Abstract

Ram spermatozoa are sensitive to extreme changes in temperature during the freeze-thawed process. The present study was conducted to examine the effects of addition of various sugars in Tris extender on sperm cryosurvival of Garut ram. Semen was collected using an artificial vagina from three mature rams once a week. Immediately after initial evaluation, semen was divided into five parts and diluted with Tris extender (control), Tris extender + 0.4% dextrose, Tris extender + 0.4% raffinose, Tris extender + 0.4% trehalose, and Tris extender + 0.4% sucrose, respectively. Semen was loaded in to 0.25 ml mini straw with the concentration of 200 million or 800 million motile spermatozoa per ml. Semen was equilibrated at 5oC for three hours, then frozen and stored in liquid nitrogen for seven days. Quality of processed-semen including percentages of motile spermatozoa (MS), live spermatozoa (LS), intact acrosome cap (IAC), and intact plasma membrane (IPM) were evaluated after dilution, equilibration, and thawing, respectively. Data were analyzed using completely randomized design with five treatments and six replicates. Means were compared significant difference test at 0.05 significant level. Results of this research showed that there was no significantly difference (P>0.05) between treatments for all sperm quality parameters after dilution and equilibration. Mean percentages of post thawing MS, LS, IAC, and IPM for dextrose (54.00; 68.00; 66.60, and 57.83%), raffinose (50.00; 64.33; 61.80, and 61.75%), trehalose (50.83; 65.67; 61.40 and 57.75%), and sucrose (49.00; 66.80; 58.50 and 58.50%) were significantly (P<0.05) higher than control (40.83; 52.67; 54.60, and 49.40%) respectively. In conclusion, addition of 0.4% dextrose, raffinose, trehalose or sucrose in Tris extender are effective in improving frozen semen quality of Garut ram. Abstract

Cauda epididymal spermatozoa could be used as an alternative source of gamete in the application of various reproductive technologies, because the spermatozoa is motile and has ability of fertilizing the oocyte. The purpose of this research was to examine the effect of addition of seminal plasma on the quality of ram cauda epididymal spermatozoa preserved at 3–5oC. Collected-spermatozoa was divided into five parts then centrifuged at 3,000 rpm for 30 min and the supernatant was removed. One-fifth was diluted with Tris extender (Tris) and the remaining was added 0.5 ml ram seminal plasma then diluted with Tris extender (PS-Tris), 15% AndroMed + 85% distilled water (PS-AM15), 20% AndroMed + 80% distilled water (PS-AM20), and 25% AndroMed + 75% distilled water (PS-AM25), respectively. Quality of collected-spermatozoa including spermatozoa concentration (SC), percentages of motile spermatozoa (MS), live spermatozoa (LS), abnormal spermatozoa (AS), cytoplasmic droplet (CD), and intact plasma membrane (IPM) were evaluated. Percentages of MS, LS, and IPM of diluted-spermatozoa was evaluated every day during preserved at 3–5oC for three days. Results of this study showed that mean SC, MS, LS, AS, CD, and IPM of fresh spermatozoa were 11,660 million/ml, 65, 81, 7.6, 10.2, and 82.2%, respectively. Addition of seminal plasma in cauda epididymal spermatozoa prior to dilution could maintain the quality of spermatozoa during preserved at 3–5oC for three days. At day-4 of storage, percentages of MS, LS, and IPM for PS-Tris (43, 58, and 59.2%), PS-AM20 (40.5, 53.75, and 53.75%), and PS-AM25 (40, 54.8, and 55.2%) were significantly (P<0.05) higher than Tris (21, 34.8, and 33.6%) and PS-AM15 (20, 40, and 42.2%). In conclusion, addition of seminal plasma in ram cauda epididymal spermatozoa prior to dilution with Tris or 20 and 25% AndroMed extenders could maintain the quality of spermatozoa during preserved at 3–5oC for three days, and could be used for artificial insemination or in vitro embryo production programs. Abstract

Integrasi usahatani ternak merupakan pilihan untuk menunjang peningkatan pendapatan petani baik yang ada di pedesaan maupun di daerah pingiran kota yang masih memiliki lahan pertanian khususnya persawahan. Penelitian ini bertujuan (1) mengetahui tingkat pendapatan petani padi sawah dengan usaha tunggal di Kawasan Persawahan Amohalo (2) menganalisis potensi peningkatan pendapatan petani dengan mengintegrasikan usaha tani padi sawah dengan usaha ternak itik, (3) menganalisis keberhasilan dan kemanfaatan secara ekonomi pada usaha tunggal dan usaha integrasi padi sawah dengan usaha ternak itik. Responden dalam penelitian ini adalah petani padi sawah yang memiliki ternak dengan jumlah 33 petani.  Penentuan responden dilakukan  secara acak sederhana yang selanjutnya dilakukan analisis pendapatan. Hasil penelitian menunjukkan bahwa(1) pada usahatani tunggal, jumlah pendapatan bersih yang diperoleh petani padi sawah di Kawasan Persawahan Amohalo adalah sebesar Rp7.716.295,58,- per tahunnya,- atau hanya sebesar Rp643.024.63,- per bulanya, (2) Pada usahatani integrasi, jumlah pendapatan bersih  yang diperoleh petani di Kawasan Persawahan Amohalo adalah sebesar Rp14.464.495,58,- per tahunnya,- atau sebesar Rp1.205.374,63,-,dan (3) pola usahatani padi sawah yang terintegrasi dengan ternak itik, lebih efisien dan menguntungkan dari pada usahatani tunggal. Pada usahatani tunggal diperoleh nilai R/C ratio sebesar 1,69 sedangkan usahatani yang terintegrasi diperoleh nilai R/C ratio sebesar 2,05.Kata kunci: Pengembangan, Integrasi usahatani ternak, pendapatan

