Abstract

Physical characterization and equivalence testing of generic and branded names of phenylbutazone tablets have been done. The information about quality of generic drugs is expected to increase the use of generic drugs by health practitioners and public. Dissolution test carried out by in vitro had correlation with bioequivalence by in vivo test. Sample of dissolution testing were generic (G) and branded (A and B) names of 200 mg phenylbutazone tablets. Physical characteristic test performed by uniformity weight test, hardness and disintegration time. Dissolution test were conducted by using basket and spectrophotometry ultra violet method in 200-400 nm wavelength. The result of uniformity weight of generic drug and branded drug was 352,02 mg and 352,82 mg, the hardness of generic was 9,58 kgf and branded was 9,50 kgf, and disintegration test result of generic was 16,1 minutes and branded was 3 minutes. The result of dissolution testing were dissolution profile and dissolved concentration at 30 minutes that compared to qualify of USP XXXII. Relative bioavailability testing of generic to A brand was 101.580 and generic to B brand was 105.275. Based on statistical test, there was no significant different or pharmaceutical equivalent. The generic tablets were equivalent to A and B branded names tablets with similar factors 82.120 and 74.271.Keywords: Equivalence, Dissolution, Salbutamol tablet; generic name, branded name

Abstract

Pengujian aktivitas antimalaria dengan menggunakan ekstrak air bunga Carthamus tinctorius L. pada biakan in vitro plasmodium falciparum strain 3D7 yang sensitif terhadap klorokuin telah dilakukan. Uji aktivitas dilakukan dengan cara melarutkan bahan uji dalam DMSO kemudian dibuat serial pengenceran dalam media RPMI sampai diperoleh konsentrasi akhir sebesar 100 µg/ml, 10 µg/ml, 1 µg/ml, 0,1 µg/ml dan 0,01 µg/ml. Pada larutan uji ditambahkan suspensi parasit dengan kadar parasitemia ±1% dan hematokrit 5%. Kultur diinkubasi selama 48 jam pada suhu 37oC. Hasil menunjukkan pengujian aktivitas antimalaria dari ekstrak air bunga Carthamus tinctorius L. memiliki daya hambat terhadap pertumbuhan Plasmodium falciparum strain 3D7 sebesar 52,68%.Kata Kunci : Bunga Carthamus tinctorius L, Antimalaria, Plasmodium falciparum 3D7, dan Ekstrak air

Abstract

Bunga Kasumba turate (Carthamus tinctorius Linn) dari suku Asteraceae merupakan tumbuhan obat tradisional etnis Sulawesi Selatan yang secara empiris digunakan dalam pengobatan campak. Ekstrak etanol dari kasumba turate memberikan peningkatan aktivitas imunoglobulin G (IgG) dan  aktivitas imunoglobulin A (IgA) yang sangat signifikan. Penelitian menunjukkan ekstrak etanol bunga C. Tinctorius L memiliki potensi sebagai antimalaria secara in vitro dengan nilai IC50 sebesar 1,06 µg/ml. Sampel bunga C. tinctorius L. kering sebanyak 3 Kg dicacah hingga menjadi serbuk sebanyak 870 gram kemudian diekstraksi secara maserasi dengan menggunakan pelarut etanol 80%. Hasil maserasi dipekatkan dengan vacuum rotary evaporator pada suhu 50°C sehingga diperoleh ekstrak kental sebanyak 178,7gram. Profil fitokimia menunjukkan ekstrak etanol bunga C. tinctorius L. mengandung senyawa metabolit antara lain saponin, terpenoid, flavonoid, tanin dan antrakuinon.Kata kunci: Skrining, ekstrak, kasumba turate, obat tradisional, antimalaria

Abstract

Theophylline is a methyl xanthine of alkaloid derivative compound has been used as a bronchodilator in the treatment of chronic lung disorders, especially asthma. Oral administration of theophylline causes inconvenient for patients including bitter taste, narrow therapeutic range, and indigestion due to increased of gastric acid secretion. Therefore, other administration of theophylline has been developed as transdermal delivery system. This study disscused about preparation of theophylline in ethosome as vesicular carrier. Preparation of ethosome was used hot method (40oC) and cool method (30oC). Characterization of ethosomes includes observation of shape and size of vesicles by microscopic method and the vesicles were fractionated by centrifugation then entrapment efficiency (% EE) were measured by spectrophotometric method. Then optimazed theophylline that entrapped in ethosome. Based on the results, ethosomes preparation by hot method were produced Small Unilamellar Vesicle (SUV), the size were 0,0522-0,100 µm, and the highest value of % EE was 98.95%, while ethosomes preparation by cold method were produced Small Unilamellar Vesicle (SUV), the size were 0.0522-0.100 ?m, and the highest value of % EE was 99.80%. The optimum concentration of theophylline was entrapped in etosom was 0.1%. It was concluded that cold method was the appropriate method for preparation of theophylline in ethosome as vesicular carrier.Keywords: Etosome, theophylline, vesicular, transdermal
