculture (IMTA)” di wilayah pesisir dimasa mendatang merupakanprospek yang menjanjikan untuk pengembangan teknologi budidaya perikanan berkelanjutan, karena teknologi ini selain lebih produktif juga merupakan teknologi bersih (green technology) danberwawasan lingkungan karena teknologinya bersifat Zero Emition atau bebas limbah. Abstract

The environmental characteristics of seed and broodstock of the grouper, a reef fish species was investigated in the Pangkep water area, South Sulawesi to understand the water quality of the physical and chemical parameters condition. The environment of the Pangkep area was found in a good condition and it is suitable for habitat of the grouper as a reef fish though in some area the reef has degraded due to the utilization of an illegal fishing devise such bomb and cyanide. But application of the local wisdom in some area of Pangkep has succeeded in protecting the reef and fish population to serve the life of the islands people.Keywords: The environmental characteristics, seed and broodstock, grouper, Pangkep

Abstract

Vaksin DNA Streptococcus iniae adalah vaksin generasi ketiga yang mengandung gen penyandi antigen vaksin. Pgm adalah protein DNA-binding yang mengaktifkan ekspresi gen virulensi, termasuk polisakarida kapsul protein. Konstruksi gen pgm yang dikendalikan oleh promoter MBA telah berhasil dibuat dan merupakan kandidat vaksin DNA untuk mengendalikan infeksi S. iniae pada ikan nila. Tujuan dari penelitian ini adalah untuk produksi dan aplikasi vaksin DNA S. iniae untuk meningkatkan imunitas ikan nila. Produksi vaksin dilakukan dengan metode isolasi plasmid dari bakteri Escherichia coli dH5α yang membawa konstruksi pMBA-pgm. Verifikasi vaksin DNA dilakukan dengan metode PCR menggunakan primer Fstrep dan Rstrep dengan ukuran target 1713 bp. Aplikasi vaksin DNA dilakukan pada dosis 25, 50,75, dan 100 ng/ml melalui metode suntik intraperitonial dan uji tantang menggunakan bakteri S. iniae dengan kepadatan 108 cfu/ml. Parameter yang diamati adalah persentase kelulushidupan (Survival rate/ SR), persentase RPS (Relative Percentage Survival), MTD (Mean Time To Death), keberadaan gen pgm, dan histologi organ hati, mata, otak, limpa dan ginjal. Produksi vaksin berhasil dilakukan menggunakan metode isolasi plasmid dan mengandung gen pgm dengan ukuran 1713 bp. Aplikasi vaksin DNA S. iniae menunjukkan bahwa dosis vaksin optimal adalah 50 ng/ml dengan nilai SR 96,667%; nilai RPS 88,461% dan MTD 4,6 hari. Histologi ikan nila yang terinfeksi S. iniae menunjukkan mata ikan nila mengalami opacity dan exopthalmia serta jaringan organ (hati, limpa dan ginjal) mengalami necrosis. Vaksin DNA telah berhasil diproduksi menggunakan metode isolasi plasmid dan telah diaplikasikan pada ikan nila dengan dosis optimum 50 ng/ml.

