Abstract

Abstrak Teknologi pemurnian senyawa dengan metoda kromatografi penting dilakukan agar didapat senyawa murni yang mempunyai aktivitas lebih tinggi. Metoda pemurnian dilakukan dengan menggunakan fasa diam silika dan fasa gerak merupakan campuran n-heksana dan etil asetat serta campuran etil asetat metanol dengan kepolaran yang dinaikkan secara gradien. Dari fraksi n-heksana  dari tumbuhan Calophyllum macrophyllum didapat  satu senyawa murni yang mempunyai aktivitas antioksidan. Kata kunci : Kromatografi, Calophyllum macrophyllum, aktivitas antioksidan.

Abstract

The soulatro coumarin compound was isolated and elucidated from the stem bark of Calophyllum soulattri Burm F, the samples were collected from Jayapura Papua Irian Island in  Indonesia.  Isolation  process  was  done  by  maceration  at  room  temperature  in  methanol, than  partitioned  in  a  mixture  of  n  hexane-water  (1:1),  followed  by  dichloromethane-water (1:1)    and  ethyl  acetate-water  (1:1).  A  portion  of  ethyl  acetate  extract  was  subjected  to column chromatography over silica gel packed and eluted with n-hexane a gradient of ethyl acetate to 100% followed by CHCl3  in MeOH (20:1, 10 :1, 5:1, 1:1). Fraction  B (CHCl3 in MeOH 20:1) was subjected to column chromatography  over silica gel 300 mesh  and eluted with  EtOAc-MeOH  mixtures  of  increasing  polarity.  Faction  with  the  same  Rf  valeus  were combined  and  eluted  with  EtOAc-MeOH    (19:1)  showed  one  spot  on  TLC.  They  were combined  and  evaporated  to  yield  a  solid  than  was  recrystallized  in  mixture  of  CH2Cl2-methanol  to  give  soulatro  coumarin  compound.  The  structure  was  determinated  by spectroscopic analysis, in particular by 1D and 2D NMR techniques, from these spectra data conclution  that  compound  is  soulatro  coumarin.  Antimalarial  assay  was  tested  against Plasmodium berghei parasite as in vivo using 18 mices rodent wich was infected by  Plasmodium berghei  parasite.  The  soulatro  coumarin    showed  activity  against  P.  berghei  with  dosage 0.0005867  mM/1  kg  body  weight  ;  0.005867  mM/1  kg  bw;  0.05867  mM/1  kg  bw;  0.5867 mM/1  kg  bw  5.867  mM/1  kg  bw  and  58.67  mM/1  kg  bw  could  inhibite  growth  rate  of parasite = 57.32%; 63.37%; 43.02%; 53.49%; 47.67% respectively.   Keywords : Antiplasmodial activity, coumarin, Calophyllum soulattri Burm. Abstract

The  anticancer  activities  of  medicinal  plants  from Indonesia’s tropical rainforests were investigated  against  Hela  cell  line.  Maytenfoliol  from  leaves  of  Calophyllumtetrapterum  3-epibetulinic  acid  from  stem  bark  of    C.  tomentosum Wight  and  D.A-friedo-oleanan-3-on  from stem bark of C. moonii showed anticancer activity. The compound 3-epibetulinic acid showed the more potent anticancer activity than maytenfoliol  and D.A-friedo-oleanan-3- with thean IC50  value were 3.17 μg/mL, 4.89 μg/mL and 5.63 μg/mL, respectively.Keywords: Anticancer  evaluation,  Hela  cell  line, Calophyllumtetrapterum.  C.  tomentosum Wight and  C. moonii

Abstract

Most of Calophyllum genus have been researched and proven as medicinal plants. Calophyllum macrophyllum Scheff is an Indonesia original plant growing on Sumatera and we found in Kerinci mount. One flavanal has been isolated from ethyl acetate fraction from the stem-bark of Calophyllum macrophyllumScheff. The compound is 5,7,2’,5’- tetrahedrons flavan-3-ol, brown crystal with melting point > 300oC. The compound was extracted using maceration technique, then fractionated using chromatography and purified by crystallization using two solvents. Molecule structure was determined using physical data and spectroscopy methods including UV, IR and 1H and 13C-NMR. Ethyl acetate fraction and isolate were assayed for inhibitory activity on -glycosidase enzyme. The potential inhibition effect is got each other with IC50 26,198 =g/ml and 12,19 =g/ml.

Abstract

Guaiacol, a phenolic compound is known as an anticancer. Dimerization of guaiacol has been done by biotransformation using peroxidase enzyme as biocatalyst. This enzyme was isolated from Indonesian plant, kailan (Brassica oleraceae var. alboglabra). Analysis of dimerization product was carried out by TLC, IR, LC-MS, and NMR. Whilst analysis of in-vitro cytotoxic activity was carried out by MTT method against breast cancer T47D and MCF7 cells. The result showed that the dimerization reaction gave O-para dehydroguaiacol. The in-vitro cytotoxic activity analysis showed that O-para dehydroguaiacol compound has potency as anti-breast cancer.

Abstract

Calophyllum flavoranulum (Clusiaceae family) is a big tree and medical plants from the tropical rain forest of the Indonesian continent. The organic extracts from stem bark yielded phenyl coumarin. The structure was elucidated with the aid of spectroscopic technique. Antiplasmodial activities of isolated compound was tested in vivo against Plasmodium berghei parasite and showed the best antiplasmodial activity. New flavoranulum coumarin compound showed activity against P. berghei parasite with inhibition growth 0, 31.33, 40, 30.67, 46 and 34% at dosage 1.1x10-10, 1.1x10-9, 1.1x10-8, 1.1x10-7, 1.1x10-6, and 1.1x10-5 mg/mL, respectively. Flavoranulum coumarin very active as antiplasmodial, and at 3 and 4 day incubation all of parasite were died.

Abstract

Calophyllum bicolor (Clusiacea) is a big tree from Indonesian rain forest in Palangkaraya, Central Kalimantan. Calophyllum or bintagor is one of many sources of natural bioactive compounds that can be used in the fields of health and pharmaceuticals. The aim of this research was to explore the antiplasmodial activity of methanol extract of Calophyllum bicolor P.F. Steven against Plasmodium falciparum. The methanol extract was purified by colomn chromatography system, hexane – ethyl acetate was used as solvent with increasing polarity. One pure compound was obtained and was elucidated based on the 1H-&13C-NMR and 2D-NMR, [COSY, HMBC and HMQC] data and the isolated compound was identified as xanthone. Methanol extract showed antiplasmodial activity growth inhibition against P. falciparum with IC50 5.2 ppm and the new 5-methoxy trapezifolixanthone compound have maximum inhibition at concentration 0.11 nMol.

Abstract

Several of Calophyllums genus have been searched and proven as medicinal plants and Calophyllum macrophyllum is one of its genus. We have been isolated a compound from ethyl acetate fraction of the stem-bark. The compound was flavan-3-ol (5,7,2’,5’-tetrahydroxy flavan-3-ol). This research aimed to determine inhibition antidiabetic activity and affinity of its compound on α-glycosidase enzyme. In vitro antidiabetic effect shown by IC50 9.10 µg/ml and docking by Arguslab 4.01 shown by ΔG -10,24 kkal/mol as in silico test. The inhibition activity of flavan-3-ol against the α-glucosidase enzyme that is very strong shows its promising potential as a candidate for antidiabetic drugs.
