Abstract

ABSTRACT Curcumin, isolatedfrom Curcuma longa L, long enough being used for medicinal purposes in Asia to prevent cancer disease. Numerous studies have indicated that curcumin has an antioxidant and anti-inflammatory activities, chemopreventive and chemotherapeukc effects. Curcumin structure consists qf two methoxylated phenol and an en01 form of @-diketone. The phenolic group, the main important structure and its ortho substitution compound with methoxy or methyl group will enhance the activity. Curcumin can blocks tumor initiation induced by benzo[a]pyrene and 7/22dimethylbenz[a]anthracene. The anti-inflammatory properties of curcumin decreases due to the inhibition of overexpression prostaglandin synthesis by COX-2 enzyme. The role of COX-2 in suppression of apoptosis and tumor cell proliferation has been studied. Curcumin induced downregulation of NF-KB is mediated through suppression of 1~Baki nase activation. Abstract

Teknik immunoassay berdasarkan reaksi antara antigen dan antibodi dapat digunakan untuk mendeteksi berbagai jenis mikroba dan protein toksin dalam bahan pangan atau analisis pengawasan lingkungan dengan spesifisitas dan sensitifitas yang cukup tinggi. Teknik ini banyak digunakan dalam bidang pertanian, kesehatan, pengawasan lingkungan dan keamanan pangan. Dalam tulisan ini dibahas prinsip-prinsip dasar teknik immunoassay, terutama aspek kimia dan penerapannya dalam analisis keamanan pangan dan pengawasan lingkungan dari segi mikrobiologisnya. Selain itu juga dibahas faktor-faktor yang dapat mempengaruhi reaksi-reaksi kimia dan hasil analisis tersebut.   Kata kunci : Immunoassay, antigen, antibody, pangan, protein toksin, reaksi kimia.

Abstract

A research on anti-microbial bioactivity of protein fraction isolated from several species of macro algae of barang lompo island in South Sulawesi by using polar solvent buffer of 0.1 M Tris-HCl with pH 8.3  containing 2 M NaCl 0.01 M CaCl2, 1 % b-mercaptoethanol, 0.5 % Triton X-100 has been conducted. The concentration of protein was determined by the Lowry method and the bioactivity test was investigated  by a gel diffusion method. Results showed that the concentration of protein in the crude extracts isolated from 1000 g of fresh samples of three species of macro algae (Turbinaria decurens bory, Sargassum echinocarpum J.G Agardh dan  Laurencia  cartilaginea Yamada) was 1,734 mg/mL;  1,086 mg/mL;  and 3,200 mg/mL respectively. Initial purification of protein uses conducted by using the fractionation method with ammonium  sulfate, followed by a dialysis process. All protein fractions showed anti-microbial bioactivity, where the highest inhibition zone was found in the crude extract from Laurencia  cartilaginea with the inhibition zone of  24,82 mm to E. coli and the protein fraction with the saturation level of ammonium sulfate of 40-60 % from Sargassum echinocarpum J.G Agardh with the inhibition zone of  22,54 mm to Malassezia furfur. The results indicated that bioactive protein from macro algae  is very potential as the basic material for the new anti-microbial drug, especially to E. coli and  Malassezia furfur. Abstract

For better understanding of DNA replicating-coupled chromatin assembly and transcription regulation in eukaryotes, we studied biochemical and genetic analysis of nuclear-related proteins from chicken DT40 cell lines. The genetic analysis of some nuclear proteins, such as HIRA and CAF-1, indicated that these proteins could play overlapping roles in chromatin dynamics and is consistent with the finding that HIRA protein exhibited binding ability to histones and ASF-1, as also ASF-1 bound directly with CAF-1p60. In this study, revealed not only that the N-terminal and C-terminal halves of HIRA mediate individually transcription repressions but also that even one of the seven WD dipeptide motifs and the LXXLL motif of HIRA are required for these mediations in vivo.  Finally, we found that HIRA-mediated repression is sensitive to TSA and it co-represses transcription together with HDAC-2. We believe our findings will contribute to a major break-through in future studies on the specific, individual roles of HIRA involved in numerous DNA-utilizing processes, through the formation and/or maintenance of the chromatin structure in higher eukaryotes.   Keywords: Histone regulator, HIRA, Transcriptional repression, Chromatin, WD dipeptide motif.

Abstract

A research on the ability of protein fraction from red algae Gelidium amansii to inhibit the growth of Salmonella thypi and Staphylococcus aureus has been conducted. Proteins was fractionated from the crude extract by salting out method with  0 – 20 %, 20 – 40 %, 40 – 60 % and 60 – 80 % ammonium sulfat saturation. Proteins was purified by a dialysis method using a selophan membrane. The protein level was determined by Lowry’s method, the highest protein concentration, 5.6 mg/mL was found in the 40 – 60 % fraction  of the red algae. Antibacterial activity test was performed using a layered jelly diffusion method on the  MHA medium. Results indicated that crude extract and the protein fraction of the red algae had antibacterial effect to Salmonella thypi and Staphylococcus aureus. The highest antibacterial activity to Staphylococcus aureus was found in the 0-20% fraction with the inhibition diameter of 14.43 mm after incubated for 24 hours. These research might provide a significant results for the following research on the antibacterial action in molecular  level of protein compounds from marine algae.

