Abstract

This research aimed to evaluate the phenotype diversity of Asian swamp eel from West Java and the biometric responses on salinity for aquaculture development. Fish were collected from three different locations, i.e., Cianjur, Sukabumi, and Karawang with body size of 23.05+0.63 cm and weight of 7.58+1.04 g. Truss morphometric measurement was conducted for 30 samples from each location and biometric response on salty water media for 200 individuals. The experiment was performed in the media with different level of salinity, no substrate and with some shelters. Completely randomized design (CRD) with three levels of salinity (i.e. 0, 6, 12 g L-1) and density 1 kg m-2 was used in this study. Rearing was carried out for 30 days with 100% of media was changed every day and fish were fed with silk worms once a day with at satiation manner. The results showed that phenotypic diversity of Asian swamp eels from Karawang and Cianjur was similar, but differ from Sukabumi population. The biometric responses of Asian swamp eel from Karawang showed the superior performance and the best salinity of media for culture without substrate was 6 g L-1.AbstrakPenelitian ini bertujuan untuk mengevaluasi keragaman fenotipe belut asal Jawa Barat serta respons biometriknya dalam pemeliharaan pada media air bersalinitas untuk pengembangan budi daya. Belut ditangkap dari tiga lokasi yaitu Cianjur, Sukabumi, dan Karawang dengan ukuran tubuh berkisar antara 23,05+0,63 cm dan bobot 7,58+1,04 g. Sebanyak 30 ekor setiap populasi digunakan untuk pengukuran truss morfometrik dan 200 ekor untuk pengujian respons biometrik pada media air bersalinitas. Pemeliharaan dilakukan pada media air bersalinitas tanpa substrat dengan pemberian pelindung. Percobaan menggunakan rancangan acak lengkap dengan perlakuan perbedaan salinitas media pemeliharaan yaitu 0, 6, dan 12 g L-1 dengan padat penebaran 1 kg m-2. Pemeliharaan belut dilakukan selama 30 hari dengan penggantian air 100% setiap hari dengan pemberian pakan berupa cacing sutera Tubificidae sebanyak satu kali per hari secara at satiation. Hasil penelitian menunjukkan bahwa belut asal Karawang dan Cianjur mirip berdasarkan keragaman fenotipenya dibandingkan populasi Sukabumi. Respons biometrik belut asal Karawang paling unggul dan media terbaik untuk pemeliharaan belut tanpa substrat adalah media dengan salinitas 6 g L-1.

Abstract

Interactions in fish community in an ecosystem can be assessed through food habits analysis. This study aims to assess the trophic structure of fish communities in Cisadea River. Fishes were collected using active and passive fishing gear at six locations representing upstream, midstream, and downstream areas at the dry season (June-November 2012). During the observation, sample was caught of 666 individuals consists of 48 species. A total of 250 individuals were analyzed for their food composition and digestive tract morphology to determine their trophic groups. Based on large group of organisms, fish community in Cisadea River consists three guild is carnivorous, omnivorous and herbivorous fish which dominated by carnivore and omnivore fish. Based on food organism taxon, the fishes were assigned to four groups i.e. insectivore, phy-toplanktivore, crustacivore, and molluscivore, with eight groups of food organisms (phytoplankton, insects, macrophytes, crustaceans, mollusc, fish, protozoa, and rotifers). In the upstream and midstream areas, the primary diet was insects and algae, whereas in the downstream the diet consists of algae, crustaceans, and insects. AbstrakInteraksi yang terjadi pada suatu komunitas ikan dapat dikaji melalui analisis makanan alaminya. Penelitian ini bertu-juan untuk mengkaji struktur trofik komunitas ikan di Sungai Cisadea. Ikan dikoleksi dengan menggunakan alat tang-kap aktif dan pasif di enam lokasi yang mewakili wilayah bagian hulu, tengah dan hilir selama musim kemarau (Juni-November 2012). Selama pengamatan, sampel yang diperoleh sebanyak 666 individu yang termasuk dalam 48 spesies. Sebanyak 250 individu dianalisis komposisi makanannya dan morfologi saluran pencernaannya untuk mengetahui ke-lompok trofik pada komunitas ikan tersebut. Berdasarkan golongan besar organismenya, komunitas ikan di Sungai Cisadea terdiri atas tiga kelompok, yaitu karnivora, omnivora, dan herbivora dengan kelompok ikan yang mendominasi adalah ikan omnivora dan karnivora. Berdasarkan takson organisme makanannya, komunitas ikan di Sungai Cisadea dibagi menjadi empat kelompok, yaitu insektivora, fitoplanktivora, krustasivora, dan moluskivora. Kelompok takson organisme makanannya ialah fitoplankton, insekta, makrofita, krustase, moluska, ikan, protozoa, dan rotifer. Di wilayah sungai bagian hulu dan tengah makanan utama ikan terdiri atas insekta dan alga, sedangkan di bagian hilir terdiri atas alga, krustase, dan insekta.

