Abstract

Myelocytes blood cell tumor in chicken is a disease caused by retrovirus, avian leukosis virus subgroup J (ALV-J). The virus has the same group as human retrovirus (HIV) which caused AIDS, but the avian type possesses oncogenic properties, that could induce cell transformation and tumor formation. ALV-J stimulates the bone marrows myelocytes and transforms them into tumor cell myelocytoma. The tumor cells then metastasis through the circulatory system causing myeloid leukosis and tumor cells accumulation in various internal organs or myelocytomatosis. This study was done on the base of the leukosis behavior of the tumor. The finding of the metastasis tumor cell in the blood smear examination is thought could be use as the diagnostic clue of the disease. Blood smear from sick chickens are collected and stained with some chemical staining substance such as may grunwald-giemsa, hematoxyllin eosin, periodic acid Schiff, congo red, toluidine blue, and sudan black B. Cytochemistry character of the blood cells was observed using light microscope. The result showed that myelocytes granules were best observed using hematoxyllin eosin, periodic acid Schiff, congo red and toluidine blue while may grunwald-giemsa, and sudan black B could not differentiate the granules. By this method, the field veterinarian will able to screen the suspected chicken flock for myeloid leukosis earlier than the occurrence of tumor formation.

Abstract

Study of the pathogenicity of gumboro virus (very virulent Infectious Bursal Disease virus, vvIBDV) oflocal isolates was performed in broilers. The chickens were grouped into four: i) infected with vvIBDV; ii)vaccinated with commercial vaccine + challenge with vvIBDV; iii) vaccinated using locally produced vaccine+ challenged with vvIBDV; and iv) control group (unvaccinated animals). Pathogenecity was analyzedbased on the sequence of infection; the distribution and degree of gross pathology and histopathologylesions on bursa Fabricius, spleen and thymus in relation to the presence of the antigen (IBDV Ag) whichwas detected using immunohistochemistry technique. During the acute phase (1-3 days following theexperimental infection) hyperemia and exudation on the bursa Fabricius were observed in the infectedgroup and the two vaccinated-challenged groups. Similarly, hyperemia was also observed on the spleenand thymus. Whilst during the chronic stages (at 7 and 14 days following the experimental infection) thebursa showed atrophy. Upon histological examination, the bursa showed interstitial edema, hemorrhage,infiltration of heterophyl cells and fibroblast hyperplasia. The bursa lymphoid follicles were depleted dueto cells necrosis and apoptosis. Plica epithelial layer was wrinkled and the Goblet cells metaplasia formedhyper plastic cysts. In the spleen and thymus severe reticulo endothelial cells proliferation was observedduring the acute stage and tended to decline during the chronic stage. During the acute and chronic stagescells containing IBDV Ag were detected in the bursa Fabricius, distributed within the mantle zone andgerminal center of lymphoid follicles in the two vaccinated-challenge groups. Whereas in the infectedgroup, the IBDV antigen were detected up to 7 days post the infection. Nevertheless, neither IBDV antigenwas detected in the spleen nor in the thymus in all the experimental groups. The results of this studyclearly explained the ferocity of vvIBDV of local isolates.

Abstract

ABSTRACTStress and glucocorticoids are two interrelated aspects and both have a strong influence on brain function,the system of the hupothalamic-pituitary-adrenal axis, and a number of target tissues in the central andperipheral nervous system. Low and high levels of glucocorticoid is affected by stress. The higher levels ofstress glucocorticoid production will be higher, and vice versa. Stres and glucocorticoids can causeinterference neuropathology that induces the emergence of signs of aging or accelerated aging.Neuropathology disorders caused by stress and glucocorticoids can be a disruption in learning and memoryneurons, the process of neurogenesis, synaptic plasticity between neurons, atrophy and death of neurons.Aging caused by stress and glucocorticoids can be observed on behavioral aspects, the electrophysiologic,histologist, and anatomic (Sains Medika, 2(2):207-220).Key words: Stress, glucocorticoids, neuropathology, aging, learning and memory neurons, synaptic plasticity,neurogenesis, atrophy, death of neuronsABSTRAKStres dan glukokortikoid merupakan dua aspek yang saling berkaitan dan keduanya mempunyai pengaruhyang sangat kuat terhadap fungsi otak, sistem aksis hipotalamus-pituitari-adrenal dan sejumlah jaringantarget pada sistem saraf pusat dan perifer. Rendah dan tingginya produksi glukokortikoid sangatdipengaruhi oleh tingkat stres. Semakin tinggi tingkat stres, produksi glukokortikoid akan semakin tinggi,demikian pula sebaliknya. Stres dan glukokortikoid dapat menyebabkan gangguan neuropatologis yangmenginduksi munculnya tanda-tanda penuaan atau mempercepat penuaan. Gangguan neuropatologiyang disebabkan oleh stres dan glukokortikoid dapat berupa gangguan pada neuron learning and memory,proses neurogenesis, plastisitas sinaptik antar neuron, atropi, dan kematian neuron. Abstract