Abstract

Penelitian ini dilakukan untuk mengkaji performa ikan nila hasil sex reversal (SRV), genetically male tilapia (GMT), dan YY pada fase pendederan pertama di akuarium. Benih ikan dipelihara selama 22 hari, dari umur 6 hari hingga 28 hari. Parameter yang diamati meliputi tingkat sintasan, persentase ikan jantan, laju pertumbuhan, dan biomassa. Hasil penelitian menunjukkan bahwa tingkat sintasan tidak berbeda (P>0,05) antar ketiga kelompok ikan dan kontrol (KN), berkisar antara 85,30%--86,20%. Persentase ikan jantan antara SRV (94,5% ± 1,32%) vs. GMT (93,8% ± 1,25%) dan GMT vs. YY (90,2% ± 1,83%) tidak berbeda (P>0,05), sedangkan antara SRV lebih tinggi daripada YY (P<0,05). Persentase ikan jantan pada ketiga kelompok ikan tersebut lebih tinggi (P<0,05) dibandingkan dengan KN (56,9% ± 3,62%). Pertumbuhan ikan YY dan GMT lebih cepat (P<0,05) dibandingkan dengan ikan SRV dan kontrol (KN). Bobot rata-rata ikan YY pada akhir penelitian mencapai 485 mg, ikan GMT 456 mg, ikan SRV 379 mg dan kontrol 342 mg. Produksi biomassa ikan YY, GMT, dan SRV masing-masing sebesar 41,3%; 32,9%; dan 10,3% lebih tinggi dibandingkan dengan KN. Dengan performa yang tinggi dan pertimbangan teknis di lapangan, benih GMT merupakan alternatif yang baik untuk dibudidayakan dalam rangka meningkatkan produksi ikan nila.The experiment was conducted to determine the performance of sex reversed (SRV), genetically male tilapia (GMT), and YY tilapia on first nursery phase in aquarium. Fry were reared for 22 days, from 6 to 28 days-old. Survival rate, percentage of male fish, growth rate and biomass were observed. The result of the study showed that survival rate among fish group and control were similar (P>0.05), ranged from 85.30%-86.20%. Percentage of male fish between SRV (94.5% ± 1.32%) versus GMT (93.8% ± 1.25%) and GMT versus YY (90.2% ± 1.83%) were also similar (P>0.05), while SRV is higher than YY (P<0.05). Percentage of male fish in the three fish groups was higher than that of control (56.9% ± 3.62%). Growth of YY fish and GMT were higher compared to SRV and control fish (KN). The mean weight of YY fish at the end of the experiment reached 476 mg, GMT fish 447 mg, SRV fish 379 mg and control 342 mg. Biomass of YY, GMT and SRV fish were respectively higher by 41.3%, 32.9%, and 10.3% compared to control. With high performance and technical consideration in farm, GMT fish can be a potential alternative to be cultured in fish farm in order to increase aquaculture production of nile tilapia.

Abstract

Salah satu spesies ikan yang menjadi target produksi perikanan budidaya nasional adalah ikan kerapu tikus (Cromileptes altivelis). Ikan kerapu tikus merupakan ikan laut budidaya komoditas ekspor, namun laju pertumbuhannya sangat lambat. Oleh karena itu, perlu dilakukan usaha budidaya yang mampu menaikkan laju pertumbuhan ikan kerapu tikus. Pendekatan nutrisi melalui penggunaan hormon pertumbuhan (Growth Hormone, GH) pada usaha budidaya diyakini mampu meningkatkan kecepatan tumbuh ikan budidaya. Pada penelitian ini, melalui teknologi DNA rekombinan telah dilakukan isolasi dan identifikasi gen GH yang selanjutnya dilakukan produksi protein rekombinan GH (recombinant Growth Hormone, rGH) dengan memanfaatkan penggunaan bakteri E. coli. Dalam pelaksanaan penelitian dilakukan konstruksi rGH yang menghasilkan bakteri E. coli BL21 (DE3) yang mampu memproduksi protein rGH. Produksi rGH dilakukan pada skala bioreaktor. Proses isolasi produk rGH-nya dalam bentuk pellet inclusion bodies yang selanjutnya dicampur dengan pelet pakan komersil hingga konsentrasi akhir protein dalam pakan mencapai 1 ng/6 mg pakan, di mana setelah dikering-anginkan, pelet pakan protein rekombinan GH dapat diaplikasikan untuk budidaya ikan kerapu tikus dan ikan budidaya lainnya.

Abstract

An evaluation of the hydro-oceanography characteristics and fisheries resources was conducted in the Seribu Island area, an offshore area of the Jakarta Bay. The area was influenced by monsoonal system that affected the oceanography and water quality situation. The bathymetry within this area was a range of 3 – 50 m with shallow water depth in and surround the islands. While the wave, current speed and tidal range were around 0,5-3,0 m; 5-49 cm/sec and 80 cm, respectively. Water quality status was varied among the area and it is depend on the distance from Jakarta Bay. In the middle and upper part of the area, water quality was within the moderate to the excellent and this condition has promoted the area as potential area of fisheries for both fish culture and fishing. As indication, an abundance of the benthic organism (sea urchin), milk fish, reef fish, seaweed and pelagic fish was found within this area. Keywords: Hydro-oceanography, fisheries resources, Kepulauan Seribu