Abstract

Objective of the research was to analysis food habits of beardless barb (Cyclocheilichthys apogon) with observing a composition of prey items. Research was carried out in June 2006, August 2006, and January 2007 in Musi River alongside the upstream and downstream using cast net and experimental gillnet. Stomach content analysis was based on index of preponderance and niche breadth. Beardless barb analyzed during research about 159 individuals fish with total length ranging from 57-175 mm and weight ranging from 1.88-71.79 gram. Beardless barb is omnivorous having the character of euryphagic with plant (54.98) as basic food; detritus (19.05), worm (9.30), phytoplankton (8.22), insect (4.89) as secondary food; and zooplankton (3.57%) as supplement food. Difference of sex, size, habitat, and research time did not cause change and difference of resources utilization by beardless barb in the Musi River. Female of beardless barb in upstream Musi River has different resources utilization with increasing of resources utilization of animal organism which is estimated to has relation of requirement of reproduction energy. Equality of resources (food organism) which utilization by beardless barb in various sex and size, was conducive existence of emulation when food resources in limited condition.

Abstract

This study aimed to evaluate the physiological responses of Asian swamp eel, Monopterus albus during the fasting period at different salinity levels. Fasting period was three days at four levels of salinity, i.e., 0 g L -1 , 3 g L - 1 , 6 g L - 1 , and 9 g L - 1 . The eels were stocked at a density of 200 kg m - 3 . Physiological parameters studied were blood plasma cortisol which was measured by radioimmunoassay method, blood plasma glucose and liver glycogen which were measured by spectrophotometer, blood plasma sodium which was analyzed by Atomic Absorption Spect rophotometer, and osmolality which was measured using osmometer. The results showed that salinity affected the physiological status of the eels. Fasting the eels could increase the blood plasma cortisol levels, but in at higher salinity levels the hormone was lower. Blood plasma sodium levels of the eels increased in line with higher salinity. Fasting the eels at salinity of 0 and 3 g L - 1 was increasing the glucose levels significantly compared to the eel at a salinity of 6 and 9 g L - 1 . In contrary, the liv er glycogen levels were statistically lower in salinity of 0 and 3 g L - 1 compared to the eels at a salinity of 6 and 9 g L - 1 . Fasting the fish for three days at all salinity treatments did not change the condition factor of the eels. The eels were exhibite d the best physiological performance during the three - days fasting period at a salinity of 9 g L - 1 .AbstrakPenelitian ini bertujuan untuk meng evaluasi respons fisiologis ikan belut Monopterus albus pada pemberokan dengan tingkat salinitas yang berbeda. Pemberokan dilakukan pada ikan belut yang berukuran panjang 26,64 ± 2,94 cm dan b o - bot 12,57±4,99 g , selama tiga hari dalam media air dengan perlakuan empat tingkat salinitas, yaitu 0 g L - 1 , 3 g L -1 , 6 g L - 1 , dan 9 g L - 1 . B iomassa belut adalah 1 kg dalam 5 L air atau pada densitas 200 kg m - 3 . Parameter fisiologis yang diu - kur meliputi kortisol plasma darah dengan metode radioimunoasai, glukosa plasma darah dan glikogen hati dengan spektrofotometer, natrium plasma darah diana lisis dengan Atomic Absorption Spectrophotometer , dan osmolalitas de - ngan menggunakan osmometer. Hasil penelitian menunjukkan bahwa p emberokan dan salinitas mengakibatkan peru - bahan kondisi fisiologis ikan belut. Pemberokan mengakibatkan k adar kortisol meningkat , n amun peningkatan salinitas dapat menekan kadar hormon tersebut. Kadar natrium plasma darah ikan belut mengalami peningkatan sejalan dengan semakin tingginya salinitas. Kadar glukosa belut yang diberok pada salinitas 0 dan 3 g L - 1 secara statistik lebih tinggi dan berbeda sangat nyata di b andingkan dengan belut pada salinitas 6 dan 9 g L - 1 . Hal sebaliknya t erjadi pada kadar gli - kogen hati, yaitu secara statistik lebih rendah pada salinitas 0 dan 3 g L - 1 dibandingkan dengan belut pada salinitas 6 dan 9 g L - 1 . P emberokan selama tiga hari pada semua perlakuan salinitas t idak mengakibatkan perubahan nilai faktor kondisi belut . Ikan belut memberikan respons fisiologis yang paling baik pada pemberokan tiga hari dengan salinitas 9 g L - 1.

