Abstract

It is known that hormone addition in media helps in vitro maturation of oocyte. This research was aimmed to determine the effect of PMSG in media to maturation rate and nucleous developvement of cow oocyte. Ovaries were obtainned from local slaughterhouse. The media used for in vitro maturation of oocyte was TCM- 199 and the treatment was 3 levels of PMSG: 0, 10 and 20 mg/ml. Result of this research showed that the dose of PMSG in maturation media was significantly affected (P<0.05)  nucleolus development of oocytes and maturation rate. The average of germinal vesicle (GV) stage in 3 levels of PMSG 0, 10 and 20 mg/ml were 38.33; 12.64 and 9.64%, respectivelly. There was no germinal vesicle breakdown (GVBD) found in 3 levels of PMSG addition. The nucleous development of metaphase–I (M-I) were 7.64; 20.2 and 22.00%, but the average of maturation rate (M-II)  was 16.32; 48.10 and 35.34% for 3 levels of PMSG: 0, 10 and 20 mg/ml, respectivelly. It is concluded that 10 mg/ml PMSG in media of in vitro maturation resuls in the highest maturation rate of cow oocyte. Key Words: Oocyte, Maturation, In vitro, Hormone, PMSG

Abstract

The study was aimed to define the morphogenetic characteristics of native song fowl chickens of West Sumatra called Kokok Balenggek. Sixty Kokok Balenggek chickens raised by farmers in Tigo Lurah sub district, Solok district were used as samples. They consisted of 40 males and 20 females. Both qualitative and quantitative morphogenetic data were collected and measured, including: color of feather and shank; form of comb; length of tars metatarsus, tibia, femur; body weight and distance between pelvis bone by females. By males, feather color of Kokok Balenggek chickens were dominated by wild feather type (e+) and silver (S) of 42,5 % and 22,5 %, respectively. Comb were mostly single form (90 %). While feather color of females were dominated by black (S) and white (25 % each), pea comb (90 %), shank color of white yellow (Id) (85 %). Length of metatarsus for male and females of 100,2 + 11,05 mm, 80,03 + 8,64 mm; tibia of 134,40 + 18,73 and 117,14 + 10,24 mm; femur of 103,36 + 16,94 and 85,63 + 11,47 mm; body weight of 1,74 + 0,46 kg and 1,1 + 0,62 kg/head, respectively. Data of distance length between pelvis bone by females were found 41,17 + 4,32 mm.

Abstract

A field survey has been conducted to study the parameter achievements of artificial insemination (AI) by 4 different breeds of cows in IV Angkat Candung sub district of Agam District, West Sumatra. Data collected included conception rate (CR), service per conception (S/C) and calving rate from 496 cows which were divided into 4 offspring breeds: Simmental, Ongol, Charolais and Brahman. Results showed that the achievements of AI in the surveyed area were relatively high. The CR, S/C and Calving rate were found 76.68 %, 1.30 and 72.57 %, respectively. The most fertile cow was indicated by Ongol.

Abstract

This study  was conducted to determine the response of superovulation by giving 16 ml dosage  of FSH hormone to female Pesisir cattle.  The estrus schedule of 15 Pesisir cows was set by inserting CIDR (Controlled Internal Drug Release) into the vagina for 12 days. At day 10, all cattles were injected with FSH for three consequent days but with decreasing dosage. On the 3rd day, FSH injection was accompanied by PGF2α injection and CIDR was removed. The detection of estrus was performed at  day 13. Natural mating was proceeded after the estrus signs visible. Collection of donor embryos was done on the 6th and 8th day after mating. The variables  measured were the response of superovulation, total number of corpus luteum, number of embryos and sex ratio. The results obtained were all  Pesisir cows responded to superovulation. The  average number of of corpus luteum and embryoes per cow were  5.93±3.17 and -----, respectively,  while the total of transferable embryoes were 90, with an average of 6.00 or 61.64%. The sexing of embryoes obtained in this study from 146 embryoes awere 76.03% males (111 embryoes) and 23.97% females (35 embryoes). Based on total of transferable embryoes, there were 51.37% male embryos and 11.28% of females embryos. The result of this study showed that the sex ratio of male embryos was higher than female embryos.