Abstract

Penelitian ini dilaksanakan dengan tujuan untuk mengetahui karakteristik habitat burung maleo di TNRAW, yang berlangsung pada bulan Maret-April 2013 di Resort Langkowala Desa Watu-watu, kecamatan Lantari Jaya, TNRAW Kabupaten Bombana, Sulawesi Tenggara. Survei lapangan dilakukan untuk mengamati kondisi habitat maleo, mengidentifikasi flora dan fauna, mengambil sampel yang diperlukan untuk pengamatan laboratorium dan melakukan wawancana langsung dengan petugas TNRAW dan masyarakat di sekitar lokasi pengamatan. Pengamatan suhu dan kelembaban udara dilakukan 3 kali sehari, yaitu pagi, siang dan sore hari. Hasil penelitian menunjukkan bahwa: (1) bahwa habitat burung maleo di kedua lokasi terdiri dari pohon-pohon dengan ketinggian yang berkisar antara 15-30 meter dan semak belukar, tetapi kedua lokasi memiliki perbedaan jenis dan jumlah vegetasi, (2) pada lokasi pertama, kedalaman lubang bertelur rata-rata 43.33 cm dan lokasi kedua rata-rata 42.66 cm, (3) pada pukul 08.00, sarang bertelur maleo di lokasi pertama memiliki temperatur 28,930C dengan kelembaban 94,75%, sedangkan lokasi kedua adalah 29,960C, dengan kelembaban 94,04%. Pada pukul 13.00, temperatur di dua lokasi bertelur tidak jauh berbeda, yaitu sekitar 31,86oC, dengan kelembaban 94%. Pada pukul 17.00, di lokasi peneluran pertama memiliki temperatur sekitar 32,110C dengan kelembaban 93,98%, sedangkan pada lokasi kedua adalah 31,970C, dengan kelembaban 93,59%.Secara umum dapat disimpulkan bahwa karakteristik habitat burung maleo di TNRAW dicirikan oleh adanya pepohonan dengan tinggi berkisar 15–30 meter dan semak belukar. Disamping itu, tekstur tanah yang dominan adalah berpasir, dengan kedalaman lubang peneluran berkisar 35–55 cm dan rata-rata 43 cm.Kata kunci : maleo, karateristik habitat, TNRAW

Abstract

This study aims to improve the genetic quality of local chickens through genetic approaches and phenotypic characterization for selection purposes. The cGH gene (Chicken Growth Hormone) is one of the genes responsible for local chicken growth traits. Tolaki chicken is a local chicken from Southeast Sulawesi. This study uses the phenotyping method of production and genotyping traits with PCR technique. A total of 50 parents of tolaki chicken were kept 4 weeks. The results showed average body weight of tolaki chicken ranged from 1,395 kg - 1,910 kg, average body weight gain was 58,78 (g / head / week), feed consumption was 81,37 (g / head / day) and ration conversion 9,68.  Body weight gain in males was slightly higher (60.30 g / head / mg) compared to females (58.42 g / head / mg). In this study, the presence of the tolaki chicken cGH gene was identified through DNA extraction and amplification through a PCR machine with a length of DNA section of 399 bp.

Abstract

ABSTRAK Penelitian ini bertujuan untuk mengetahui daya tetas dan lama menetas telur ayam tolaki pada mesin tetas dengan sumber panas yang berbeda. Penelitian dilaksanakan selama 5 bulan di kandang pembibitan Fakultas Peternakan UHO. Ayam tolaki yang digunakan terdiri atas 5 ekor jantan dan 15 ekor betina umur sekitar 20 bulan. Perkawinan ayam dilakukan dengan cara IB. Parameter yang diamati adalah: fertilitas, daya hidup embrio, daya tetas, bobot tetas dan lama menetas. Hasil penelitian menujukkan: (1) rataan fertilitas telur ayam tolaki pada mesin tetas PL adalah 58,57% dan mesin tetas PLM adalah 46,88%, namun kedua mesin tetas secara statistik tidak berbeda nyata, (2) rataan DHE pada mesin tetas PL adalah 96,67% dan mesin tetas PLM adalah 89,58%, (3) rataan daya tetas pada mesin tetas PL adalah 45,61% dan mesin tetas PLM adalah 64,81%, (4) rataan bobot tetas, pada mesin tetas PL adalah 26,47 g, sedangkan mesin tetas PLM 26,96 g, dan (5) lama menetas telur pada mesin tetas PL adalah 21.05 hari dan PLM adalah 21,09 hari. Secara statistik, penggunaan mesin tetas dengan sumber panas berbeda tidak berpengaruh nyata terhadap semua parameter yang diamati.Mesin tetas dengan sumber panas berbeda tidak berpengaruh nyata terhadap fertilitas, DHE, daya tetas, bobot tetas dan lama menetas telur ayam tolaki. Untuk meningkatkan daya tetas direkomendasikan menggunakan mesin tetas dengan sumber panas kombinasi listrik dan lampu minyak. Kata Kunci : Fertilitas, DHE, Fertilitas Lama Menetas, Mesin Tetas dan Ayam Tolaki.