Abstract

A research to explore morpho-anatomcal changes and energy storage on various organs during gonadal development of twospots catfish, Mystus nigriceps was carried out in Klawing River, Purbalingga, Central Java. Morpho-anatomical parameter e.g. condition factor, viscera somatic index, adipose fin index, hepato somatic index, gonado somatic index; and energy content of muscle of dorsal region, visceral organ, adipose fin, liver, and gonad tissue were measured based on gonad development stages. The results showed that morpho-anatomy and energy content in tissues have changed during the period of gonadal development. Moreover, there were a process of storage, transfer and transformation of energetic substances for gonad maturation and spawning activities. AbstrakSuatu penelitian dengan tujuan untuk mengkaji perubahan nilai indeks morfo-anatomi dan penyimpanan energi pada beberapa organ tubuh ikan senggaringan selama masa perkembangan gonad telah dilaksanakan di Sungai Klawing, Purbalingga, Jawa Tengah. Parameter morfo-anatomi meliputi faktor kondisi (CF), indeks jaringan viseral (VSI), indeks sirip lemak (AFI), indeks jaringan hati (HSI), indeks jaringan gonad (GSI), dan kandungan energi pada organ/jaringan otot punggung, sirip lemak, organ viseral, hati, dan gonad telah diukur berdasarkan tingkat perkembangan gonadnya. Hasil penelitian menunjukkan bahwa selama masa perkembangan gonad terjadi perubahan nilai-nilai parameter morfo-anatomi dan kandungan energi pada organ yang diukur, terjadi proses penyimpanan, perpindahan dan perubahan materi berenergi baik untuk keperluan pematangan gonad maupun untuk aktivitas pemijahan.

