Abstract

The purpose of this research was to know the effect Bawang Dayak (Eleutherine palmifolia Merr) in Saprolegnia sp infection of nile tilapia (Oreochromis sp) with smear method. This research used Completely Randomized Desin consisted one factors with three treatments and three replicates (A: 10 ppt/ind), (B: 20 ppt/ind), and (C: control). The research using aquarium and nile tilapia (5-8 cm body length) with density of 2 ind/aquarium. Parameters observe were inhibitory test, recovery time, survival rate, and water quality. The result showed  that optimal consentration Bawang Dayak (Eleutherine palmifolia Merr) is 20 ppt.

Abstract

Tujuan penelitian ini untuk mengetahui tingkat protein dan energi pakan terbaik pada ikan sepat. Manfaat penelitian ini untuk dapat dijadikan informasi dasar mengenai tingkat protein dan energi yang sesuai bagi kebutuhan konsumsi pakan dan pertumbuhan benih ikan sepat. Ikan uji yang digunakan ikan sepat (Trichogaster pectoralis) dengan bobot individu berkisar antara 5±2 gram dan panjang baku berkisar 5±3 cm. Penelitian dilakukan secara eksperimen, menggunakan Rancangan Acak Lengkap (RAL) dengan 6 macam perlakuan dan masing-masing diulang sebanyak 3 kali. Perlakuan yang diberikan terdiri atas tingkat  energi protein yang berbeda, yaitu perlakuan A (2800 kkal/35%), perlakuan B (3150 kkal/35%), perlakuan C (3150 kkal/40%), perlakuan D (3600 kkal/ 40%), perlakuan E (3600 kkal/45%), dan perlakuan F (4050 kkal/45%). Parameter yang diamati meliputi pertumbuhan mutlak, laju pertumbuhan relatif, konversi pakan, efisiensi pakan, kelangsungan hidup, dan kualitas air. Hasil penelitian menunjukkan bahwa pakan dengan tingkat protein 40% dan energi 3150 kkal/kg menghasilkan pertumbuhan mutlak, laju pertumbuhan relatif, konversi pakan, dan efisiensi pemberian pakan yang terbaik dibandingkan dengan perlakuan lainnya. Kelangsungan hidup ikan sepat berkisar antar 96,67-100%. Kualitas air (suhu, pH, DO, NH3, dan CO2) pada tiap perlakuan masih memenuhi standar untuk  kehidupan ikan sepat.

Abstract

Abstract. The purpose of this study was to examine the Acriflavine doses and immersion time on the sex ratio and survival rate of Bagrid catfish (Hemibagrus nemurus. The completely randomized design of factorial was applied in this research (4x2) with 3 replications. The tested doses were 0 mg/L, 2.5 mg/L, 5 mg/L, and 7.5 mg/L and immersion times were 6 hours and 12 hours.  The Anova test showed that Acriflavine doses and immersion times were not influence significantly on survival rate and sex ratio of Hemibagrus nemurus. However, the highest male sex ratio was found 5 mg/L dose and 12 hours immersion time.Keywords : Bagrid catfish, Hemibagrus nemurus, acriflavine, masculinization  Abstrak. Penelitian ini bertujuan untuk menguji pengaruh dosis akriflavin dan lama perendaman terhadap keberhasilan pembentukan kelamin jantan dan sintasan benih ikan baung yang terbaik. Rancangan yang digunakan adalah Rancangan Acak Lengkap Faktorial (4x2) dengan tiga kali ulangan. Variabel bebas yang diukur adalah kombinasi antara dosis akriflavin (0 mg/L, 2,5 mg/L, 5 mg/L, dan 7,5 mg/L) pada lama perendaman (6 jam dan 12 jam. Hasil uji Anova menunjukkan bahwa perlakuan dosis akriflavin dan lama perendaman tidak memberikan pengaruh yang nyata terhadap sintasan dan rasio kelamin ikan baung. Namun demikian hasil terbaik diperoleh pada perlakuan dosis 5 mg/L akriflavin dengan lama perendaman 12 jam, yakni 61,83%. Kata kunci : Baung, Hemibagrus nemurus, akriflavin, jantanisasi

Abstract

The purpose of the research was to give information condition and potention of mud crab. This research used survey method which are a). observation, b). direct interview c). in situ treatment and d). sampling.  Research location at Sungai Musang Village, Aluh-Aluh, Banjar Regency, South Kalimantan Province. Parameters observe were mud crab species, water quality (water temperature, salinity, water pH, NO2-N, NO3-N, NH3-N, DO, and transfarance), and biology (plankton dan mud crab larvae). The results founded that 3 mud crab species 1). Scylla serrata 2). S. transquebarica and  3) S. oceanica. The range of water temperature was (30-340C), Salinity (0-1%o), water pH (7,80-8), NO2-N (0,36 mg/L), NO3-N (2,01 mg/L), NH3-N (0,46-2,5 mg/L), DO (1,65-4,20 ppm), transparance (32 cm), diversity index of phytoplankton (0,89), and  zooplankton (1,58).  Ecology and biology, showed that water quality was suitable to mud crab culture. Mud crab larvae was trap zoae and megalopa stadia with density of 20 ind/L. Little number of mud crab trapped, was assumed due todisfunction condition of  mangrove forest.

Abstract

Objectives of this study were to find out the protein of Aeromonas hydrophila for vaccine, and to evaluate the efficacy of the protein to control Motile Aeromonas Septicemia (MAS) in catfish (Pangasius hypophthalamus). A. hydrophila was isolated from the kidney of diseased catfish. The isolate was cultured, sonicated and sentrifuged into cell-free extract and debris. The protein in the cell-free extract was precipitated with ammonium sulphate, dialized, then fractionated by gel filtration column chromatography on Sephadex G150.The concentration of protein were estimated by spectrophotometer at 280 nm. Fractions obtained from the gel filtration with high concentration of protein were analized by sodium dodocyl sulphate-polyachrylamide gel electrophoresis (SDS-PAGE). The protein bands obtained from SDS-PAGE with molecular weight more than 10 kDa were tested for immunogenicity to mice. Futhermore, the most immunogenic protein was used for fish vaccination. The vaccinated and unvaccinated fishes were chalenged with A. hydrophila.The results indicated that vaccination with the protein significantly increased (P<0.01) the antibody titer either in mice or catfish. Relative Percent Survival of catfish vaccinated with the protein at 5; 7.5; and 10 µg/fish were 61.54%, 80.77% and 76.92%, respectively. The optimum dose of vaccine was 8.33 µg/fish with maximum RPS of 82.05%.