Primary lung cancers, or tumors originating in the lung, are relatively uncommon in dogs. The objectiveof this study was to describe the canine lung carcinoma that serially transplanted into severe combinedimmunodeficiency (SCID) mice, in order to established cell line from this tumor cell.  Morphology andcharacteristic of this canine lung carcinoma in SCID mice by histopathological and ultrastructuralexaminations with metastatic lesion in lung were also examined.  Histopathologically, the tumor masswere consisted of cuboidal to columnar cells with papillary pattern, uniform in size, the nuclei were oftenvariable in size, and some cells have vacuole on their cytoplasms.  Glandular forms were predominant withlobulated pattern, ductal pattern with papillary injected into tube-like structure were also encountered.Mitotic figures commonly found with inflammatory reaction were sometimes present in the interstitiumand lumen gland.  Ultrastructural analysis of the tumor cells showed round to oval cells with one or moreprominent nucleoli.  The cells possessed numerous mitochondria, smooth endoplasmic reticulum, andindividual cells which were interconnected via desmosomes.  Tonofilament characterize by long cytoplasmicmaterial was encountered.  Positive reaction of the round to oval tumor cells to anti keratin antibodyconfirmed that their epithelial cell nature.  Lung metastatic lesions were found in SCID mice aftertransplantation and this phenomenon indicated that canine lung carcinoma is tumorigenic to SCID mice.

Abstract

ABSTRACT. The aim of this research was to produce cincau copper-derivative chlorophyll powder and to observe its phYSico-chemical properties and the effect on blood lipid profile of5 month male New Zealand White rabbits. The blood lipid profile observed were total cholesterol, triglyceride, HDL (high-denSity lipoprotein) cholesterol, and LDL (low-density lipoprotein) cholesterol. A two months treatment was conducted A randomblock design was applied with the following treatment on each experimental unit: PO (basal diet, negative control), PI (basal diet with cholesterol 0.1% (wlw) added, positive control), P2 (basal diet with cholesterol 0.1% (wlw) and 16.7 mglkg-body weight/d natural chlorophyll powder added), P3 (basal diet with cholesterol 0.1% (wlw) and 16.7 mglkgbody weight/d copper-derivative of chlorophyll powder added), P4 (basal diet withcholesterol 0.1% (wlw) and 50.1 mglkg-body weightld copper-derivative of chlorophyll powder added}, P5 (basal diet with cholesterol 0.1% (wlw), and 16.7 mglkg-body weight/dcommercial chlorophyll powder added The results of study showed that cincau leaves chlorophyll extract solution with Cu1+ 100 mglL level produce the cincau copper-derivate chlorophyll powder with highest pH, solubility, and greenness compared to other copper levels. Feeding with copper-derivative of chlorophyll cincau powder (P3) most likely decreased total cholesterol, triglycerides, LDL cholesterol increased the HDL cholesterol,and decreased atherosclerosis risk after 2 month as compare to feeding contain of natural chlorophyll (P2). Abstract

Background: Physiological aging or aging due to oxidative stress causes to disruption of glutathione homeostasis of the hippocampus. Disruption of glutathione homeostasis characterized by a decrease in hippocampus glutathione levels below the normal threshold. Decreased levels of glutathione in the hippocampus was demonstrated decreased hippocampus function. One way to increase glutathione levels in the hippocampus and the function of hippocampus is to increase the concentrations of glutathione precursor. One of the glutathione precursors is alanine-glutamine dipeptide. This research was designed to obtain the most effective level of alanine-glutamine dipeptide supplementation to improve glutathione synthesis in the hippocampus of aged and oxidative-stressed rats. The highest glutathione concentrations in the hippocampus showed the best improvement on the hippocampus function in physiological aging or oxidative-stress aging rats.Design and Methods: The experimental rats were assigned into a completely randomized design with 2x2x4 factorial arrangement. The first factor was the age of the experimental rats, consisted of two levels i.e., 12 and 24 months. The second factor was oxidative stress consisted of two levels, i.e., without or with oxidative stress. The third factor was the level of alanine-glutamine dipeptide administration consisted of 4 concentrations, i.e. 0%, 3%, 5%, and 7%.Results: The results showed that administration of 7% alanine-glutamine dipeptide has resulted the highest levels of glutathione hippocampus in younger (0.0154 mg/mg tissue) or aged (0.0140 mg/mg tissue) rats or in normal (0.0150 mg/mg tissue) and in oxidative-stressed (0.0144 mg/mg tissue) rats. The increased hippocampus glutathione levels were associated to the improved functions of the hippocampus. Conclusion: alanine-glutamine dipeptide administration of 7% consentrations gave the best results on repair function of the hippocampus and has the potential to slow aging, both physiological aging or oxidative-stress aging rats (Sains Medika, 4(1):1-12).