Abstract

DNA barcoding of genus Kryptopterus catfish from Mahakam River, East Kalimantan by using mitochondrial DNA COI gene has been performed to obtain genetic marker (barcoding DNA), genetic characteristics and genetic tree. Amplification of mitochondrial COI gene regions was conducted by using COI Fish F1 and COI Fish R1 primers on the three catfish species consist of Kryptopterus apogon (N=3), K. micronema (N=6), and K. limpok (N=1). Based on partial COI mtDNA multiple alignment, we obtained three specific nucleotide site as genetic marker. Meanwhile, genetic characteristic analysis revealed 13.58% of nucleotide variation. Nucleotide substitution was greater at the third codon and transitional substitution was greater than transvertion. Genetic tree reconstruction based on p-distance separated the three catfish species with perfect bootstrap value. AbstrakPenelitian kode batang DNA spesies ikan lais genus Kryptopterus asal Sungai Mahakam Kalimantan Timur mengguna-kan gen COI DNA mitokondria dilakukan dengan tujuan untuk mengungkap penanda genetik (kode batang DNA), ka-rakterisasi genetik dan pohon genetik. Amplifikasi daerah gen COI DNA mitokondria dilakukan dengan menggunakan pasangan primer COI FishF1 dan COI FishR1 terhadap tiga spesies ikan lais yang terdiri atas Kryptopterus apogon (N=3), K. micronema (N=6), dan K. limpok (N=1). Hasil penelitian menunjukkan penyejajaran berganda pada gen par-sial COI DNA mitokondria antara spesies K. apogon, K. micronema, dan K. limpok diperoleh tiga situs nukleotida spe-sifik sebagai penanda genetik (kode batang DNA). Analisis karakterisasi genetik pada genus Kryptopterus berdasarkan gen parsial COI DNA mitokondria menunjukkan situs nukleotida yang bervariasi sebesar 13,58%, substitusi nukleotida lebih besar pada kodon ketiga dan bersifat transisi dari pada transversi. Konstruksi ulang pohon genetik berdasarkan ni-lai p-distance dapat memisahkan kelompok spesies K. apogon, K. micronema,dan K. limpok dengan nilai bootstrap sempurna.

Abstract

The objective of the research is to know the relationship between the altitude and biological aspects of swamp eel. It is expected that the result of the research can be used as basic information for its culture development. Swamp eel sample were collected from many locations at Subang regency, West java eg: Nanggerang (17 m asl), Gunung Sari (43 m asl), Suka Melang (150 m asl), Gunung Tua (400 m asl) and Tambak Mekar (600 m asl) villages during April to Mei 2001. In every sampling sites were measured physical-chemical and biological parameters of water and substrat texture were examined. Biological aspects of swamp eel e.g.: fish abundance, condition factors, maturity index, fecundity and food habits were analysed in laboratory. The result of the research indicated that altitude affected physical, chemical and biological parameters of water and substrate texture. The altitude also influenced the fish abundance, food habits and fecundity of the fish ell.AbstrakTujuan dari penelitian ini adalah untuk mengetahui hubungan antara ketinggian tempat dan aspek-aspek biologi belnt sawah Hasil penelitian ini diharapkan dapat digunakan sebagai informasi dasar untuk pegembangan budidaya Contoli ikan belut sawah ditangkap dari beberapa lokasi di Kabupaten Subang, Jawa Barat yakni di Desa Nanggerang (ketinggian 17 m dpi), Gunung Sari (43 m dpi), Suka Melang (150 m dpi), Gunung Tua (400 m dpi) dan Tambak Mekar (600 m dpi) dari bulan April - Mei 2001. Di setiap lokasi pengambilan contoh diukur parameter-parameter fisika-kimia air dan tekstur substrat. Aspek biologi ikan belnt sawah yang dianalisis adalah kelimpahan, faktor kondisi, indeks kematangan gonad, fekunditas dan kebiasaan makanan. Hasil penelitian menunjukkan bahwa ketinggian tempat mempengaruhi parameter fiskia, kimia dan biologi serta tekstur substrat. Ketinggian tempat juga mempengaruhi kelimpahan, kebiasaaan amakanan dan fekunditas belut sawah.