Abstract

A research about potential of 2-phenylethyl pentanoate ester in order to be used in an effort to increase the activity of antibiotics INH, SM and ETA against M. tuberculosis strain H37Rv had been done. The 2-phenylethyl pentanoate ester was synthesized by Fischer esterification reaction of 2-phenylethanol and pentanoate acid in mole ratio 1:2, 2:1, 3:1 and 5:1. Characterization of ester as product was conducted using refractometer, FTIR and H-NMR. The product of the synthesis of 2-phenylethyl pentanoate obtained as colorless liquid. From this research was known that optimum mole ratio of 2-phenylethanol and pentanoate acid in Fischer esterification reaction to obtained 2-phenylethyl pentanoate was 5:1 in 39,59 % yield. The antibiotic activity essay of INH, SM, and ETA by 2-phenylethyl pentanoate against M. tuberculosis was done by measuring the MIC with 9 variations of antibiotic and ester concentration in MGIT medium. The 2-phenylethyl pentanoate could to increase antibiotic activity of SM and ETA. MIC value to SM 1 μg/mL in combination with 2-phenylethyl pentanoate 0.1 mM and value to ETA  0,5 μg/mL in combination with 2-phenylethyl pentanoate 1 mM.

Abstract

Senyawa turunan oleanan 3-asetil-12-oleanen-28-oat diisolasi dari fraksi n-heksan kulit batang M.umbellata (Houtt) Stapf var. degrabrata K. Struktur molekul ditentukan melalui analisis data spektroskopi IR, NMR 1D dan NM R 2D ( 1 H, 13 C, DEPT, COSY, HMQC dan HMBC). Hasil uji bioakt ivitas menunjukkan bahwa senyawa tersebut bersifat toksik terhadap A. salina dengan nilai LC 50 361,93 μg/mL. Pada konsentrasi 1000 μg/ mL senyawa oleanan 3-asetil-12-o leanen-28-oat memberikan hambatan tertinggi terhadap bakteri B. subtilis dan jamu r C. albicans, dengan diameter zona hambat 15,8 mm dan 15,2 mm.

Abstract

Red algae Eucheuma cottonii and Eucheuma spinosum which one alternative source of antioxidant and anticancer because contains polysaccharide compound. This research aims to isolate and examine the activity of antioxidant and toxicity polysaccharide extract from red algae Eucheuma cottonii and Eucheuma spinosum . The study was carried out by isolating the polysaccharide extract using water and methanol-ethanol precipitation. Antioxidant activity of crude extract was examined using 1,1-Difenil-2-pikrilhidrazil (DPPH) method, and toxicity test was carried out using Brine Shrimp Lethality Test (BSLT). The research results showed that the crude extract polysaccharide for Eucheuma cottonii and Eucheuma spinosum have strong antioxidant activity with IC50 value of 72,49 ppm and 75,98 ppm. The result of BSLT assay showed that the crude extract polysaccharide has a highest toxicity with LC50 value of 165,88 ppm and 337,21 ppm there are classified as toxic. The crude extract polysaccharide Eucheuma cottonii and Eucheuma spinosum has a potential to be developed as an alternative antioxidant and anticancer agent.

Abstract

A research has been on determining the origin of forest honey quality Mallawa Maros based on the levels of 5-(hydroxymethyl)furfural-2-karbaldehida (HMF)is quantitatively based on the variation of heating temperature and duration of storage with White spectrophotometric method. The results showed that the levels of forest honey HMF origin Mallawa on heating temperature 35°C,90°C and 110°C respectively is 42.2023 mg/Kg, 46.0274 mg/Kg and 62.2520 mg/Kg. While the storage duration for 3,4, and 5 months had higher levels of HMF each 27.4309 mg/Kg, 42.5158 mg/Kg and 48.9405 mg/Kg. Heating temperature and storage time a big influence on levels of HMF and show a strong relationship to the water content, pH and free acid, reducing sugar content and diastase enzyme activity. Based on the research results, levels of HMF at the heating temperature of 110oC has passed the standards set by the SNI (up to 50 mg/Kg). Whereas the standard set by IHC for honey originating from thetropics (80 mg/ Kg)

Abstract

Isolation, structure elucidation, activity test against Salmonella thypi, Staphylococcus aureus of secondary metabolite from hydroid Aglaophenia cupressina Lamoureoux, were conducted. This study was aimed to identify and isolate the compound with antibacterial properties of hydroid Aglaophenia crupessina Lamoureoux. This study used stepwise method with the following sequences: extraction, isolation, purification, structure elucidation, and bioactivity test. Study results indicated three compounds found: (1)carboxylate acid compound: hexadecanoic acid and has antibacterial properties; (2) alkaloidcompound which was considered as a new compound: aglaounhas and has antimicrobialproperties; (3) steroid compound: β-sitosterol with no antimicrobial properties.Keywords: Isolation, characterization, secondary metabolite, Aglaophenia cupressina, Antibacterial.