Abstract

Plants lindur (Bruguiera gymnorrhiza) is one of the plants that have a potential as asource of bioactive compounds for the antioxidant. This study aimed to determine thechemical composition, bioactive compound and determine antioxidant activity of leaves,bark and roots of plants lindur (B. gymnorrhiza). This research was conducted throughseveral stages proximate test, stratified extraction, phytochemical test, and antioxidantactivity test with DPPH. The highest yield value resulting by ethanol extract of the leavesis 12,85 % and the lowest was produced by n-hexane root extract that is 0,18 %. The leavescontain high levels of protein, fat, and water which is the higher of the two parts from theother samples. Bark containing ash content (4,12%) and carbohydrates (46,02%). Root hasthe highest levels of carbohydrates that is 25,91 %. Bioactive components were detected onethanol and ethyl acetate extracts of the leaves are flavonoids, tannins, phenols, saponins,steroids and tritepenoid. Bark extract ethanol, ethyl acetate and ethanol root barkhas bioactive components there are flavonoids, tannins, phenols, saponins, andtritepenoid.Keywords : Porphyridium cruentum, photoperiod, growth,biomass, polysaccharides

Abstract

The background of this study was to determine the safety of pauh kijang extract which is orally given to mice while the aim of the study was to evaluate the liver and kidney histopathological changes after receiving various dose of the extract mango (Irvingia malayana Oliv. Exe Benth.) ethanol extract. There were five group of treatments in this study : mice received extract with the concentration of (I) 1000mg /kg BW, (II)100 mg/kg BW, (III) 10 mg/kg BW, (IV) 1 mg/kg BW, and (V) received carboxy methyl cellulose-sodium (CMC-Na) as negative control. Each treatment consisted of five mice. After seven days of treatment, mice were sacrified. The liver and kidney were sampled and processed to prepare histopathology slides. The histopathology parameters were examined by counting the degeneration and necrotic cells of hepatocytes and renal epithelial tubule. The lesion of glomerulus such as atrophy and protein sedimentation were also observed. The result also showed that extract concentration of > 10 mg/kg BW result in damaging liver cells significantly, while kidney cell damage occurred significantly in the exposure of extract > 1 mg/kg BW.Key words : Irvingia malayana, liver, kidney

Abstract

Physiological aging or aging due to oxidative stress decrease glutathione level in the hippocampuswhich impacts the respons impaired hippocampus celuller. Hippocampus cellular respons disorderscharacterized with decreased viability, increased mortality, and the shortening of the axons of neurons.One way to improve hippocampus cellular respons is to  increase the levels of glutathione and theconcentration of glutathione precursor. One compound that provides glutathione precursors is alanine-glutamine dipeptide. This research was designed to obtain the improve of hippocampus cellular responsresult from the administration of 7% alanine-glutamine dipeptide concentration of aged or oxidative-stressed rats. The improvement of hippocampus cellular respons affect  the improvement of the hippocampus function. The experimental rats were assigned into a completely randomized design consisted of threefactors with 2x2x2 factorial arrangement. The first factor was the age of the experimental rats, consistedof two levels i.e., 12 and 24 months. The second factor was oxidative stress consisted of two levels, i.e.,without and with oxidative stress. The third factor was alanine-glutamine dipeptide administrationconsisted of 2 concentrations, i.e. 0% and 7%. The results showed that  administration of 7% alanine-glutamine dipeptide improved level of glutathione in the hippocampus either in younger (58,76%) or aged(125,81%) rats or in normal (76,47%) and in oxidative-stressed rats (97,26%). These antioxidant hadmediated the respons improve viability, mortality, and long axons responses of neurons at younger (4,11%,37,07%, and 12,58%) or aged (6,91%, 37,85%, and 32,84%) rats, in normal (3,25%, 29,21%, and 21,04%)and oxidative stress (7,80%, 43,01%, dan 25,56%) rats. This research concluded that the alanine-glutaminedipeptide 7% increased glutathione levels.  This increased level affected the improvement of cellularresponds in aging hippocampus, physiological aging, or aging due to oxidative stress in rats.
