Abstract

Phytoestrogen is the chemical compound contains in plant which has estrogenic like effect. Estrogen has important function on woman’s sexual organ, such as proliferation of uterine and vaginal cornification. Phytoestrogen are weak agonists for estrogen and illicit statistic significantly increases in uterine wet weight, at definite dose, in uterothrophic bioassay. Biji Klabet or Fenugreek (Trigonella foenum-graecum L.) contains steroidal sapogenins such as diosgenin, tigogenin, gitogenin, yamogenin and trigoneoside, that suspected having an estrogen-likeeffect or as phytoestrogen. The aim of this research was to investigate the estrogenic effect of fenugreek’s ethanolic extract in immature rat models. These models represent the climacteric/menopause phase, where estrogen level is verylow because ovary produces no estrogen. The testing animals were divided into five groups, namely normal group, estradiol control group and three level doses of fenugreek extract (30mg/200gBW; 60mg/200gBW and 120mg/200gBW). Theresult indicated that on 120mg/200gBW, fenugreek extract significantly (=0.05) increasing wet weight uterine and proliferation of uterine, such as uterine thickness and uterine diameters. Fenugreek extract, start on 60mg/200gBW, also induces vaginal lubrication and estrus cycle.

Abstract

Trigonella  foenum-graecum  (TFG)  is  one  of  medicinal  plants  contains  some steroidal sapogenin  such as diosgenin, yamogenin, gitogenin, tigogenin and trigoneoside, also  alkaloid  trigonellin,  which  is  have  many  activity  as  antidiabetic,  estrogenic  and  also as  anti  cancer.    This  experiment  was  done  to  explore  the  activity  of  some  extract  of TFG on some cell lines such as MCF7 (Human Breast Cancer Cell-line), T47D (Human Breast  Cancer  Cell-line),  PC3  (Human  Prostate  Cell-line)  and  SKOV  (Human  Ovarian Carcinoma Cell-line).  This assay was done using MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium)  methods.    Results  showed  that  ethyl  acetate  fraction  gives  the lowest  IC50  than  another  extracts.  IC50  for  PC3  is  66.24  ppm,  IC50  for  MCF7  is  41.81 ppm,  IC50  for  T47D  is  58.63  ppm.    These  datas  can  be  used  for  further  research  to isolate the active compound from TFG. The testing animals were divided intonormal group, ovariectomized control group, estradiol control group and three level doses of fenugreek extract (30mg/200gBW; 60mg/200gBW and 120mg/200gBW).The result indicated that start on 60mg/200gBW, fenugreek extract significantly could increasing mammary gland proliferation. Empirically fenugreek containingdiosgenin, that caused breast enhancement. Abstract

Biji Klabet or Fenugreek (Trigonella foenum-graecum L.) contains steroidal sapogenins such as diosgenin, tigogenin, gitogenin, yamogenin and trigoneoside, that are suspected having an estrogen-like effect or as phytoestrogen. The objective of this study was to evaluate the estrogen-like effect of biji klabet alcohol extracts. This research used immature Wistar female rats, age of 19 days, which were represent the climacteric/menopause phase. The testing animals were divided into five groups, namely normal group, estradiol control group and three level dose groups of fenugreek extract (30 mg/200 gBW; 60 mg/200 gBW and 120 mg/200 gBW). The result showed that on 120 mg/200 gBW fenugreek extract significantly (a=0.05) increased estradiol and FSH level on immature rat. Analysis of vaginal smear showed vaginal lubrication effect, surprisingly on 120 mg/200 gBW could induce estrous cycle.

Abstract

Recombinant protein Fim-C S.typhi is a potential protein that can be used as an alternative typhoid vaccine and produced on a large scale. However, before entering into a large-scale stage, laboratory optimum data on the factors that affect the production process of Fim-C Salmonella typhi proteins are required. This study aims to obtain information regarding the effect of host cell number E.coli BL21 (DE3) pLysS and overexpression time on production of recombinant protein Fim-C Salmonella typhi as the basis for developing candidate of typhoid vaccine. The optimization process of overexpression was done by adding 0.5 mM IPTG (Isopropyl-β-D-thiogalactopyranoside) inducer into bacterial cultures of 2%, 4%, and 6% with 4, 5, and 6 hours over expression. The measurement of the concentration of Fim-C recombinant protein extracted samples were done by a spectrophotometric method used BCA Kit Assay Thermo ScientificTM with wavelength 590nm. The characterization of those protein extracts was performed using SDS-PAGE method. The results from the study concluded that the number of 4% E.coli bacterial cells and four-hour overexpression time are the optimum condition of Fim- C Salmonella typhi characterized by the presence of high-intensity bands at ± 31 kDa.