Abstract

Mercury is a dangerous heavy metal to fish and human who consumed that contaminated fish. The objective of the research was to analysis the effect of mercury to physiological condition of milkfish which reared in various level of salinity. Experiment was divided into four treatments: A (0% + 0 mg Hg L-1); B (0%o + 0.012 mg Hg L-1); C (10%o + 0.012 mg Hg L-1); and D (20%o + 0.012 mg Hg L-1), each with three replications. Milkfish of 7-8 cm in length was reared in aquarium of 60 x 40 x 40 cm3 in size, filled with 40 L of water. Twenty milkfish were added into each aquarium, fed by artificial food up to 5% biomass, 3 times per day during 30 day of experiment. Physiological parameters were observed by measuring osmotic gradient, oxygen consumption and blood glucose. Within freshwater conditions, the results showed that osmotic gradient and blood glucose increased, while oxygen consumption decreased in response to the present of mercury. Milkfish that reared in 10%o salinity showed better physiological conditions than other treatment, i.e. osmotic gradient 0.237±0.088 Osm kg-1 H2O, oxygen consumption 0.257±0.037 mgO2 g-1 hour-1, and blood glucose 11.77±1.30 mg mL-1. It was shown that the toxicity level of mercury were the lowest at 10%o salinity. AbstrakMerkuri merupakan logam berat yang sangat berbahaya bagi kehidupan ikan, bahkan dapat membahayakan kesehatan bagi yang mengkonsumsi ikan yang telah terkontaminasi logam berat tersebut.Penelitian ini bertujuan untuk mengkaji pengaruh merkuri terhadap kondisi fisiologis ikan bandeng yang dipelihara pada berbagai tingkat salintas. Percobaan terdiri atas empat perlakuan yakni: A (0% + 0 mg Hg L-1), B (0%o + 0,012 mg Hg L-1), C (10%o + 0,012 mg Hg L-1), dan D (20%o + 0,012 mg Hg L-1).Masing-masing perlakuan memiliki tiga ulangan. Ikan bandeng yang berukuran 7-8 cm dipelihara dalam akuarium berukuran 60 x 40 x 40 cm3 dengan volume air 40 L. Masing-masing akuarium diisi dengan 20 ekor ikan bandeng dan dipelihara selama 30 hari. Pakan buatan berbentuk pellet diberikan tiga kali per hari sebanyak 5% dari bobot biomassa. Parameter yang diukur meliputi beban osmotik, tingkat konsumsi oksigen, dan kadar glukosa darah. Hasil penelitian menunjukkan bahwa pada ikan bandeng yang dipelihara di media air tawar, keberadaan merkuri menyebabkan naiknya beban osmotik dan kadar glukosa darah serta menurunkan tingkat konsumsi oksigen. Kondisi fisiologis ikan bandeng yang dipelihara pada salinitas 10%o lebih baik dibandingkan perlakuan lainnya dengan nilai beban osmotik 0,237±0,088 Osm kg-1 H2O, tingkat konsumsi oksigen 0,257±0,037 mgO2 g-1 jam-1 dan kadar glukosa darah 11,7±1,30 mg.mL-1. Dampak toksisitas merkuri terhadap kondisi fisiologis ikan bandeng berkurang ketika dipelihara pada media dengan salinitas 10%o.

Abstract

This experiment was conducted to determine the effect of alkalinity on the survival rate and daily growth rate of Barbodes sp. The fishes were reared in 12 aquariums with four different alkalinity levels 48, 78, 108 and 138 ppm. Each aquarium filled with 10 fish at the average weight of 12.25 to 12.64 g attempt. The treatment lasted for 75 days. During the experiment, the fish were given commercial diet (pellet) with the dose of 5% of biomass weight at the frequency of three times a day. Result of the experiment showed that the survival rate for all treatments was 100%, while the alkalinity which suitable for daily growth rate of the fish was 80 ppm C’aCOr

Abstract

The observation was done in Palabuhanratu bay from June 2005 until February 2006, The observation aimed to describe fecundity and development of the gonad. Samples were caught using hand lines with 6-9 mesh size and “kincang” boat. Total sample were 108 with range of length are 210 - 812 mm and total weight between 62 - 6434 gram. Fecundity was between 205.547 - 3.028.159 and shown that P. multidens had a large reproductive potency. The development of gonad (testes and ovary) in morphology and histology could explained the first size sexual maturity, spawning season and spawning type.