Abstract

Fenugreek seed or bijiklabet (Trigonellafoenum-graecum L.) was known to have activity against degenerative diseases such as diabetes mellitus, hypercholesterolemia and postmenopausal symptoms.  This study was conducted to investigate the safety of ethanolic extract of fenugreek on white rat, by determining Lethal Concentration 50 value (LC50).  This in-vivo assay referred to the WHO protocols for the toxicity assay of natural medicines. We used Spraquedawley white rats, female and male, 6 weeks of age, which divided into a normal group and fivetreatment groups (1g/kgBW, 4g/kgBW, 8g/kgBW, 12g/kgBW, 16g/kgBW). Test sample was given once orally then the animal were monitored for two weeks.  Observation of toxic effect includedphysical symptom of central nerve system, autonom nerve system and digestive system.  Alldeathanimal were counted and LC50 were calculated.  Result showed that there was no toxic effect and no death animal until 16g/kgBW dose of treatment.  We concluded that ethanolic extract of Fenugreek is practically non toxic.

Abstract

Trigonella foenum-graecum (TFG) is one of medicinal plants containing several steroidal sapogenins, such as diosgenin, yamogenin, gitogenin, tigogenin and trigoneoside, and also alkaloid trigonellin and some flavonoids such as vitexin, isovitexin, orientin, isoorientin, which has many activities, such as antidiabetic, estrogenic, and also anticancer.  As phytoestrogen, TFG was predicted to have potency as Selective Estrogen Receptor Modulators (SERMs) which is used for hormonal-dependent breast cancer treatment. This experiment was carried out to investigate interaction of some sapogenin steroids and flavonoids in TFG to estrogen receptor alpha (ERα) and its activity to breast cancer cell line as confirmation. In silico prediction was carried out to investigate their estrogenic activity by analyzing their binding affinity to ERα using AutoDock Vina program. In vitro activity confirmation of TFG extract and its fractions were carried out using MTT assay on Erα-positive human breast cancer cell line, MCF-7.  Results showed that free binding energies of diosgenin and yamogenin were -6.4 kcal/mol, estradiol was -6.0 kcal/mol, and tamoxifen was -5.1 kcal/mol.  While cytotoxicity assay showed that ethyl acetate fraction gave the lowest IC50 of 41.81 ppm, with total steroid content of 20.03 ppm.  From these results, we can conclude that diosgenin and yamogenin have greater binding affinity to ERα comparing to estradiol and tamoxifen.  In vitro assay confirmation showed that ethyl acetate fraction has cytotoxic effect on MCF-7 cells.Keywords: Trigonella foenum-graecum, sapogenin steroids, MCF-7, estrogen receptor alpha, binding affinity

Abstract

Indonesian medicinal herbs have so many biological activity, including anticancer activity. This assay aims to evaluate three Indonesian medicinal plants (Trigonella foenum-graecum L. (TFG), Aglaia elliptica Blume. (AE) and Foeniculum vulgare Mill. (FV)) which have potency in anticancer activity. The experiment was done by MTT assay to observe the cytotoxicity activity on breast cancer cell lines MCF7.  Apoptotic potency was evaluated based on % population of MCF7 cell in subG0/G1 phase.  The results showed that IC50 from ethanolic extract of TFG was 241.24 ppm, ethanolic extract of AE was 19.442 ppm and  ethanolic extract of FV was 69.41 ppm.  Apoptotic potency for TFG was 6.46%,  AE was 11.94% and for FV was 8.73%. Thus, it is concluded that ethanolic extract of AE gives the lowest IC50 and the higher potency of apoptotic by significantly (p<0.05) increase % population of cells in subG0/G1 phase on 100ppm concentration.Keywords :  Trigonella foenum-graecum L., Aglaia elliptica Blume. and Foeniculum vulgare Mill., MCF-7, Apoptotic, Cytotoxic
